LHRH promotes the synthesis and release of a 87,000 Da protein (GP-87) by enriched gonadotrophs.

Protein secretion by cultured pituitary cells from 14-day-old female rats was estimated using [35S]methionine incorporation followed by either one- or two-dimensional electrophoresis and autoradiography. Stimulation of total cells or gonadotrophs by LHRH promoted the synthesis and release of a specific polypeptide (apparent molecular weight 87,000, pI = 4.6). Silver staining of cellular proteins from both gonadotroph-enriched and gonadotroph-depleted populations prepared by centrifugal elutriation revealed a high concentration of this polypeptide in the gonadotrophs and a very low level in the other cell population. This species was thus called Gonadotrope Polypeptide GP-87. Release of labeled GP-87 by gonadotrophs was both time dependent (up to 4 h) and LHRH dose dependent (from 10(-9) M to 10(-7) M) as was the release of LH. Attempts to precipitate GP-87 from the incubation medium with anti-LH antiserum were unsuccessful suggesting that GP-87 is not a 'big' form of LH. TRH neither stimulated the release of GP-87 from gonadotrophs nor from lactotrophs though it did stimulated PRL release. From these data, we conclude that gonadotrophs in culture synthesize a specific polypeptide (GP-87), LHRH stimulates both the synthesis and release of GP-87, and the pituitary cell response is peptide specific. The LHRH-induced synthesis and release of GP-87 could be an important step in the molecular processes that regulate gonadotrophin secretion.

[The methodology and accuracy of MRI-based orbital volume calculations].

OBJECTIVE: The aim of the present work was to establish a method for orbital volume calculation based on MR scanning data for the sake of better radiation hygiene. MATERIALS AND METHODS: The orbital volumes of 35 ophthalmologically healthy children were calculated on the basis of MRI scans. After data transfer to a separate workstation, volumetric analysis was carried out by two independent radiologists using semi-automated software. The accuracy of the calculated values was compared with orbital volumes measured by anatomic preparations and given in studies by various authors. RESULTS: Volume calculation was possible in all patients using MRI data. There is an acceptable agreement with the presented anatomic facts and the measured values of Bentheley. In the Wilcoxon test there was not a big difference between the courses of the values (p = 0.507). CONCLUSION: Even though we can obtain a better image of the bizarre structure of the bony orbits with CT, MR-based volumetry of the orbit is a reliable method and is not burdened by radiation exposure. It can thus be an important condition for the planning and the controlling of modern therapeutic concepts in treating anophthalmos and microphthalmos.

14-3-3beta binds to big mitogen-activated protein kinase 1 (BMK1/ERK5) and regulates BMK1 function.

Big mitogen-activated kinase 1 (BMK1/ERK5) is a member of the MAPK family activated by growth factors that mediates cell growth and survival. Previous data show that BMK1 can be activated by steady laminar flow and is atheroprotective by preventing endothelial cells from undergoing apoptosis. The primary structure of BMK1 is distinct from other MAPK members by virtue of a unique long C-tail, suggesting specific mechanisms of regulation. To characterize regulatory mechanisms for BMK1 function, we identified binding proteins by yeast two-hybrid analysis. Among these proteins, the scaffolding protein 14-3-3 was identified. BMK1 bound to 14-3-3beta in vitro and in vivo as demonstrated by glutathione S-transferase (GST)-14-3-3beta fusion protein pull-down assays and coimmunoprecipitation. Phosphorylation of BMK1 was most likely required for this interaction. GST-14-3-3beta pull-down assays using truncated constructs of BMK1 and site-directed BMK1 mutants demonstrated that the interaction requires serine 486 within the C terminus of BMK1. BMK1 bound to 14-3-3beta basally, and the interaction was greatly abrogated when BMK1 was activated. The interaction of 14-3-3beta and BMK1 inhibited kinase activities stimulated by constitutively active (CA)-MEK5 and epidermal growth factor. Mutation of serine 486 (BMK1-S486A) prevented the interaction with 14-3-3beta and enhanced BMK1 activity upon epidermal growth factor stimulation. These data demonstrate an inhibitory function for 14-3-3beta binding to BMK1 and show that serine 486 phosphorylation represents a novel regulatory mechanism for BMK1.

