Effect of optical tissue clearing on spatial resolution and sensitivity of bioluminescence imaging.

In vivo bioluminescence imaging (BLI) is a powerful method of in vivo molecular imaging based on the use of optically active luciferase reporter genes. Although this method provides superior sensitivity relative to other in vivo imaging methods, spatial resolution is poor due to light scattering. The objective of this study was to use hyperosmotic agents to reduce the scattering coefficient and hence improve spatial resolution of the BLI method. A diffusing fiber tip was used to simulate an isotropic point source of bioluminescence emission (550 to 650 nm). Mouse skin was treated in vitro and in vivo with glycerol (50%, 30 min) and measurements of optical properties, and imaging photon counts were made before, during, and after application of glycerol to the skin sample. Glycerol application to mouse skin had little effect on the absorption coefficient but reduced the reduced scattering coefficient by more than one order of magnitude. This effect was reversible. Consequently, the spot size (i.e., spatial resolution) of the bioluminescence point source imaged through the skin decreased by a factor of 2 (550-nm light) to 3 (650-nm light) after 30 min. Simultaneously, an almost twofold decrease in the amount of light detected by the BLI system was observed, despite the fact that total transmission increased 1.7 times. We have shown here that multiply scattered light is responsible for both observations. We have shown that applying a hyperosmotic clearing agent to the skin of small rodents has the potential to improve spatial resolution of BLI owing to a reduction in the reduced scattering coefficient in the skin by one order of magnitude. However, reducing the scattering coefficient reduces the amount of light reaching the camera due to a reduction in the amount of multiply scattered light that reaches the camera aperture and thus reducing the sensitivity of the method.

Self-absorption correction for carbon-14.

A new and simple technique has been developed (10) for correcting C(14) radioactivity measurements for loss of radiation due to self-absorption. Results are discussed which show the applicability of the technique for many different counting situations. It is found that the absorption of radiation follows a hyperbolic law much more closely than an exponential one. Furthermore, the absorption coefficient for the sample itself has been found to be a function of weight rather than a constant, as had been assumed for the derivation of the law of exponential absorption.

Short-duration low-direct-current electrical field treatment is a practical tool for considerably reducing counts of gram-negative bacteria entrapped in gel beads.

Application of a direct-current electrical field for very short times can serve as a practical nonthermal procedure to reduce or modify the microbial distribution in gel beads. The viability of Escherichia coli and Serratia marcescens entrapped in alginate and agarose beads decreases as the field intensity and duration of electrical field increase.

Correlating Streptococcus mutans with dental caries in young children using a clinically applicable microbiological method.

The present study used a clinically applicable microbiological method to correlate Streptococcus mutans counts and dental caries prevalence in young children. The study population consisted of 37 subjects, between 16 and 60 months of age. Using a sterile tongue depressor, samples of unstimulated saliva were obtained from subjects and inoculated onto elevated agar plates containing a selective medium. Plates were incubated anaerobically at 37 degrees C for 48 h. chi 2 and multiple regression analyses gave highly significant relationships (p less than 0.01) between dental caries prevalence and the number of S. mutans colonies.

Effect of topically-applied solutions of iodine, sodium fluoride, or chlorhexidine on oral bacteria and caries in rats.

Antibacterial levels of iodine, chlorhexidine or sodium fluoride were topically applied to the molar teeth of rats. The effect of these agents on buccal and sulcal caries, as well as on certain oral bacterial populations was assayed. All three agents exerted a significant cariostatic effect which was site specific. Buccal caries incidence and reductions appeared correlated to S. mutans counts, while sulcal caries was better correlated to total cultivable counts. These studies suggested that the simultaneous application of iodine and NaF might exert an additive cariostatic effect.

A geographic information system for characterizing exposure to Agent Orange and other herbicides in Vietnam.

