Chemical aspects of propranolol metabolism. Synthesis and identification of 3-(4-hydroxy-1-naphthoxy)propane-1,2-diol as a metabolite of propranolol in the dog, in man and in the rat liver 9000g supernatant fraction.

4-Hydroxypropranolol glycol (2), a suspected metabolite of propranolol was synthesized from 4-allyloxy-1-naphthaldehyde (4). Osmium tetroxide oxidation of 4 afforded a glycol (5) and subsequent Baeyer-Villiger rearrangement afforded 2. Its presence as a biliary metabolite in a dog maintained on a constant infusion of pseudoracemic propranolol (made up of equal molar 2S-propranolol-3',3'-d2 and 2R-propranolol-d0) into the portal vein was confirmed based on GC-MS data of the TFA and TMS derivatives of the standard and biliary metabolites. Greater amounts of 2 arose from 2R-propranolol than from 2S-propranolol (1.5:1). Similarly, 2 was formed as a urinary metabolite in one subject maintained on oral propranolol, 80 mg every 6 hours. Compound 2 was also formed when propranolol and propranolol glycol were incubated in the presence of the rat liver 9000g supernatant fraction. In addition to 2, an isomeric ring-hydroxylated propranolol glycol, perhaps 7-hydroxypropranolol glycol, was formed when propranolol glycol was the substrate.

Reduction of chronic doxorubicin cardiotoxicity in dogs by pretreatment with (+/-)-1,2-bis(3,5-dioxopiperazinyl-1-yl)propane (ICRF-187).

Adult beagle dogs were given doxorubicin (1.0 mg/kg body weight i.v.) either alone or 30 min after ICRF-187 (NSC 169780) (12.5 mg/kg body weight i.p.) at weekly intervals. Control dogs received 0.9% NaCl solution i.v. 30 min after ICRF-187 i.p. (12.5 mg/kg body weight). One week after the 15th injection (300 mg/sq m total dose), the animals were sacrificed. The frequency and extent of cellular lesions were graded on a scale of 0 to 4+. Such lesions, consisting mainly of vacuolization and myofibrillar loss, were noted in the hearts of all six dogs given doxorubicin alone. The lesions were severe (4+) in five of these animals and moderate (2+) in one. In contrast, no abnormalities were noted in the hearts of four of the six dogs pretreated with ICRF-187 before doxorubicin administration; the remaining two animals in this group had minimal alterations (1+). At the dosage regimen used in the present experiments, doxorubicin did not induce lesions in lungs, liver, kidney, diaphragm, small intestine, or skeletal muscles. Comparable decreases in white blood cell count, red blood cell count, hemoglobin, and serum iron concentration were found in animals receiving doxorubicin with or without ICRF-187. Concurrent administration of ICRF-187 offers a promising means of reducing the chronic cardiotoxicity induced by doxorubicin.

[Pharmacokinetics of 1-(4'-oxyphenyl)-2-(3'-phenyl-thiopropyl-amino) propane-1-01 hydrochloride (Istiod) in the rat].

The pharmacokinetics (serum, urinary and biliary concentrations) of Isotiod have been studied in rats. Doses exploited the property of the molecule in an alkaline medium with a diazonium salt to form coloured azo compounds. In rats Isotiod determines blood concentrations whose intensity and duration are proportional to the dose and route of administration; notable quantities of the drug are eliminated by the urinary way and a small quantity by the bile way with a personal technique.
