Linear relationship between the maintenance of hippocampal long-term potentiation and retention of an associative memory.

The hypothesis that the maintenance or decay of an associative memory trace after an extended retention interval is a function of the residual strength of the synapses originally strengthened during learning was examined in a classical conditioning paradigm in which high-frequency stimulation of a hippocampal input--the medial perforant path--served as a conditioned stimulus. Rats received perforant path stimulus-foot shock pairings while engaged in a previously acquired food-motivated lever-pressing task. Conditioned suppression of lever pressing was the behavioral measure of learning and retention of the association. Stimulus trains to the perforant path at an intensity above the threshold for eliciting a population spike induced long-term potentiation of synaptic transmission in the dentate gyrus. Synaptic potentials recorded extracellularly in the dentate gyrus were subsequently monitored for 31 days to examine quantitatively the decay of synaptic potentiation, a period after which retention of the learned association was assessed. All rats learned the association to a similar extent and displayed equivalent amounts of long-term potentiation by the end of conditioning. A slowly decaying function of synaptic potentiation was observed in remembering rats, i.e., rats with high retention performance after the 31-day learning-to-retention interval, while forgetting was associated with a rapid decay of long-term potentiation. Behavioral performance at the long-term memory test was linearly correlated with the amplitude of long-term potentiation maintained just prior to the retention test. The results favor the hypothesis that long-term associative memory depends, at least in part, on the maintenance of elevated synaptic strengths in the pathway activated during learning and suggest a role for the lasting component of long-term potentiation in the maintenance of memory.

Variation in eggshell characteristics and in intrauterine egg retention between two oviparous clades of the lizard Lacerta vivipara: insight into the oviparity-viviparity continuum in squamates.

The concept of the oviparity-viviparity continuum refers to the wide range in the length of intrauterine egg retention and, hence, in the stage of embryonic development at oviposition existing in squamates. The evolutionary process underlying this continuum may involve not only a lengthening of egg retention in utero, but also a marked reduction in the thickness of the eggshell. The idea that there may exist a negative correlation between the developmental stage reached by the embryo at oviposition and the eggshell thickness within squamates, although supported by the comparison of oviparous vs. viviparous species, has seldom been evaluated by comparing eggshell thickness of oviparous forms with different lengths of intrauterine egg retention. Eggs of two distinct oviparous clades of the lizard Lacerta vivipara were compared. The eggs laid by females from Slovenian and Italian populations have thicker eggshells, contain embryos on average less developed at the time of oviposition, and require a longer incubation period before hatching than the eggs laid by females from French oviparous populations. Our data and several other examples available from the literature support the idea that the lengthening of intrauterine retention of eggs and the shortening of the subsequent external incubation of eggs are associated with reduction in the thickness of the eggshell, at least in some lineages of oviparous squamates. The current hypotheses that may account for this correlation are presented and a few restrictions and refinements to those hypotheses are discussed. In particular, other changes, such as increased vascularization of the oviduct and of the extraembryonic membranes, may play the same role as the decrease of eggshell thickness in facilitating prolonged intrauterine egg retention in squamates. Future studies should also consider the hypothesis that the length of intrauterine retention might directly depend on the extent of maternal-fetal chemical communication through the eggshell barrier.

Studies of in vitro skin permeation and retention of a leukotriene antagonist from topical vehicles with a hairless guinea pig model.

