Isolation and characterisation of genes for androgen-responsive secretory proteins of rat seminal vesicles.

Under the influence of testosterone, rat seminal vesicles synthesise large amounts of a tissue specific protein, S. Recombinant lambda clones have been isolated containing overlapping sequences covering a 27.5 kilo base region of the rat genome within which the gene for protein S is located. Recombinant plasmids bearing cDNA sequences for protein S were constructed in pBR328. One (pcS2) contains a 690 nucleotide insert and is probably full length. Detailed restriction maps of the S-gene are presented and the structure was confirmed by analysis of R-loops and heteroduplexes. The S-gene covers a 2 kbp region of the genome and consists of a 5' intron (490 bp) separating a leading exon (120 bp) containing the 5' untranslated region from a central exon (310 bp) containing most of the coding sequence and part of the 3' untranslated region. A larger intron (1100 bp) lies within the 3' untranslated region. The cloned gene is representative of the native gene but the S gene may be heterogeneous. Using pcS2, the hormonal control of S-specific mRNA was examined and a pronounced differential response to testosterone was observed.

Age-related changes in purinergic and adrenergic components of sympathetic neurotransmission in guinea-pig seminal vesicles.

1. Purinergic and adrenergic components of sympathetic neurotransmission and contractile responses to exogenous alpha,beta-methylene ATP and noradrenaline have been investigated in the seminal vesicles of 1-day (new-born), 2-weeks (young), 12-weeks (adult) and 2-years old (aged) guinea-pigs. 2. In seminal vesicles of new-born guinea-pigs electrical field stimulation (EFS; 80 V, 0.5 ms for 30 s, 2-32 Hz) evoked tonic frequency-related contractions. In 2-weeks old guinea-pigs the tonic contraction masked an initial phasic component of the neurogenic responses, whereas in 12-weeks and 2-years old guinea-pigs, neurogenic responses were biphasic, a phasic response being followed by a tonic contraction. In all experimental groups, prazosin (10(-6) M) blocked the tonic contraction while desensitization of P2X receptors by alpha,beta-methylene ATP (10(-4) M) abolished the phasic responses. 3. The phasic purinergic component of the neurogenic response was significantly higher in 12-weeks and 2-years old animals, compared with 2-weeks old guinea-pigs. At 32 Hz phasic contractions were (mN mg(-1) tissue): 0.047+/-0.012, 0.018+/-0.040 and 0.147+/-0.026 in 2-weeks, 12-weeks and 2-years old guinea-pigs, respectively. In contrast, the tonic adrenergic component of the neurogenic contraction significantly declined at 12-weeks and 2-years compared with 2-weeks old guinea-pigs. 4. Contractile responses (mN mg(-1) tissue) to the highest concentration of alpha,beta-methylene ATP tested were significantly higher in 2-weeks (0.248+/-0.022) than in 1-day old animals (0.113+/-0.012) and decreased in 12-weeks (0.163+/-0.016) and 2-years old guinea-pigs (0.200+/-0.008). The pD2 values for the purinoceptor agonist were also significantly lower in adult (4.74+/-0.20) and aged guinea-pigs (5.22+/-0.08) compared with 2-weeks old animals (5.91+/-0.27). Conversely, responses to the highest concentration of noradrenaline gradually decreased with age, without significant changes in the pD2 values. Contractile responses to KCl (240 mM) did not differ significantly between the experimental groups. 5. These results demonstrate age-related changes in purinergic and adrenergic components of sympathetic neurotransmission in the guinea-pig seminal vesicles. The purinergic component is absent in new-born animals and it appears fully developed in adult and old guinea-pigs, while the adrenergic component decreases with age. Pre- and postjunctional mechanisms contributing to the age-related changes of sympathetic neurotransmission are discussed.

The protein conformation and a zinc-binding domain of an autoantigen from mouse seminal vesicle.

The protein conformation of a mouse seminal vesicle autoantigen was studied by circular dichroism spectroscopy. At pH 7.4, the spectrum in the UV region appears as one negative band at 217 nm and one positive band at 200 nm. This together with the predicted secondary structures indicates no helices but a mixture of beta form, beta turn, and unordered form in the protein molecule. The conformation is stable even at pH 10.5 or 3.0. The spectrum in the near-UV region consists of fine structures that are disturbed in acidic or alkaline solution. The environments around Trp2 and Trp82 of this protein were studied by intrinsic fluorescence and solute quenching. They give an emission peak at 345 nm, and about 87% of them are accessible to quenching by acrylamide. Correlating the quenching effect of CsCl and Kl on the protein fluorescence to the charged groups along the polypeptide chain suggests the difference in the "local charge" around the two tryptophan residues. The presence of ZnCl2 in the protein solution effects no change in the circular dichroism but perturbs the fluorescence due to Trp82. Analysis of the fluorescence data suggests a Zn(2+)-binding site on the protein, which cannot coordinate with both Ca2+ and Mg2+. The association constant for the complex formation is 1.35 x 10(5) +/- 0.04 x 10(5) M-1 at pH 7.4.

