Improved equipment for abdominal fetal electrocardiogram recording: description and clinical evaluation.

Reliable computer-based equipment for transabdominal or indirect recording of fetal electrocardiogram (FECG) is described. The proposed equipment allows a real-time displaying of the signals (fetal + maternal ECG) without averaging procedures and it does not require the employment of a shielded room; moreover, it is user-friendly to medical personnel. An elementary form of semi-automatic computation of the fetal heart rate (FHR) was also implemented. The equipment simultaneously acquires three signals from seven electrodes, six placed on the maternal abdomen following the three space axes, and one placed on the left leg as a indifferent electrode. The signals are magnified and analogically filtered before undergoing digital finite impulse response (FIR) filter. Then the signals are displayed on the screen of a personal computer (PC). The PC also provides the possibility of storing the acquired signals for further analysis of elaborations. The quality of the recordings allows the analysis of both the rapid and slow electrical phenomena of the fetal heart, and it is not significantly influenced by the occurrence at the same time of uterine contractions. The performance of this method was assessed in 140 pregnant women with gestation periods of 29-42 weeks. In 131 cases (93.6%) the fetal QRS complex was detected and the FHR was obtained. A reliable evaluation of P and QRS waves and of ST interval, in spite of the interference of the maternal complexes, was possible in 102 cases (72.8%).

Occupational asthma in sawmills of eastern Canada and United States.

Eleven individuals with a history of work-related asthma are described. They were employed in 10 different sawmills of northwestern and southeastern Quebec and Maine where coniferous trees (spruces, firs, and pines) are cut into boards. Duration of exposure and symptomatology varied from 1.5 to 40 years and 0.5 to 10 years, respectively. Ten subjects were atopic and seven demonstrated immediate skin reactivity to mixed tree pollens. The diagnosis of occupational asthma was confirmed by significant and sustained changes in serial peak expiratory flow rates at work as compared with a period off work in every individual and significant changes in bronchial responsiveness to histamine at work as compared with a period off work in eight individuals or significant changes in FEV1 at work in the three other subjects. Specific inhalation tests by exposing four workers to sawdust in our laboratory were negative. We conclude that working in this specific type of sawmills of eastern Canada and northeastern United States can cause occupational asthma. Although the causative agent is unknown, the presence of atopy and/or immediate skin reactivity to tree pollens and/or bronchial hyperresponsiveness might be risk factors.

An optimal linear filter for the reduction of noise superimposed to the EEG signal.

In the present paper a procedure for the reduction of super-imposed noise on EEG tracings is described, which makes use of linear digital filtering and identification methods. In particular, an optimal filter (a Kalman filter) has been developed which is intended to capture the disturbances of the electromyographic noise on the basis of an a priori modelling which considers a series of impulses with a temporal occurrence according to a Poisson distribution as a noise generating mechanism. The experimental results refer to the EEG tracings recorded from 20 patients in normal resting conditions: the procedure consists of a preprocessing phase (which uses also a low-pass FIR digital filter), followed by the implementation of the identification and the Kalman filter. The performance of the filters is satisfactory also from the clinical standpoint, obtaining a marked reduction of noise without distorting the useful information contained in the signal. Furthermore, when using the introduced method, the EEG signal generating mechanism is accordingly parametrized as AR/ARMA models, thus obtaining an extremely sensitive feature extraction with interesting and not yet completely studied pathophysiological meanings. The above procedure may find a general application in the field of noise reduction and the better enhancement of information contained in the wide set of biological signals.

Sister-chromatid exchanges in Picea abies--a test for genotoxicity in forest trees.

A genotoxicity test, based on the evaluation of sister-chromatid exchange frequencies, has been developed for the spruce fir. The basic frequency was 36.9 SCEs/cell. Mitomycin C treatment (MMC, 5 x 10(-6) M, 0.5 h) doubled the 'spontaneous' SCE frequency, maleic hydrazide treatment (MH, 5 x 10(-4) M, 0.5 h) increased it nearly 7-fold. This corresponds to data obtained previously for Vicia faba. Chromatid-type aberrations were induced by the same mutagens (MH, 5 x 10(-4) M, 0.5 h; MMC 2 x 10(-5) M, 1 h) or by triethylenemelamine (TEN, 2 x 10(-4) M, 0.5 h). MH treatment resulted in aberration yields comparable to those observed in Vicia faba, MMC and TEM were less efficient aberration inducers in P. abies. While SCEs may be counted for single chromosomes, for reliable evaluation of chromatid aberrations large numbers of complete and well spread metaphases have to be inspected.

