Coccidiosis immunization: effects of mushroom and herb polysaccharides on immune responses of chickens infected with Eimeria tenella.

An experiment was conducted to investigate the effects of polysaccharide extracts (E) of two mushrooms, Lentinus edodes (LenE) and Tremella fuciformis (TreE), and an herb, Astragalus membranaceus (AstE), on the immune responses of chickens infected with Eimeria tenella. A total of 180 broiler chickens were assigned to nine groups: three groups were fed with each of the extracts (LenE, TreE, and AstE), three groups were fed with the extracts and immunized with live oocyst vaccine (LenE+V, TreE+V, and AstE+V), a group was immunized with the vaccine only, and there were two controls (E. tenella-infected and noninfected groups). The oocyst vaccine was given at 4 days of age, and the extracts (1 g/kg of the diet) were supplemented from 8 to 14 days of age. At 18 days of age, all birds except those in the noninfected group were infected with 9 x 10(4) sporulated oocysts. The results showed that at 7 days postinfection (p.i.), birds fed the extracts without vaccination had lower body weight (BW) gain than those given the vaccine only. However, the extracts in conjunction with the vaccine significantly enhanced BW gain of the infected chickens compared with the vaccine group. Of the three extracts, LenE and TreE showed a better growth-promoting effect. The extracts largely increased oocyst excretion of droppings during the primary response postvaccination. The cecal peak oocyst output and lesion scores measured at 7 days p.i. were higher in the groups fed the extracts than in the group immunized with the vaccine only, whereas those of the groups fed with the extracts and immunized with the vaccine were not significantly different from the vaccine group. Of the three extracts, both LenE- and AstE-fed groups showed lower cecal oocyst output. Thus, as compared with the extracts, the live, attenuated vaccine showed better results with significantly increased immune response in coccidial infected birds. The polysaccharide extracts may prove useful against avian coccidiosis, and, particularly when they are used in conjunction with vaccine, they have shown preliminary promise against the experimental coccidial infection.

Effects of herbal preparation Equiguard on hormone-responsive and hormone-refractory prostate carcinoma cells: mechanistic studies.

The Equiguard is a dietary supplement comprised of standardized extracts from nine herbs, respectively, Herba epimedium brevicornum Maxim (stem and leaves), Radix morindae officinalis (root), Fructus rosa laevigatae michx (fruit), Rubus chingii Hu (fruit), Schisandra chinensis (Turz.) Baill (fruit), Ligustrum lucidum Ait (fruit), Cuscuta chinensis Lam (seed), Psoralea corylifolia L. (fruit), and Astragalus membranaceus (Fisch.) Bge (root). This proprietary product, formulated according to Chinese traditional medicinal concepts, is aimed at restoring harmony in the of the kidney, an organ which Chinese medicinal principles consider to be vital for invigorating as well as maintaining balance of the entire urological system. As the prostate is an integral component of the urological system, we performed in vitro studies to test the effects of ethanol extracts of Equiguard to modulate prostate growth and gene expression. These studies used prostate cancer cells mimicking the androgen-dependent (AD) and androgen-independent (AI) states of prostate carcinogenesis. Results show that Equiguard significantly reduced cancer cell growth, induced apoptosis, suppressed expression of the androgen receptor (AR) and lowered intracellular and secreted prostate specific antigen (PSA), and almost completely abolished colony forming abilities of prostate cancer cells. These data support the interpretation that this herbal formulation contains ingredients that collectively may be efficacious in preventing or treating AD and AI prostate carcinoma. The anti-prostatic activities of Equiguard may stem from its complex composition capable of targeting multiple signal transduction/metabolic pathways, to effectively correct, counteract or circumvent the impaired or dysfunctional mechanisms accompanying different stages of prostate carcinogenesis.

[Study of unified analytical method of compatibleness for traditional Chinese medicines. I. Separation, recognition of characteristic components and fingerprint analysis for single and complex medicines].

It is difficult to separate, identify and quantify multi-component in complex traditional Chinese medicines (TCM). Many analytical methods for TCM have been established, but only a few components can be separated and quantified. For analyzing the multi-component and finding new method of compatibility analysis, a strategy--the multi-mode and multi-column chromatographic system and unified analytical method has been proposed by us. A part of the results from the study of astragalus root(A), Chinese angelica root(Ca.) and their complex prescription (Danggui Buxue Tang, DBT) with the analytical mode of reversed-phase HPLC/photodiode array detector as an example of the method for unified separation and recognition of characteristic components and fingerprint analysis was introduced. This is the first part of the results of compatibleness analysis for TCM.

