Chiral copper complexes of phosphino sulfenyl ferrocenes as efficient catalysts for enantioselective formal Aza Diels-Alder reactions of N-sulfonyl imines.

In the presence of a catalytic amount of silver perchlorate, copper(I) bromide complexes of planar chiral 1-phosphino-2-sulfenylferrocenes behave as very efficient chiral Lewis acids catalysts in the formal Aza Diels-Alder reaction of N-arylsulfonyl aldimines with electron-rich dienes (Danishefsky's and related dienes). Mixing of equimolar amounts of the readily available enantiopure ferrocenyl P,S-bidentate ligand and CuBr quantitatively affords the precatalyst Cu complex as an air-stable solid. This catalytic asymmetric procedure has a broad structural scope: aldimines of aromatic, alpha,beta-unsaturated, and even enolizable aliphatic aldehydes have been successfully used. The corresponding 2,3-dihydro-4-pyridones were obtained in good yields (57-90%) and with homogeneously high enantioselectivity (82-97% ee).

Enantioselective hydrogenation of imines in ionic liquid/carbon dioxide media.

The enantioselective hydrogenation of N-(1-phenylethylidene)aniline using cationic iridium complexes with chiral phosphinooxazoline ligands was studied as a chemical probe to assess the potential of ionic liquid/carbon dioxide (IL/CO2) media for, multiphase catalysis. The biphasic system leads to activation, tuning, and immobilization of the catalyst that would be impossible in classical organic solvent systems or in either of the two unconventional media separately. In particular it is demonstrated that (i) the presence of CO2 can be beneficial or even mandatory for efficient hydrogenation in the IL; (ii) the precursor is activated in the IL by anion exchange allowing one to use in situ catalysts; (iii) the anion of the IL greatly influences the selectivity of the catalyst; (iv) the products are readily isolated from the catalyst solution by CO2 extraction without cross contamination of IL or catalyst; and (v) the IL leads to enhanced stability of the catalyst. These results are corroborated and rationalized on the basis of the physicochemical properties of the biphasic medium and the chemical characteristics of the catalytic systems.

Structure and activity of hydrogenated derivatives of (+)-5-methyl-10,11-dihydro-5H-dibenzo[a,d]cyclohepten-5,10-imine (MK-801).

Several hydrogenated derivatives of the potent NMDA antagonist 1 have been prepared and evaluated as competitive inhibitors of [3H]-1 binding. These compounds were also tested for their ability to act as noncompetitive antagonists of NMDA in vitro. These studies indicate that two aromatic rings are not strictly required for high-affinity binding or NMDA antagonism.

Synthesis of isoquinolines and pyridines by the palladium/copper-catalyzed coupling and cyclization of terminal acetylenes and unsaturated imines: the total synthesis of decumbenine B.

Monosubstituted isoquinolines and pyridines have been prepared in good to excellent yields via coupling of terminal acetylenes with the tert-butylimines of o-iodobenzaldehydes and 3-halo-2-alkenals in the presence of a palladium catalyst and subsequent copper-catalyzed cyclization of the intermediate iminoalkynes. In addition, isoquinoline heterocycles have been prepared in excellent yields via copper-catalyzed cyclization of iminoalkynes. The choice of cyclization conditions is dependent upon the nature of the terminal acetylene that is employed, as only aryl and alkenyl acetylenes cyclize under the palladium-catalyzed reaction conditions that have been developed. However, aryl-, vinylic-, and alkyl-substituted acetylenes undergo palladium-catalyzed coupling and subsequent copper-catalyzed cyclization in excellent yields. The total synthesis of the isoquinoline natural product decumbenine B has been accomplished in seven steps and 20% overall yield by employing this palladium-catalyzed coupling and cyclization methodology.

An enantioselective ketene-imine cycloaddition method for synthesis of substituted ring-fused 2-pyridinones.

Previously, a method for the stereoselective synthesis of beta-lactams, starting from 2H-Delta(2)-thiazolines and Meldrum's acid derivatives, has been reported from our laboratory. We now report a new method for the synthesis of optically active, highly substituted ring-fused 2-pyridinones. This was discovered when 2-alkyl-Delta(2)-thiazolines and Meldrum's acid derivatives were treated with HCl(g) in benzene at 5 --> 78 degrees C. Further refinement of the synthetic protocol revealed that use of 1,2-dichloroethane as solvent at 0 --> 64 degrees C led to the desired 2-pyridinones in good yields and with excellent enantioselectivity. Use of these conditions allowed preparation of 2-pyridinones from several different Delta(2)-thiazolines and Meldrum's acid derivatives and may be a general route to 2-pyridinones.

