Blue- and green-absorbing visual pigments of Drosophila: ectopic expression and physiological characterization of the R8 photoreceptor cell-specific Rh5 and Rh6 rhodopsins.

Color discrimination requires the input of different photoreceptor cells that are sensitive to different wavelengths of light. The Drosophila visual system contains multiple classes of photoreceptor cells that differ in anatomical location, synaptic connections, and spectral sensitivity. The Rh5 and Rh6 opsins are expressed in nonoverlapping sets of R8 cells and are the only Drosophila visual pigments that remain uncharacterized. In this study, we ectopically expressed Rh5 and Rh6 in the major class of photoreceptor cells (R1-R6) and show them to be biologically active in their new environment. The expression of either Rh5 or Rh6 in "blind" ninaE(17) mutant flies, which lack the gene encoding the visual pigment of the R1-R6 cells, fully rescues the light response. Electrophysiological analysis showed that the maximal spectral sensitivity of the R1-R6 cells is shifted to 437 or 508 nm when Rh5 or Rh6, respectively, is expressed in these cells. These spectral sensitivities are in excellent agreement with intracellular recordings of the R8p and R8y cells measured in Calliphora and Musca. Spectrophotometric analyses of Rh5 and Rh6 in vivo by microspectrophotometry, and of detergent-extracted pigments in vitro, showed that Rh5 is reversibly photoconverted to a stable metarhodopsin (lambda(max) = 494 nm), whereas Rh6 appears to be photoconverted to a metarhodopsin (lambda(max) = 468 nm) that is less thermally stable. Phylogenetically, Rh5 belongs to a group of short-wavelength-absorbing invertebrate visual pigments, whereas Rh6 is related to a group of long-wavelength-absorbing pigments and is the first member of this class to be functionally characterized.

Ultraviolet polarized light study on isolated myocardial trabeculae and skeletal fibers.

The main components of the excitable membranes are protein and lipoproteins absorbing in the ultraviolet. Contractile proteins in muscle also absorb ultraviolet light. If polarized light is used, it becomes possible to distinguish the orientation of the absorbing groups either at rest or during activity. A system of microspectrophotometry in vivo using polarized ultraviolet light has been established to study the portion of myocardial trabeculae or of skeletal fibers in the central compartment of a double sucrose gap apparatus. Thus the system also allows electrophysiological studies. On both preparations, a relation between the absorbance change at 280 nm and contractile activity has been measured. These preparations also contain a fraction of oriented compounds absorbing at 280 nm and another at 260 nm. During mechanical activity, the amount of oriented compounds increases at 280 nm and decreases at 260 nm. These changes were assigned to an increase in the ratio of oriented amino acids, tyrosine and tryptophan, and to a decrease of oriented adenosine triphosphate.

Paint and tape: collection and storage of microtraces of paint in adhesive tape.

The collection and preservation of microtrace evidence with the aid of an adhesive tape is a method of choice in forensic science. This technique is rapid and easy and allows the concentration of microtraces on a carrier, which facilitates further investigations in the laboratory. Adhesive tapes are currently used to secure microtraces of fibers and glass, but hardly for traces of automotive paint and other lacquers for fear of interference with the analysis of binders. A collection of automotive paint consisting of original and repair lacquers collected by tape has been evaluated. After various times of storage within the tape, these samples were compared with untreated references by microscope FT-IR and microspectrophotometry (MSP). Another set of paints was collected in 1984, stored within the tape until 1995, and examined the same way. About 170 layers of lacquer with various types of binder were examined. With the exception of one clear lacquer no difference between treated samples and references was detected. This small difference observed could be correlated to the exposure to xylene (extractant) and was not caused by the storage within the adhesive tape.

Two cases of mélanose neurocutanée with development of malignant melanoma: a microspectrophotometric and electron microscopic study.

Two cases of neurocutaneous melanosis with development of malignant melanoma in the Japanese are presented. The first case was a 4-year-old boy in whom a retroperitoneal melanoma appeared with giant nevi, and cerebral and spinal melanosis. The second case was a 39-year-old man, in whom a primary leptomeningeal melanoma developed with leptomeningeal melanosis and smaller pigmented nevi. Microspectrophotometric and electron microscopic studies were made on the neoplastic and non-neoplastic melanotic tissues to elucidate the histogenesis of this rare disorder. Two different patterns of nuclear DNA histograms, corresponding to melanosis and melanoma, were obtained by microspectrophotometry, Considerable variation in the ultrastructure of the melanocytes was seen by electron microscopy.

