Allele frequencies for 15 STR loci in Van-Ağri districts of the Eastern Anatolia region of Turkey.

Allele frequencies of 13 tetrameric short tandem repeat (STR) loci (D3S1358, TH01, D21S11, D18S51, D5S818, D13S317, D7S820, D16S539, CSF1PO, vWA, D8S1179, TPOX, FGA) and 2 pentameric short tandem repeat loci (Penta E and Penta D) included in the PowerPlex 16 kit were obtained from a sample of 116 unrelated individuals in Van-Ağri districts of the Eastern Anatolia region of Turkey. The expected performance of these loci for personal identification and paternity testing in this population was estimated.

STR frequencies for the PowerPlex 16 System Kit in a population from Northeast Spain.

The aim of this paper, was to obtain the frequencies for the 15 STR loci included in the PowerPlex 16 System Kit on a population of 204 unrelated Caucasian individuals living in Northeast of Spain in order to use for forensic purposes. Statistical analyses were performed using the programs GENEPOP version 3.3 and Powerstats version 1.2. The results showed that all the loci met Hardy-Weinberg expectations.

Y-chromosome STR haplotypes in the Açores Archipelago (Portugal).

Allele and haplotype frequencies of 10 Y-chromosome STR loci were co-amplified in a sample from the Açores Islands (Portugal). We found high haplotype diversity in the Açores sample (0.998). The genetic profile of this population revealed to be statistically different from that of Madeira Island and from North Portugal, two related populations with already a fairly amount of published data. This result stresses the importance of using local databases in forensic genetics.

Allelic frequencies at 12 STR loci in Colombian population.

Three hundred and twenty-six to four hundred and fifty-one individuals were typed for 12 STR markers and revealed by silver stain method and assisted by a computer analysis. Forensic and Hardy-Weinberg values were found. The Kodak ds 1D Image Analysis Software V 2.0.3 EDAS DC 120 System was a useful tool for determining the correct size of pattern bands that were out allelic ladders, alleles 18 and 17 from F13A01, 15 from TPOX and 4 from TH01 were determined by this method and some corroborated by automatic analysis.

Development of guidelines to designate alleles using an STR multiplex system.

We have carried out extensive validation of a multiplex system of 6 STR loci and the Amelogenin sex test to investigate the following characteristics: relative peak areas of heterozygote peaks; non-specific artefacts; stutters caused by slippage of the Taq enzyme; pull-up (an electronic artefact often associated with software). The purpose of this paper is to demonstrate how interpretation guidelines can be formulated by experimentation and analysis using casework samples. Automation allows data relating to band position (in base pairs), peak area and peak height to be easily collected and manipulated in spreadsheets or computer programs. This paves the way to designing expert systems to carry out the interpretative process.

Additional variability at the D12S391 STR locus in an Austrian population sample: sequencing data and allele distribution.

The highly polymorphic STR locus D12S391 was investigated in an Austrian population sample (N = 150) by PCR-amplification, comparative detection on native and denaturing polyacrylamide gels and solid phase single stranded sequencing of three size variant alleles and several additional alleles. A total of 15 alleles, distinguishable by size under denaturing conditions, could be detected. No deviations from Hardy-Weinberg equilibrium were observed in the population investigated (P = 0.52). Sequencing of size variants designated 17.3 and 18.3 showed an incomplete (GAT) repeat unit at position two of the tandem region. Additional new sequence variants due to varying compositions of the number of (AGAT) and (AGAC) repeats could be identified. Due to distinct electrophoretical mobilities of alleles of the same size but different sequence structures, denaturing detection conditions should be employed when the aim is standardization.

Analysis and interpretation of mixed forensic stains using DNA STR profiling.

The use of multiplex PCR and fluorescent dye technology in the automated detection and analysis of short tandem repeat loci provides not only qualitative information about the profile--i.e. which alleles are present--but can also provide quantitative information on the relative intensities of the bands, and is therefore a measure of the amount of amplified DNA. The availability of this quantitative information allows for the interpretation of mixtures in a detailed way which has not been previously possible with many other human identification systems. In this paper we present a simple approach to the resolution and analysis of mixed STR profiles resulting from the testing of mixed biological stains in forensic casework and highlight factors which can affect it. This approach requires a detailed knowledge--gained through a mixture of experiments and validation studies--of the behaviour of each locus within the multiplex systems described. We summarise the available data from previously published experimental work and validation studies to examine the general principles underlying this approach.

Report of the European DNA profiling group (EDNAP): an investigation of the complex STR loci D21S11 and HUMFIBRA (FGA).

This paper describes a collaborative exercise which was intended to demonstrate whether uniformity of DNA profiling results could be achieved between European laboratories using two complex short tandem repeat (STR) loci. The loci D21S11 and HUMFIBRA (FGA) were chosen because they are commonly used by different European laboratories. D21S11 has approximately 14 common alleles (f > 0.001), whereas HUMFIBRA has 19 common alleles. Laboratories were asked to test seven blood stains, one of which was a known control, and to report the results to the coordinating laboratory. The exercise demonstrated that complex STRs were amenable to standardisation.

Analysis of the short tandem repeat (STR) locus HumVWA in a Qatari population.

Polymerase chain reaction (PCR), amplified DNA fragments and denaturing polyacrylamide gel electrophoresis were used to investigate the STR locus HumVWA in a Qatari population sample (n = 200). Seven alleles were identified with frequencies ranging from 0.0125 to 0.3175. The distribution of genotypes fits the Hardy-Weinberg equilibrium. The HumVWA system shows a power of discrimination of 0.9290 and a chance of paternity exclusion of 60.1% in the population studied.

Report of the European DNA Profiling Group (EDNAP)--an investigation of the hypervariable STR loci ACTBP2, APOAI1 and D11S554 and the compound loci D12S391 and D1S1656.

This paper describes the results of three collaborative exercises which continues the EDNAP theme to explore whether uniformity of DNA profiling results could be achieved between European laboratories using STRs. In an earlier exercise, complex hypervariable AAAG-repeat STR loci were investigated, but reproducibility was found to be poor because of the variation of techniques used by participating laboratories. In the exercise reported here, an internal allelic ladder composed of ACTBP2 and D11S554 fragments was distributed. This ladder was used to size ACTBP2 analysed by a "singleplex" PCR amplification and D11S554 combined with APOAI1 in a separate "duplex" reaction. Laboratories were asked to test 7 blood stains, one of which was a known control, and to report the results to the co-ordinating laboratory. The exercise demonstrated that ACTBP2 showed good reproducibility between laboratories, whereas further testing would be needed to validate APOAI1 and D11S554 for interlaboratory comparisons. In separate exercises, the simple loci D12S391 and D1S1656 were tested; both of these showed excellent reproducibility between laboratories.
