Saccharomyces cerevisiae mutants bearing mutations at the cyc4 locus are partially deficient in cytochrome synthesis. Although the mutation is not in the structural gene for delta-aminolevulinic acid (Alv) synthase, the mutants are deficient in Alv synthesis in vivo as indicated by abnormally low intracellular Alv concentrations. The cyc4 mutation causes cells to grow very slowly in minimal glucose medium, but not in yeast extract-peptone-glucose medium. A simple nutritional defect caused by the cyc4 mutation is not involved because cytochrome deficiency is enhanced by growing cyc4 cells in yeast extract-peptone medium. A regulatory role for CYC4 is indicated. Evidence for negative feed-back control of Alv synthase by heme is provided by the observation of enhanced intracellular Alv accumulation in yeast mutants partially deficient in decarboxylation of uroporphyrinogen and coproporphyrinogen, respectively.
Delta-aminolaevulinic acid is well known to be an intermediate in the synthesis of haeme and small amounts are normally excreted in urine (ALA-U). An increased excretion of this substance occurs in porphyria and in lead poisoning. The urine specimens of 670 children (362 boys and 308 girls), aging from less than one to fifteen years and who had no evidence of abnormal exposure to lead, have been collected in order to find out the range of normal values of ALA-U expressed in mg/l for the different age and sex group. Our data can be usefull, together with zincprotoporphyrin (ZPP), for screening of anemias and for monitoring lead exposure.

The biosynthesis of porphobilinogen by homogenates of the lungs, heart, liver, kidneys, spleen, pancreas and small intestinal tissues was studied in 77 intact and in 68 rats with marked lead poisoning. The activity of ALA dehydratase in the lungs was decreased by 40.3%, in the kidneys by 40%, in the heart by 38.2% and was normal in homogenates of the liver, pancreas and small intestine. In spleen homogenates, the activity was increased by 26.3%. Potential mechanisms of different ALA dehydratase reactions in organs tissues of rats with lead poisoning are discussed.
The circadian rythm concerning the excretion of urinary metabolites ALA and CP was investigated. More ALA and CP are excreted in the urine during the day than the night. This phenomenon is more evident with CP. The sampling of urine for biochemical control of saturnism is discussed and the sample collected after 24 h was chosen.
In our experimental conditions, the autoradiographic study of the 3H deltaaminolevulinic and 55Fe incorporation allows us to show selectively the red blood cells of Lineus lacteus actively synthetizing haemoglobin.