Hyperviscosity and hypofunction of the seminal vesicles.

The study was designed to determine whether hyperviscosity of the semen sample is related to dysfunction of the male accessory glands. It was carried out on men who consecutively attended an infertility clinic between June 1989 and June 1991, and the men were grouped according to viscosity of semen samples (normal viscosity or higher viscosity). Semen samples from 229 infertility patients were studied. From these, 155 had normal viscosity and 74 showed hyperviscosity. The effect of hyperviscosity of semen samples on seminal quality and the function of the prostate was evaluated by acid phosphatase measurement, and the seminal vesicles by measurement of corrected fructose. Sperm motility (grades II-III), sperm vitality, and corrected fructose were significantly reduced in samples with high viscosity (p < .05). A high prevalence of hyperviscosity in semen samples was associated with only hypofunction of the seminal vesicles. In fact, 36.5% of subjects with hyperviscosity showed reduced levels of corrected fructose. The same association with hyperviscosity was not observed when only hypofunction of the prostate was present, or when hypofunction of both prostate and seminal vesicles was present (P:NS). Further analysis showed that high viscosity is observed mainly when corrected seminal fructose levels were below 1.5 mg/mL x 10(6) spz/mL. It would appear that hyperviscosity affects sperm motility and is associated with hypofunction of the seminal vesicles.

[Primary seminal vesicle abscess: diagnosis and treatment by transrectal ultrasound].

A 38 years old man with the only past medical history of bilateral orchiopexy 15 years ago is diagnosed of left primary seminal vesicle abscess. The patient is sent to our emergency service with an ultrasound finding of either dilated vas deferent or seminal vesicle abscess with hiperecogenic material inside it. The examination was performed after a 5 month history of purulent ejaculation. The diagnose of seminal vesicle abscess is confirmed by TRUS and, in the same procedure, drainage of the abscess was completed by needle-aspiration puncture. After this treatment the patient stay 12 hours at the observation room with intravenous antibiotics and is discharged home with oral antibiotics. We are interested in this case because of the low incidence of this pathology and the easy diagnosis and treatment by TRUS.

Echinococcal cyst of the seminal vesicles: a case-report and literature review.

Solitary echinococcal cysts of the seminal vesicles are very uncommon. The use of ultrasonography, computed tomography and magnetic resonance imaging aids considerably in the differential diagnosis. Surgical removal of pelvic echinococcal cysts should be performed with great caution to avoid accidental puncture and parasite spillage in the retroperitoneal space. We report on a case of a male patient presenting with urinary retention owing to a large hydatid cyst of the seminal vesicles. The literature regarding this rare clinical entity is also reviewed.

A hitherto undescribed case of "collision" tumour: liposarcoma of the seminal vesicle and prostatic carcinoma.

A hitherto undescribed type of the rare "collision" tumours is reported: necropsy in a 77-year-old male patient revealed liposarcoma of the right seminal vesicle in association with anaplastic carcinoma of the prostate. The carcinoma was fused with the liposarcoma as a result of its invasive local growth and had formed deposits in the lymph nodes, bones and lungs. The left seminal vesicle showed localized amyloidosis. A review of the published cases of mesenchymal tumours of the seminal vesicles is presented.

Isolated vasculitis of the seminal vesicle.

We present 2 cases of histologically identified necrotizing vasculitis found in the seminal vesicles of radical prostatectomy specimens containing prostatic adenocarcinoma. In neither case did the patient suffer from symptoms nor show signs of systemic vasculitis. In 1 case, the vasculitis correlated clinically with dense adhesions noted at surgery that simulated extraprostatic tumor extension. Isolated vasculitis of the seminal vesicle is similar to reports of isolated vasculitis limited to other organs, in that local clinical findings may or may not be present, but the patient is free of systemic disease. Careful clinical follow-up is warranted to exclude the subsequent development of systemic vasculitis in these patients.

Signals of seminal vesicle autoantigen suppresses bovine serum albumin-induced capacitation in mouse sperm.