Shi-quan-da-bu-tang (ten significant tonic decoction), SQT. A potent Chinese biological response modifier in cancer immunotherapy, potentiation and detoxification of anticancer drugs.

Shi-Quan-Da-Bu-Tang (Ten Significant Tonic Decoction), or SQT (Juzentaihoto, TJ-48) was formulated by Taiping Hui-Min Ju (Public Welfare Pharmacy Bureau) in Chinese Song Dynasty in AD 1200. It is prepared by extracting a mixture of ten medical herbs (Rehmannia glutinosa, Paeonia lactiflora, Liqusticum wallichii, Angelica sinesis, Glycyrrhiza uralensis, Poria cocos, Atractylodes macrocephala, Panax ginseng. Astragalus membranaceus and Cinnamomum cassia) that tone the blood and vital energy, and strengthen health and immunity. This potent and popular prescription has traditionally been used against anemia, anorexia, extreme exhaustion, fatigue, kidney and spleen insufficiency and general weakness, particularly after illness. In order to restore immunity in cancer patients, potentiate the therapeutic effect and ameliorate adverse toxicity of anticancer agents, 116 Chinese herbal formularies (Kampo) have been screened and evaluated. Fifteen compounds were found to have such actions. Among these, SQT was selected as the most effective as a potent biological response modifier. During the last eight years, animal models and clinical studies have revealed that SQT demonstrates extremely low toxicity (LD50 > 15 g/kg op murine), self-regulatory and synergistic actions of its components in immunomodulatory and immunopotentiating effects (by stimulating hemopoietic factors and interleukins production in association with NK cells, etc.), potentiates therapeutic activity in chemotherapy (mitomycin, cisplatin, cyclophosphamide and fluorouracil) and radiotherapy, inhibits the recurrence of malignancies, prolongs survival, as well as ameliorate and/or prevents adverse toxicities (GI disturbances such as anorexia, nausea, vomiting, hematotoxicity, immunosuppression, leukopenia, thrombocytopenia, anemia and nephropathy, etc.) of many anticancer drugs. The application and mechanistic studies of SQT in future development have potential importance in basic and clinical research of the traditional Chinese therapeutic approach of "toning the blood and strengthening Qi (vital energy)" in cancer immunotherapy.

Phytochemicals potentiate interleukin-2 generated lymphokine-activated killer cell cytotoxicity against murine renal cell carcinoma.

The success of adoptive immunotherapy using recombinant interleukin-2 (rIL-2) and lymphokine-activated killer (LAK) cells in several cancers has been hampered by severe toxicity associated with high doses of rIL-2. Methods that reduce the dosage of rIL-2 without loss of clinical efficacy are needed. In this study we determined the in vitro effect of a phytochemical immune modulator, Astragalus membranaceus (AM), and two fractions isolated by high-performance liquid chromatography on the cytotoxicity of rIL-2-generated LAK cells against a murine renal cell carcinoma. Our results indicated a 10-fold potentiation of rIL-2-generated LAK cell cytotoxicity manifested by tumor cell lysis of 88% in the group with 100 U/ml of rIL-2 plus AM versus 86% in the group with 1,000 U/ml of rIL-2 alone. Potentiation was obtained with the purified fractions as well. A significantly reduced number of LAK cells was required to achieve the tumor cytotoxicity after LAK cell generation with rIL-2 plus the phytochemicals as compared with rIL-2 alone. Our data indicate that AM is an effective immune modulator, capable of potentiating in vitro the antitumor activity of rIL-2-generated LAK cells.

[Experimental study on yiqi-huoxue therapy of liver fibrosis].

UNLABELLED: The experimental hepatic fibrosis was treated with YiQi (reinforcing Qi, YQ) Huoxue (activating blood circulation, HX) principle which was consisted of astragalus membranaceus, Ligusticum wallichii, paeonia lactiflora, etc. After stimulation with CCl4 over four months, the Wistar rat developed liver fibrosis. Rats were divided into the normal control, the toxifying control, YQHX group and HX group. The experimental period lasted over four months. RESULTS: (1) Mortality of animal: Both toxifying control and HX group reached 50%, while YQHX group was 16% only. These results suggest that YQHX agents could strengthen the body resistance; (2) The determination of serum SGPT: The mean levels in toxifying control were 39.3 +/- 39, in HX group 43.7 +/- 12.9, while in YQHX group 29.0 +/- 7.6 (units/Lserum). These results indicated that YQHX agents had the function of protecting the liver and lowering the activity of SGPT (P less than 0.01); (3) Measuring the contents of hepatic collages: The mean levels in toxifying control was 38.9 +/- 3.3 (mg/g liver), while in HX group and YQHX group 28.7 +/- 2.2 and 22.7 +/- 1.1 (mg/g liver) respectively. The results indicated that the YQHX agents had the best results in treating hepatic fibrosis; (4) Observation with hepatic histopathology: The degree of hepatocyte degeneration and necrosis in YQHX group was milder than that in toxifying and HX group. These observations revealed that YQHX agents possessed the function of protecting liver. There was severe liver fibrosis in toxifying control, but the degree of liver fibrosis in YQHX group was significantly milder than that in toxifying control.(ABSTRACT TRUNCATED AT 250 WORDS)