The impact of Psychiatric Reform on inpatient services in a metropolitan area of Southern Italy: the Naples case.

Six years after the Psychiatric Reform, phasing out of Mental Hospital and implementing a community based Mental Health System, i.e. the main goals of the Reform, are not yet performed in Southern big cities of Italian peninsula. In Naples metropolitan area this new Mental Health policy faces with a dramatic lack of community services. This paper summarizes data concerning the trends in psychiatric admissions to some inpatient public settings of Naples before and after the Reform, with the intent of pointing out the short-time outcomes of this law in a highly populated area. The Authors stress some positive results of the Reform in the study area, as well as the factors which have contributed to the inadequate application of the new law.

Personality and affectivity characteristics associated with eating disorders: a comparison of eating disordered, weight-preoccupied, and normal samples.

The purpose of this study was to assess the relative contribution of personality and emotional experience to self-reported eating attitudes in a group of patients with clinically diagnosed eating disorders, a weight-reduction training group (Weight Watchers), and a control group without body weight problems. Participants in this study (N = 114) completed Estonian versions of the Eating Disorder Inventory-2 (EDI-2; Garner, 1991), NEO Personality Inventory (Costa & McCrae, 1989), and Positive Affect and Negative Affect Schedule, Expanded Form (Watson & Clark, 1994). Data demonstrated validity of the Estonian version of EDI-2 in its ability to identify problems on a continuum of disordered eating behavior. Among the Big Five personality dimensions, Neuroticism made the largest contribution to EDI-2 subscales. Two other dimensions, Openness to Experience and Conscientiousness, also predispose individuals to eating problems. Personality traits made a larger contribution to the self-reported eating pathology than the self-rated effects experienced during the last few weeks. It was argued that personality dispositions have a larger relevancy in the etiology of eating disorders than emotional state.

[Ecological correlation studies on standardized incidence ratios of malignant neoplasms contracted from different sources of drinking water supply].

Ecological correlation analysis was made between the source of drinking water and standardized incidence ratios (SIRs) of malignant neoplasms registered in Niigata, Japan. The purpose of this study was to find out some clues for generating a working hypothesis that the contamination of drinking water with environmental chemicals especially of endocrine disrupting chemicals in big rivers may be related to the occurrence of cancers. Sources of drinking water in the year 1974 were classified into two categories; one from the Shinano and Agano and their down-stream rivers, and the other from small rivers flowing into these big rivers, rivers originating from the mountain areas, underground and reservoirs (The other group). After excluding registered cases under 20 years of age, the registry data (1992-94) were sub-divided into two categories according to the source of drinking water. The SIRs were calculated by using two different standard populations; Niigata total and "The other group". All of the sites were examined in the former standard. In the latter case, however, 12 and 9 sites in males and females, respectively were selected based on the assumption that the expected numbers in each age group totaled 5 or over when there were more than 200 registered patients as a total. Concerning the judgment for the significant association in the former standard, we considered the presence of positive associations when both of the SIRs in the two categories were significantly different from the Niigata total population (p < 0.05 by chi 2 test). Of the 33 sites in males, the following sites were correlated with the source of drinking water; tongue, colon, rectum, gallbladder, etc., prostate, kidney, etc. and all sites. Of 35 sites in females, those were the colon, gallbladder, etc., bronchus and lung, breast, corpus uteri, thyroid and all sites. Of the 12 and 9 respective sites subjected to the second analysis, positive associations were observed in the following: esophagus, colon, rectum, liver, gallbladder, etc., bronchus and lung, prostate, bladder and all sites in males, and colon, rectum, gallbladder, etc., bronchus and lung, breast and all sites in females. Final conclusion, however, should be waived until the consistency of association can be confirmed by using more reliable incidence data, because the difference in the death certificates only (DCO) rate between the two groups might have played as a bias. The use of mortality data may be an alternative way to evaluate the present findings in terms of the consistency of association.

Ectopic production of big ACTH in carcinoma of the lung. Its clinical usefulness as a biologic marker.