A leukotriene antagonist [Ro 23-3544; 6-acetyl-7-[5-(4-acetyl-3-hydroxy-2-propylphenoxy)pentyloxy] -3,4-dihydro-2H-1-benzopyran-2-carboxylic acid; 1] was studied in vitro for its permeation through and retention in hairless guinea pig skin from various topical vehicles. Both the free acid and the sodium salt forms of the drug were used. The vehicles evaluated were polyethylene glycol 400, propylene glycol, dimethyl sulfoxide (DMSO), C12-C15 alcohol lactates, dimethyl isosorbide, butyrolactone, methylpyrrolidone, hexyl laurate, isopropyl myristate, and caprylic/capric triglyceride (Neobee M5). For the salt form of the drug, the highest permeability coefficient and retention were obtained from DMSO and methylpyrrolidone, respectively. For the acid form, however, the highest permeability coefficient and retention were obtained from hexyl laurate and DMSO, respectively. The highest permeation and retention values were not obtained from the same vehicle for either the salt or the acid form of the drug. This observation questions the validity of using permeation (flux) measurements to screen topical drugs and formulations. Although the precise reasons for this lack of correlation between permeation and retention are not known at this time, this study has shown that the solubility parameters of the drug and the vehicles used may play an important role. It seems logical to conduct skin retention studies rather than flux measurements in evaluating drug delivery from dermatological products.

Effects of verapamil, ouabain, and ethacrynic acid on calcium retention in perfused rat kidneys.

A Sprague-Dawley rat kidney perfusion technique was used in situ to study the effect of verapamil, ouabain, and ethacrynic acid on renal calcium retention. The technique involves perfusion of the kidneys via the abdominal aorta and then through the left and right renal arteries and dorsal aorta. Verapamil (1 mM) in Krebs-improved Ringer solution increased calcium retention in the kidneys by approximately 117% compared with controls. With Na-free Krebs-improved Ringer solution, calcium retention increased by only 92%. However, in Krebs-improved Ringer solutions containing 59 and 122 mequivalents of Na, calcium retention in the kidney increased by 46 and 43%, respectively, compared with controls. With Krebs-improved Ringer solution containing 15 mM ouabain, calcium retention in the kidney decreased by 29.5%, whereas with 15 mM ouabain plus 1 mM ethacrynic acid in the perfusate, the effect on calcium retention in the kidney was insignificant compared with controls. These results suggest that two sodium-dependent calcium-transporting systems exist at the peritubular side of the kidney tubules: (1) a Na(+)-Ca+2 countertransport system sensitive to verapamil and (2) a Na(+)-Ca+2 cotransport system sensitive to the intracellular concentration of sodium.

Quantification of naphthalenes in jet fuel with GC x GC/Tri-PLS and windowed rank minimization retention time alignment.

Comprehensive, two-dimensional gas chromatography (GC x GC) is used in conjunction with trilinear partial least squares (Tri-PLS) to quantify the percent weight of naphthalenes (two-ring aromatic compounds) in jet fuel samples. The increased peak capacity and selectivity of GC x GC makes the technique attractive for the rapid, and possibly less tedious analysis of jet fuel. The analysis of complex mixtures by GC x GC is further enhanced through the use of chemometric techniques, including those designed for use on 2-D data such as Tri-PLS. Unfortunately, retention time variation, unless corrected, can be an impediment to chemometric analysis. Previous work has demonstrated that the effects of retention time variation can be mitigated in sub-regions of GC x GC chromatograms through the application of an objective retention time alignment algorithm based on rank minimization. Building upon this previous work, it is demonstrated here that the effects of retention time variation can be mitigated throughout an entire GC x GC chromatogram with an objective retention time alignment algorithm based on windowed rank minimization alignment. A significant decrease in calibration error is observed when the algorithm is applied to chromatograms prior to construction of Tri-PLS models. Fourteen jet fuel samples with known weight percentages of naphthalenes (ASTM D1840) were obtained. Each sample was subjected to five replicate five-minute GC x GC separations over a period of two days. A subset of nine samples spanning the range of weight percentages of naphthalenes was chosen as a calibration set and Tri-PLS calibration models were subsequently developed in order to predict the naphthalene content of the samples from the GC x GC chromatograms of the remaining five samples. Calibration models constructed from GC x GC chromatograms that were retention time corrected are shown to exhibit a root mean square error of prediction of roughly half that of calibration models constructed from uncorrected chromatograms. The error of prediction is lowered further to a value that nearly matches the uncertainty in the standard percent weight values (ca. 1% of the median percent volume value) when the aligned chromatograms are truncated to include only regions of the chromatogram populated by naphthalenes and compounds of similar polarity and boiling point.

