Benzo(a)pyrene diolepoxide-DNA adducts detected by synchronous fluorescence spectrophotometry.

Using benzo(a)pyrene (BP) as a model carcinogen we are currently applying a fluorescence technique to detect the very low levels of carcinogen-DNA adducts in human populations due to environmental exposure. In synchronous fluorescence spectrophotometry for detection of BP-diol epoxide-DNA, excitation and emission wavelengths are scanned simultaneously with a fixed wavelength difference (delta lambda) of 34 nm. Compared to conventional fluorescence methods only one peak emerges because excitation and emission peaks have to match delta lambda to show. Because of the quenching effect of DNA, samples are hydrolyzed by acid. After this, BP-diol epoxide (BPDE)- -modified DNA gives a peak at the same wavelength and of the same fluorescence yield as BP-tetrols. When DNA from peripheral blood lymphocytes of 44 coke oven workers were analyzed, 10 had a sharp peak at 379. Among 36 coke oven workers from another factory, 4 had detectable levels of adducts. A much smaller percentage of samples was positive in a group of aluminum plant workers. We have also found BPDE-DNA adducts in DNA from pulmonary alveolar macrophages and peripheral blood lymphocytes from tobacco smokers and some of the nonsmokers.

Determination of nitrite by flow injection spectrophotometry using a home-made flow cell detector.

A simple, rapid, low cost and accurate determination using flow injection spectrophotometry with a home made flow cell detector, in combination with green LED and photodiode, had been developed for nitrite based on the Griess-Saltzman reaction, measuring the absorbance at 525 nm. The proposed system is able to monitor nitrite in freshwater samples at a throughput of 60 samples per hour with a relative standard deviation (%RSD) of less than 2.4% and a detection limit of <7 microg dm(-3). The calibration graph was linear between 0.010 - 1.0 mg dm(-3).

Simultaneous spectrophotometric determination of Bi(III) and Sb(III) based on their complexes with iodide in acidic media using the H-point standard addition method and first-derivative spectrophotometry.

The H-point standard addition method (HPSAM) was applied to the simultaneous determination of Bi(III) and Sb(III). This method is based on the difference in the absorbance of complexes of Bi(III) and Sb(III) with iodide in acidic media in different wavelength pairs. The results showed that Bi(III) and Sb(III) can be determined simultaneously with concentration ratios of 15:1 to 1:20. First-derivative spectrophotometry was also used for the simultaneous determination of Bi(III) and Sb(III) based on their complexes with iodide. The proposed methods were successfully applied to the simultaneous determination of Bi(III) and Sb(III) in synthetic samples.

On-line collection/concentration and detection of sulfur dioxide in air by flow-injection spectrophotometry coupled with a chromatomembrane cell.

A simple and rapid procedure for SO2 determination in air was developed by using a flow injection analysis (FIA) system coupled with a 3-hole chromatomembrane cell (CMC). The CMC was applied for the on-line collection/concentration of SO2 from air into a solution of 2 g l(-1) triethanolamine (TEA) solution as an absorbing solution: SO2 was converted to SO3(2-) in the alkaline absorbing solution. The solution containing absorbed SO2 was introduced into the carrier stream of the FIA system. The amount of SO3(2-) in the absorbing solution was measured by spectrophotometry with a mixed reagent of pararosaniline and formaldehyde, and was converted to the concentration of SO2 in the air sample. A calibration graph prepared by using standard sodium sulfite aqueous solutions was adopted for the determination of SO3(2-) in the absorbing solution. The SO2 concentration in indoor air examined was found to be 22.7 +/- 0.2 ppbv using 20 ml of air sample with the air flow rate of 5 ml min(-1), where the relative standard deviation was 1.7%. The detection limit for aqueous solutions and air samples were 6.9 x 10(-8) M and 0.48 ppbv, respectively. The measuring time for one sample was about 10 min when a 20 ml air sample was used. The interferences from common anionic species, formaldehyde and acetaldehyde, were also examined.

Determination of anion-exchange resin performance based on facile chloride-ion monitoring by FIA-spectrophotometry with applications to water treatment operation.

Based on a flow-injection spectrophotometry, an automatic analytical method for determination of ppb-level chloride-ion has been established. By use of this method, a novel FIA method for the determination of SBAER performance has also been developed. In this paper, the effects of concentration, dosage, and flow rate of the regenerant on BEC of SBAER were first investigated dynamically by the FIA method. In addition, the flow rate of the sample water and the temperature of the ion exchange resin were also examined. The optimum conditions were obtained: the volume of the regenerant (sodium hydroxide) was 50 mL (0.15 g resin), and its concentration was 3% (w/v); the volumetric flow rates of the regenerant and the sample water were 0.5 ml/min (4.3 m/h) and 1.5 ml/min (13 m/h), respectively. The exchanging temperature was 25 +/- 5 degrees C. The method is characterized by the use of a micro resin-column, shorter testing cycle, easy operation, and high reproducibility. The proposed method is approximately 30 times more efficient than the manual method, and it can be used for the exchange performance comparison of various SBAER.