[Some quantitative characteristics of the frog retinal rod outer segments].

Retinal rod outer segments in frogs were studied by means of light microscopy, refractometry, microspectrophotometry, and electron microscopy. Analysis of the data obtained shows that an unidentified substance, which makes up about 50% of outer segment dry weight, is lost during routine biochemical investigations. The protein parts of the rhodopsin molecules make up 85% of the outer segments proteins and 25% of outer segment dry weight. Rhodopsin molecules can be arranged in a square array with a unit cell side of about 7 nm on one side of each disk membrane. Lipids in a single membrane occupy only 2 nm, and disk membranes are strongly hydrated.

Norepinephrine in the rat cortex before and after occlusion of chronic arteriovenous fistulae: a microdialysis study in an animal model of cerebral arteriovenous malformations.

OBJECTIVE: In an animal model, we tested the hypothesis that intracerebral, extracellular norepinephrine (NE) influences cerebral blood flow (CBF) in the presence of cerebral arteriovenous malformations (AVMs). METHODS: A microdialysis (MD) probe was implanted into the cortices of Sprague-Dawley rats 3 months after the creation of extracranial arteriovenous fistulae (AVFs; n = 19), carotid ligation (n = 10), and cervical vessel preparation (sham operations; n = 8). Ipsilateral regional cortical oxygen saturation (microspectrophotometry) and regional cerebral blood flow (laser Doppler flowmetry) were scanned with simultaneous MD sampling for NE (high-performance liquid chromatography). Local CBF and cortical capillary oxygen saturation (SO2) were recorded continuously with serial MD sampling after fistula occlusion for 100 minutes. The scanning protocol was repeated after 120 minutes. Values were compared by analysis of variance and Duncan's test (P < 0.05; values expressed as mean +/- standard deviation). RESULTS: NE was highest in sham-operated animals and lowest after carotid ligation as compared with before and after the creation of the AVFs (sham operations, 5.9 +/- 2.3 nmol/L; common carotid artery ligation, 2.4 +/- 1 nmol/L; before creation of AVF, 4.1 +/- 1.9 nmol/L; after creation of AVF, 4.1 +/- 2.3 nmol/L). Regional cortical oxygen saturation differed significantly between all groups; regional cerebral blood flow was lower only in sham-operated animals (sham operations, 210 +/- 75 laser Doppler units [LDU], 71 +/- 13% SO2; common carotid artery ligation, 250 +/- 82 LDU, 62 +/- 19% SO2; before creation of AVF, 249 +/- 96 LDU, 65 +/- 14% SO2; after creation of AVF, 256 +/- 82 LDU, 74 +/- 10% SO2). After fistula occlusion, a sustained CBF increase for 100 minutes up to 40% above baseline (i.e., regional cerebral blood flow and regional cortical oxygen saturation before occlusion) was observed. SO2 increased to 16% after 120 minutes. Peaks of NE concentrations occurred 40 minutes and 100 minutes after fistula occlusion. CONCLUSION: The model seems to be valid for the simulation of events that occur before and after the obliteration of human cerebral AVMs in most cases, but not in those prone to hyperperfusion syndromes. The results provide evidence that cerebral sympathetic nervous system activity can be monitored by in vivo MD and that NE may play a role in CBF control before and after the obliteration of AVFs.

Colorectal carcinoma. DNA ploidy pattern and prognosis with reference to tumor DNA heterogeneity.

Nuclear DNA content was measured in 72 colorectal carcinomas using single-cell microspectrophotometry on Feulgen-stained smears. Four samples were analyzed from each tumor. Patients were followed for 41-65 months (average, 53). DNA heterogeneity (both aneuploid and nonaneuploid patterns) was present in 44% of the cases. Sixty-eight percent of the tumors showed an aneuploid DNA pattern in at least one of the samples. Patients with nonaneuploid tumors tended to have a survival advantage over patients with homogeneously aneuploid tumors and demonstrated a significantly longer disease-free survival. The DNA ploidy pattern is of potential value in conjunction with histopathologic prognostic parameters in colorectal carcinoma. Since colorectal tumors exhibit pronounced DNA heterogeneity, multiple samples are required from each tumor to permit a proper evaluation of its DNA pattern. The DNA heterogeneity may represent tumor progression and can partly explain the conflicting results reported concerning DNA pattern and prognosis in colorectal carcinoma.