Asymmetric synthesis of aziridine 2-phosphonates from enantiopure sulfinimines (N-sulfinyl imines). Synthesis of alpha-amino phosphonates.

An aza-Darzens reaction, involving the addition of chloromethylphosphonate anions to enantiopure sulfinimines, has been developed for the asymmetric synthesis of aziridine 2-phosphonates. Best results involve cyclization of the syn and anti diastereomerically pure alpha-chloro-beta-amino phosphonates to cis- and trans-N-sulfinyl aziridine 2-phosphonates, respectively, with n-BuLi. A transition-state hypothesis is proposed wherein the chloromethylphosphonate anion adds to the C-N bond on the side that is opposite the bulky p-tolyl sulfinyl group. The N-sulfinyl group is easily removed by treatment with MeMgBr or TFA/MeOH, which affords the NH-aziridines in good yield. Using transfer hydrogenation conditions, the NH-aziridines were regioselectively opened to the corresponding enantiopure alpha-amino phosphonates without N-activation and in excellent yield.

A CASPT2 and CASSCF approach to the cycloaddition of ketene and imine: a new mechanistic scheme of the Staudinger reaction.

We report a new theoretical study of the mechanism of the parent Staudinger reaction between ketene and formaldimine. This mechanism, in contrast with previously reported computational studies, involves the formation of two different intermediates, gauche and trans, which can be interconnected on the potential energy surface depending upon reaction conditions. This novel CASPT2/CASSCF mechanistic scheme may be considered a new starting point to rationalize stereochemical outcome and solvent effects in these reactions.

Theoretical study of the ruthenium-catalyzed cyclocotrimerization of alkynes with isocyanates and iothiocyanates: cemoselective formation of pyridine-2-one and thiopyrane-2-imine.

The cyclocotrimerization of acetylene with isocyanate HNCO and isothiocyanate HNCS, mediated by CpRuCl, is theoretically investigated on the basis of DFT/B3LYP calculations. By these means, the experimental result can be rationalized as to why, with HNCO, a nitrogen-heterocycle is formed, but with HNCS a sulfur-heterocycle is formed. According to the proposed mechanism, the key reaction step is the addition of a double bond to a metallacyclopentatriene formed by oxidative coupling of two acetylene ligands coordinated to CpRuCl, giving a bicyclic carbene intermediate. This double-bond-addition is initiated by eta1 attack at the ruthenium center, and it is just the attacking atom that is going to be incorporated into the cycle. Thus, the chemoselectivity originates from the fact that, for HNCO, N attack is preferred over O, but for HNCS, S attack is preferred over N. The onward reaction is a reductive elimination to give a coordinatively unsaturated metallaheteronorbornene intermediate finally rearranging to a ligated heterocycle. Completion of the cycles is achieved by an exothermic displacement of the respective heterocyclic product by two acetylene molecules which regenerates the bisacetylene complex.

Irradiation of imine-group VI carbene complexes in the presence of alkynes. 2. Control of product distribution.

The photoreactivity of iminecarbene complexes in the presence of alkynes has been explored. Up to four different reaction paths are available depending on the alkyne and carbene complex substituents, although in each case only one type of product is isolated. 2H-Pyrrole derivatives are formed mainly from aryl alkynes. When alkyl alkynes are used, the method affords a new type of aza-dendralene product in good yields. Isoquinoline derivatives can also be formed in a two-step one-pot photochemical process when the appropriate substituents are present. Finally, indene derivatives are also available through a benzannulation reaction. To explore the underlying mechanism, we carried out computations using DFT methods. Experimental and theoretical results compare well, which allows control over the reaction and product distribution.

An asymmetric nitro-Mannich reaction applicable to alkyl, aryl, and heterocyclic imines.

[reaction: see text] A protocol for the enantioselective nitro-Mannich coupling between alkyl, aryl, and heterocyclic p-methoxybenzylimines and trimethylsilylnitropropanate catalyzed by a chiral tBu-BOX Cu(II) catalyst is described. It uses the lowest reported loading of commercially available metal catalyst and chiral ligand, and gives the highest yields and selectivities for a broad substrate range including nonaromatic aldimines. The resultant beta-nitroamines are obtained in 70-94% enantiomeric excess in good yield and can be readily reduced to synthetically useful 1,2-diamines.
