[Obtaining iodinated luteinizing hormone while preserving its biological properties].

The author describes a method of obtaining biologically-active 125I-labeled luteinzing hormone in iodation with chloramine T. The hormone and chloramine T concentration ratio of 1:2, and the reaction time of 20 sec was used. LH-125I with the specific activity of 27--30 microCi/microgram was bound with the receptor strictly specifically.

[Practical treatments of suppurative skin mycoses and allergic skin mycoses due to Candida].

In the inflammatory mycoses the author recommends an oral treatment consisting in griseofulvin, and, in case of severe inflammation, prednisone per os at the same time with a local treatment (painting with alcohol iodate 1%, followed by the application of a cream with cortisone associated with an antimicrobial antibiotic and pincer epilation). This way the evaluation is 3-4 weeks shorter and the scars and alopecia are avoided. In the allergic manifestations due to Candida (urticaria, eczema, itching), after discussing their peculiarities the author recommends the desensitization with an extract of Candida cultivated right from patient's primitive focus. This type of desensitization is twice specific: specific anti-Candida and at the same time specific to patient's own strain, even in the case of saprophyte Candida. The very good results obtained after 2-3 months of treatment are presented.

[Anti-arrhythmic effect of adaptative activation of the vagus nerve system and a new synthetic acetylcholine analog].

A higher vagal tone or its stimulation under certain conditions is known to increase the threshold of cardiac fibrillation and even to arrest developing arrhythmias. This effect is usually evaluated as a result of limiting the excessive adrenergic effect on the heart, which is observed in stress and ischemia. The following two facts have been first identified: 1) adaptation to moderate continuous stress may induce tonic excitation of parasympathetic regulation of the heart and enhance its resistance to ischemic and reperfusion arrhythmias; 2) this antiarrhythmic effect is completely reproducible by the recently synthesized acetylcholine analogue EDIHYP, ethyl-3/2-ethyl-2,2-dimethylhydrazinium/propionate iodate.

Studies on retinotoxic potential of a novel antitumor antibiotic--sparsomycin--in rats.

Sparsomycin (Sm) is a potent inhibitor of protein synthesis with an anticancer potential. Two years after its discovery in 1962 a phase I clinical trial revealed serious drug-induced retinotoxicity. The mechanism of this toxicity still remains unresolved; however, its understanding is important for the reintroduction of Sm or one of its analogues in clinical practice. If Sm penetrates the retina, its toxic effect should be seen as inhibition of a protein(s) vital for the visual function. Treatment of healthy albino rats and Royal College of Surgeon (RCS) rats with subtoxic doses of Sm was unable to produce ocular toxic effects. Disruption of the blood-retina barrier with sodium iodate allowed Sm to decrease opsin content by only 27%. These results strongly indicate that Sm might become retinotoxic solely upon extreme conditions such as permeabilization of the blood-retina barrier which may happen only in some rare pathological situations.

Choriocapillaris regeneration in the rabbit. Ultrastructure of new endothelial tube formation.

After experimental destruction of the retinal pigment epithelium (RPE) by intravenous injection of sodium iodate, the rabbit choriocapillaris (CC) atrophies. This report studies the subsequent regeneration of the CC with the goal of identifying sources of new CC and mechanisms by which new endothelial tube is formed. Light and transmission electron microscopic methods were used. The CC appeared to regenerate from three sources: choroidal venules, remnant CC and newly produced cells. Whatever the source, new endothelial tube was formed by extensions of the endothelium that sealed off new lumenal space by junctional complexes. This process was always confined to the tissue space within remnant sleeves of basement membrane left behind by initial atrophy, and which served as a substrate for endothelial movement. The endothelium of new endothelial tube was initially thick and unfenestrated. It became thinner and fenestrated by a process of cavitation, in which cavities continuous with the lumen pushed into the thick endothelial cytoplasm towards its abluminal side. At this site new fenestrae were formed. Fenestrae were initially scattered about the endothelial tube, but with its maturation became localized on the side facing the regenerated RPE.

Electrical responses from detached retina inside the intact rabbit eye.

The local ERG (LERG) from detached retina within the intact rabbit eye has been recorded. Retinal detachments were made in Dutch rabbit eyes by injecting Hanks' balanced salt solution into the subretinal space through a glass micropipette. Electrical responses were recorded with a double-barrelled microelectrode that was advanced through the vitreous until the tip penetrated the retina to enter the subretinal space while the other tip remained in the vitreous. Recordings from detached retina showed an initial fast positive, fast negative and late slow positive wave. The waveform and stimulus response characteristics of these waves (inverted) corresponded to the a-, b-wave and slow PIII respectively. None of the LERG waves was affected by intravenous sodium iodate and only the slow positive wave was extinguished by intravitreal barium chloride. This system for recording the LERG may be useful in investigating physiological changes in detached retina in vivo.

[Does a iatrogenic damage caused by dacryography exist?].