Ectopic expression of ultraviolet-rhodopsins in the blue photoreceptor cells of Drosophila: visual physiology and photochemistry of transgenic animals.

We have generated transgenic flies expressing R7 cell-specific opsins in the major class of photoreceptor cells of the Drosophila retina and characterized their spectral properties using high-resolution microspectrophotometry and sensitivity recordings. We show that the Rh3 and Rh4 opsin genes encode UV-sensitive opsins with similar spectral properties (lambda max = 345 nm and 375 nm), and that Rh3 corresponds to the R7p and R7marg class of visual pigments. We have also generated Rh3 and Rh4 isoform-specific antibodies and present an R7 cell map of the Drosophila retina. In a related set of experiments, we show that it is possible to coexpress two different visual pigments functionally in the same cell and produce photoreceptors that display the summed spectral response of the individual pigments. These findings open up the possibility of tuning an animal's visual behavior by targeted expression of combinations of opsin genes to selective types of photoreceptors.

Chemical and photo-oxidative hair damage studied by dye diffusion and electrophoresis.

Microspectrophotometric and electrophoretic methods were used to characterize and quantify the effects of primary damage to hair from chemical and photochemical oxidative processes. The diffusion of molecules proceeding from the fiber surface to the center of untreated and modified (by chemical and photochemical oxidative processes) hair fibers was mapped by fluorescence microscopy and quantified by calculating diffusion coefficients of a fluorescent molecule. In addition, an electrophoretic separation technique, namely, SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis), was used not only to substantiate the results obtained in the microfluorometric study, but also to show how the main classes of proteins of unaltered hair are modified by cosmetic chemical treatments, light exposure, and combinations of these two processes. UV microspectrophotometry is an alternate analytical method to evaluate photo-oxidative damage in hair, and supports the results obtained by microfluorometry.

[Ultrastructural characteristics of nephroblastoma and its relation to DNA ploid].

Fifty-one cases of nephroblastoma were studied by light and electron microscopy as well as microspectrophotometry. Under light and electron microscope, the tumor appeared to be carcinosarcoma structurally, consisting of epithelial, sarcomatous and undifferentiated tumor cells. Their ultrastructural feature was similar to that of renal tubular epithelial cells and fibroblasts. It could be differentially diagnosed from renal mesoblastic tumors by electron microscopy when the tumor was chiefly composed of sarcomatous cells. Microspectrophotometer measurement showed an increased average nuclear DNA content (DI greater than 1.04), belonging to aneuploid tumor with polyploid cells, serving as one important feature of malignant tumors. In addition, the poorer the tumor differentiation the higher was the DNA content of the tumor cells, suggesting that the measurement of nuclear DNA of tumor cells may also help ascertain the degree of differentiation and malignancy of nephroblastomas.

DNA distribution pattern in colorectal carcinoma. A methodologic study comparing two different biopsy techniques.

DNA content was measured in 59 samples from 20 colorectal adenocarcinomas and 5 adenomas by single cell microspectrophotometry. Samples were obtained simultaneously by fine needle aspiration (FNA) and touch imprints of punch (IMP) biopsies. The results showed that specimens obtained by the two biopsy methods displayed similar DNA ploidy patterns in 46 of 59 samples (78%). The DNA patterns in six samples were aneuploid in IMP and nonaneuploid in FNA and vice versa in seven samples. When DNA patterns of the individual tumors were classified according to the most aggressive one, the two biopsy methods showed 88% concordance, with similar DNA patterns in 22 of 25 patients. The DNA patterns in three patients were classified as aneuploid in FNA specimens and nonaneuploid in IMP. The results indicate that imprints of punch tissue biopsies can be used for a reliable evaluation of the DNA pattern in colorectal tumors.

Effect of thromboxane receptor blockade on oxygen supply/consumption variables during reperfusion in the anesthetized dog.