The contribution of ammonia emissions from agriculture to the deposition of acidifying and eutrophying compounds onto forests.

In the vicinity of a large ammonia emission area, dry and wet deposition of acidifying and eutrophying compounds onto Douglas Fir forests was studied by sampling throughfall, stemflow and bulk precipitation. Deposition amounts of NH(4)(+) and SO(4)(2-) were recognised to be among the highest of Central Europe, resulting in extremely high inputs of (potential) acid to the forest soils (13.1 kEq ha(-1) year(-1)). The contribution of NH(3) emissions from agriculture to the total acid deposition to the forests was 52%. The total nitrogen deposition amounted to 115.0 kg ha(-1) year(-1), 83% originating from NH(3) emissions and 17% from NO(x) emissions. Calculated mean dry deposition velocities of NH(3) and SO(2) were much larger than reported in the literature. A synergistic effect between NH(3) and SO(2) in the process of dry deposition is suggested and evidence for this effect is discussed. When deposition models do not take this interaction into account, they will underestimate NH(3) and SO(2) deposition amounts in areas with intensive animal husbandry.

General methods and materials.

In the winter of 1982-83 an experiment was initiated at the University of Hohenheim to investigate the long-term effects of near ambient concentrations of O(3), SO(2), and simulated acidic rain and fog, alone and in combination, on mineral cycling, physiology, biochemistry and anatomy of Norway spruce (Picea abies L. Karst.), Silver fir (Abies alba Mill.) and European beech (Fagus silvatica L.) seedlings in model ecosystems. Open-top chambers above soil lysimeters were protected with shelters 1 m above the open-top against intrusion of ambient dust and rain. The soil, a replaced acid brown earth with a base saturation of 20-30% was from the Black Forest region of the Federal Republic of Germany. This paper describes the technical design aspects of the study, including chamber construction and ventilation, SO(2) and O(3) application and monitoring, simulated precipitation forms and dispensing features, and the environmental parameters monitored. A short description of the plants used and the soil status is also presented. Chamber climatic and edaphic conditions allowed tree seedling normal growth over the five years of study. The pollutant levels within the appropriate chambers were comparable to ambient concentrations found at southwest German forest sites and varied with weather conditions.

Flow rates of ions in waters percolating through a model ecosystem with forest trees.

From 1983-88 the long-term effects of low level exposure with O(3), SO(2) and simulated acid rain on mineral cycling in model ecosystems with spruce, fir and beech seedlings were investigated. Systems consisting of open-top chambers built above lysimeters were protected against the intrusion of ambient rain and dust. As part of the investigations on mineral cycling the fluxes of elements with water input and output of the canopy and soil compartments are presented. During the 5 year duration of the experiment, pronounced effects on canopy deposition and cation leaching were observed. Most noticeable were throughfall enrichment with sulfate through dry deposition of SO(2) as influenced by duration of needlewetting and factors promoting SO(2) oxidation. Depending on sulfur deposition, leaching of calcium, magnesium, manganese, zinc and ammonium from canopies was elevated, in total leading to enhanced soil input of acid. After 15 months, the water percolating the soils in the lysimeters of these treatments was acidified, with elevated flowrates of sulfate, manganese, calcium and magnesium. The results on canopy/soil leaching are compared to those from old conifer stands in the field.

Mineral content in the soil and tree foliage.

Seedlings of fir (Abies alba Mill.) and spruce (Picea abies L. Karst.) were fumigated with SO(2), O(3) and SO(2) + O(3) in open-top chambers (OTCs) for almost 5 vegetation periods. As background stress, simulated rain of pH 4.0 was applied. Nutrient content of soil, soil solutions, and trees was investigated and balanced. In the upper partition of the soil high concentrations of exchangeable Ca(2+) were found in all chambers. The SO(2) and SO(2) + O(3) treatments led to increased Ca(2+), Mg(2+) and Mn(2+) concentrations in soil solution and the pool of exchangeable protons increased. This response was most evident in the SO(2) and SO(2) + O(3) chambers and less clear in the filtered pH 5.0 control chamber. In the SO(2) treatment increased Mn and S levels were found in the needles. Ca content in the needles showed a decreasing trend. O(3) alone had no consistent effect on needle nutrient content.