This report considers the potential usefulness of adrenocorticotropic hormone (ACTH) determinations in diagnosis and in prognosis for therapy of patients with carcinoma of the lung but without clinical Cushing's syndrome. The report is based on radioimmunoassay data from 129 patients, including 62 with lung cancers and 67 with nonmalignant pulmonary conditions. Elevated plasma ACTH was found in 21 of 24 patients with untreated cancer and the hormone was detected in tumor extracts and/or bronchial washings from the remaining 3. Elevation of plasma ACTH was found in only 10 of 38 treated patients. Absence of clinical Cushing's syndrome in spite of high plasma ACTH concentrations is explained by the observation that the predominant form of ectopic ACTH in plasma is immunoreactive but nonbioactive 'big' ACTH. Prolonged survival, for longer than 19 months, was observed in only 5 patients: all patients with low plasma ACTH after resection of the lung tumor and 2 of 3 patients with low plasma ACTH without therapy. ACTH was found in all available malignant tissue, primary and metastatic, from the lung carcinoma group,but not in normal lung or in 5 tumors metastatic to the lung. Of the 39 patients diagnosed initially to have chronic obstructive pulmonary disease, 14 showed plasma ACTH elevation. However, 3 of these patients with the highest concentrations subsequently manifested carcinoma or carcinoma in situ.

Activation of the hepatic endothelin-system in rats with biliary liver fibrosis.

Circulating plasma endothelin-1 (ET-1) is elevated in liver cirrhosis, in a disease-stage-dependent manner. However, ET-1 exerts its effects mainly via paracrine and autocrine pathways. Therefore, the aim of the present study was to analyze the hepatic endothelin (ET) system in liver cirrhosis resulting from bile duct obstruction (BDO). Wistar rats were subjected for 6 weeks to either sham operation (control) or BDO. Thereafter, hepatic ET-1 concentrations were elevated 7.2-fold in BDO compared to control (p <0.001), whereas big ET-1 was unchanged. The density of both ET receptor subtypes was upregulated in BDO (ETA: 7.4-fold and ETB: 4.9-fold vs control, p < 0.001, respectively). The affinity of both receptor subtypes was significantly reduced in BDO. In conclusion, our data demonstrated for the first time that the hepatic ET system in liver cirrhosis is characterized by a simultaneous upregulation of both ET-1 tissue concentration as well as the density of hepatic ETA- and ETB-receptors, suggesting a synergistic activation of the hepatic ET system in rats with BDO. The increased ET-1 tissue concentration is not a result of an altered big ET-1 synthesis in biliary liver fibrosis, suggesting an increased activity of endothelin-converting enzyme (ECE) in liver cirrhosis.

Studies on the constituents of Clematis species. VII. Triterpenoid saponins from the roots of Clematis terniflora DC. var. robusta Tamura.

From the roots of Clematis terniflora, nine new oleanolic acid 3,28-O-bisdesmosides called clematernosides A, B, E, F, G, H, I, J and K, and two new hederagenin 3,28-O-bisdesmosides called clematernosides C and D, have been isolated together with two known saponins, huzhangoside B and clematichinenoside C. The structures of the new saponins have been elucidated based on chemical and spectral evidence. Among the new spaonins, clematernosides I and J have a nonasaccharide moiety and a total of twelve monosaccharide moieties in the molecule. This is the first report of the isolation and structure elucidation of such "big" glycosides.

[The treatment of severe head-brain injuries in Austria] .

INTRODUCTION: We performed this study in order to assess epidemiology and current practice of treatment of severe traumatic brain injury in Austria. Our survey followed the methods of a study published by J. Ghajar et al in the USA in 1995 and we compared the results to the Brain Trauma Foundation's "Guidelines For The Management Of Severe Head Injury". METHODS: The collected data represent answers to telephone interviews of 60 surgical intensive care units. We were able to evaluate data from all departments which treat severe brain traumas (Glasgow Coma Scale < or = 8) in Austria. RESULTS: At the time the treatment modalities of severe head injuries are not homogeneous and there are also big interdisciplinary management differences (trauma surgeons versus neurosurgeons). CONCLUSIONS: Results showed that there is a need for a brain trauma databank in Austria. We also recommend formation of an interdisciplinary brain trauma working group in order to control whether guidelines and standardized therapeutic modalities are being followed.

Glaucoma in China: how big is the problem?

