[Morphological characteristics of the thymus gland of albino rats after prenatal administration of indomethacin].

By means of histological, histochemical and electron microscopical methods the thymus of white rat fetuses and offspring has been investigated during various age periods after indomethacin++ influence (2.5 mg/kg). In the fetuses retardation in separation of the gland parenchyma into lobules has been revealed. During the first two weeks of life the section area of the medulla decreases. Amount of lymphoid cells decreases; small and degenerating lymphocytes decrease in their number, while the part of lymphoblasts, middle lymphocytes and figures of mitosis increases. Enzymatic activity in the nervous structures of the organ is inhibited. Some essential disturbances of the intracellular structures are revealed; they demonstrate certain destructive changes. The data obtained show a decreasing function of the thymus after the prenatal influence of indomethacin++ during the first month of life, which is especially manifested during the first two weeks.

[The role of thymus gland in the regulation of the activity of peripheral T-lymphocytes in the process of spontaneous blastogenesis in mice].

The total count, spontaneous proliferation and proliferative response of thymic and spleen T-cells in mixed lymphocyte culture and Con A-induced suppressor activity do not reveal significant disorders in 10-14-month A/Sn and C3H/He mice with spontaneous mammary tumors (weight under 2-3 g). However, these indices are quite different in varying age (2, 6, 12 month) A/Sn mice and C57Bl/6 mice with low rates of spontaneous tumors. The analysis of thymus-dependent immunity changes observed with age shows that relatively intensive migration of nonmature thymocytes, T-suppressor precursors is noted in mice with high cancer incidence. This phenomenon is considered to be one of major mechanisms regulating immune response in spontaneous-carcinogenesis.

Detection of HTLV-I tax-rex and pol gene sequences of thymus gland in a large group of patients with myasthenia gravis.

We report in this study the use of polymerase chain reaction (PCR) for the amplification of the genomic DNA, isolated from thymic tissue, using the primers flanking HTLV-I/II tax-rex genes and the sequence method to analyze the HTLV-I pol sequence of 27 Italian patients with myasthenia gravis. These molecular methods showed that 92.5% of patients tested positive for tax gene and 55% for pol genes; 55.5% samples were positive for both the tax gene of HTLV-I/II, and the pol gene of HTLV-I. Histologic investigation of the thymus showed that 15 samples had thymic hyperplasia, 93% tested positive for the tax gene, and 40% tested positive for both the tax and pol genes of HTLV-I. In contrast, 91.6% of thymoma-positive samples were positive for tax gene I/II and 75% positive for both genes, tax and pol type I. The sequence analysis of PCR product for tax and pol genes confirmed that these amplified products were HTLV-I, with minimal variations. Our date suggested that either HTLV-I or part of the virus genome is involved in the etiopathogenesis of myasthenia gravis.

Histological changes associated with enlargement and regression of the thymus glands of the red-billed quelea Quelea quelea L. (Ploceidae: weaver-birds).

Thymic lobes form over 200 red-billed queleas, Quelea quelea L., were examined histologically. Samples were taken from embryos about to hatch, juveniles and adults. The lobes varied in size from very small to very enlarged (1- greater than 5 mm long). The constituent cell types are described in detail and the occurrence of these cells in different sized lobes is discussed. A cycle of events is proposed which accounts for the observations presented here. It is suggested that the large numbers of erythroid cells found in the cortex of some individuals were developing in situ. The significance of erythropoiesis within the thymus is discussed.

[The role of the thymus gland in the development of adaptation reactions of the immune system during fractionated low-level gamma irradiation].

In experiments with normal and thymectomized rats it has been revealed that exposure of normal animals to 0.35 Gy of gamma-radiation induces changes in blood cells subsets, depression of NK functional activity, decrease in polymorphonuclear leukocyte basal chemiluminescence level, suppression of the delayed type of hypersensitivity reaction and stimulation of local IgE-synthesis in respiratory organs. At the same time irradiation of adult thymectomized animals did not resulted in significant fluctuations in cellular and humoral immunity indices as well as blood cells functional activity level. Some mechanisms of radiation-induced immune system disturbances mediated via thymus are discussed.

[Effect of the lining of the thymus gland on hematopoietic stem cell kinetics in a suspension of embryonic mouse liver].

A study was made of the kinetics of hemopoietic stem cells in the organic culture of a suspension of mouse embryonic liver cultivated directly on the filters and on the preliminarily grown embryonic thymic feeder. It was shown that stem hemopoietic cells were retained in the course of one month of cultivation in the embryonic liver suspensions; during the first two weeks hemopoiesis in the suspension practically failed to differ from such in the fragments of the same age and cultivation period, whereas during the subsequent periods hemopoiesis proved to be more active in the suspension cultivated on thymic feeder. A possible participation of the thymus in the hemopoiesis is discussed.