Measurement of dry deposition of ammonia on a forest.

Ammonia concentration gradients above a Douglas fir canopy were measured from 16 August to 31 December 1989 by two automated high-precision thermodenuders at the location Speulderbos in The Netherlands. Concentration gradients were used to calculate the dry deposition flux of ammonia via flux-gradient theory. Meteorological data were obtained from a nearby tower. Ammonia concentrations were highly variable with highest values during the night. Concentration gradients were very small during daytime and quite large at night. Median values of the calculated deposition flux and deposition velocity were 0.1 microg m(-2) s(-1) and 3.2 cm s(-1) respectively (N=1624).

Comparative impacts of forest harvest and acid precipitation on soil and streamwater acidity.

Annual H+ budgets were calculated for a catchment with a 65-year-old spruce-fir forest, and for an adjacent catchment at three years after a whole-tree harvest. In the harvested catchment sinks for H+ due to weathering and mineralization reactions were 85% greater, and sources of H+ due to biomass uptake were 60% greater than in the undisturbed catchment. There was an overall net consumption of 550 eq H+ ha(-1) year(-1) at year 3 after harvest compared to a net generation of 520 eq H+ ha(-1) year(-1) for the 65-year-old forest. Soil pH increased by 0.2-0.4 units soon after harvest, but there was little change in pH of streamwater from the harvested watershed. The whole-tree harvest resulted in a total production of about 30,000 eq H+ ha(-1) due to biomass removal. In contrast, wet and dry deposition at rates measured in this study could add more than 50,000 eq H+ ha(-1) in the 65-year period before the next harvest. Reducing the intensity of harvest may lessen long-term impacts of these sources of H+ on acidification of soils and streams.

Effects of L-alanine injection and post-hatch deprivation of feed and water on glycogen reserves in turkey poults.

1. Glycogen concentration (mg/g) in liver, epiphyseal bone (proximal femoral chondyle) and toe (first medial phalanges) declined during post-hatch holding of poults fir up to 72 hr without feed and water. 2. Muscle (gastrocnemius) glycogen concentration was unchanged by post-hatch holding. 3. Glycogen concentration (mg/g) in the whole body was unchanged while whole body glycogen (total mg) content was decreased by holding. 4. Injections of 1 ml of 1 M L-alanine did not enhance whole body glycogen stores.

Evaluation of methods for detecting ecological effects from genetically engineered microorganisms and microbial pest control agents in terrestrial systems.

This report summarizes and evaluates research from several laboratories that deals with the detection of ecological effects induced through exposure of microbes or plants to genetically engineered microorganisms (GEMs) and microbial pest control agents (MPCAs). Some 27 potential endpoints for measuring effects have been studied. Perturbations induced by GEMs have been detected in about one-half of these endpoints. Detectable effects have been recorded for over half of the 16 species of bacteria and fungi studied. The effects caused by GEMs and MPCAs include inhibition of beneficial mycorrhizal fungi growing on Douglas fir seedling roots, depression in plant root and shoot growth, inhibition of predatory soil protozoa, accumulation of a toxic metabolite during biodegradation that inhibits soil fungi, increased microbial community respiration due to rapid lignin breakdown in soil, and the displacement of a broad group of gram-negative bacteria that inhabit the root surface of cereal crops. These effects were usually, but not always, of short duration. However, some of the changes were irreversible during the observation time of days, weeks, or in one case, months.

Computationally efficient sub-band coding of ECG signals.

A data compression technique is presented for the compression of discrete time electrocardiogram (ECG) signals. The compression system is based on sub-band coding, a technique traditionally used for compressing speech and images. The sub-band coder employs quadrature mirror filter banks (QMF) with up to 32 critically sampled sub-bands. Both finite impulse response (FIR) and the more computationally efficient infinite impulse response (IIR) filter banks are considered as candidates in a complete ECG coding system. The sub-bands are threshold, quantized using uniform quantizers and run-length coded. The output of the run-length coder is further compressed by a Huffman coder. Extensive simulations indicate that 16 sub-bands are a suitable choice for this application. Furthermore, IIR filter banks are preferable due to their superiority in terms of computational efficiency. We conclude that the present scheme, which is suitable for real time implementation on a PC, can provide compression ratios between 5 and 15 without loss of clinical information.

Altering product outcome in Abies grandis (-)-limonene synthase and (-)-limonene/(-)-alpha-pinene synthase by domain swapping and directed mutagenesis.

