[Effects of baicalin on hepatocyte apoptosis induced by TNF-alpha and act D in rats].

OBJECTIVE: To study the effects of baicalin on rat hepatocyte apoptosis induced by tumor necrosis factora (TNF-alpha) and actinomycin D (Act D) in vitro. METHODS: Hepatocytes were cultured with different concentrations of baicalin and the cell apoptosis was induced by TNF-alpha and Act D in vitro. After culturing for 24 h, the activity of hepatocysts was examined by MTT assay and the function of hepatocytes for secreting albumin was detected by bromocresolum Viride method. Agarose gelelectrophoresis and flow cytometric analysis were used to determine the apoptotic cells. RESULTS: The actively of the hepatocyte (A) and content of albumin (ALB) in the supernatant of cultured cells in the groups of 0.2 microgram.ml-1 and 2.0 micrograms.ml-1 of baicaline were significantly higher than those in apoptotic model group (A: P < 0.01; ALB: P < 0.01); respectively Even the ALB value in the group of 0.2 microgram.ml-1 baicalin was higher that of black control group (P < 0.01); The clear "ladder-like" streak-band of DNA fragments was found only in the apoptosis model group by the agarose gel electrophoresis. The flow cytometry showed that all the apoptosis rates in the three baicalin groups were lower than that of the apoptotic model group (P < 0.01). CONCLUSION: Baicaline can restrain rat hepatocyte apoptosis induced by TNF-alpha and ActD in vitro.

[The effect of salvia miltiorrhiza and shengmai on inflammatory mediator and renal function of post-operative patients with obstructive jaundice].

OBJECTIVE: To investigate the effect of salvia miltiorrhiza and shengmai (SS) on inflammatory mediator and renal function in patients with obstructive jaundice (OJ). METHODS: This study included three groups: the OJ patients receiving SS for 6 days after operation (the SS group, n = 15); the OJ patients not receiving SS (the OJ group, n = 15); the patients with other hepato-biliary disorders (the Non-OJ group, n = 15). The levels of ET, LPs, TNF-alpha, IL-6, IL-8 and urine RBP, TFR, ALB were measured one day before operation and 1, 4, 7 days after operation. RESULTS: The SS group's LPs, TNF-alpha, IL-6, IL-8, ET, RBP, TFR and ALB levels were obviously lower on Day 7 after operation as compared with those on Day 1 after operation (P < 0.05). The urine RBP and ALB levels were significantly lower in the SS group than in the OJ group (P < 0.05). CONCLUSION: The post-operative use of salvia miltiorrhiza and shengmai for patients with obstructive jaundice may effectively decrease the postoperative levels of plasma LPs, ET, TNF-alpha, IL-6 and IL-8. At the same time, salvia miltiorrhiza and shengmai may protect renal function by inhibiting inflammatory mediator and improving blood dynamics.

[Comparison of different lysis buffers in the assessment of the efficiency of single cell polymerase chain reaction for preimplantation genetic diagnosis].

OBJECTIVE: To set up a highly efficient cell lysis system to increase the efficiency of single cell polymerase chain reaction (PCR) for preimplantation genetic diagnosis (PGD). METHODS: First we picked up one single lymphocyte or blastomere and lysed it in alkaline lysis buffer (ALB), proteinase K buffer (PK) and a new proteinase K lysis buffer compounded by ourselves. Then we amplified the 3rd exon of Gene ALD using nested PCR and compared the amplification efficiencies. RESULTS: The amplification efficiencies of the single lymphocyte in ALB, PK and the lysis buffer compounded by ourselves were 90%, 74%, and 87%; those of the single blastomere were 98%, 77%, and 96%. CONCLUSION: ALB and the lysis buffer compounded by ourselves are superior to PK in the lysis of cells in clinical PGD.

[Inflammatory mediator in patients with obstructive jaundice and its effect on renal function].