Shape retention in porcine-septal cartilage following Nd:YAG (lambda = 1.32 microm) laser-mediated reshaping.

BACKGROUND AND OBJECTIVE: Photothermal heating of mechanically deformed cartilage accelerates stress relaxation and results in sustained shape change. In this study, shape retention was measured in Nd:YAG laser reshaped porcine septal cartilage. MATERIALS AND METHODS: Specimens were laser reshaped either 4 (Group I) or 28 hours (Group II) following extraction from the crania. Specimens were bent into approximately semicircular shapes and irradiated half way between the endpoints of the semicircle. Resultant bend angle was calculated based on linear measurements. Shape retention was calculated by comparing resultant curvature with pre-irradiation measurements. RESULTS: Mechanical deformation alone resulted in initial bend angles varying from 188 degrees to 229 degrees. Resultant bend angles varied from 84 degrees to 194 degrees corresponding to shape retention varying from 58 to 75%. Non-irradiated cartilage retained less than 46% of the original bend. Shape retention was greater in Group II, compared to Group I. In Group I, no cephalocranial difference in shape retention was observed, though in Group II greater shape retention was observed in rostral specimens. CONCLUSION: While laser heating does significantly reshape cartilage, clinical use of this technology will require "overbending" of the cartilage graft to compensate for this memory effect. The degree of overbending is likely to vary with cartilage type and location.

Discriminating the xanthones in an extract of Swertia franchetiana by retention parameters.

The quantitative structure--retention relationship is one of the most actively studied topics in the field of chromatography. In this paper, retention parameters of components were used to discriminate the xanthones in a methanol extract of Swertia franchetiana The extract was analysed by HPLC under two different multistage linear gradient conditions and the retention parameters calculated from these retention data. It was found that the retention parameters of xanthones are in a specific region in the plot of log k(w) vs. S and the xanthones in the extract could be distinguished from other components by this feature. Furthermore, xanthone aglycones and xanthone glucosides could also be discriminated by retention parameters.

New technique for increasing retention of arginine on an anion-exchange column.

A method is described for enhancing retention of arginine on a pellicular anion-exchange column. Arginine exhibits adjustable increases of retention time that are dependent on the acidity of standard or sample matrix. This effect is based on interactions of the protonated form of arginine with the residual cation-exchange groups on the core beads of pellicular particles. The relative magnitude of retention time shift of arginine is evaluated for identical concentrations of hydrochloric, sulfuric, and perchloric acids. Although the direct addition of acid is very effective in influencing the retention of arginine, it affects peak shapes and retention of other peaks in the chromatographic separation. The new technique-acid coinjection-achieves a similar retention enhancement for arginine with only a minimal effect on the rest of the separation. Detection limits, reproducibility results, and calibration data are presented for the chromatography of amino acids with acid coinjection. Improved resolution of arginine is demonstrated with chromatograms of soybean hydrolysate and cell culture samples.

Prevention of superficial wound separation with subcutaneous retention sutures.

The objective of this prospective study was to determine the efficacy of subcutaneous retention sutures in the prevention of superficial wound separation in obese women undergoing gynecologic surgery. From 1985 to 1990, 80 women with subcutaneous tissue of 5 cm or greater undergoing laparotomy were randomly assigned to either standard wound closure (37 patients) or closure including subcutaneous retention sutures (43 patients). We examined the relationship of the following factors to the development of superficial wound dehiscence: malnutrition (total protein < 6.0 mg/dl, albumin < 3.0 mg/dl), minidose heparin (5000 U subcutaneously every 8 hr for 5 days), diabetes mellitus, prolonged operative time (> 180 min), intraoperative spillage of enteral contents, and subcutaneous retention sutures. In a univariate analysis, malnutrition (P = 0.02), the use of heparin (P = 0.03), and the absence of retention sutures (P = 0.02) were associated with an increased risk of separation. In a multivariate analysis, malnutrition (P = 0.02), low-dose heparin (P = 0.03), and the absence of retention sutures (P = 0.04) retained their independent association with superficial wound separation. This study suggests that the use of subcutaneous retention sutures reduces the incidence of superficial wound separation in obese women undergoing gynecologic surgery.