AIMS: To derive preliminary estimates for the number of adults in China suffering from glaucoma, and project the burden of visual morbidity attributable to primary and secondary glaucoma. METHODS: Age and sex specific data from two population surveys were applied to US Census Bureau population estimates for urban and rural China. It was assumed that data from Singapore were representative of urban China, and those from Mongolia were representative of rural China. RESULTS: It was estimated that 9.4 million people aged 40 years and older in China have glaucomatous optic neuropathy. Of this number, 5.2 million (55%) are blind in at least one eye and 1.7 million (18.1%) are blind in both eyes. Primary angle closure glaucoma (PACG) is responsible for the vast majority (91%) of bilateral glaucoma blindness in China. The number of people with the anatomical trait predisposing to PACG (an "occludable" drainage angle) is in the region of 28.2 million, and of these 9.1 million have significant angle closure, indicated by peripheral anterior synechiae or raised intraocular pressure. CONCLUSIONS: This extrapolation of data from two east Asian countries gives an approximate number of people in China suffering from glaucoma. It is unlikely that this crude statistical model is entirely accurate. However, the authors believe the visual morbidity from glaucoma in China is considerable. PACG is probably the leading cause of glaucoma blindness in both eyes, and warrants detailed investigation of strategies for prevention.

[Health Promotion and Quality of Life among mothers of preadolescents in Chile: a focused ethnography].

The aim of this ethnographic trial was to identify cultural patterns present in mothers' actions when protecting their children from environmental risks like drugs and violence. Data collection consisted in a tape-recorded structured interview with 10 mothers of 9 to 13-year-old preadolescents, complemented with pictures, field notes and participant observation. Mothers conceptualize their child as a "Big Boy" who wants to make his own decisions and be independent, generating conflicts in the family as a whole. As a conclusion, mothers perceive their pre teenagers as more vulnerable, so a close guiding and control is necessary for their developing activities. They believe it will help their children to recognize hazardous situations and stay away from them.

GABI-Kat SimpleSearch: an Arabidopsis thaliana T-DNA mutant database with detailed information for confirmed insertions.

Insertional mutagenesis approaches, especially by T-DNA, play important roles in gene function studies of the model plant Arabidopsis thaliana. GABI-Kat SimpleSearch (http://www.GABI-Kat.de) is a Flanking Sequence Tag (FST)-based database for T-DNA insertion mutants generated by the GABI-Kat project. Currently, the database contains >108,000 mapped FSTs from approximately 64,000 lines which cover 64% of all annotated A.thaliana protein-coding genes. The web interface allows searching for relevant insertions by gene code, keyword, line identifier, GenBank accession number of the FST, and also by BLAST. A graphic display of the genome region around the gene or the FST assists users to select insertion lines of their interests. About 3500 insertions were confirmed in the offspring of the plant from which the original FST was generated, and the seeds of these lines are available from the Nottingham Arabidopsis Stock Centre. The database now also contains additional information such as segregation data, gene-specific primers and confirmation sequences. This information not only helps users to evaluate the usefulness of the mutant lines, but also covers a big part of the molecular characterization of the insertion alleles.

Endothelin-1 stimulates cyclic GMP formation in porcine kidney epithelial cells via activation of the L-arginine-dependent soluble guanylate cyclase pathway.

Endothelin-1 (ET-1) elevated cyclic GMP levels in cultured porcine kidney epithelial cells (LLC-PK1) in a concentration-dependent manner with an EC50 value of about 5 x 10(-10) M. NG-methyl-L-arginine and NG-nitro-L-arginine inhibited cyclic GMP responses to 10(-8) M ET-1 with IC50 values of 1.2 x 10(-6) and 7.6 x 10(-8) M, respectively, and the inhibition was prevented with L-arginine. ET-1-induced cyclic GMP accumulation was enhanced with superoxide dismutase and diminished with oxyhemoglobin and methylene blue. Furthermore, the effect of ET-1 on the cyclic GMP levels was totally dependent on extracellular Ca2+. ET-3, but not big ET-1 and ET C-terminal hexapeptide16-21, elicited similar cyclic GMP responses as observed with ET-1 at the same concentration range. These data strongly suggest that, in LLC-PK1 cells, ET-1 stimulates formation of an endothelium-derived relaxing factor-like substance from L-arginine in a Ca(2+)-dependent fashion, which in turn activates soluble guanylate cyclase to elevate cellular cyclic GMP levels. The effects of ET on cyclic GMP accumulation in the kidney epithelial cells may be related to the natriuretic effects of ET in vivo.