OBJECTIVE: To investigate the effect of inflammatory transmitter on renal function in obstructive jaundice. METHODS: 15 patients with obstructive jaundice (OJ) as the experimental group and 15 non-joundiced patients as the control group were involved in this study. The plasma ET, LPS, TNF-alpha, IL-6, IL-8 and urine REP, TFR, ALB were measured in 15 patients with OJ before and 1, 4, 7 days after surgery and in 15 patients without jaundice as controls. RESULTS: The levels of plasma ET, LPS, TNF-alpha, IL-6, IL-8 and urine RBP, TFR, ALB increased significantly at multiple time points as compared with those in the non-OJ patients (P < 0.05), and the changes of plasma ET, LPS, TNF-alpha, IL-6, IL-8 are the same as those of urine RBP, TFR, ALB. CONCLUSION: These data suggest that there is interaction between ET, LPS, TNF-alpha, IL-6, IL-8 and the damage to renal function with OJ after surgery.

Effects of mebendazole, albendazole, and praziquantel on alkaline phosphatase, acid phosphatase, and adenosine triphosphatase of Echinococcus granulosus cysts harbored in mice.

Mice infected with protoscoleces of Echinococcus granulosus for 12-14 months were treated ig with mebendazole (Meb) 25-50 mg.kg-1 x d-1 for 7-14 d, albendazole (Alb) 200 mg.kg-1 x d-1, cr praziquantel (Pra) 500 mg.kg-1 x d-1 for 14 d. The mice were killed 24 h after the last medication, and acid phosphatase (ACP), alkaline phosphatase (AKP), and adenosine triphosphatase (ATPase) including (Na, K, Mg)-ATPase, (Na, K)-ATPase, and (Mg)-ATPase were determined and compared with those of untreated control group. The results showed that ACP activities of cyst wall in treated groups were lower than the control group. Whereas AKP activity of cyst wall in Pra group increased markedly, this is not the case in Meb and Alb groups. Three ATPase activities of cyst wall were inhibited in both Meb and Alb groups, Meb being more potent. No apparent changes in the ATPase activities were seen in Pra group.

Metabolic effects of a novel bioartificial liver on serum from severe hepatitis patients: an in vitro study.

OBJECTIVE: To establish a novel bioartificial liver (BAL) consisting of spheroids of porcine hepatocytes in a hollow-fiber bioreactor, and to perform an in vitro study on its metabolic effects on the serum from severe hepatitis B patients. METHODS: Hepatocytes were isolated from pup pigs and cultured as aggregate spheroids through rotation and vibration. Phase-contrast microscopy, transmission electron microscopy, and scanning electron microscopy were used for morphological detection of hepatocyte spheroids. The hepatocyte spheroids were then transferred into the shell of a polysulfone hollow-fiber bioreactor, creating a novel BAL. Diluted serum samples of severe hepatitis B patients were circulated for 3 hours each into the bioreactor, by using an extracorporeal circulatory system. Every half hour, including both before and after perfusion, serum samples were collected to assay total bilirubin (TBIL), total protein (TP), albumin (ALB), and globulin (GLB) concentrations in order to judge the metabolic effects of this novel BAL. RESULTS: Most hepatocytes had formed spheroids with high viability after 24 hours in culture. After 3 hours of perfusion, when compared with the control group, the serum concentration of TBIL in the treatment group decreased significantly (P < 0.01), but the serum concentrations of TP and ALB increased significantly (P < 0.05). CONCLUSIONS: Hepatocytes can be conveniently cultured as aggregate spheroids through a rotation and vibration method. The novel BAL is efficient in removing bilirubin from the serum of severe hepatitis B patients, and in supplying the serum with ALB. Thus, the BAL might provide effective therapy for patients with severe hepatitis B.

[Effects of pregnancy and blood components on solubility of fluoride volatile anesthesia].