Determination of oligoelements in parenteral formulations by planar chromatography and spectrophotometry.

The authors have studied the chromatographic behavior of parenteral preparations for pediatric use containing inorganic cations. After separation and identification by thin-layer chromatography, Mn(2+), Zn(2+), and Cu(2+) were analyzed by a method based on reaction with an appropriate reagent and extraction with an organic solvent which yielded elution and preconcentration, resulting in an appropriate solution for colorimetric quantitation. Cr(3+) cation was determined by atomic absorption spectrophotometry after appropriate chromatographic separation, using microcrystalline cellulose (adsorbent) and an acetone:water:hydrochloric acid mixture (80:5:8) as the mobile phase.

The use of infrared spectrophotometry for measuring body water spaces.

BACKGROUND: The conventional method of measuring total body water by the deuterium isotope dilution method uses gas isotope ratio mass spectrometry (IRMS), which is both expensive and time-consuming. We investigated an alternative method, using Fourier transform infrared spectrophotometry (FTIR), which uses less expensive instrumentation and requires little sample preparation. METHOD: Total body water measurements in human subjects were made by obtaining plasma, saliva, and urine samples before and after oral dosing with 1.5 mol of deuterium oxide. The enrichments of the body fluids were determined from the FTIR spectra in the range 1800-2800 cm-1, using a novel algorithm for estimation of instrumental response, and by IRMS for comparison. RESULTS: The CV (n = 5) for repeat determinations of deuterium oxide in biological fluids and calibrator solutions (400-1000 micromol/mol) was found to be in the range 0.1-0.9%. The use of the novel algorithm instead of the integration routines supplied with the instrument gave at least a threefold increase in precision, and there was no significant difference between the results obtained with FTIR and those obtained with IRMS. CONCLUSION: This improved infrared method for measuring deuterium enrichment in plasma and saliva requires no sample preparation, is rapid, and has potential value to the clinician.

[Determination of eliminating ratio of Chinese traditional medicine for hydroxyl radical by fluorescent spectrophotometry].

Benzoic acid with weak fluorescence may react on .OH, and products with intense fluorescence are made. Extractives of Chinese traditional medicine may eliminate .OH in solution, and make amounts of the products to reduce. Then, increase level of fluorescence of products in solution will be lowered. Based on this principle, a new method is developed to determine eliminating ratio of Chinese traditional medicine for .OH. It is shown that productivity of .OH tends to saturation when H2O2 is shown more than 20 min by 280 nm UV light; .OH may react on benzoic acid completely when molar ratio of H2O2 and benzoic acid is 30:1; linear response range of products fluorescence is 2.2-80 mmol.L-1 with concentration of H2O2. IC50 of elimination .OH with magnoliae and eucommia are 1.025 and 515.3 mg.L-1 respectively. There are no remarkable difference between these results and that of spectrophotometry.

[Simultaneous determination of iron, cobalt and nickel in oil by multiwavelength linear regression spectrophotometry].

A method for simultaneous determination of iron, cobalt and nickel, using multiwavelength linear regression spectrophotometry, is proposed. Since there were no interferences caused by cobolt and nickel complexes at the peak of iron complex compound formed with 5-Br-PADAP at 750 nm, the content of iron was determined by working-curve method at 750 nm. Cobalt and nickel were simultaneously determined by multiwavelength linear regression method over the range 550-600 nm. The method was applied to the analysis of iron, cobalt and nickel in synthetic mixtures and oil samples with satisfactory results. The recovery was 97.5%-104.3% and the RSD% was 1.4%-3.8%.

[Study on determining the composition of ternary complex (AI-CAS-CTMAB) with the ratio of double peak values at double wavelengths by UV spectrophotometry].

In the present paper, the composition of complex Al-CAS-CTMAB was studied with the ratio of double peak values at dual-wavelengths by UV spectrophotometry. First, the composition of complex of Al-CAS was determined in this method, Al:CAS = 1:2. The absorption of ternary complex(Al-CAS-CTMAB) was determined with reference reagent of the complex Al-CAS. The epsilon values of Al-CAS and Al-CAS-CTMAB were determined with water reference. The composition of complex Al (CAS)n1 (CTMAB)n2 was calculated with the formula: [formula: see text] CAS:CTMAB = 1:1; Al:CAS:CTMAB = 1:2:2. The calculated value and that obtained by traditional method are agreeable. The method is reliable for determining composition of ternary complexes.