[Osteosarcoma in rats receiving long-term PTH injection].

Daily injection of human PTH (1-34) to osteoporotics has been shown to increase bone mineral density and reduce fracture risk within 18 months. Despite its impressive effects on human bone, safety became a big issue when it was revealed that up to 53% of rats received hPTH (1-34) for 2 years developed osteosarcoma. On the interpretation of the data, most experts agreed that this is unlikely to predict an increased risk of osteosarcoma in human osteoporotic patients. Some of the reasonings are as follows: (1) fundamental differences in bone metabolism between rat and human, (2) rats received PTH at a dose 12-25 folds of the human dosage, for 80-90% of their life span whereas in human PTH would be used for only 2-3% of the life span.

ACB-PCR measurement of K-ras codon 12 mutant fractions in livers of Big Blue rats treated with N-hydroxy-2-acetylaminofluorene.

K-ras codon 12 GGT-->GAT and GGT-->GTT mutations are the most frequently observed K-ras point mutations in human and rodent tumors and therefore are implicated in carcinogenesis for many tissues. Measurement of these mutations in rat models and human tissue could facilitate a more logical extrapolation of rodent tumorigenesis data to human disease. We have developed allele-specific competitive blocker PCR (ACB-PCR) assays for rat K-ras codon 12 GGT-->GTT and GGT-->GAT mutations that parallel the already published assays for human K-ras codon 12 mutations. Liver K-ras codon 12 mutant allele fractions were measured in vehicle-treated and N-hydroxy-2-acetylaminofluorene (N-OH-AAF)-treated Big Blue rats. The average K-ras codon 12 GGT-->GTT mutant fraction (MF) for four control rats was 50 x 10(-6) (95% CI: 27 x 10(-6), 95 x 10(-6)) and for four treated rats was 165 x 10(-6) (95% CI: 87 x 10(-6), 312 x 10(-6)), indicating a 3.3-fold increase with treatment (95% CI: 1.3-8.1). The average MF of K-ras codon 12 GGT-->GAT for control rats was 1320 x 10(-6) (95% CI: 498 x 10(-6), 3500 x 10(-6)) and for treated rats was 8450 x 10(-6) (95% CI: 3180 x 10(-6), 22 400 x 10(-6)), indicating a 6.4-fold increase with treatment (95% CI: 1.6-25.4). These transgenic rats were part of a study that included analysis of liver lacI mutations. Although data from lacI determinations show that this compound induces mostly G-->T mutations, using the ACB-PCR method both K-ras codon 12 GGT-->GTT and GGT-->GAT MFs were significantly increased in treated rats versus control rats. This data raises the possibility that N-OH-AAF may not only induce mutations by a genotoxic mechanism, but also by amplification of both de novo and pre-existing K-ras mutation.

[Survey of the reproductive features and insemination/mating results in full blood Arabian and Haflinger mare herds on stud farms in west and middle Anatolia].

Within a German-Turkish university partnership documentation of reproductive data of brood-mares was performed as a part project of the cooperation contract. In the study Arab, Haflinger and cross-breed mares were included. The mares mainly were housed in big studfarms and a smaller part was kept under small private farms. Almost three quarters of both the Arab and Haflinger mares exhibited an estrous length of 1-4 days, whereas the others showed a heat duration of a period of 5-10 days. In the same group of probands a mean length of sexual cycle of 18-24 days could be observed in 38.2% of the Haflinger. The majority of the other mares in both races documented a cycle duration of more than 25 days. The remarkable short duration of heat is discussed as caused by the sharp change from winter to spring environment conditions in that area. An explanation of the marked long mean cycle interval is difficult and might be caused by inadequate documentation of the data. Conception results and foaling data as 83.4% resp. 78.8% in the Arabs and 76.6% resp. 74.8% in the Haflinger mares are within the international standard. Conception rate and foaling data in the cross-breed (72.3% resp. 55.3%) are low compared with data (96.9% 70.8%) of Arab brood-mares kept under stud condition in that area. These data point out the necessity of improving pre- and postconceptional care of the mares under stud conditions as well as in private keeping.