OBJECTIVE: To determine the effect of pregnancy on blood/gas partition coefficient (B/G) of volatile anesthetics and to evaluate the relation between B/G and blood component concentration. METHODS: Blood samples were obtained from four groups of fasting women: 8 nonpregnancies, 8 first-trimester pregnancies, 12 mid-trimester pregnancies, 12 third-trimester pregnancies. B/G of desflurane, isoflurane and halothane in each subject was measured by double equilibration technique. Red blood cell (RBC), hemoglobin concentration (Hb), hematocrit (HCT) and the serum concentration of cholesterol (Chol), triglyceride (TG), albumen (Alb), globulin (Glob) were measured. RESULTS: 1. There was no significant difference in B/G of desfluraneand and in B/G of halothane among the four groups. There was significant difference in B/G of isoflurane; the lowest B/G was in the mid-trimester and the highest B/G was in the third trimester of pregnancy. The trend direction of halothane among four groups was similar to that of isoflurane, although no significant difference was observed therein. 2. RBC, Hb and HCT showed statistically significant difference among the four groups, which decreased to the lowest level in the mid-trimester and came up slightly in the third trimester. There was statistically significant difference in Alb, A/G, TG and Chol among the four groups. Alb and A/G were the lowest and TG, Chol were the highest in the third-trimester of pregnancy. 3. There was a correlation between the serum TG and the B/G of halothane. The B/G of isoflurane correlated directly with serum TG and Hb. The B/G of desflurane correlated directly with Chol. CONCLUSION: The above findings indicated that pregnancy brought on alterations in the blood components and serum components, thus inducing the difference in B/G between the flaoride volatile anesthetics. During pregnancy, the B/G of isoflurane altered significantly more than that of halothane did, whereas no significant alteration in the B/G of desflurane was noted.

[A transgenic mouse that targets the expression of Cre recombinase in hepatocyte].

OBJECTIVE: To construct a mouse that specifically expresses Cre recombinase in hepatocyte. METHODS: A hepatocyte specific transgenic construct containing mouse albumin promoter, the Cre recombinase gene and the poly (A) of human growth factor gene was generated. The linearized constructs were introduced into the fertilized eggs by microinjection to obtain the transgenic mice. The transcriptional specificity of Cre recombinase was detected by reverse transcription polymerase chain reaction (RT-PCR). The expression and function of Cre recombinase were detected by PCR and Southern Blot after crossing the Alb-Cre transgenic mice with the Smad4 conditional knockout mice. RESULTS: The linearized constructs were microinjected into 837 fertilized eggs, and then the 797 effective eggs of microinjected eggs were implanted into the oviducts of 27 pseudo pregnant mice. In the 53 offspring, there were 6 mice carrying the transgene identified by polymerase chain reaction (PCR) and Southern Blot. Cre recombinase transcripts were detected in the livers and testis of the Alb-Cre transgenic mice using RT-PCR. The Cre recombinase was expressed in the livers of the double heterozygous for Alb-Cre and Smad4 floxed allele, and the exon 8 floxed by loxP site was deleted. CONCLUSION: A hepatocyte-specific Cre transgenic mouse was generated successfully. The Cre recombinase expressed specifically in liver and could mediate the recombination between loxP sites in vivo.

[Comparison of serum biochemical features between SARS and other viral pneumonias].

OBJECTIVE: To identify blood chemistry changes in coronavirus of severe acute respiratory syndrome (SARS). METHODS: Biochemical changes in SARS patients were summarized and compared with other viral pneumonias. Serum total protein (TP), albumin (Alb), total cholesterol (TC), triglyceride (TG), calcium (Ca), ferrum (Fe), lactate dehydrogenase (LDH), creatine kinase (CK), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) between SARS and other viral pneumonias were examined by Roche Diagnostics assay, HITACHI7600 automatic analyzer. Clinically confirmed SARS patients, patients with other viral pneumonias, and healthy controls were included in the study. RESULTS: Compared with healthy person, the levels of serum TC, Fe, Ca, Alb were significantly lowered (P<0.05 or P<0.01), while the activity of LDH, CK, ALT, AST were elevated, the increase of CK and the decrease of Fe were the most significant (P<0.05 or P<0.01), the changes of TP and TG were not obvious. In the other viral pneumonias patients, ALT, AST, LDH were elevated slightly than those of healthy person, while Fe, Ca, Alb, TC, CK were a little reduced, but there was no significant difference between the two groups. In convalescent stage, all the tests were returned to normal ranges except ALT, AST were still elevated in SARS patients. CONCLUSION: The changes in serum biochemistry are more marked in SARS patients compared with patients suffering from other viral pneumonias, the decrease of Fe as well as the inhibition of TC may be caused by the treatment of anti-virus.

[Population analysis of electrophoretic variation in blood serum albumins of European (Acipencer ruthensis L.) and Siberian (A. ruthensis marsiglii Brandt) sterlet].