Capacitation is the prerequisite process for sperm to gain the ability for successful fertilization. Unregulated capacitation will cause sperm to undergo a spontaneous acrosome reaction and then fail to fertilize an egg. Seminal plasma is thought to have the ability to suppress sperm capacitation. However, the mechanisms by which seminal proteins suppress capacitation have not been well understood. Recently, we demonstrated that a major seminal vesicle secretory protein, seminal vesicle autoantigen (SVA), is able to suppress bovine serum albumin (BSA)-induced mouse sperm capacitation. To further identify the mechanism of SVA action, we determine the molecular events associated with SVA suppression of BSA's activity. In this communication, we demonstrate that SVA suppresses the BSA-induced increase of intracellular calcium concentration ([Ca2+]i), intracellular pH (pH(i)), the cAMP level, PKA activity, protein tyrosine phosphorylation, and capacitation in mouse sperm. Besides, we also found that the suppression ability of SVA against BSA-induced protein tyrosine phosphorylation and capacitation could be reversed by dbcAMP (a cAMP agonist).

[The seminal vesicle dynamic emptying test as an interventional echographic control of the spermogram in the examination of male sterility].

Ultrasound imaging to verify effective emptying of seminal vesicles is proposed, during spermograms included in investigation of couple sterility. The aim is to detect distal obstructive seminopathies, often unilateral and inaccessible to enzymatic analysis of sperm. A review of recently drafted case reports of 41 sterile men showed that in half of them an insufficient ejaculate volume always corresponded to poor emptying of seminal vesicles as seen on ultrasound images. If an obstruction is present it is usually in the ejaculatory canal zones. Two patients with azoospermia were treated by endoscopic resection of terminal part of ejaculatory canals with only one good result in the spermogram. With respect to counting of spermatozoa, it is not possible in patients with decreased emptying of seminal vesicles, a part from those with azoospermia, to establish any correlation between type of obstructive seminopathy and its effect on the spermogram. Of the 41 men in this series, a half had problems with their partner and the other half another known sterility factor.

Role of high molecular weight seminal vesicle proteins in eliciting the uterine inflammatory response to semen in mice.

Mating evokes a characteristic pattern of molecular and cellular events in the rodent reproductive tract, including an infiltration of the endometrial stroma and uterine lumen with activated macrophages and granulocytes, which closely resembles a classic inflammatory response. Previous studies in mice indicate that these cellular changes are associated with, and are largely a consequence of, an upregulated synthesis and release of granulocyte-macrophage colony-stimulating factor (GM-CSF) from the uterine epithelium in response to seminal fluid. The aim of this study was to investigate further the origin and nature of the factors present in seminal fluid that trigger the GM-CSF response. It was found that the characteristic increase in uterine expression of mRNA encoding GM-CSF and release of GM-CSF bioactivity from uterine epithelial cells into the luminal cavity seen after mating with intact or vasectomized males was no longer evident in matings with male mice from whom the seminal vesicles had been surgically removed. The extent of inflammatory leucocyte infiltration into the endometrium was also reduced; the most notable effect was a complete absence of the exocytosis of neutrophils into the luminal cavity normally seen after matings with intact or vasectomized males. Bioassay of the GM-CSF output of oestrous endometrial cells after culture with crude or Sephacryl S-400 chromatographed fractions of seminal vesicle fluid showed that the GM-CSF stimulating activity was predominantly associated with protein moieties in seminal vesicle fluid of approximately 650,000 M(r) and 100,000-400,000 M(r). These data confirm the presence in seminal vesicle fluid of specific factors that initiate an inflammatory response in the uterus after mating through upregulating GM-CSF synthesis in the uterine epithelium. The significance of the cytokine release and cellular changes induced by seminal plasma for implantation of the conceptus and pregnancy outcome remain to be determined.

Localized amyloidosis of seminal vesicles: report of three cases in surgically obtained material.

Localized amyloidosis of the seminal vesicles (ASV) is reported as an incidental finding in surgical specimens from three elderly men. In two cases, the amyloid deposits were bilateral, subepithelial, and clinically inapparent, features similar to other cases in the literature. In one case, the diagnosis was made on a transrectal prostatic needle biopsy that included a small portion of seminal vesicle; to our knowledge, this has not been previously reported. Electron microscopy in one case demonstrated nonbranching fibrils characteristic of amyloid, and pretreatment of tissue sections using the permanganate method in two cases showed almost complete ablation of congophilia. Evidence suggests that ASV is a permanganate-sensitive, non-AA (amyloid, protein A) type of amyloid that may be different from all other types of amyloid previously characterized.

Schwannoma of the seminal vesicle.

A 79-year-old man presented with a history of nocturia. On rectal examination a solid mass above the prostate gland was discovered. Computerized tomography (CT) and ultrasound (US) showed that the mass was solid and was arising from the seminal vesicle. After a needle biopsy, pathology and immunocytochemistry confirmed the mass to be Schwannoma. Tumors of the seminal vesicle are rare. To our knowledge Schwannoma of the seminal vesicle has not been described before.