(-)-(4S)-limonene synthase (LS) and (-)-(4S)-limonene/(-)-(1S, 5S)-alpha-pinene synthase (LPS) from grand fir (Abies grandis) exhibit nearly 91% sequence identity (93% similarity) at the amino acid level, yet produce very different mixtures of monoterpene olefins. To elucidate critical amino acids involved in determining monoterpene product distribution, a combination of domain swapping and reciprocal site-directed mutagenesis was carried out between these two enzymes. Exchange of the predicted helix D through F region in LS gave rise to an LPS-like product outcome, whereas reciprocal substitutions of four amino acids in LPS (two in the predicted helix D and two in the predicted helix F) altered the product distribution to that intermediate between LS and LPS, and resulted in a 5-fold increase in relative velocity. These results, in conjunction with modeling of the two enzymes, suggest that amino acids in the predicted D through F helix regions are critical for product determination.

Mutational analysis of a monoterpene synthase reaction: altered catalysis through directed mutagenesis of (-)-pinene synthase from Abies grandis.

Two monoterpene synthases, (-)-pinene synthase and (-)-camphene synthase, from grand fir (Abies grandis) produce different product mixtures despite having highly homologous amino acid sequences and, presumably, very similar three-dimensional structures. The major product of (-)-camphene synthase, (-)-camphene, and the major products of (-)-pinene synthase, (-)-alpha-pinene, and (-)-beta-pinene, arise through distinct mechanistic variations of the electrophilic reaction cascade that is common to terpenoid synthases. Structural modeling followed by directed mutagenesis in (-)-pinene synthase was used to replace selected amino acid residues with the corresponding residues from (-)-camphene synthase in an effort to identify the amino acids responsible for the catalytic differences. This approach produced an enzyme in which more than half of the product was channeled through an alternative pathway. It was also shown that several (-)-pinene synthase to (-)-camphene synthase amino acid substitutions were necessary before catalysis was significantly altered. The data support a model in which the collective action of many key amino acids, located both in and distant from the active site pocket, regulate the course of the electrophilic reaction cascade.

MCNP study for epithermal neutron irradiation of an isolated liver at the Finnish BNCT facility.

A successful boron neutron capture treatment (BNCT) of a patient with multiple liver metastases has been first given in Italy, by placing the removed organ into the thermal neutron column of the Triga research reactor of the University of Pavia. In Finland, FiR 1 Triga reactor with an epithermal neutron beam well suited for BNCT has been extensively used to irradiate patients with brain tumors such as glioblastoma and recently also head and neck tumors. In this work we have studied by MCNP Monte Carlo simulations, whether it would be beneficial to treat an isolated liver with epithermal neutrons instead of thermal ones. The results show, that the epithermal field penetrates deeper into the liver and creates a build-up distribution of the boron dose. Our results strongly encourage further studying of irradiation arrangement of an isolated liver with epithermal neutron fields.

Measurement of free beam neutron spectra at eight BNCT facilities worldwide.

Eight epithermal neutron beams, constructed for clinical or preclinical studies of NCT, have been dosimetrically characterized by in-air measurements with a set of activation foils for the determination of the neutron energy spectra in free beam. Measurements have been made on the already closed epithermal BNCT facility at the BMRR of the Brookhaven National Laboratory, on the HFR at JRC in Petten, The Netherlands, on the epithermal mode beam at KURRI, Japan, on the fission converter beam at MIT, USA, on the epithermal beam of the RA-6 facility in Bariloche, Argentina, on the epithermal beam at WSU, USA, on the mixed mode beam at JRR-4 at JAERI, Japan, as well as on the epithermal beam at FiR 1 at VTT, Espoo, Finland.

Spatial characterization of BNCT beams.

The space distribution of the epithermal neutron flux was determined for the epithermal neutron beams of several NCT facilities in USA (FCB at MIT), Europe (HFR at JRC, Petten; FiR at VTT, Espoo; LVR-15 at NRI, Rez) and Japan (JRR-4 at JAERI, Tokai). Using p-n diodes with (6)Li radiator and the set of Bonner sphere spectrometer (BSS) the beams were quantified in-air. Axial beam profiles along the beam axes and the radial distributions at two distances from the beam aperture were measured. Except for the well-collimated HFR beam, the spatial characteristics of the other studied beams were found generally similar, which results from their similar designs.
