Silica and silicates in femoral lymph nodes of barefooted people in Ethiopia with special reference to elephantiasis of the lower legs.

Electron microscopy of femoral lymph nodes of barefooted Ethiopians show the presence of numerous particles of colloid-size and electron-density in the lysosomes of the macrophages in the gland. On diffraction analysis, the particles are found to be amorphous. Elemental microanalysis of the X-ray spectrum indicate the predominance of Si, Al and Fe. In elephantiasics some particles contain silicon alone, presumably silica. The distribution of the Al/Si ratios of the particles in subjects with elephantiasis of the lower legs show a difference from that of non-elephantiasics which is statistically "highly significant" at p less than 0.001. The importance of this as a possible aetiological factor in the disease is discussed. The hypothesis that the disease is discussed. The hypothesis that the disease is a silicosis of the peripheral lymphatics of the lower limbs is supported by the present study.

Preparation and characteristics of a calcium magnesium silicate (bredigite) bioactive ceramic.

In this study, new bredigite (Ca7MgSi4O16) ceramics were prepared by sintering sol-gel-derived bredigite powder compacts at 1350 degrees C for 8 h. The bending strength, fracture toughness and Young's modulus were about 156 MPa, 1.57 MPa m(1/2) and 43 GPa, respectively. The in vitro bioactivity of the bredigite ceramics was evaluated by investigating the apatite-formation ability in simulated body fluid (SBF) and the effect of ionic products from bredigite dissolution on the mouse fibroblasts cell line L929. In addition, the in vitro biocompatibility of the bredigite ceramics was evaluated by osteoblasts adhesion and proliferation assay. The results showed that bredigite ceramics could induce HAp formation in SBF. The products from bredigite dissolution significantly promoted cell growth at a certain concentration range. Furthermore, osteoblasts adhered and spread well on bredigite ceramics, and osteoblasts proliferation on bredigite ceramics was obvious.

Co-immobilization of glucose oxidase and hexokinase on silicate hybrid sol-gel membrane for glucose and ATP detections.

The co-immobilization of glucose oxidase (GOD) and hexokinase/glucose-6-phosphate dehydrogenase (HEX) in the silica hybrid sol-gel film for development of amperometric biosensors was investigated. The silica hybrid film fabricated by hydrolysis of the mixture of tetraethyl orthosilicate and 3-(trimethoxysiyl)propyl methacrylate possessed a three-dimension vesicle structure and good uniformity and conformability, and was ready for enzyme immobilization. The electrochemical and spectroscopic measurements showed that the silica hybrid sol-gel provided excellent matrice for the enzyme immobilization and that the immobilized enzyme retained its bioactivity effectively. The immobilized GOD could catalyze the oxidation of glucose, which could be used to determine glucose at +1.0 V without help of any mediator. The competition between GOD and HEX for the substrate glucose involving ATP as a co-substrate led to a decrease of the glucose response, which allowed us to develop an ATP sensor with a good stability. The fabricated silica hybrid sol-gel matrice offered a stage for further study of immobilization and electrochemistry of proteins.
