Chemiluminescence microflow injection analysis system on a chip for the determination of uric acid without enzyme.

A new microflow injection analysis (microFIA) system on a chip coupled with chemiluminescence (CL) for the non-enzymatic determination of uric acid is described. The microFIA system produced by using two transparent poly(methylmethacrylate) (PMMA) chips measured 50 x 40 x 5 mm, the microchannels, etched by CO2 laser, were 200 microm wide and 100 microm deep, and the volume of the reaction area (RA) was about 1.2 microL. The injection pump, with accurate time control, monitored all reagents, including the sample. The uric acid was sensed by the chemiluminescence reaction between luminol and ferricyanide. The linear range of the uric acid concentration was 0.8-30 mg/L and the detection limit was 0.5 mg/L (S/N = 3). The relative standard deviation was 4.42% for 5 mg/L uric acid (n = 8). The proposed method has been successfully applied to the non-separation determination of uric acid in human serum and urine.

Nonlinear systems analysis of the hippocampal perforant path-dentate projection. I. Theoretical and interpretational considerations.

1. Nonlinear systems analytic procedures, based on an orthogonalized functional power series approach, were developed for study of the transformational properties of the hippocampal formation. As a testing stimulus, the procedures utilize a train of electrical impulses with randomly varying interimpulse intervals. The specific case was considered of applying such a stimulus to the perforant path, a major afferent to the hippocampal dentate gyrus that arises from the entorhinal cortex. Resulting field potentials evoked within the dentate gyrus are recorded to all impulses in the train. Computational algorithms based on cross-correlations determine the relationship between the interimpulse interval within the random train and amplitude of the evoked dentate potentials. The calculations, which reduce to averaging procedures, were derived for first- and second-order terms, or kernels, of the orthogonalized functional power series. 2. It is proposed that such an approach can be applied to a single component of the complex field potential evoked in the dentate gyrus. This component, the population spike, reflects the action potential discharge of dentate granule cells. Thus, a field potential component for which the underlying neuronal generator is well-known can be analyzed with respect to the transformational characteristics of the network of neurons that influence that generator. Other components of the complex field potential produced by other generators can be ignored. It is shown that this adaptation has the effect of greatly simplifying both the computation and presentation of kernels. 3. As a further consequence of this adaptation, the resulting first- and second-order kernels were shown to have specific interpretations. The first-order kernel represents the average response of the orthodromically driven granule cells to the set of stimuli comprising the random impulse train. The second-order kernel quantitatively characterizes the nonlinearity of the granule cell response, and may be interpreted as a generalized recovery function; i.e., the first input of any pair of stimuli in the train activates the newtork, and the second input tests the modulatory influence of the network excited by the initial input. 4. Most past investigations of nonlinearities of the perforant path-dentate projection have utilized pairs of stimulus impulses. We show here that, for a second-order system, the expected results from paired impulse experiments may be predicted from second-order kernels. Disagreement between the measured and predicted results reflects interactions of a higher order, and thus, greater system complexity.(ABSTRACT TRUNCATED AT 400 WORDS)

Genucom knee analysis system: reproducibility and database development.

The Genucom is a relatively new computerized system that performs a multidimensional analysis of knee laxity. Therefore, the purposes of the present investigation were twofold: 1) to estimate the reproducibility of the testing system in its ability to obtain similar test values on the same test subject within a back-to-back two-test protocol and 2) to develop a database of normal values as a standard against which values of suspected ligament injury can be compared to reliably determine whether, in fact, ligament injury has occurred. In the reproducibility phase of the study, 20 subjects were tested in two trials each of eight tests, yielding 20 separate test values per trial, accounting for each angle of flexion and plane of force tested. Both legs were tested, providing an N of 40. Test-retest correlations of the two trials and dependent t-tests indicated that the Genucom provided generally stable and reproducible results. In the normal value database phase of the study, 218 different subjects were given one trial of the same eight tests. Testing of both legs provided an N greater than 400. Means and standard deviations were calculated for each test.

Systems analysis of possible mechanisms of mammalian aging.

A comparison is made between how bacteria and men use their DNA to accomplish any major biological function. To make this comparison, a modified cybernetic analysis was used. This new type of cybernetic analysis makes a distinction between controlled and regulated systems and may permit an extension of cybernetics into new areas of biology. Prokaryotes have only regulated systems for all major functions, whereas eukaryotes can have both regulated and controlled systems. Several lines of reasoning, including the new cybernetic analysis, suggest that death, as opposed to aging, may be a regulated function in mammals. By analogy with other major regulated developmental functions in mammals, it is also suggested that the hypothalamic-pituitary axis may be the site for the regulation of the time of death. An example of how the pituitary might act on the DNA of peripheral cells to cause possible, severe loss of functional capacity is given by some recent experiments with RNA polymerase activity of liver nuclei from rats of different ages and with different endocrine states.

[Systems analysis of colour music corrective effect].

In the context of P. K. Anokhin's theory of functional systems, the corrective effects of various combinations of medical therapeutical resonance music (MTRM) and dynamic colour exposure were analyzed. As compared to rehabilitative music programmes, MRTM was shown to have a more pronounced relaxing effect as manifested both in the optimization of emotion and in the activity of autonomic regulation of cardiovascular functions. On combined MRTM and dynamic colour flow exposures, the relaxing effect is most marked. In the examinees, the personality and situation anxieties diminish, mood improves, cardiovascular parameters become normal, the rate of metabolic processes and muscular rigidity reduce, the spectral power of alpha-rhythm increases, these occurring predominantly in the anterior region of the brain. The findings suggest the high efficiency of the chosen way of normalizing the functional status of man.