We studied blood serum albumins in European (Acipencer ruthensis L.) and Siberian (A. ruthensis marsiglii Brandt) sterlet using disk electrophoresis in polyacrylamide gel. The albumins were shown to be controlled by three codominant alleles of a single locus (ALB*a, b, c). In European sterlet, all three theoretically possible genotypes were described, one of which (ALB*c/c) occurred extremely rarely (one individual). Siberian sterlet was found to be monomorphic for albumins: all fish examined had the ALB*a/a genotype. There was no correlation between albumin patterns and fish fatness. In a number of samples from the Volga River basin, spatial and temporal differentiation was found and analyzed. The results suggest that construction of hydroelectric plants may provoke massive and prolonged starlet migrations.

Differentiation of hepatocytes from mouse embryonic stem cells and its significance.

BACKGROUND: There have been lots of important discoveries in the research on embryonic stem (ES) cells; but few studies have been involving in the differentiation and differentiation ratio of hepatic stem cells. To provide a new source of hepatocytes in hepatocytes transplantation and liver tissue engineering, we investigated the differentiation and differentiation ratio of hepatocytes in the ES cells differentiating system in a certain length of time. METHODS: BALB/c ES cells from mice were cultured for differentiation in medium without leukemia inhibitory factor (LIF). During the period of differentiation of ES cells, dual immunofluorescence was performed to analyze the expression of hepatic proteins in cytoplasm such as alpha-fetoprotein (AFP) and albumin (ALB). The count of relative hepatocyte-like cells was analyzed by flow cytometry (FCM) and the hepatic differentiation ratio was obtained. RESULTS: The expression of AFP was first determined at day 7 and that of ALB at day 13. The expressions of AFP and ALB became stronger with the extention of the differentiation time. The hepatic differentiation ratio at day 13 and 21 was 5.5% and 10.4%, respectively. CONCLUSION: ES cells could differentiate into hepatocytes, which should be a new source of hepatocytes in transplantation of hepatocytes and liver engineering.

Tolerance and efficacy of combined diethylcarbamazine and albendazole for treatment of Wuchereria bancrofti and intestinal helminth infections in Haitian children.

This randomized, placebo-controlled trial investigated the tolerance, efficacy, and nutritional benefit of combining chemotherapeutic treatment of intestinal helminths and lymphatic filariasis. Children were infected with Ascaris (30.7%), Trichuris (53.4%), and hookworm (9.7%) with 69.9% having more than one of these parasites. A total of 15.8% of the children had Wuchereria bancrofti microfilariae. Children were randomly assigned treatment with placebo, albendazole (ALB), diethylcarbamazine (DEC), or combined therapy. The combination of DEC/ALB reduced microfilarial density compared with placebo, ALB, or DEC (P < or = 0.03). Albendazole and DEC/ALB reduced the prevalence of Ascaris, Trichuris, and hookworm more than placebo or DEC (P < or = 0.03). Among Trichuris-infected children, those receiving ALB and DEC/ALB demonstrated greater gains in weight compared with placebo (P < or = 0.05). Albendazole and DEC/ALB were equally efficacious in treating intestinal helminths and for children with W. bancrofti microfilaremia, DEC/ALB was more effective than DEC, with no increase in severity of adverse reactions.

Human interleukin-1 alpha crosses the blood-testis barriers of the mouse.

Interleukin-1 alpha (IL-1 alpha) has been shown to have direct effects on the gonads, affecting steroidal secretion, DNA synthesis by spermatogonia, and the immune function of the testes. It is unclear, however, how IL-1 alpha exerts these effects because the testis is partitioned into basal and adluminal compartments by both a vascular and a Sertoli cell barrier. The authors used a highly sensitive method to quantify the unidirectional flux rates (Ki) into the testis of technetium pertechnetate-labeled human albumin (T-alb), a compound that does not readily cross the vascular barrier, and human IL-1 alpha radioactively labeled with 125I (I-IL). The entry rate (Ki) was almost six times greater for I-IL than for T-alb. Part of the enhanced entry of I-IL was due to a saturable transport system. Nearly 0.2% of the total injection had entered the testes 60 minutes after intravenous administration, and more than 75% of that amount was not accounted for by the albumin space. Collection of testicular interstitial fluid from the basal compartment and seminiferous tubule fluid from the adluminal compartment showed preferential entry of I-IL into these compartments. Analysis by high-pressure liquid chromatography or radioactivity recovered from the testis showed that intact I-IL was entering the testis. The leakiness of the blood-testis barrier was measured by the rate of entry for T-alb, which was not altered by injection of unlabeled human IL-1 alpha in doses of up to 50 micrograms/kg (5 x 10(6) U/kg), and by the wet weight of the testes. The results show that circulating IL-1 alpha can have direct access to the testis, supporting previous studies suggesting a direct effect of IL-1 alpha on gonadal function.

