[Study on quantitative test on the DNA damage of Aspergillus flavus caused by citral with a comet analysis system].

Comet assay provided a object method for DNA damage caused by physical and chemical factors. A new system of SCGE image analysis, which combines microphotodensitometry and microphotography with image analysis have been developed. Compared with the international popular, it has well at speed, convenience, exchange face of Chinese and English and print. We used it to directly determine the A. flavus' DNA damages for citral. The results show that test time was shorted 2/3 and accuracy was improved. It is probable that cell structural parameter, cell molecule and membrane could be test long, successive, movably and instantly. So the new system has a wide application.

A sequential injection analysis system for potassium clavulanate determination using two potentiometric detectors.

Two almost independent potentiometric methods were implemented in a multi-task flow system based on the concept of sequential injection analysis for the analytical control of potassium clavulanate in pharmaceutical formulations. In addition to in-line preparation of the solutions submitted to the analysis, the system simultaneously detected the clavulanate anion and the potassium cation using two potentiometric detectors selective to each of the species referred. For the two determinations, the system developed presented an analytical range of 2 mM-0.1 M. Relative standard deviations of 0.5 and 0.6%, respectively, for clavulanate and potassium, were calculated from the signals obtained for 10 consecutive injections of a solution of 5x10(-3) M potassium clavulanate. The frequency of sampling was 53 samples h(-1). The quality of results supplied was evaluated by comparison with the reference procedure revealing relative errors less than 2% for the two determinations. The simultaneous attainment of two measurements permitted the standardisation of results in real time, the detection of faults in the procedure and monitoring the chemical stability of clavulanate.

Morpho-functional principle of neuroendocrine system analysis.

A new approach to the analysis of the neuroendocrine system (NES) is suggested. It is based on the fact of structural and metabolic determination of any effect on cell and cell aggregates. The principle of a common communication channel in the NES is formulated and a possible method of its formalization is proposed.

Global asymptotic stability of bone remodeling theories: a new approach based on non-linear dynamical systems analysis.

Mathematical tools for the analysis of nonlinear dynamical systems are applied to the study of stability of bone remodeling theories. As a practical application, the same problem studied by Harrigan and Hamilton (1992) and Cowin et al. (1994b) is analysed using these tools, and their findings on the necessary and sufficient conditions to ensure local asymptotic stability are easily confirmed. Using a general approach based on Lyapunov's method the same condition has been found to be necessary and sufficient also for the global asymptotic stability, thus confirming a result obtained by Harrigan and Hamilton (1994) by variational methods applied to finite-element models. The proof is based on the discretization of the spatial domain but the results for the continuum can be easily extrapolated.

The Oxford modular cataract image analysis system.

A modular system of acquisition and analysis of Scheimpflug, retro-illumination and fluorescence images of the in vivo human crystalline lens is described. Image analysis is directed towards the following goals: Scheimpflug slit-images are analysed for: (1) The optical density of nuclear cataract present; (2) The dimensions of the lens and the lenticular zones; (3) The curvatures of the lens and lenticular zones. Retro-illumination images are analysed for: (1) The percentage area which is occupied by cataract; (2) A combined measure (weighted integral) describing both the amount of cataract present and its optical density. Lenticular auto-fluorescence images are analysed for the mean density (fluorescence) of the lens as a whole. A pilot study of the repeatability of the methods is presented.

[Usefulness of a dose distribution analysis system with a common scanner].

Although one-dimensional densitometry has been widely performed for linac dosimetry, it is a time-consuming, labor-intensive procedure. We studied a new densitometry system (DD-System) that consists of a personal computer and common flatbed-type scanner, with regard to its usefulness for daily QC checks in comparison with conventional densitometry. The spatial resolution of the DD-System is very high, and its accuracy of measurements of field size and flatness are equivalent to those of a conventional system at an optical density <2.0. Although the performance of the image scanner limits the maximum optical density of our system, this system can acquire high-resolution two-dimensional dose distributions quickly and easily. In conclusion, the DD-System is very useful for reducing the amount of time and effort required in daily QC activities. We consider this system applicable to the analysis of complicated dose distributions in this era of IMRT.

Overlapping functional systems: a theory for vertebrate central nervous system function in terms of informal systems analysis.

An holistic theory of the functional organization of the central nervous system, a system at the level of the organ, in vertebrate organisms is presented as an alternative to localization of function by using two sets of complementary rules for systems designation derived from systems theory. These rules reveal three types of systems according to levels of operation and the origins of goals. These are: (1) the teleogenic or primary systems (reproductive, food-water intake-use, thermoregulative, immune, oxygen intake-use); (2) the teleozetic or subsystems (internal sensory and motor, external motor, external sensory); (3) the teleonomic systems or echelons (field, local circuit, basic functional unit, basic excitation unit). The systems, subsystems, and echelons are discussed with regard to their hierarchical relationships, the nature of their goals, and the supporting experimental evidence. The theory is discussed as an explicit statement of multileveled relationships for the analysis of the vertebrate CNS and, therefore, as presenting a paradigm for, or a way of thinking about, local and global brain theories, brain dysfunction, and brain evolution.

LAMP. A data management and analysis system for patient registries and clinical studies.