The thromboxane A2/PGH2 receptor antagonist SQ 30,741 has been previously shown to reduce infarct size and to improve subendocardial reflow. The purpose of this study was to determine the effect of SQ 30,741 on reperfusion O2 supply/consumption variables. Anesthetized open-chest dogs treated with vehicle or 1 mg/kg + 1 mg/kg/hr SQ 30,741, i.v. (starting 10 min after the onset of ischemia) were subjected to 90 min left circumflex coronary occlusion and 3 hr reperfusion. Regional myocardial blood flow (radioactive microspheres) and arterial and venous O2 saturations (microspectrophotometry) were determined. Animals treated with SQ 30,741 had significantly higher subendocardial reflow at 3 hr (48 +/- 6 ml/min/100 g) compared with vehicle (27 +/- 10 ml/min/100 g). At 3 hr postreperfusion, O2 extraction was significantly higher in the reperfused region compared with the nonischemic region, although extraction was not at maximal values. O2 extraction was similar in vehicle- and SQ 30,741-treated animals despite the near doubling of reflow into the subendocardial region with SQ 30,741. O2 consumption was significantly reduced in the reperfused subendocardial region (1.65 +/- 0.9 ml O2/min/100 g) in vehicle controls compared with the nonischemic subendocardial region (10.2 +/- 2.6 ml O2/min/100 g). SQ 30,741 significantly improved subendocardial reperfused regional O2 consumption (4.03 +/- 0.41 ml O2/min/100 g) compared with vehicle and this increase was proportional to the flow increment (no change in the O2 supply/consumption ratio). SQ 30,741 is thus increasing subendocardial reflow secondary to an increase in O2 consumption and the increased O2 consumption may be due to preservation of myocardial tissue.

Video fluorescence microscopy as a tool for the study of cellular heterogeneity in epithelia.

The use of functional fluorescent dyes has allowed us to monitor intracellular pH in individually identified cells in renal epithelia. Using video microscopy we simultaneously measured the change in intracellular pH in several contiguous cells in response to various maneuvers. The video equipment included a silicon intensified target camera, a VHS videocassette recorder, a high resolution monochrome monitor, a video photometric analyzer and a 2-channel chart recorder. This equipment had a spatial resolution of 1 micron by light microscopy and a response time of less than 200 ms; it allowed us to perform double fluorescent labeling and obtain reliable measurements of intracellular pH, independent of gain, regardless of the location of the image on the screen. Using this video system we have shown that there is substantial heterogeneity in activity of H+/HCO3- transport pathways among adjacent cells in a monolayer of cells cultured from the rat renal inner medullary collecting duct. In isolated perfused rabbit renal cortical collecting ducts, video microscopy allowed us to show that there are two different types of intercalated cells: one that exhibits apical Cl-/HCO3- exchange and one that does not. Both show alkaline intracellular pH with respect to non-acid-base transporting epithelia. Video microscopy has several advantages over conventional microspectrophotometry. It provides rapid data acquisition along with increased sensitivity and the capacity for some subcellular analyses. One is able to analyze several individually identified cells during an experimental maneuver. The present video system was assembled for less than $15,000 and permits a more complete analysis of an epithelium than either single-cell photometry or spectrophotometric analysis of thousands of cells in suspension or monolayers.

[Relation of cytochemistry changes in the peripheral blood ANAE in chronic glomerulonephritis and differentiation-syndromes].

The ANAE reaction of the peripheral blood from 24 patients of chronic glomerulonephritis (grouped according to the identification of TCM) and 30 normal individuals were observed by cytochemistry and microspectrophotometry. It was found that the percentages of positive lymphocytes of the deficiency of Kidney-Yin was 47.8 +/- 7.6%, among which the percentages of lymphocytes of spot granular pattern and scattered granular pattern were 42.4 +/- 6.3% and 5.4 +/- 2.2% respectively, the ANAE relative quality of monocytes of the deficiency of Kidney-Yin was 51.26 +/- 2.59. They were 39.7 +/- 7.4%, 31.2 +/- 5.8%, 8.5 +/- 2.7% and 41.84 +/- 2.66 respectively in the deficiency of Kidney-Yang. The results of normal group were 65.5 +/- 6.8%, 53.6 +/- 6.7%, 11.9 +/- 3.4% and 54.78 +/- 3.34 in their given order. The differences in three groups each other were marked significantly. The authors have found the difference between the patients of chronic glomerulonephritis and the normal individuals was significant statistically.

Acute effects of 3'-azido-3'-deoxythymidine on the cell cycle of HL60 cells.