Criteria for the design of fetal heart rate analysis systems.

Criteria are described for the automated analysis of fetal pulse intervals, fetal movements and of uterine contractions measured externally, antenatally and interactively on-line, for implementation on a personal computer interfaced to an appropriate fetal monitor, and tested on 10,000 records. Measurements of short and longer term fetal heart rate variation are compared; both are required to identify sinister records. Recall and display of records acquired on the same patient over several weeks has proved a useful diagnostic aid.

Application of computer-assisted sperm analysis system to elucidate lack of effects of cyclophosphamide on rat epididymal sperm motion.

A computer-assisted sperm analysis (CASA) system was used to examine the motion of epididymal spermatozoa derived from cyclophosphamide (CP)-treated male rats. Male rats were orally dosed daily for 1 week with 20 mg/kg of CP. Males were euthanized or were mated 3 times with untreated females at 1 day, 3 weeks, and 8 weeks after the final treatment. Significant decreases in testicular and epididymal weights and epididymal sperm counts of the treated animals were noted after 8-week recovery. Histopathological morphometry of the testis revealed minimal damage to spermatogonia at 1 day after the final treatment and to spermatocytes after 3-week recovery in the CP-treated group. On Caesarian section, increased post-implantation losses were found in females mated with CP-treated males in matings starting 1 day and 3 weeks after the final treatment. On the other hand, none of the sperm motion parameters of treated males derived from the CASA system exhibited significant changes at any time points, although the spermatozoa of treated males at 1 day and 3 weeks after the final treatment were damaged at the DNA level, and the spermatozoa of males after 8-week recovery had been the target cells of CP when they were spermatogonia in the testis. It was thus found that damaged spermatozoa could exhibit no changes on their motion when the damage was confined to the nuclei, and that the effect of CP on sperm nuclei was reversible.

Comparison of three multiplex cytokine analysis systems: Luminex, SearchLight and FAST Quant.

Multiplex cytokine analysis technologies have become readily available in the last five years. Two main formats exist: multiplex sandwich ELISA and bead based assays. While these have each been compared to individual ELISAs, there has been no direct comparison between the two formats. We report here the comparison of two multiplex sandwich ELISA procedures (FAST Quant and SearchLight) and a bead based assay (UpState Luminex). All three kits differed from each other for different analytes and there was no clear pattern of one system giving systematically different results than another for any analyte studied. We suggest that each system has merits and several factors including range of analytes available, prospect of development of new analytes, dynamic range of the assay, sensitivity of the assay, cost of equipment, cost of consumables, ease of use and ease of data analysis need to be considered when choosing a system for use. We also suggest that results obtained from different systems cannot be combined.

A new method for evaluation and dietary therapy of congenital: deficiencies of amino acid metabolic enzymes. Linear system analysis and optimization of feedback inputs for the metabolic pathways of lysine, methionine and isoleucine.

We intended to elucidate an integrated mathematical model of amino acid metabolism and we propose a system for optimization treatment of disturbed metabolic states caused by congenital enzyme deficiencies. Our analysis focused on the metabolic pathway starting at asparaginic acid proceeding to isoleucine, methionine and lysine. The rate of change in the concentration of the biochemical species was expressed as 21 linear rate equations. We obtained the rate constants and the magnitude of feedback from reported experimental data. Linear systems analysis revealed that the metabolic system under study was stable but uncontrollable. These properties were insensitive to changes in the magnitude of feedback. To show the effect of optimizing the feedback so that it minimizes the square of the concentration of the species and the control input, we analyzed the impulse response of the species, transient response and the singular value of the system for four cases; (1) at the physiological state without optimizing the feedback, (2) at the physiological state attained after optimizing the feedback, (3) at the pathophysiological state attained with enzyme deficiency states for lysine and methionine metabolism without optimizing the feedback, and (4) at the pathophysiological state attained after optimizing the feedback for enzyme deficiencies. In the enzyme deficient model, the impulse response oscillated and lasted longer than that in the physiological state. These changes appeared even in the species on other branched pathways. The singular value was elevated in the enzyme deficient state. By optimizing the feedback, all the impulse responses in the enzyme deficient state recovered to nearly those in the normal physiological state. Similarly, the transient response and the singular value in the enzyme deficient state recovered to nearly the normal physiological values. We elucidated the numerical value of the feedback gain for this optimization. The present analysis is useful for the evaluation of the integrated properties of amino acid metabolism and the optimization technique is potentially of use for determining a treatment course for congenital metabolic enzyme deficiencies.

Reduced-system analysis of the effects of serotonin on a molluscan burster neuron.

The mathematical model described in Bertram (1993) is used to carry out a detailed examination of the manner in which the neurotransmitter serotonin modifies the voltage waveform generated endogenously by burster neuron R15 of Aplysia. This analysis makes use of a reduced system of equations, taking advantage of the slow rate of change of a pair of system variables relative to the others. Such analysis also yields information concerning the sensitivity of the neuron to brief+ synaptic perturbations.
