Transcription of telomere repeats in protozoa.

The telomerically located variant cell surface glycoprotein (VSG) gene expression sites of the protozoan parasite Trypanosoma brucei are transcribed by an unusual alpha-amanitin resistant RNA polymerase. We show that the telomere GGGTTA repeats located at the chromosome ends of T. brucei and the related protozoan T. equiperdum are also transcribed by alpha-amanitin resistant RNA polymerases. This transcription predominantly proceeds unidirectionally towards the end of the chromosome, in both bloodstream and insect form trypanosomes and results in the generation of heterogeneously sized steady state RNA. We postulate that telomere repeat transcription results from readthrough downstream of telomeric genes. Telomere repeat transcription was found in all seven protozoan species tested, but was alpha-amanitin resistant only in trypanosome species which exhibited antigenic variation. The data indicate that in some trypanosome species a subset of telomeres is transcribed by a different type of RNA polymerase.

Cell surface control in Blepharisma intermedium (Protozoa Ciliata). Blocking of gamone II-induced homotypic interacting ligands.

PHA, Con-A, or anti-tubulin antibodies inhibit homotypic pair formation, in B, intermedium mating type-I cells in the presence of suboptimal concentrations of gamone II. The inhibition is dependent on the dose of gamone added; the structural conformation and the relative concentration of the inhibitor; and the time of addition of the inhibitor. The block can be selectively prevented by competitive inhibitors of each ligand. The receptors for the inhibitors are distinctive and there is no cross-reaction between the ligands. It is concluded that ligand binding and subsequent receptor-ligand aggregation must induce a change within the cell-surface membrane, which distorts the distribution and/or affects an optimal conformational aspect of a specific membrane-receptor system for the gamon, a prerequisite for cell pair formation.

Form and pattern in ciliated protozoa: analysis of a genic mutant with altered cell shape in Tetrahymena pyriformis, Syngen 1.

A single cell isolated from the sexual progeny of mutagenized parents gave rise to a clone of cells with an abnormal, conical shape. Breeding analysis revealed that this shape results from the action of a single recessive gene, co (conical). Homozygous mutant cells are shorter and wider than wild type cells, and have their widest point at a more posterior position. Nonetheless, cortical parameters such as number of ciliary rows, number of ciliary units within these rows, and positions of contractile vacuole pores remain essentially unchanged in conical cells, suggesting a considerable degree of mutual independence of pattern and form. Shape changes prior to cell division bring about some convergence in form of dividing conical and wild type cells. However, in conical cells the new oral apparatus and fission line form well posterior to the cell equator, so the opisthes are invariably smaller than proters. Macronuclei nonetheless undergo constriction at the normal central location, and the characteristic inequality in the DNA content of the two macronuclear division products is not increased by the conical condition. Generation times are, on the average, nearly the same in the two wild type daughter cells and in conical proters, while the small conical opisthes have generation times averaging one-third longer. This prolongation explains why population doubling times are always somewhat longer in cultures of conical cells than in parallel cultures of wild type cells. The unusually long generation times of conical opisthes allows for the compensation of initial size differences. Observations on shape changes in conjugating cells of various genotypic combinations suggest that the wild type gene product is not freely exchangeable across the conjugation bridge. The implications of the conical phenotype for problems of cellular patterning and positioning of organelle systems are considered in the discussion.

Infraciliature and myoneme system of Campanella umbellaria (Protozoa, Ciliophora, Peritrichida).

The infraciliature and myoneme system of Campanella umbellaria were revealed using the protargol impregnation technique. The main characteristics of the infraciliature are the peristomial ciliary rows (haplokinety and polykineties), which make four and a half turns around the peristomial disc before plunging into the infundibulum, and the aboral infraciliature, which is made up of the aboral ciliary wreath (trochal band) and the scopula. The myoneme system is composed of: 1) longitudinal fibers, which include 60-84 (mean 72.3) short longitudinal fibers, 40-56 (mean 45.8) medium-length longitudinal fibers, and numerous long longitudinal fibers; and 2) circular fibers, which include 8-12 (mean 9.3) peristomial ring fibers, linking fibers, support fibers, and peristomial disc fibers. The various fibers in C. umbellaria are interconnected to form a single myoneme system that may act as a cell skeleton as well as providing the mechanism by which the zooid contracts and relaxes.

DNA polymerases from a parasitic protozoa Leishmania donovani UR6: evidence of presence of a novel kind of DNA polymerase.

DNA polymerases of Leishmania donovani have been isolated and purified. The cell extract has been chromatographed on a phosphocellulose column that separated into three peaks. The activity peak 1 was further purified to homogeneity. The DNA polymerase is a 64 KDa polypeptide, resistant to N-ethylmaleimide and aphidicolin. It requires MnCl2 and a high concentration of KCl (0.5 M) for maximal activity. It has both 3' to 5' and 5' to 3' exonuclease activities that reside in the same polypeptide.
