Fluorometric measurement of glycosylated albumin in human serum.

A simple and rapid fluorometric assay of glycosylated albumin was developed using dansylated phenyl boronic acid (N-(5-dimethyl amino-1-naphthalene sulfonyl)-3-aminobenzene boronic acid). The emission spectrum of the reagent changes upon binding to cis-diols present on glycosylated albumin in serum, allowing quantitation of glycosylated albumin. Results from the new procedure correlated well with those from commercially prepared affinity chromatography (r = 0.85).

Measurement of 5-aminolaevulinic acid by reversed phase HPLC and fluorescence detection.

A new method for sensitive measurement of delta-aminolaevulinic acid (ALA) in biological material is described. ALA is derivatized with dansyl chloride, separated by HPLC and estimated using a fluorescence detector. The pretreatment of biological samples includes desamination of L-alpha-aminoacids with L-aminoacid-oxidase before dansylation. The sensitivity of the method is slightly below 1 pmol/injection for standards and the lower limit of quantification is 0.1 mumol/l for plasma and 10 nmol/l for cerebrospinal fluid. Reference values in plasma are 3.53 +/- 1.75 (SD) (n = 43) mumol/l and in packed erythrocytes they ranged from 6 to 26 mumol/l (mean: 14.0 +/- 5.5 mumol/l). In cerebrospinal fluid of non-porphyric individuals less than 2 nmol/l were recovered.

Distribution and occurrence of amphetamine and p-hydroxyamphetamine in tissues of the rat after injection of d-amphetamine sulfate.

The distribution of amphetamine and the formation and distribution of its principal metabolite p-hydroxyamphetamine after the injection of relatively small amounts of d-amphetamine was examined in the rat. Amphetamine entered all organs readily and was rapidly eliminated. p-Hydroxyamphetamine was rapidly synthesised and it is suggested that substrate inhibition of hepatic hydroxylating enzymes occurs. In the brain, amphetamine was homogeneously distributed between the seven brain regions examined whereas p-hydroxyamphetamine was predominantly concentrated in the striatum.

A role for endothelin ETA receptors in regulation of renal function in spontaneously hypertensive rats.

While there is evidence to suggest that endothelin-1 is involved in regulation of kidney function and blood pressure, the importance of endothelin ETA receptors in this area has not been clearly defined. The novel, non-peptide endothelin ETA receptor antagonist, BMS-182874, (5-(dimethylamino)-N-(3,4- dimethyl-5-isoxazolyl)-1-naphthalene sulfonamide) was used to examine effects of endothelin ETA receptor blockade on renal function in spontaneously hypertensive rats. Preliminary studies were conducted to determine an effective dose of BMS-182874. Infusion of BMS-182874 (10 mumol/kg/min, i.v.) inhibited effects of exogenous endothelin-1 on glomerular filtration rate, renal blood flow, and mean arterial pressure in Sprague-Dawley rats. Administration of BMS-182874 (10 mumol/kg/min, i.v.) to anesthetized, male, spontaneously hypertensive rats decreased renal blood flow by approximately 50% (1.2 +/- 0.11 ml/min/100 g body weight) compared to vehicle (2.7 +/- 0.23). There was no effect of BMS-182874 on glomerular filtration rate (0.5 +/- 0.05 ml/min/100 g body weight; vehicle: 0.7 +/- 0.06). Mean arterial pressure decreased significantly after BMS-182874 (123 +/- 3.8 mm Hg; vehicle: 162 +/- 4.8). Urine flow and renal vascular resistance were unchanged by BMS-182874. Endothelin ETA receptor density was increased approximately 50% in spontaneously hypertensive rat kidneys compared to normotensive kidneys, with no change in equilibrium dissociation constant. Endothelin ETB receptor density and equilibrium dissociation constant were similar in the two rat strains. Plasma immunoreactive endothelin was higher in hypertensive (5.9 +/- 0.31 fmol/ml) than normotensive rats (2.8 +/- 0.15). The results suggest endothelin ETA receptors may play a role in the regulation of renal function in this model of hypertension.

Cytoplasmic lipid droplets as the possible eventual cellular fate of active forms of cyclosporin.

The activity of cyclosporins can be defined with regard to their ability to inhibit the proliferation of susceptible lymphoblastoid cell clones. All active cyclosporins cause the emergence of highly refringent globular bodies, independently of cell susceptibility to cyclosporin. Cyclosporins devoid of proliferation-inhibition capability do not cause such alterations in cell morphology. The use of dansylated active cyclosporins suggests that within a few hours of treatment in vitro, most if not all cyclosporin is contained within those globular bodies. By using cytochemical methods allowing differential staining, we show here that these cyclosporin-containing structures are neither normal mitochondria (shown by use of rhodamine 123) nor normal lysosomes (shown by use of acridine orange under stringent staining conditions) but most probably lipid droplets (shown by use of perylene and various dansylated hydrophobic probes).