A computer system for the management and analysis of clinical data is described. The system is implemented in the ANSI standard MUMPS language and runs on popular minicomputers. The system is designed to allow clinical investigators or research assistants to define a data base, enter and edit data, produce patient-specific reports, and perform a variety of analyses on user-specified groups of subjects. Programmer intervention is not required at any stage of the management or analysis process, and on-line documentation provides a friendly environment for inexperienced users.

Sequence analysis of Chinese and Japanese Curcuma drugs on the 18S rRNA gene and trnK gene and the application of amplification-refractory mutation system analysis for their authentication.

The botanical origins of Chinese and Japanese Curcuma drugs were determined to be Curcuma longa, C. phaeocaulis, the Japanese population of C. zedoaria, C. kwangsiensis, C. wenyujin, and C. aromatica based on a comparison of their 18S rRNA gene and trnK gene sequences with those of six Curcuma species reported previously. Moreover, to develop a more convenient identification method, amplification-refractory mutation system (ARMS) analysis of both gene regions was performed on plants. The ARMS method for the 18S rRNA gene was established using two types of forward primers designed based on the nucleotide difference at position 234. When DNAs of four Curcuma species were used as templates, PCR amplification with either of the two primers only generated a fragment of 912 base pairs (bp). However, when DNAs of the purple-cloud type of C. kwangsiensis and C. wenyujin were used, PCR amplifications with both primers unexpectedly generated the fragment, suggesting that these two were heterozygotes. The ARMS method for the trnK gene was also established using a mixture of four types of specific reverse primers designed on the basis of base substitutions and indels among six species, and common reverse and forward primers. C. phaeocaulis or the Chinese population of C. zedoaria, the Japanese population of C. zedoaria or the purple-cloud type of C. kwangsiensis, the pubescent type of C. kwangsiensis or C. wenyujin, and C. aromatica were found to show specific fragments of 730, 185, 527 or 528, and 641 or 642 bp, respectively. All species including C. longa also showed a common fragment of 897-904 bp. Using both ARMS methods, together with information on producing areas, the identification of Curcuma plants was achieved. Moreover, the ARMS method for the trnK gene was also useful for authentication of Curcuma drugs.

Computer-assisted semen analysis systems: comparison with routine evaluation and prognostic value in male fertility and assisted reproduction.

Semen analysis was performed by employing a computer-assisted semen analysis (CASA) system (SM-CMA), in comparison with visual estimation by microscope. There was a significant relationship between the values obtained by both methods, but a large range of differences in individual values was observed. Results of semen analysis in 407 men complaining of reduced fertility were investigated for their relationship to fertility outcome. The parameters obtained by the CASA system were analysed in relation to time to conception, applying the Cox proportional hazards model of regression. In univariate regression analysis, all examined CASA parameters were shown to have a significant effect on cumulative hazard function. However, applying a multivariate step forward approach, only percentage of motile spermatozoa and log-transformed values of sperm count remained significant predictors of later fertility due to the close intercorrelations among the examined covariates. We concluded that the determination of elaborate motility characteristics as obtained by CASA systems is of limited value to optimizing the evaluation of male fertility status.

The multi-purpose camera: a new anterior eye segment analysis system.

A new device for the analysis of the anterior eye segment was developed. With this device, a Scheimpflug slit image and a retroillumination image can be taken separately using a CCD camera. The quality of the images obtained was satisfactory enough for analysis. The data obtained were stored and analyzed on an almost real-time level using an on-line computer. The menu for the data analysis provided for this device includes multilinear densitometry, with planimetry, area densitometry, measurement of anterior chamber depth and thickness of lens layers, determination of chamber angle and positioning of implanted intraocular lenses, and measurement of opaque areas in the retroillumination image.

The neurobiology of conditioned and unconditioned fear: a neurobehavioral system analysis of the amygdala.

A neurobehavioral system approach to conditioned and unconditioned fear is presented. By employing reproducible fear behaviors in Pavlovian conditioning and unconditioned fear paradigms, it has been possible to delineate some differences in neural circuitry and cellular biology for conditioned and unconditioned fear. It is suggested that the basolateral complex of the amygdala and the central nucleus of the amygdala are part of the neural circuitry for fear conditioning but not for unconditioned fear to a predator odor. Furthermore, changes in expression of the transcription factor early growth response gene 1 in the lateral nucleus of the amygdala are shown to be important for contextual fear conditioning but not for unconditioned fear to a predator odor. In addition, data suggest that although conditioning to a synthetic predator odor, trimethylthiazoline, has been difficult to demonstrate, conditioning can occur by modifying by the environment. Finally, the relevance of the animal studies to human anxiety disorders is discussed.

[The interoceptive properties of the vestibular apparatus (a systems analysis of vestibular-oculomotor interaction)].

Modern views about physiological functions of vestibular system are submitted, their contradictions are picked out. The result of development of experimental and clinical vestibulometry till last decades at the foundation of critical estimation of effectiveness of different methods of vestibular receptor's stimulation are summed up. It is proposed to consider vestibular apparatus only as structural sensory element of eye's moving functional system at the foundation of cybernetics' principles of physiological function's regularity. It is proposed to estimate vestibular function as a part of the whole afferent stream in system, which hasn't their own way at effector neurone once which determines together with proprioceptive afferentation internal system's status. Methodological principles of further research of vestibular apparatus and vestibular function are determined.