Between 1961 and 1971, U.S. military forces dispersed more than 19 million gallons of phenoxy and other herbicidal agents in the Republic of Vietnam, including more than 12 million gallons of dioxin-contaminated Agent Orange, yet only comparatively limited epidemiologic and environmental research has been carried out on the distribution and health effects of this contamination. As part of a response to a National Academy of Sciences' request for development of exposure methodologies for carrying out epidemiologic research, a conceptual framework for estimating exposure opportunity to herbicides and a geographic information system (GIS) have been developed. The GIS is based on a relational database system that integrates extensive data resources on dispersal of herbicides (e.g., HERBS records of Ranch Hand aircraft flight paths, gallonage, and chemical agent), locations of military units and bases, dynamic movement of combat troops in Vietnam, and locations of civilian population centers. The GIS can provide a variety of proximity counts for exposure to 9,141 herbicide application missions. In addition, the GIS can be used to generate a quantitative exposure opportunity index that accounts for quantity of herbicide sprayed, distance, and environmental decay of a toxic factor such as dioxin, and is flexible enough to permit substitution of other mathematical exposure models by the user. The GIS thus provides a basis for estimation of herbicide exposure for use in large-scale epidemiologic studies. To facilitate widespread use of the GIS, a user-friendly software package was developed to permit researchers to assign exposure opportunity indexes to troops, locations, or individuals.

Analysis of pharmacist and pharmacist-extender workforce in 1998--2000: assessing predictors and differences across states.

OBJECTIVE: To examine the impact of supply and demand factors on filled positions for pharmacists and pharmacist extenders (pharmacist technicians and aides) and assess differences across states through analysis of state-level pharmacist labor market data. DESIGN: Cross-sectional analysis. SETTING: United States. PARTICIPANTS: Not applicable. INTERVENTIONS: Not applicable. MAIN OUTCOME MEASURES: State-level counts of filled pharmacist and pharmacist-extender positions, wages, and various available demographic, health, policy, and other factors related to the pharmacist labor market. RESULTS: Across states, the total population and the number of community pharmacy prescriptions were very accurate predictors (R2 = 0.99) of the number of pharmacist and pharmacist-extender positions, and all other variables were insignificant after these two variables were controlled for. Pharmacists and pharmacist-extenders were positively correlated, and the ratio of the two was not related to observable policy-related variables. Outlying states, in terms of simple pharmacist-to-population ratios, were difficult to categorize. CONCLUSION: Future changes in prescriptions are likely to affect the pharmacist and pharmacist-extender labor markets. Across states, pharmacists and extenders relate as complements rather than substitutes. The number of pharmacist graduates and state-level regulations regarding technician-to-pharmacist ratios appears to have a small effect on filled positions across states.

Bromide kinetics and distribution in the rat. II. Distribution of bromide in the body.

The distribution of 82Br-bromide in 15 different organs and tissues of rats has been determined by high-resolution gamma-ray spectrometry and by the scintillation counting technique at different times after the application of Na 82Br, either by subcutaneous injection or by continuous administration in the drinking water. The amount of 82Br-bromide in the various tissues reached its largest uptake within a few hours, and the concentration ratio of 82Br in the tissues to blood remained practically constant between 8 and 396 h after the application. The whole stomach of rats was the only organ of those investigated that had a larger uptake of 82Br than blood. Contrary to some previous findings, the concentration of radiobromide in the thyroid was found not to exceed that in the blood. A remarkably high concentration of 82Br was found in the skin, which represented, because of its large mass, the most abundant depot of bromide in the body of rats. The demonstrated excretion of bromide was mainly renal, at a rate of approximately 5% of the administered dose per 24 h.

Treatment of root caries lesions with chlorhexidine-containing varnishes and dentin sealants.

PURPOSE: To evaluate the effects of chlorhexidine-containing varnishes and dentin sealants on the progress of demineralization, cariogenic bacteria and clinical appearance of root caries lesions. METHODS: 68 lesions in 22 adult patients, with a mean age of 51.3 +/- 13.8 years were enrolled in this study. After the removal of soft, infected dentin, all lesions were randomly sealed with (1.) Cervitec, used as control (CE), (2.) EC 40 (EC), (3.) Prime and Bond NT (PB) or (4.) Seal & Protect (SP). Clinical appearance, laser fluorescence diagnosis (DIAGNOdent) and bacteriological status were recorded at intervals of 1 and 3 months. RESULTS: Regarding alteration of surface texture and color of the lesions, differences between the groups were not detected. Application of EC resulted in suppressed MS counts after 3 months (P < 0.05). Initial lower MS counts were observed in the CE group (P = 0.053). Laser fluorescence values increased in the chlorhexidine groups (CE = 27.9 +/- 17.5, EC = 28.3 +/- 26.0) but remained stable in the sealant groups (PB = 15.1 +/- 22.0, SP = 10.2 +/- 7.3), (P < 0.05).