Diazepam blocks the interfering effect of post-training behavioral manipulations on retention of a shuttle avoidance task.

Rats were submitted to a training and a test session of shuttle avoidance. Exposure to a session of extinction of this task either 2 or 24 h after training interfered with retention test performance. Exposure to an open field 2, but not 24 h after the avoidance training also interfered with retention. Diazepam blocked the deleterious effect of extinction and of the open field on retention of the avoidance task. Diazepam alone had no effect when given after avoidance training; it did, however, also interfere with retention when given prior to training. It is likely, therefore, that diazepam cancelled the effect of the extinction or of the open field on avoidance retention because of anterograde amnesia (i.e., it prevented the recording of these tasks). The deleterious effect of the open field on retention of shuttle avoidance can be explained by retroactive interference caused by the addition of information. It is not due to a direct influence on retrieval, it is not due to extinction, and it had to be recorded 2 h after training in order to the effective.

Influence of sous vide processing, steaming and boiling on vitamin retention and sensory quality in broccoli florets.

In this study the effect of sous vide processing, steaming and traditional boiling of broccoli florets on retention of ascorbic acid, vitamin B6 and folacin was investigated. Freshly prepared samples were also sensorily evaluated. In all heat treatments ascorbic acid was found to have the highest retention, and was therefore judged not to be suitable as an indicator for vitamin retention in general. When the three types of heat treatment were compared, boiling showed lowest retention of all the vitamins examined (45-64% in 5 min), whereas sous-vide processing gave the highest retentions (97-100% in 5 min). Retentions during steaming were a little lower (83-100% in 5 min). Sensory evaluation showed that sous-vide cooked and steamed broccoli florets generally had higher acceptability than boiled.

Cocaine enhances retention of avoidance conditioning in rats.

The effects of post-training cocaine administration were tested on retention of a one-way active avoidance task in rats. A 5.0 mg/kg IP dose of cocaine enhanced retention of the avoidance task, in three separate experiments, as indicated by an increase in the number of avoidances made when animals were tested 24 h after training, while both a lower (2.5 mg/kg) and a higher (7.5 mg/kg) cocaine dose had no effect. Lidocaine (4-8 mg/kg) administered post-training did not reliably affect retention in the same task. Cocaine's ability to enhance retention depended on the interval between training and drug injection such that only cocaine administered directly after training enhanced retention the following day. The results show that post-training cocaine administration enhances retention of an active avoidance task in rats, and that this effect is probably independent of the anesthetic properties of the drug.

The retention catalogue: an instrument for quality management.

UNLABELLED: GOAL AND MATERIAL: We evaluated the clinical use of the "retention catalogue" employing a standardized questionnaire. RESULTS: Statistical evaluation of the standardized questionnaire demonstrated that the retention catalogue (RC) is an effective method in daily practice for orthodontists to convey patient information on the causes and prevention of relapses in crowding. CONCLUSIONS: Routine utilization of the retention catalogue is recommended. It both contributes to successful retention management and offers the orthodontist a certain degree of legal protection. Retention behavior of large polycyclic aromatic hydrocarbons on metalloprotoporphyrin-silica stationary phases.

The retention behavior of large polycyclic aromatic hydrocarbons (LPAHs) (> or = 7 rings) on newly developed metalloplotoporphyrin (MProP)-silica stationary phases is examined and the results are compared to previously reported data for retention of the same solutes on commercially available phases. HPLC columns packed with FeProP-silica are shown to exhibit unique shape selectivity for LPAH retention, with the planar LPAHs always retained much longer than corresponding non-planar solutes. Solute planarity, length to breadth ratio (L/B value), and number of carbon atoms within the LPAHs are all demonstrated to contribute to the retention sequence observed. Further, the retention of LPAH solutes on FeProP-silica phases is shown to be more predictable than on other reversed-phase columns, with the elution sequence constant regardless of the mobile phase composition. Due to the extremely high planar selectivity of FeProP-silicas with respect to LPAH retention, it is envisioned that columns packed with these phases could be used in conjunction with existing commercial columns to devise Inethods for more efficient separation of complex mixtures of LPAHs in environmental and other samples.

