The purpose of this study was to investigate the cellular composition of peritoneal fluid during post-surgical re-epithelialization and to determine the metabolism of arachidonic acid by these cells. Rabbits underwent a midline laparotomy followed by abrasion of the broad ligament. The presence of adhesions was graded and the peritoneal exudative cells collected up to 14 days thereafter. Ascitic fluid and cells were separated by centrifugation and the cellular percipitate incubated with [14C]arachidonic acid. The aliquot was separated by silica Gel G thin-layer chromatography and the specific radioactivity of each strip determined. To evaluate the reformation of adhesions 14 days after the first abrasion, rabbits underwent reabrasion of the same area and the pattern of arachidonic acid metabolism by the ascites cells was similarly evaluated. Six hours after the abrasion, PMNs comprised 87.5% of the peritoneal exudative cells (total 0.03 X 10(7) cells/rabbit). On Day 3, the total cell number increased to 2.92 X 10(7), 97.6% of which were large mononuclear cells. No significant change in the type of distribution of adhesions was evident from 6 hr through Day 2. After Day 7, the total number of adhesions was minimal; however, those that were present were primarily severe. The second-look evaluation of adhesion formation was not found to be consistent prior to the 7th postoperative day, since many filmy bands present prior to that time were not present later. The in vitro formation of 5-HETE by these cells increased from 6 hr through Day 11. Production of di-HETE increased beginning Day 3 and maintained high steady state levels thereafter.(ABSTRACT TRUNCATED AT 250 WORDS)

Intraperitoneal (IP) injections of either 1, 3 or 9 ml of neurotensin-containing solutions (NTCS) with 5.4, 54, 540 or 5400 nM of neurotensin (NT) were found to cause concentration-dependent, but volume-independent, increases of blood pressure (BP) and heart rate (HR) in anesthetized, close-abdomen guinea pigs. The duration of both effects varied between 15 to 30 min depending both on the NT concentration and volume of NTCS utilized. Indirect evidence suggested that NT inactivation within the peritoneal cavity contributed to shorten the duration of NT effects. Animal pretreatment with a ganglion blocker, adrenoceptor antagonists, clonidine or capsaicin, reduced the BP and HR increases caused by IP injection of NTCS whereas both effects were either unaffected or slightly potentiated by animal pretreatment with atropine, morphine or captopril. Addition of a local anesthetic to NTCS inhibited the hemodynamic effects of NT whereas acute bilateral cervical vagotomy was without significant effect. These results suggest that NT has the ability to trigger cardiovascular reflexes following its IP injection in guinea pigs. The activation of peritoneal, sympathetic, capsaicin-sensitive primary afferents appears to be at the basis of these reflexes, the amplitudes of which seem poorly related to the volume of NTCS utilized (at least within the range of volume examined).

UNLABELLED: Changes of the mucosa of the small intestine in animal experiments can be produced by a variety of methods. The direct application of numerous chemical agents on the epithelium of the mucosa and parenteral administration of cytostatics as well as an indirect damage by experimental intestinal lymphatic obstruction lead to mucosal lesions with atrophy of the villi. Our paper presents a simple indirect method for induction of structural mucosa changes in rats using experimentally produced peritoneal alterations as a precursor. MATERIALS AND METHODS: The first experimental group with 108 male wistar rats was divided into 5 subgroups and treated as follows: 43 rats got continuous daily intraperitoneal injections of 3-3 ml/kg body weight sesame oil DAB 6 which contains 50% oleic acid, 36% linolic acid, 1% free fatty acids, 0.1% lecithin and portions of stearic, palmitic and arachic acid; 6 rats got 6.6 ml sesame oil per kg i.p., 43 animals received daily doses of 3-3 ml oil/kg per orally by a pharyngeal tube, 6 rats of each group were sacrificed after 68 and 92 days, all the rest were killed after 100 days of continuous daily treatment. Aortic blood was used for determination of the total cholesterol serum concentration. Blocks of tissue of every organ were fixed in 4% formalin, sections were stained with hematoxylin and eosin, hematoxylin-sudan and by the periodic acid Schiff (PAS-) method. For the purpose of autoradiographic determination of the 3H-thymidine labelling index the rats were administered i.p. 2 mCi 3H-thymidine/kg 40 minutes prior to death. Autoradiographs were prepared from sections of the small intestine by using liquid photoemulsion Ilford K5. In the second experimental group 7 rats got a single i.p. injection of 5 ml, 5 rats of the 10 ml and 9 rats of 20 ml sesame oil per animal; 4 rats were administered 8 ml daily for 5 days...

OBJECTIVE: To determine the response of adhesion and peritoneal fibroblasts to hypoxia. DESIGN: Prospective experimental study. SETTING: University medical center. PATIENT(S): Primary cultures of fibroblasts established from the peritoneal and adhesion tissues of the same patients (n = 2) to minimize genetic variations. INTERVENTION(S): Hypoxia treatment of the primary cultured fibroblast. MAIN OUTCOME MEASURE(S): Analyze the expression of extracellular matrix (ECM) components, metalloproteinases and their tissue inhibitors, growth factors, and cytokines in adhesion and peritoneal fibroblasts under normal and hypoxic conditions by reverse transcriptase/polymerase chain reaction analysis. RESULT(S): Compared to peritoneal fibroblasts, adhesion fibroblasts had a significant increase in the basal mRNA levels for collagen I, fibronectin, MMP-1, TIMP-1, TGF-beta 1, TGF-beta 2, and IL-10. Hypoxia resulted in a further increase in collagen 1, fibronectin, TIMP-1, TGF-beta 1, TGF-beta 2, IL-10, and IFN-gamma mRNA levels in both peritoneal and adhesion fibroblasts. The increase was more profound in adhesion fibroblasts. CONCLUSION(S): Hypoxia induces molecular changes in both peritoneal and adhesion fibroblasts, creating a milieu that favors adhesion development. The effect of hypoxia was more profound on adhesion fibroblasts.

Solute and water transport from blood to peritoneal cavity occur by diffusion and osmotic ultrafiltration, whereas absorption to blood via lymphatics negatively affects these two processes. This article delineates the physiology of peritoneal membrane and numerous factors that influence mass transport during peritoneal dialysis, thereby affecting its therapeutic efficacy. Benefits and limitations of continuous ambulatory peritoneal dialysis (CAPD) are discussed and compared to those of hemodialysis. Survival on CAPD, its complications and imperfections are reviewed in light of the widespread acceptance of the procedure.
