Homology modelling of the nuclear receptors: human oestrogen receptorbeta (hERbeta), the human pregnane-X-receptor (PXR), the Ah receptor (AhR) and the constitutive androstane receptor (CAR) ligand binding domains from the human oestrogen receptor alpha (hERalpha) crystal structure, and the human peroxisome proliferator activated receptor alpha (PPARalpha) ligand binding domain from the human PPARgamma crystal structure.

We have generated by homology the three-dimensional structures of the ligand binding domain (LBD) of several interrelated human steroid hormone receptors (SHRs). These are the oestrogen receptor beta (hERbeta), the pregnane-X-receptor (PXR), the Ah receptor (AhR) and the constitutive androstane receptor (CAR). They were produced by homology modelling from the human oestrogen receptor alpha (hERalpha) crystallographic coordinates [Nature 389 (1997) 753] as a template together with the amino acid sequences for hERbeta [FEBS Lett. 392 (1996) 49], PXR [J. Clin. Invest. 102 (1998) 1016], AhR [Proc. Natl. Acad. Sci. U.S.A. 89 (1992) 815] and CAR [Nature 395 (1998) 612; Mol. Cell. Biol. 14 (1994) 1544], respectively. The selective endogenous ligand, in each case, was docked interactively within the putative ligand binding site using the position of oestradiol in hERalpha as a guide, and the total energy was calculated. In each receptor model a number of different ligands known to fit closely within the ligand binding site were interactively docked and binding interactions noted. Specific binding interactions included combinations of hydrogen bonding and hydrophobic contacts with key amino acid sidechains, which varied depending on the nature of the ligand and receptor concerned. We also produced the human peroxisome proliferator activated receptor alpha (PPARalpha) by homology modelling using the human PPARgamma (hPPARgamma) LBD crystallographic coordinates summarised in [Toxicol. In Vitro 12 (1998) 619] as a template together with the amino acid sequence for hPPARalpha [Toxicol. In Vitro 12 (1998) 619; Nature 395 (1998) 137]. The models will provide a useful tool in unravelling the complexity in the physiologic response to xenobiotics by examining the ligand binding interactions and differences between the steroid hormone receptors activation or inactivation by their ligands.

Synthesis and crystal structure determination of a new pressure-induced iridium ditelluride phase, m-IrTe2, and comparison of the crystal structures and relative stabilities of various IrTe2 polymorphs.

The new monoclinic IrTe2 phase m-IrTe2 was synthesized under pressure, and its structure was determined by X-ray powder diffraction. The relative stabilities of the three known and three hypothetical IrTe2 polymorphs were discussed on the basis of tight binding electronic band structure calculations. m-IrTe2 exhibits structural features of both CdI2- and pyrite-type IrTe2 phases and is expected to be nearly as stable as that of the CdI2-type IrTe2. The hypothetical IrS2- and ramsdellite-type IrTe2 phases are predicted to be more stable than the CdI2-type IrTe2.

Novel cation [N(SO2NMe3)2]+ and its synthesis and crystal structure. Dichloride of imido-bis(sulfuric) acid HN(SO2Cl)2. Part 1. Crystal structures of KN(SO2Cl)2.(1/2)CH3CN, KN(SO2Cl)2.(1/6)CH2Cl2, and [PCl4][N(SO2Cl)2].

Several salts of bis(chlorosulfonyl)imide HN(SO2Cl)2 (1), namely, two solvates of its potassium salt, KN(SO2Cl)2.(1/2)CH3CN (1K1), KN(SO2Cl)2.(1/6)CH2Cl2 (1K2), and its tetrachlorophosphonium salt, [PCl4][N(SO2Cl)2] (2), were prepared and structurally characterized. The reaction of HN(SO2Cl)2 with Me3N gives the [N(SO2Cl)2]- salt of a novel cation, [N(SO2NMe3)2]+. This cation is analogous to the [HC(SO2NMe3)2]+ cation, but in contrast to the latter, it is fairly stable to hydrolysis. The salt [N(SO2NMe3)2]+[N(SO2Cl)2]- (3) can be converted into salts of other anions by being treated with diluted aqueous solutions of the respective acids, and thus NO3-, Cl-.H2O, SeO3(2-), CH3COO-, HSO4-, (COO)2(2-) salts were prepared. Treatment of 3 with concentrated HNO3 gave the [N(SO2NMe3)2]+ [O2NO-H-ONO2]- salt, and the addition of an HCl-acidified FeCl3 aqueous solution yielded the FeCl4- salt. Methanolysis resulted in the formation of MeOSO3- and [MeOSO2NSO2OMe]- salts. All salts have been characterized by chemical analysis, vibrational spectroscopy, and X-ray structure determinations.

Single-Crystal, Solid-State, and Solution (113)Cd and (77)Se NMR and X-ray Single-Crystal Study of a [Cd(SeR)(2)(N-donor)(2)] Complex.

[Cd(Se-2,4,6-i-Pr(3)-C(6)H(2))(2)(bpy)] (1) has been characterized by X-ray crystallography and studied by solution and solid-state (113)Cd and (77)Se NMR to serve as an analog for biologically occurring [M(S-Cys)(2)(His)(2)] centers. The unit cell parameters for 1 are as follows: a = 21.99(2) Å, b = 21.43(4) Å, c = 16.72(3) Å, V = 7881.3(4) Å(3), Z = 8, orthorhombic space group, Pccn. Two chemically inequivalent cadmiums and seleniums are found per unit cell. The principal values of the cadmium chemical shift tensors are (delta(11) = 752 ppm, delta(22) = 570 ppm, delta(33) = 93 ppm) and (delta(11)= 733 ppm, delta(22) = 547 ppm, delta(33) = 100 ppm) with respect to 0.1 M aq. Cd(ClO(4))(2). Those for selenium are (delta(11) = -950 ppm, delta(22) = -1040 ppm, delta(33) = -1599 ppm) and (delta(11) = -933 ppm, delta(22) = -1069 ppm, delta(33) = -1586 ppm) with respect to solid (NH(4))(2)SeO(4). The orientation of the cadmium chemical shift tensor is similar to that of the previously published sulfur analog.

Mineralomimetic sodalite- and muscovite-type coordination frameworks. Dynamic crystal-to-crystal interconversion processes sensitive to ion pair recognition.

A flexible sodalite-type metal organic framework [Cu(pyrimidine-2-olate)2]n (1R) is obtained in a self-assembly process involving Cu2+ ions and pyrimidine-2-olate ligands. 1R suffers a series of sequential and reversible structural changes upon solid-liquid sorption processes of metal nitrates. The 1C-to-1O transformation occurs only with large cations.

Intermolecular proton transfer in solid phase: a rare example of crystal-to-crystal transformation from hydroxo- to oxo-bridged iron(III) molecule-based magnet.

Intermolecular proton transfer in solid phase from the hydroxo bridge to a water molecule occurs in a new mu-hydroxo iron(III) compound of formula {EtNH3[Fe2(ox)2Cl2(mu-OH)].2H2O}n leading to a still crystalline compound in which the mu-oxo bridge replaces the mu-hydroxo one. Both three-dimensional compounds exhibit magnetic ordering at Tc ca. 70 K due to a spin canting.