Iodized oil retention due to postbiopsy arterioportal shunt: a false positive lesion in the investigation of hepatocellular carcinoma.

BACKGROUND: Iodized-oil computed tomography (CT) is useful for the diagnosis of hepatocellular carcinoma, but there may be false-positive results in patients who have undergone some percutaneous transhepatic interventional procedures. The purpose of the present study was to verify the correlation between subsegmental hepatic parenchymal retention of iodized oil on CT and the arterioportal shunt caused by liver biopsy. METHODS: Iodized-oil CT scans were reviewed in 24 patients with hepatic tumors who had liver biopsy and subsequent iodized oil chemoembolization. Iodized oil chemoembolization was performed shortly after biopsy (1-10 days; mean = 2.6 days). The results were correlated with hepatic arteriography, with a special emphasis on the presence of hepatic arterioportal shunt. RESULTS: Wedge-shaped subsegmental retention of iodized oil along or adjacent to the biopsy needle path was observed in iodized-oil CT in 17 of the 24 patients. In three patients, there was subsegmental enhancement on prebiopsy helical dynamic liver CT at the same area of iodized oil retention, and therefore iodized oil retention was considered to be due to hepatocellular carcinoma. In the remaining 13 (54%) patients, the peripheral iodized oil retention was considered to be due to biopsy-induced arterioportal shunt. In all these patients, arterioportal shunt was confirmed by hepatic arteriography. CONCLUSION: Wedge-shaped hepatic parenchymal retention of iodized oil is commonly observed in iodized-oil CT due to biopsy-induced arterioportal shunt, and this appearance should not be confused with a hepatocellular carcinoma.

Patterns of retention in a preventive intervention with ethnic minority families.

This study examined socio-demographic, child, and family-level correlates of retention patterns among parent participants of Familias Unidas/SEPI (Coatsworth, Pantin, & Szapocznik, 2002), a randomized, controlled trial of a family-focused preventive intervention. The current study's aim was to identify variables that could be used to classify ethnic minority (African American and Hispanic) caregivers (N = 143) into their known patterns of retention across 30 sessions of the intervention. Person-centered analyses identified three broad attendance pattern groups: (a) non-attenders; (b) variable-attenders; (c) consistent-high-attenders. Subgroups of the variable-attender group included: (a) dropouts; (b) variable-low-attenders; (c) variable-high-attenders. Four socio-demographic indicators were significant discriminators of the broad retention patterns. Three family-level factors were significant discriminators of the variable-attender subgroups. Additional significant mean/rate differences among retention pattern groups on correlates are reported. Implications for how retention is examined in preventive interventions and for developing intervention strategies for improving retention rates are discussed.

Arterial underfilling does not cause sodium retention in cirrhosis.