This study was aimed at investigating the changes in corneal epithelium caused by dacryocystography. Such changes can be either chemical--from iodate contrast media (iodized oil and water-soluble non-ionic cm)--or physical--from needle or catheter. Twenty-five patients (50 eyes) were studied using BUT, Schirmer tests, biomicroscopy, and pachymetry. Follow-ups immediately after dacryocystography demonstrated superficial keratitis, which was less severe with water-soluble non-ionic cm. Such lesions were hardly ever observed in patients who had been treated with dacriosol ointment on the cornea just before the X-ray exam. Follow-ups at 24 hours showed the superficial keratitis to have disappeared in all patients, independent of the kind of cm employed and of the ophthalmic ointment applied to the cornea.

Proteolysis in human erythrocytes is triggered only by selected oxidative stressing agents.

Human erythrocytes have been treated with different agents producing oxidative stress. Diamide, tetrathionate, chromate, cystamine and iodate preferentially influenced the cellular redox state by oxidation of free and protein thiol groups leaving the redox state of hemoglobin virtually unaffected. None of these compounds was able to stimulate the proteolysis. By contrast, phenylhydrazine, nitrite, hydrogen peroxide, ter-butylhydroperoxide, cumene hydroperoxide and copper-ascorbate caused a noticeable oxidation of hemoglobin to methemoglobin. These latter agents, except nitrite and copper-ascorbate, triggered proteolysis. Identical results have been obtained in a ghost-free hemolysate. The fraction containing the proteolytic activity was isolated from hemolysate and tested on native or oxidant-treated hemoglobin. The proteolysis was stimulated by all agents able to produce methemoglobin. It is concluded that proteolysis correlated to an unbalance of cellular redox state. The results obtained with isolated and recombined fractions suggests that increased proteolysis does not depend on the removal of the effect of protease(s) inhibitor(s). Since all agents stimulating proteolysis are able to generate free radicals, it seems that protein breakdown is triggered by the direct effect of these intermediates on proteins (mostly hemoglobin) without the involvement of radical species produced in the membranes by action of organic hydroperoxides. In addition, since nitrite and copper-ascorbate, which also oxidize hemoglobin by radical generation, are unable to stimulate proteolysis, it should be concluded that protein degradation induced by oxidative stress is a complex event induced only by specific agents.

Erythrocyte aging: a comparison of model systems for simulating cellular aging in vitro.

Senescent cell antigen (SCANT) is a "neo antigen" that appears on the surface of normal old cells and initiates IgG binding and cellular removal. To investigate the mechanism by which SCANT is generated from its parent molecule, band 3, we subjected intact human erythrocytes to treatments that have been reported to result in changes in band 3 and/or to mimick aging in vitro. The validity of these treatments as model systems for erythrocyte aging was evaluated using a "red cell aging panel" that provides a biochemical profile of a senescent red cell. Treatments were assessed for their ability to induce in vitro the following changes observed in normal erythrocytes aged in vivo: 1 increased breakdown of band 3 as detected by immunoblotting, 2 decrease in anion transport efficiency as detected with a sulfate self-exchange assay, 3 decrease in total glyceraldehyde 3-phosphate dehydrogenase activity with an increase in membrane-bound activity, and 4 increase in the binding of autologous IgG as detected with a protein A binding assay. Neither incubation with the free radical-generating xanthine oxidase/xanthine system, nor treatment with malondialdehyde, and end product of free radical-initiated lipid (per)oxidation, results in age-specific changes. Loading of the cells with calcium and oxidation with iodate results in increased breakdown of band 3, but does not lead to increased binding of autologous IgG. Only erythrocytes that have been stored for 3-4 weeks show the same structural and functional changes as observed during aging in vivo.

[Determination of americium-241 in urine].

A technique has been developed for the determination of americium 241 in urine by a radiochemical purification of the nuclide from uranium (upon co-precipitation of americium 241 with calcium and lanthanum), plutonium, thorium, and polonium 210 (upon co-precipitation of these radionuclides with zirconium iodate). alpha-Radioactivity was measured either in a thick layer of the americium 241 precipitate with a nonisotope carrier or in thin-layer preparations after electrolytic precipitation of americium 241 on a cathode.

[Alimentary iodine deficiency in East Germany and its reflection in thyroid parameters following the initiation of struma prevention].

By regional and countrywide studies carried out in the past 10 years the occurrence of endemic goitre and iodine deficiency (ID) of grade II has been demonstrated in 1981. In 1983 till 1985 an iodized salt (25 mg KI/kg) was offered mainly in the southern counties of the G.D.R. This measure has been followed by the production of 84% of the whole paket salt iodized with stable potassium iodate (32 mg KIO3/kg) since July 1985 and delivered in all the G.D.R. Nevertheless, in the end of 1985 the persistence of iodine deficiency of grade II has yet to be stated by low renal iodine excretion, low iodine content of milk and human milk post partum. The reference values of TT3 (0.84-3.76 nmol/l) and TT4 (41.8-137.3 nmol/l) assessed in 1985 for the G.D.R. are to be interpreted before the background of an iodine deficiency of grade II.

Effects of intraocular pressure and other factors on subretinal fluid resorption.