In vivo evaluation of serotonergic agents and alpha-adrenergic blockers on premature ejaculation by inhibiting the seminal vesicle pressure response to electrical nerve stimulation.

OBJECTIVE: To evaluate the effect of drugs on premature ejaculation using a rat animal model in which the seminal vesicle was electrically stimulated via its lesser splanchnic nerve and changes in the pressure response monitored. MATERIALS AND METHODS: Male Wistar rats (aged 12-14 weeks) were injected intravenously with prazosin and serotonergic agents (serotonin, clomipramine, fluoxetine, imipramine and indatraline) at various concentrations 10 min before electrical nerve stimulation (ENS) of the lesser splanchnic nerve; the initial increase in seminal vesicle pressure in response to ENS was then compared. RESULTS: The pressure response to ENS was reduced in the presence of prazosin or serotonergic agents. The mean (SEM) maximum inhibition values were 84.1 (8.9%) by fluoxetine at 0.1 mg/kg, 67.9 (8.7)% by prazosin at 0.1 mg/kg, 60.9 (11.0)% by serotonin at 3 mg/kg, 54.9 (4.6)% by clomipramine at 3 mg/kg, 30.0 (11.0)% by imipramine at 0.1 mg/kg, and 20.9 (4.3)% by indatraline at 0.1 mg/kg. From the concentration-response curve, the potency of prazosin was lower than that of fluoxetine, but was higher than that of serotonin or clomipramine. CONCLUSIONS: Like serotonin, fluoxetine and clomipramine can reduce the pressure response of the seminal vesicle to ENS. Among these inhibitory agents, including prazosin, fluoxetine was the most effective and may be valuable for the clinical treatment of ejaculatory dysfunction in man.

In vitro translation of major abundant class poly(A+)-mRNA from rat seminal vesicle.

Total poly (A)-containing messenger RNA [poly(A)-mRNAT] was isolated from seminal vesicles of adult male rats. By preparative 5%-20% linear sucrose gradient centrifugation in 1% SDS, a single peak was seen in the 11S region [poly(A)-mRNA11S]. Poly (A)-mRNAT and poly(A)-mRNA11S were translated in the wheat germ S-30 in vitro translation system. Laemmli-SDS-polyacrylamide slab gel electrophoresis of translation products of poly(A)-mRNAT revealed three major protein bands at approximately 18,500, 15,000, and 14,000 daltons. In vitro translation of poly(A)-mRNA11S across the 11S peak revealed partial purification of the mRNAs for the three predominant proteins. The lower molecular weight part of the 11S peak enriched for the mRNAs coding for the 15,000 and 14,000 dalton polypeptides, and the higher molecular weight side of the peak enriched for the 18,500 dalton mRNA. The highly abundant class of mRNA from rat seminal vesicle can be purified with one sucrose gradient centrifugation.

[A seminal vesicle cyst treated by transurethral resection: a case report].

A 40-year-old man with dysuria was referred to our department on November, 18, 2004. A transabdominal ultrasonography revealed a cystic mass arising from his bladder neck. Computed tomography and magnetic resonance imaging revealed the cyst to be homogeneous and ovally shaped with a clear margin. Bilateral vasography showed the compression of the left seminal vesicle by the cyst and there was communication between the cyst and the left seminal vesicle for diagnosis of the seminal vesicle cyst, percutaneous needle aspiration was performed. The aspirated fluid included microscopical spermatozoa and proved negative for cytology. Absolute ethanol was injected into the cyst and transurethral unroofing of the cyst was performed on January 14, 2005. The prostate was resected until we reached into the cyst.

Vasoactive intestinal peptide stimulates apical secretion in hamster seminal vesicle epithelial cells in culture.

In this study, we investigated the presence of vasoactive intestinal peptide (VIP) receptors in the epithelial cells of the hamster seminal vesicle, by using cell clusters isolated from the gland and cultivated in a serum-free bicameral culture system. An immunocytochemical approach and autoradiographic and biochemical binding experiments with 125I-VIP as radioligand were performed. The effect of this neuropeptide on cultured cell proliferation and protein secretory activity was also analysed. The release of trichloroacetic-acid-precipitable radioactive material by epithelial cells to the apical and basal compartments of the bi-chamber was estimated in absolute and relative terms. The results of this work indicate that: (1) VIP receptors are present in the membranes of clusters of epithelial cells from seminal vesicles and are further maintained in cultured cells; (2) VIP does not exert any mitogenic effect in these cells; (3) VIP affects the directionality of secretion, favouring the absolute and relative amounts of protein released to the apical compartment of the bi-chamber. The expression of VIP receptors in the epithelial cells of the hamster seminal vesicle and the direct secretagogue-like activity of this neuropeptide in these cells might be affected by other factors, namely, androgens.