PURPOSE: To test the peripheral arterial vasodilation hypothesis of sodium retention in cirrhosis. This states that sodium retention is triggered by arterial underfilling and predicts that development of sodium retention will be associated with significant and related declines in indices of arterial filling that reverse when sodium retention resolves. DESIGN: Longitudinal evaluation of a cohort of patients with alcoholic liver disease. PATIENTS AND METHODS: Eighteen men, 8 of whom were studied twice, 3 three times, 2 four times, and 5 five times (40 between-study comparisons). Between 23 studies, the patients were ascites-free (Group NN). Ascites spontaneously disappeared between seven studies (Group YN), appeared between six studies (Group NY), and remained present between four studies (Group YY). Between-study changes in blood volume, arterial blood pressure, cardiac output, systemic vascular resistance, left atrial volume, left ventricular diastolic diameter, aortic root diameter, aortic blood velocity, plasma norepinephrine and atrial natriuretic factor concentrations, plasma renin activity, and urinary sodium excretion were evaluated by paired t-tests. These changes were also compared among groups by analysis of variance. In addition, correlations among the changes were sought. RESULTS: Systolic, diastolic, and mean arterial pressures, left ventricular diastolic diameter, aortic root diameter, stroke volume, cardiac output, plasma norepinephrine concentration, and systemic vascular resistance were unchanged between studies. Left atrial volume increased between studies in Group NY. Pulse pressure fell more in Group NY than in Groups NN and YN, principally as a result of a decline in systolic blood pressure. Plasma norepinephrine concentration, plasma renin activity, and blood volume rose more in Group NY than in Groups NN, YN, and YY. Changes in both systolic and pulse pressures were directly correlated with the change in sodium excretion but unrelated to the change in plasma norepinephrine concentration. Changes in plasma norepinephrine concentration and plasma renin activity were unrelated to changes in blood pressure, systemic vascular resistance, and urinary sodium excretion. CONCLUSIONS: None of the indices of arterial filling tested except pulse pressure were related to sodium retention. Reduced pulse pressure is inconsistent with arterial underfilling, as peripheral vasodilation instead increases pulse pressure by increasing diastolic run-off. These data do not support the hypothesis that arterial underfilling is the stimulus for sodium retention in alcoholic cirrhosis.

Relationship between binding affinity, retention and sensitivity of human rhabdomyosarcoma xenografts to Vinca alkaloids.

Xenografts of human rhabdomyosarcoma (RMS) have been derived that differ in their degree of sensitivity to Vinca alkaloids. Lines Rh12 and Rh18 demonstrated, respectively, high and moderate sensitivity to vincristine (VCR), but showed little responsiveness to vinblastine (VLB) in vivo. Rh18/VCR-3, a subline of Rh18 selected for resistance to VCR under in situ conditions, was insensitive to further challenge with VCR. Resistance was associated with elimination of the agent in a biphasic manner, whereas sensitivity to VCR corresponded to very prolonged drug retention in sensitive neoplastic tissues. The initial half-times for drug retention in tumors in vivo (t1/2 alpha) correlated with the degree of sensitivity of tumors to Vinca alkaloids, decreasing t1/2 alpha being associated with decreased sensitivity. A single binding species was observed when membrane-free supernatant fractions were incubated at 37 degrees for 15 min with 10.4 nM [3H]VCR and analyzed by gel filtration HPLC. The protein eluted with a retention time of 57 min and corresponded to a molecular weight (Mr) of approximately 113,000 daltons, agreeing very closely with the Mr of dimeric tubulin (approximately equal to 110,000 daltons). Two fractions were collected and eluted on a one-dimensional denaturing gel. Proteins were transferred subsequently to nitrocellulose and probed with an 125I-labeled monoclonal antibody specific for beta-tubulins. Only the fraction containing bound [3H]VCR contained tubulin. Estimates for the dissociation constants (Kd) for the binding affinity of VCR and VLB in crude, membrane-free supernatant fractions from RMS xenografts were obtained by computer curve fitting using a mathematical binding model. Data fitted a two-site binding model, with Kd values for the high-affinity site ranging from 61 to 160 nM, and for the low-affinity site, from 42 to 94 microM. At physiologically achievable drug concentrations, the relationship between binding affinity, drug retention and tumor sensitivity was examined further. A close relationship was apparent between the Kd values for VCR in Rh12, Rh18 and Rh18/VCR-3 tumor supernatant fractions and VLB in Rh12 preparations, and t1/2 alpha values for drug retention. Prolonged drug retention correlated with a low binding constant. As t1/2 alpha decreased, binding affinity also decreased, as demonstrated by an increase in the Kd value. Consequently, the tightness of drug binding in tumors also correlated with the degree of sensitivity of the xenografts to Vinca alkaloids.(ABSTRACT TRUNCATED AT 400 WORDS)
