Subcellular flux of potassium and rubidium in amphibian oocytes.

To determine the net Rb+ influx and K+ efflux at a subcellular level, fully grown and a Ringer's solution where K+ was substituted for by Rb+ on a molar bases. For 40 hrs serial samples of the oocytes were cryofixed and cryosectioned for elemental analysis in mM per kg dry weight using electron probe x-ray microanalysis. Oocyte volume remained constant. Net Rb+ influx showed a slow exponential increase into the nucleus, the yolk-free cytoplasm and the yolk platelets. There was significant K+ efflux from the nucleus but not from the yolk-free cytoplasm or the yolk. The Na+ concentration remained unchanged in all compartments during the course of the experiment. There was however a slow but significant increase in the concentration of Cl- in each of the subcellular compartments but this increase was not sufficient to balance the observed increase in the sum of K+ plus Rb+. Thus Rb+ accumulates selectively in all three subcellular compartments indicating that Rb+ is not a good K+ surrogate in the oocytes. That K+ demonstrates efflux from the nucleus but not from the cytoplasmic compartments is interpreted to suggest that some of the nuclear K+ is lost in exchange for Rb+ but that essentially none of the cytoplasmic K+ is-lost in exchange for Rb+. The findings provide strong evidence for adsorption of Rb+ in the cell.

[High extracellular concentrations of potassium and rubidium modulate the synaptic transmission in the frog labyrinth].

High extracellular K or Rb levels (20 mM) produce an increase in the resting EPSP and spike frequencies recorded intra cellularly from single fibres of the posterior nerve in the isolated frog labyrinth. The afferent discharge facilitation proved to be inversely related to the fibre's initial resting activity. The K effect is systematically larger than the Rb effect. High sensitive and scarcely sensitive units may be identified with respect to K and Rb action. The present findings suggest that, according to previous models of hair cell functioning, the K and Rb effects are mediated by a raise in intracellular Ca concentration which sustains an increased transmitter release at the cyto-neural junction.

[X-ray fluorescent determination of iron, copper, zinc, bromine and rubidium in biopsies of the gastric mucosa].

A method of a combined use of fiber gastroscopy and x-ray fluorescent analysis was elaborated for a study of the chemical composition of the gastric mucosa. Fr, Cu, Zn, Br, and Rb concentrations were determined in gastric mucosa biopsy specimens obtained during fiber gastroscopy, using x-ray fluorescent analysis. The concentrations of trace elements in the gastric mucosa were determined in 13 healthy persons, in 23 patients with chronic mucosal atrophy, in 15--with gastric ulcer, and in 16--with polyps. Besides, the content of trace elements was determined in the tumorous tissue of 66 gastric cancer patients. The results of the investigation revealed a statistically significant rise of mean Fr concentrations on the ulcer edge, in polyps and tumorous tissue as compared to the unchanged mucosa. Polypous tissue differed from the normal gastric mucosa by a low level of Cu, the difference being statistically significant. A tendency to a decrease in Zn content as compared to the unchanged mucosa was noted in the edge of ulcerative defects. The method can be effectively used for intravital studies on Fr, Cu, Zn, Br, and Rb levels in the G. I. tract mucosa.

Epinephrine-stimulated maintained rubidium efflux from guinea pig hepatocytes may involve alpha 1- and alpha 2-adrenoceptors.

The epinephrine-stimulated maintained increase in 86Rb efflux from guinea pig hepatocytes may consist of both alpha 1- and alpha 2-adrenoceptor-mediated components. Both the alpha 1-selective adrenoceptor agonist phenylephrine and the alpha 2-selective agonist clonidine evoked a maintained increase in 86Rb efflux. Prazosin, an alpha 1-selective antagonist, did not inhibit the maintained increase in 86Rb efflux elicited by epinephrine, whereas yohimbine, an alpha 2-selective antagonist, did. In the absence of external Ca, no maintained increase in 86Rb efflux was observed. The results suggest that there may be two separate alpha-adrenergic sensitive Ca influx pathways into guinea pig hepatocytes, one mediated by alpha 1- and the other by alpha 2-adrenoceptor activation.

Myocardial uptake of thallium and rubidium during alterations in perfusion and oxygenation in isolated rabbit hearts.

The comparative effects of altered cellular function and coronary perfusion on myocardial 201Tl and 83Rb uptake were evaluated in three groups of isolated rabbit hearts having isovolumic contractions. Paired-indication dilution experiments were performed with 201Tl, 83Rb, and 111In-labeled albumin as an intravascular reference marker. In Group A hearts (n = 12), isotope transport was determined during control, hypoxia, and ischemia. In Group B hearts (n = 8), isotope transport was measured at control flow and again at a 50% and 80% reduction. In Group C hearts (n = 8) only 201Tl uptake was determined at control and following coronary reperfusion. Myocardial 201Tl and 83Rb transport were not significantly different and were proportional to flow. Although all interventions caused significant hemodynamic alterations, neither tracer was affected by hypoxia at constant flow. Thallium-201 permeation, however, was transiently decreased immediately after coronary reperfusion. We conclude that myocardial uptake of 201Tl and 83Rb are similar and directly related to flow, but do not reflect hypoxia induced cellular dysfunction.

[Changes in rubidium and cesium levels in the blood of patients on long-term dialysis and decreased velocity of neural conduction].

In 19 patients aged 19 to 45 years on long-term dialysis treatment during 12.6 +/- 18.2 months the concentrations of Rb and Cs were determined by atomic spectrometry in whole blood before and behind the dialyser at the beginning and end of dialysis. At the same time the concentrations of these elements were determined in the dialysing fluid. In all patients the velocity of conduction in the motor fibres in the upper and lower extremities was determined before and after dialysis. Damage to the peripheral neurons was demonstrated in the lower extremities mainly in 79% of cases. Increased velocity of motor conduction in at least one nerve related directly proportionally to the Cs concentration of the serum was demonstrated in 56-70% of the patients after one dialysis.