We observed the effects of intraocular pressure (IOP), vitreous pressure and gravity on the resorption of small retinal detachments (blebs) made with Hanks' solution or autologous serum. Raising the IOP decreased the absorption time moderately and lowering the IOP increased it. These effects were greater when the RPE had been damaged by sodium iodate or laser burns, but we conclude that IOP makes only a small, limited contribution to normal subretinal fluid absorption. Neither liquefaction of the vitreous nor retinal weight had a significant influence on fluid absorption.

Radioactive iodine exchange reaction of HIPDM: kinetics and mechanism.

In conjunction with single photon emission computed tomography (SPECT), iodine-123 (123I)-labeled N,N,N'-trimethyl-[2-hydroxy-3-methyl-5-iodobenzyl]-1,3-propanediamine (HIPDM) has been used clinically as a regional cerebral perfusion imaging agent. The [123I]HIPDM can be prepared by a simple aqueous exchange reaction in a kit form. We synthesized unlabeled HIPDM by condensation of 2-hydroxy-3-methyl-5-iodobezaldehyde and N,N,N'-trimethyl-1,3-propanediamine, followed by a sodium borohydride reduction reaction. The kinetics of the radioactive iodine exchange reaction for the preparation of [123I]HIDM is controlled by the pH, the temperature, and the presence of reductant (sodium bisulfite), and oxidant (sodium iodate). The reaction is a second order iodine-iodine exchange with an activation energy of 30.6 kcal/mole. The mechanism of this reaction probably involves the formation of an active 1+ or iodine free radical, which is sensitive to the presence of a reductant, such as sodium bisulfite.

[Use of angiopneumography with Tc-99M in 2 cases of pneumopathy caused by anatomical defect].

A parallel study, using traditional angiopneumographic technique with iodate contrast medium and the radioisotopic technique employing 99MTc has been carried out in some cases of bronchopulmonary malformations due to anatomic defect. The purpose of this is to assess the angiographic performance at pulmonary circulation level of an extremely simple radioisotopic technique. Analysis of the two cases presented suggests that although radioisotopic angiopneumography presents lower resolution capacity for viewing small and medium lumen branches of the pulmonary artery, it has the advantage of associating the results of traditional angiopneumography with statis pulmonary scintigraphy in a single exploration. It is considered that this investigation can be used particularly in those patients with severe cardiorespiratory insufficiency, for whom the invasive technique presents some by no means inconsiderable risks.

[Behavior of blood amylase and plasma deoxyribonuclease I in dogs subjected to intra-operative cholangiography with the use of B 10610].

An experimental study was conducted on the use of a new iodate-molecule, B 10610, synthesized in the Bracco research Laboratory, for intra-operative cholangiographic diagnostics. For the purpose of evaluating the possible rise and the extent of any pancreatic damage in animals subjected to intra-operative radiological exploration of the biliary ducts with the use of the said substance, the blood levels of amylase and plasmatic 1st deoxyribonuclease were assayed after various time intervals, such tests being particularly specific in the humoral diagnostics of both acute and chronic pancreatic diseases. On the basis of the tests made, it is concluded that the introduction of this new dye does not cause irreversible lesions of the pancreas, but only purely functional and transient alterations.

[The value of the basophil degranulation test on the study of hypersensitivity reactions to iodinated contrast media].

The ability of basophils and tissue mastocytes activated by sensitised IgE antibodies to release mediators in the presence of the specific antigen underlies immediate hypersensitivity reactions. This phenomenon can be explored by the in vitro basophil degranulation test adopted not only in experimental pathology, but in human allergological pathology and in the study of post-streptococcal and lupus glomerulonephritis. The test has been carried out to analyse sensitisation to iodate contrast media in 71 patients already submitted to contrastography, 37 of whom had given evidence of allergic reactions. All patients with a previous history of sensitisation presented, in vitro, a positive reaction (56 + 15.3% degranulation) in the presence of the contrast medium. Against this, none of the patients with negative test presented an allergic reaction during contrastography. Stress is laid on the practical importance of the test prior to performance of contrastography so as to predict possible sensitisation and select the most suitable medium.

A histochemical study on the adrenal components of the teleost Cirrhinus mrigala (Ham.).

The adrenal components of C. mrigala are embedded in the pronephric cephalic kidney around the post cardinal vein. The cortical cells responded positively to the lipids, ascorbic acid, delta 5-3 beta-HSD, G-6-PD, MAO, acid and alkaline phosphatase tests. The presence of intense MAO activity may suggest the possible involvement of monoamines in the adrenocortical function. Localization of lipids and delta 5-3 beta-HSD show the sites of corticosteroid synthesis. In the chromaffin cells, MAO, acid and alkaline phosphatase activity was moderate whereas they gave a strong reaction to ascorbic acid test in comparison to the cortical cells. Noradrenaline (NA) and adrenaline (A) storing cells were differentiated adopting glutaraldehyde silver, dichromate and iodate techniques. NA and A storing cells are almost totally depleted of their contents after reserpine treatment. The histochemical response of the adrenal gland of this species is largely comparable to that of higher vertebrates.