Effect of aminoguanidine and albendazole on inducible nitric oxide synthase (iNOS) activity in T. spiralis-infected mice muscles.

The aim of this study was to provide evidence for the expression of iNOS in the cells of inflammatory infiltrates around larvae in skeletal muscles of T. spiralis infected mice. The BALB/c mice (n = 8) divided into subgroups, received either aminoguanidine (AMG)--a specific iNOS inhibitor or albendazole (ALB)--an antiparasitic drug of choice in trichinellosis treatment. Control animals (n = 2 in each subgroup) were either uninfected and treated or uninfected and untreated. Frozen sections of hind leg muscles from mice sacrificed at various time intervals after infection were cut and subjected to immunohistochemistry, using monoclonal anti-iNOS antibody. The ALB-treated mice revealed stronger iNOS staining in the infiltrating cells around larvae than the infected and untreated animals. On the contrary, in the AMG-treated animals, the infiltrating cells did not show any specific iNOS reaction. These data confirm the specificity of iNOS staining in the cellular infiltrates around T. spiralis larvae and shed some light on the role of nitric oxide during ALB treatment in experimental trichinellosis.

[The research on the relationship between the polymorphism of 1082A/G, anti-inflammatory interleukin-10 gene promoter with its effect of preventing ESRD patients from microinflammation and arteriosclerosis].

OBJECTIVE: We do this investigation in order to reveal the relationship between the polymorphism of 1082A/G, anti-inflammatory interleukin-10 gene promoter, and end stage renal disease (ESRD) patients microinflammatory state and arteriosclerosis (AS). METHODS: We used PCR-RFLP to measure the various kinds of distribution of IL-10 gene-1082A/G genotype and relevant indexes of microinflammatory state and AS of 870 ESRD patients and 1000 healthy persons of control group and to analyze the mechanism of its protection effect keeping ESRD patients away from microinflammation and arteriosclerosis. RESULTS: Compared with the control group, CRP, TNF-alpha, CH50, C3, IL-10 and Alb of ESRD group were in the normal range, but still significantly higher than those of the control group, while IL-10, Alb were significant lower (P < 0.05). The genotype distribution and allele frequency of IL-10A/G gene had no significant differences between the healthy group and the control group (P > 0.05). Levels of CRP, TNF-alpha, CH50 and C3 of ESRD patients with IL-10A/A genotype were significantly higher than those of ESRD patients with G/G and G/A genotype (P < 0.05), while IL-10 and Alb were significantly lower (P < 0.01). The production of IL-10 in serum from patients with IL-10A/A genotype was significantly lower than that of patients with G/G and G/A genotype (P < 0.01). The incidence rate of AS of patients with IL-10-1082A/A genotype was significantly higher than that of patients with G/G and G/A genotype (P < 0.01). The raise of AS incidence rate was correspondent with the decline of serum IL-10 and raise of serum CRP and Fib. CONCLUSION: The IL-10A/A genotype is a predictable factor of microinflammatory state and high AS incidence rate in ESRD patients. We use IL-10G/G genotype to modulate the high production of serum IL-10, to decline inflammatory reaction and to keep away from microinflammation and AS in ESRD patients. We should work hard on improving the dialysis membrane to reduce the anti-inflammatory factors in uremia for chronic renal failure patients with high arteriosclerosis risk.

[Effects on glutathione of patients received glutamine dipeptide enriched parenteral nutrition post abdominal surgery].