[Experiences with the QBC-V hematology system at the veterinary hospitals of Zurich and Bern].

The QBC V hematology system was tested with respect to its application in veterinary medicine. PCV's and counts of total leukocytes, granulocytes, lympho/monocytes and platelets collected from 435 horses, dogs and cats were determined and compared with conventionally measured values. Precision and accuracy were found to be good for the majority of parameters. In the authors' opinion the QBC V hematology system is well suited for use in veterinary practice.

The role of endothelium in the pathogenesis of vascular diseases.

In connection with the increasing importance of the endothelium in understanding the pathogenesis of vascular diseases, new types of mediators originating in the endothelium are described, such as EDRF and endothelin. The method of endothelial cell cultivation has contributed to their detection. Besides such analytical methods, a more global in vivo aspect of endothelial function is dealt with by the method of counting circulating endothelial cell carcasses in blood applicable also clinically. The increase of endothelaemia goes hand in hand with both the tendency to thrombosis and increased permeability. Similarly, the main atherosclerotic risk factors, smoking, hypertension and hypercholesterolaemia could be effective by producing an endothelial perturbation as indicated by increased endothelaemia.

Method evaluation: automated microscopy as a compendial test for particulates in parenteral solutions.

Accurate enumeration and sizing of contaminant particles in parenteral solutions are critical to the assessment of product quality and suitability for patient use. Current manual microscopic and instrumental (light obscuration) methodologies specified by the USP both result in data of high variability. The microscopic assay for large volume parenterals is time consuming and incorporates an undesirable level of subjectivity; light obscuration counters tend to undersize larger particles and fibers and have low detection efficiency for some particle types commonly present in small volume injections. Light microscopic image analysis is proposed as a method which combines the best features of the two present methods and allows an accurate, precise, and cost effective analysis of parenteral contaminant particles. This paper briefly summarizes the principles of microscopic image analysis and discusses its application in concert with optimized sampling and counting techniques as an improved compendial methodology. Instrument performance requirements are discussed with reference to a number of currently available image analysis systems.

[Experimental studies of anemia in cattle caused by immune antibody. 1. Release of an immune antibody-induced anemia].

Subclinical moderate to medium haemolytic anaemia may be produced in cattle by intravenous slow drip infusion of erythrozyte-specific serum with immune antibody. Anisocytosis may be caused to appear in the blood count even by low-quantity intravenous application of iso-immune serum. Increase in serum quantity will lead to predominance of small erythrocytes (microspherocytes). The Price-Jones curve is likely to shift to the left and will flatten. The detection of microspherocytes actually reflects an antigen-antibody reaction caused by transmission of iso-immune antibody and resulting in partial damage of the erythrocyte membrane rather than in complete erythrocytolysis. The absence of both haemoglobinaemia and haemoglobinuria seems to support the assumption that premature degradation of the damaged erythrocytes takes place extravasally in the reticulohistiocytary system.

The quantitation of the rat gastric mucosa by morphometric methods.

Cell and tissue components are three-dimensional objects which are usually studied on thin sections, where they appear as two-dimensional profiles. Stereological methods, which permit an extrapolation from two- to three-dimensional space, are useful to quantify biological structures. After briefly reviewing the fundamental principles of stereology, their practical application as point-, intersection- and particle-counting is explained and illustrated on the gastric mucosa. To facilitate stereologic studies, an analytic model of gastric mucosa is presented. Finally, some data on the quantitative composition of different parts of the rat gastric mucosa as well as the mean volume and number of different gastric epithelial cells are presented.

Recent advances in epidemiological field techniques in filariasis.

