Thiocyanate cannot inhibit the formation of reactive nitrogen species in the human oral cavity in the presence of high concentrations of nitrite: detection of reactive nitrogen species with 4,5-diaminofluorescein.

In the human oral cavity, nitrite is reduced to nitric oxide (NO) by certain bacteria. 4,5-Diaminofluorescein (DAF-2) was transformed to a fluorescent component triazolfluorescein (DAF-2T) in a bacterial fraction of saliva in the presence of nitrite. No detectable consumption of DAF-2 and formation of DAF-2T were observed in bacterial fraction in the absence of nitrite. The nitrite-dependent transformation of DAF-2 to DAF-2T was inhibited by catalase, SCN(-), and CN(-) suggesting the participation of peroxidases in saliva in the transformation. The formation of DAF-2T, which was observed by the addition of an NO generating reagent (+/-)-(E)-4-ethyl-2-[(E)-hydroxyimino]-5-nitro-3-hexenamide (NOR 3) to bacterial fraction, was also inhibited by catalase, SCN(-), and CN(-). The degree of the inhibition by SCN(-) decreased as the concentration of nitrite or NOR 3 was increased. Superoxide dismutase (SOD) enhanced nitrite- and NOR 3-induced fluorescence increase in the bacterial fraction, and the degree of the enhancement decreased as the concentrations of nitrite and NOR 3 were increased. In whole saliva filtrate, the inhibitory effects of SCN(-) on the fluorescence increase decreased as the concentration of nitrite was increased, but the enhancement by SOD was not significantly affected by the increase in the concentration of nitrite. As salivary bacteria produce O(2)(-), H(2)O(2), and NO and as peroxidase/H(2)O(2)/nitrite systems in saliva produce NO(2), the effects of SCN(-) are discussed taking SCN(-)-dependent inhibition of NO(2) formation by peroxidases in saliva into consideration and the effects of SOD are discussed taking O(2)(-)-dependent consumption of NO into consideration. It is concluded that when the rate of the formation of NO is high, SCN(-) is not effective enough to inhibit the formation of N(2)O(3) in the oral cavity.

Three species of plagioporine opecoelids (Digenea), including a new genus and two new species, from marine fishes from off the coast of Chile.

Villarrealina peruanus (syn. Pirupalkia queulensis) is described from Cilus gilberti, off the Talcahuano region, Chile. It is considered an opecoelid and is distinguishable by the combination of its extensive uterus, which reaches to the posterior extremity, and the vitellarium restricted to the hindbody. Jerguillicola leonora n. g., n. sp. from Aplodactylus punctatus, off the Talcahuano region, Chile, is distinguished by the uterus reaching into the post-testicular region, the lack of a seminal receptacle, the eggs attached together is strings formed by a matrix, the large cirrus-sac filled mainly with fairly dense connective tissue, a long coiled seminal vesicle and a long coiled ejaculatory duct. Neolebouria georgenascimentoi n. sp. from Pinguipes chilensis and Prolatilus jugularis, off the Talcahuano region, Chile, can be distinguished from its congeners by a its elongate cirrus-sac and oblique entire to weakly lobed testes.

White piedra and Trichosporon species in equatorial Africa. III. Identification of Trichosporon species by slide agglutination test.

Fifty-two Trichosporon strains isolated from Gabonese female patients 15-60 years, were studied. The identity of these strains was established by two different methods: the method proposed by Guého et al. (1992), based on mycological criteria, and a slide agglutination method performed with monospecific antisera prepared in our laboratory. The final results show a perfect correlation between the two methods, which allowed us to identify 25 strains of T. mucoides, 21 strains of T. inkin and seven strains of T. asahii. The results of the agglutination tests performed with 24-h-old subcultures grown on Sabouraud glucose agar are available in less than 15 mins. In the light of these results, it appears that this method, which is rapid and easy to perform and reproduce, may readily be used in hospital laboratories. In addition, this method allowed us to verify the presence of antigens common to the genera Cryptococcus and Trichosporon, which were easily shown by the use of crude sera. The fact that the anti-Trichosporon monospecific sera lose their capacity to agglutinate Cryptococcus neoformans proves their high specificity.

Rab GTPases are recruited to chlamydial inclusions in both a species-dependent and species-independent manner.

Chlamydiae are obligate intracellular bacteria that replicate within an inclusion that is trafficked to the peri-Golgi region where it fuses with exocytic vesicles. The host and chlamydial proteins that regulate the trafficking of the inclusion have not been identified. Since Rab GTPases are key regulators of membrane trafficking, we examined the intracellular localization of several green fluorescent protein (GFP)-tagged Rab GTPases in chlamydia-infected HeLa cells. GFP-Rab4 and GFP-Rab11, which function in receptor recycling, and GFP-Rab1, which functions in endoplasmic reticulum (ER)-to-Golgi trafficking, are recruited to Chlamydia trachomatis, Chlamydia muridarum, and Chlamydia pneumoniae inclusions, whereas GFP-Rab5, GFP-Rab7, and GFP-Rab9, markers of early and late endosomes, are not. In contrast, GFP-Rab6, which functions in Golgi-to-ER and endosome-to-Golgi trafficking, is associated with C. trachomatis inclusions but not with C. pneumoniae or C. muridarum inclusions, while the opposite was observed for the Golgi-localized GFP-Rab10. Colocalization studies between transferrin and GFP-Rab11 demonstrate that a portion of GFP-Rab11 that localizes to inclusions does not colocalize with transferrin, which suggests that GFP-Rab11's association with the inclusion is not mediated solely through Rab11's association with transferrin-containing recycling endosomes. Finally, GFP-Rab GTPases remain associated with the inclusion even after disassembly of microtubules, which disperses recycling endosomes and the Golgi apparatus within the cytoplasm, suggesting a specific interaction with the inclusion membrane. Consistent with this, GFP-Rab11 colocalizes with C. trachomatis IncG at the inclusion membrane. Therefore, chlamydiae recruit key regulators of membrane trafficking to the inclusion, which may function to regulate the trafficking or fusogenic properties of the inclusion.