Mini transposon vector mediated foreign gene expression in Mesorhizobium huakuii subsp. rengei.

Among the transposable elements, mini-Tn5 transposon vector has proven to be of greater utility for insertion mutagenesis of variety of Gram negative bacteria. The mini-Tn5 vector containing promoter less egfp gene and gentamycin resistant gene was used for the present study. The transposon vector was introduced to M. huakuii from E. coli S17 by conjugation. The conjugants were screened for stable expression of egfp both in free-living and in nodules of Astragalus sinicus. The result showed that the conjugant #3 showed stable expression of green fluorescent both in free-living and bacteroid stage. The visualization of sym plasmid of wild strain and conjugants showed that conjugant #3 had a fragmentation of large sized plasmid into two but without affecting the nodulating ability. These results clearly indicated that mini-Tn5 vectors (Transposon vectors) the best alternate tools for plasmid vectors for integration of foreign genes in chromosomal DNA or symbiotic plasmid and expression, both in free-living and bacteroid stage of Rhizobium.

[Effect of astraglus injection on serum apoptosis relevant factors in patients with chronic heart failure].

OBJECTIVE: To investigate the effect of Astragalus injection (AGI) on serum apoptosis related factors such as soluble Fas (sFas), soluble Fas ligand (sFasL) and tumor necrosis factor alpha (TNF-alpha) in patients with chronic heart failure (CHF). METHODS: Eighty-four patients with CHF of NYHA II-IV grade were randomly divided into two groups, the treated group (42 patients) treated with AGI and the control group (42 patients) treated with routine treatment. The level of serum sFas, sFasL, and TNF-alpha were measured with ELISA before and after treatment. At the same time, patients' heart function were graded according to the NYHA classification and their indices of left ventricular function were determined. RESULTS: Patients' NYHA grade was improved in both groups after treatment (P < 0.05 or P < 0.01), and it was better in the treated group than that in the control group (P < 0.05). Moreover, in the treated group, the left ventricular end-systolic volume (LVESV) and left ventricular end-diastolic volume (LVEDV) decreased, the left ventricular ejection fraction (LVEF) increased, which was significantly different than that before treatment (P < 0.05) respectively; level of serum apoptosis related factors sFas, sFas L and TNF-alpha were significantly decreased after treatment, showing significant difference as compared with those before treatment or with those in the control group after treatment (P < 0.05 or P < 0.01). While all indices had no obvious change in the control group. CONCLUSION: AGI may be regarded as an effective remedy for treatment of CHF owing to its effects in decreasing the level of serum apoptosis related factors in patients with CHF.

Subcellular localization of copper in tolerant and non-tolerant plant.

The ability of Elsholtzia splendens Naki (E. splendens) to accumulate copper appears to be governed by its high degree of coppertolerance. However, the tolerance mechanism on the physiological basis is unknown. Using transmission electron microscope (TEM) and energy dispersive analysis of X-rays (EDX), the likely location of copper within the cells of the tolerant and non-tolerant was determined. Here the role of vacuolar and cell wall compartmentalization in this copper tolerant plant were investigated. A direct comparison of copper locations of E. splendens and the non-tolerant Astragalus sinicus L. (A. sinicus) showed that the majority of copper in the tolerant was localized primarily in the vacuolar, cell wall, on the plasmamembrane, beside lipid grains induced by copper pollution, in the chloroplasts and amyloids; but in the non-tolerant, copper precipitates only be observed on the plasmamembrane, in the chloroplasts and cytoplasm under copper exposure conditions that were toxic to both species. This revealed that the tolerant accumulates more copper in the vacuole and cell wall than the non-tolerant, where was regarded as the storage compartment of tolerant plant or hyperaccumulator for heavy metals.