There have been very few advances in epidemiological field techniques for filariasis in recent years. Advances in other areas of study of filariasis have been published and a summary of some of those which have some application to field technology will be given. Studies on vectors involving host influences on the development of filariae and the use of DNA probes for vector speciation were reported as well as the development of techniques that yielded species-specific identification of larvae in mosquito vectors. Experiments on animal transmission potentials also have been described. Mathematical models are becoming popular for various biological systems. It is postulated that models will enable workers to make predictions on prevalence of diseases and effect of control measures. A model has been proposed for bancroftian filariasis transmission dynamics and frequency distribution of microfilariae in blood samples. Other statistical methodologies have been published relating to age and sex distribution and frequency distribution and one suggested that females have increased resistance to filariasis, especially in the reproductive years. Advances in diagnostic technologies are highly relevant to the epidemiology of filariasis. The development of antibody and especially antigen detection systems using monoclonal antibodies, DNA probes, and polymerase chain reactions, etc, will make a significant impact on the identification of infected persons. Findings along other lines may also have application to field epidemiology. The quantitative blood count (QBC) acridine orange hematocrit tube has also been found to be of value in diagnosis. Elevated levels of ecdysteroids found in other filarial infections should be tested in cases of lymphatic filariasis.

[The course of Jones-mote reaction in guinea pigs after the sensitization with mycobacteria (author's transl)].

Mycobacterial antigens induce of JONES-MOTE reaction, which is well stated in 7 to 9 days after being intracutaneously sensitized (Tab. 1). The reaction is adjuvant independent; Al(OH)3 as an adjuvant stabilizes and enlarges the reaction in the interval between 7 and 9 days after the sensitization (Tab 2). The erythema size, read 6, 24 and 48 hours after the intradermal testing shows no significant differences (Tab. 3). The maximal intensity of the JONES-MOTE reaction was found after 24 hours (Tab. 4). The sensitization with 0.2 mg of bacterial mass is sufficient (Tab. 5, Fig. 1). The size of erythema increases when 0.5 mug to 10 mug of homologous sensitin are used (Tab. 6, Fig. 2). An oral application of M. fortuitum increased the total count of basophilic leukocytes and caused cross reactions (Fig. 3).

[What do we know today about neuroleptic malignant syndrome?].

Neuroleptic malignant syndrome is a complication that may occur during treatment with neuroleptic drugs, or after abrupt cessation of dopaminagonists. Although the condition is relatively rare, it has a high mortality of about 20% when untreated. The first symptoms can appear the first day after initiation of treatment with neuroleptic drugs. The syndrome manifests itself in the form of a tetralogy composed of extrapyramidal symptoms, a change in the level of conciousness, elevated body temperature and autonomic instability. It is also associated with secondary laboratory findings such as increased creatine kinase and leucocyte count. Treatment includes stopping the neuroleptic drugs, application of dopaminagonists, benzodiazepines and fluid replacement. If the patient, after an episode of neuroleptic malignant syndrome, has a psychotic relapse, neuroleptica can be recommended after a break of one to four weeks. Two cases are described to demonstrate the picture.

How mothers teach basic-level and situation-restricted count nouns.

Recent research suggests that young children assume, as a default, that a word applied ostensively to an unfamiliar object (e.g. a man riding in a car) is a basic-level count noun (e.g. person) rather than a situation-restricted count noun (e.g. passenger). In this experiment, 14 mothers and their children (mean age 3.7; range 3.0 to 4.6) participated in a story-book reading session in which the mothers taught their children both a basic-level count noun and a situation-restricted count noun for a series of object drawings. An analysis of mothers' spontaneous teaching strategies revealed that, for a given object, they typically taught a basic-level count noun before a situation-restricted count noun. Furthermore, they tended to teach basic-level count nouns exclusively through ostensive definition, especially when the objects were unfamiliar. Moreover, they were (1) more likely to use ostensive definitions and (2) less likely to provide additional information concerning the application of the word, when teaching basic-level count nouns than when teaching situation-restricted count nouns. The results provide new evidence that word teachers, like young word learners, assume that a basic-level count noun is the psychologically-privileged candidate for an ostensive definition involving an (unfamiliar) object.