OBJECTIVE: To observe the effects of glutamine (GLN) enriched parenteral nutrition (PN) on glutathione level, immune function, liver function, infectious complication and hospital stay of patients undergone abdominal operation. METHODS: Forty patients with abdominal surgical disease undergoing elective surgical treatment were randomly divided into two groups (study and control group). All patients received total parenteral nutrition (TPN) for up to 7 days perioperatively. The control group received TPN while the study group received TPN including GLN dipeptide. Both groups received isonitrogen and isocaloric nutrition. Nonprotein caloric intake is 83.6 kJ.kg(-1).day(-1), glycogen: lipid ratio is 7:5, nitrogen intake is 0.16 g.kg(-1).d(-1), calories:nitrogen ratio is 489 kJ:1 g. RESULTS: The decreasing degree of glutathione levels of plasma and red blood cell in study group is lower than that in control group postoperatively. GSH/GSSG ratio of plasma in study group is higher than that in control group on 3rd day postoperatively (53 +/- 11 vs 31 +/- 7, F = 4.725, P = 0.001). Albumin (ALB) of both groups decreased slightly postoperatively. ALB level of study group was higher than that of control group on 3rd day postoperatively [(3.8 +/- 0.4) g.dl(-1) vs (3.4 +/- 0.4) g.dl(-1), F = 2.128, P = 0.02]. No significant differences were noted between immunoglobin (IgG, IgM and IgA) and T lymphocyte subgroup (CD4, CD8, CD4/CD8) levels of both groups postoperatively. There was no infectious complication in study group, while one in control group. There was shorten trend of hospital stay in study group compared with control group [(22.3 +/- 2.1) d vs (24.9 +/- 1.7) d, t = -0.935, P = 0.32]. CONCLUSIONS: Glutamine enriched PN has the trend of maintaining glutathione level of plasma and red blood cell in patients having undergone abdominal operation, and can sustain GSH/GSSG ratio and ALB level, protect antioxidant ability, promote liver function postoperatively. Glutamine enriched PN has the trend of shortening hospital stay, decreasing incidence of infectious complication.

Albumin binding sites are expressed on the abluminal plasma membrane of capillary endothelium.

The presence of albumin binding sites on the abluminal front of vascular endothelium was examined on the capillaries of the adipose tissue. The experimental procedure consisted in injecting interstitially within the rat epididymal fat and epicardial fat, albumin (alone or bearing oleic acid) either conjugated with gold particles (Alb-Au or Alb-OA-Au) or radioiodinated [( 125I]-Alb). In controls, polyethyleneglycol-gold complex (PEG-Au) and [125I]-IgG were used as tracers. The results revealed that: a) albumin binding sites are expressed on the abluminal front of endothelium especially concentrated on plasmalemmal vesicles; b) the retrotranscytosis of albumin conjugates from the interstitium to the capillary lumen is a poorly represented process; c) the binding of the tracers used appears to be time and concentration dependent; d) albumin conjugates do not bind significantly to plasmalemmal vesicles of adipocytes, pericytes and smooth muscle cells; e) PEG-Au and [125I]-IgG do not show a binding pattern similar to that of albumin conjugates.

Changes in salivary components by drug administration in patients with heart diseases.

In this study, patients with heart diseases were classified into 2 groups: Warfarin user and Warfarin non-user, and six salivary components were determined to assess intraoral pathologic conditions. Groups of healthy subjects and patients with periodontal disease without receiving any medication were set as control groups, and they were compared with those of the 2 groups with heart diseases. In patients with heart diseases in both the groups, albumin (ALB) level was found to be significantly higher compared to that in the control groups, and it was significantly higher in the patient group receiving Warfarin user and Warfarin non-user compared to that in the patient group with periodontal disease. C-reactive protein (CRP) levels were found to be higher in both the groups with heart diseases than those in the healthy group. Correlations between various salivary components and the clinical parameters were examined, showing significant correlations between ALB and gingival index (GI) and clinical attachment level (CAL), and between alanine aminotransferase (ALT) and GI, probing depth (PlI), bleeding on probing (BOP) and CAL. Significant correlations were also found between creatine kinase (CK) and PlI, GI and BOP. Thus, it was suggested that ALB and CRP might serve as the markers of intraoral pathologic conditions, and CK and ALT might serve as those alternative to GI.