The average doubling times of HL60 cells grown in the presence of 0,5,10 and 100 microM 3'-azido-3'-deoxythymidine (AZT) for 72 h were, respectively, 51.1, 65.7, 69.0 and 76.3 h. This drug-concentration-dependent prolongation of the cell-doubling time was associated with a progressive increase in modal cell volume. The technique of combined Feulgen microspectrophotometry and 3H-thymidine autoradiography revealed that the cell cycle distribution of HL60 cells cultured in the presence of 5 and 100 microM AZT for 48 h was abnormal, with an increased percentage of cells in the S phase and a decreased percentage in the G1phase. From the cell doubling times and the cell cycle distribution data, and making a number of assumptions, upper limit estimates for the duration of the S phase in cultures containing 0,5 and 100 microM AZT were calculated to be 26.2, 35.8 and 49.6 h, respectively. The data indicate that concentrations of AZT achieved in the plasma of patients receiving this drug (i.e. 5 microM) cause a substantial prolongation of both the cell cycle time and the duration of the S phase of HL60 cells. It therefore seems likely that some of the toxic effects of AZT seen in vivo, including impairment of bone marrow function, are at least partly related to AZT-induced disturbances of DNA synthesis and proliferation in human cells.

[Thyroid microcarcinoma with a fatal outcome and 34 other unusually aggressive cases reported in the literature].

A follicular thyroid microcarcinoma was revealed by scapular metastases. Despite treatment, other metastases were the cause of death after a course of 14 years. In the medical literature there are at least thirty-four other examples of follicular or papillary carcinoma of less than 15 mm (previously called occult) that have either given rise to blood-born metastases or have been the cause of death. Present data from light and electron microscopy, immunohistochemistry and microspectrophotometry cannot differentiate between small and large carcinomas. Microcarcinomas (particularly papillary ones) are frequent in the general population and are found in 5% of thyroids. They are usually not aggressive. Those which show vascular or capsular invasion, a lack of lymphoid infiltrate and large cervical lymph node metastases are more likely to have an unfavorable course. The discovery of a microcarcinoma in a thyroid cells for an extended follow-up.

Interrelation of formalin fixation, chromatin compactness and DNA values as measured by flow and image cytometry.

The severity and consistency of the effect of formalin fixation on the quantitation of DNA by flow cytometry (FCM) and image cytometry (ICM) were studied. As compared to ethanol, formalin fixation substantially decreased the propidium iodide fluorescence from mouse hepatocyte nuclei analyzed by FCM; it was also associated with an altered 4n-to-2n signal ratio and with false aneuploid peaks by FCM, but not by ICM (microspectrophotometry). ICM, on the other hand, suffered from a dependence of the DNA signal on nuclear size, which was not seen with FCM. The DNA signal variation was related to variations in the chromatin state, as shown by differences between monocytes and lymphocytes, and between RAJI cells fixed under various ionic strengths. The dependence of the DNA signal on the chromatin state indicates a need for caution in interpreting aneuploidy in formalin-fixed cells. For FCM, pseudoaneuploidy appears avoidable by using a Feulgen fluorescence staining technique. New imaging modes may be necessary to solve the problem of cell size dependence for ICM DNA determination.

Effects of thermotherapy on the morphology and cell cycle of a transplanted ascites hepatoma in rats.

The effects of heating on the morphology and cell cycle of ascites hepatoma AH-100B cells were investigated. Five rats with AH-100B ascites cells were warmed in a water bath at 39 degrees C, 41 degrees C, or 43 degrees C, respectively. After heating, changes in the morphology of tumor cells were observed every day over a 3-day period under a light microscope and changes in the nuclear DNA content were measured by microspectrophotometry. Morphologic changes in AH-100B cells, which included swelling and vacuolization of the cytoplasm and constriction, vacuolization and destruction of nuclei, were observed. Multinucleated giant cells were characteristically observed in the cells from rats heated to 43 degrees C. In the case of rats heated to 43 degrees C, an increase was seen in the number of cells with a DNA content of 8C to 10C, and some of the cells exhibited polyploidy with a DNA content greater than 10C. It appears that one of the mechanisms of the antitumor effect of hyperthermia in vivo involves changes in the cell cycle of cancer cells and the accumulation of polyploid cells.
