Continuous subtherapeutic insulin counteracts hypothalamopituitary-gonadal alterations in diabetic rats.

In experimental animal models, gonadal axis lesions are probably responsible for reproductive disorders associated with diabetes mellitus. The pathogenesis of these disorders is not yet known, but it is assumed that insulin deficiency plays an important role. To check this hypothesis, we have investigated the hypothalamopituitary-gonadal axis of insulin-treated streptozocin-induced diabetic (STZ-D) rats and compared it with that of untreated diabetic and control animals. Insulin was delivered by subcutaneously implanted osmotic minipumps. Furthermore, to determine whether possible beneficial insulin effects are selectively limited to the gonadal axis or act generally, we also studied retinal microangiopathy. The hypothalamopituitary-gonadal axis of insulin-treated diabetic animals was almost unchanged. On the contrary, retinal microangiopathy was only slightly influenced by subtherapeutic insulin doses. In conclusion, continuous administration of insulin at subtherapeutic doses can successfully counteract most of the effects of diabetes on the gonadal axis. Thus, the gonadal-axis impairment in STZ-D animals appears to be related to the fall of plasma insulin below a critical level. Furthermore, the various organ systems may respond to different plasma insulin threshold levels.

Electrophoretic and gel chromatographic analyses of follicle-stimulating hormone in human serum.

The pleomorphism of follicle-stimulating hormone (FSH) in human serum was investigated by gel chromatography on Sephadex G-200 and zone electrophoresis in agarose suspension. The FSH activity was measured by radioimmunoassay. By gel chromatography FSH in sera from 3 males and 5 females of various ages was eluted within a narrow range after albumin and no sex hormone related pleomorphism was found. Electrophoresis in 0.17% agarose suspension was found to be a technique suitable for analysis of differences in charge of FSH in sera from men and women. A total of 31 sera from 25 individuals was investigated. The results strongly indicate that there is in man a sex hormone related pleomorphism of FSH with less acidic forms of FSH in sera of normal women of fertile age than in sera of men and post-menopausal women. Qualitative analysis of FSH in serum by electrophoresis may become an important complement to the quantitative determinations of the FSH concentration for the clinical diagnosis of reproductive disorders and of FSH producing tumours.

Insulin-dependent diabetes mellitus and menstrual dysfunction.

Disordered reproductive function has long been recognized as a prevalent problem among women of reproductive age who suffer from insulin-dependent diabetes mellitus (IDDM). Delay in menarchial age is frequently seen if IDDM develops in the peripubertal years and some form of menstrual dysfunction is found in nearly one-third of all women of reproductive age with IDDM. This review summarizes some of the prevailing views regarding the mechanisms through which uncontrolled IDDM is thought to disrupt normal hypothalamic-pituitary-gonadal function. Although animal studies have suggested that poorly controlled IDDM may adversely affect the uterovaginal outflow tract and/or ovarian function, no clinical studies have suggested that abnormal uterine or ovarian function underlies the menstrual dysfunction observed in young diabetic women. Similarly, pituitary function as assessed by basal gonadotrophins and gonadotrophin-releasing hormone (GnRH)-stimulated gonadotrophin release appears to be normal in young women with IDDM. Moreover, although there has been some suggestion that pituitary function may decline with increasing duration of diabetes, this issue has not been thoroughly investigated. It appears that the oligo/amenorrhea noted in IDDM is principally hypothalamic in origin and may represent intermittent (and perhaps reversible) failure of the GnRH pulse generator, similar to the situation observed in women who engage in endurance training or who suffer from anorexia nervosa. Although the exact pathophysiological mechanisms that subserve dysfunction of the GnRH neuronal system are not well understood, attention has focused on increased central opioidergic activity, increased central dopaminergic activity, and central glucose deprivation. In this era of emphasis on tight glycaemic control and its impact in preventing diabetes complications, the consequences of IDDM on reproductive potential appear to be important and must be included in future investigative efforts.

Prevalence of autoantibodies to thyroperoxidase in patients with various thyroid and autoimmune diseases.

An original radioimmunoassay for quantitation of circulating autoantibodies (aAb) to thyroperoxidase (TPO) proved to be well suited for large scale routine testing. The present study was aimed to assess the prevalence of aAb to TPO in patients with various thyroid and autoimmune disease and, for comparison, in women referred for reproductive disorders and indication of in vitro fertilization. Anti-TPO aAb were measured in sera from 32 healthy subjects and 262 patients thoroughly investigated for thyroid dysfunction. As determined in healthy subjects, the normal level of aAb to TPO in serum ranged from 0.30 to 3.07 mg/l (of affinity-purified) anti-TPO aAb. Anti-TPO and anti-MIC aAb levels were both normal in 115 patients and correlated well (r = 0.835, P less than 0.001) in the remaining 147 patients. Coexistence of normal level of anti-TPO aAb and abnormal level of anti-MIC aAb was found in 4 patients and ascribed to a lack of specificity or sensitivity of the test for anti-MIC aAb. Coexistence of abnormal level of anti-TPO aAb and normal level of anti-MIC aAb was found in 67 patients of whom 62 presented only slightly elevated (3.1 to 10.0 mg/l) anti-TPO aAb concentration; the 5 remaining patients, all with overt thyroid autoimmune disease, showed anti-TPO levels between 10.7 to 100.7 mg/l.(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of housing, management, and health of dairy heifers on first-lactation udder health in southwest Sweden.

Data on health, management, and housing from birth to first calving were collected for 2,126 heifers on semi-monthly visits made by project veterinarians to 107 dairy herds from southwest Sweden. Additional data were obtained from the official milk- and health-recording program. Factors associated with incidence of veterinarian-treated clinical mastitis (VTCM) in the period 7 d before (d -7) to 30 d after first calving and of elevated cow composite somatic cell count (SCC, > or =200,000 cells/mL) at first test milking after first calving, respectively, were investigated using a 2-level (animal and herd) logistic regression analysis after initial screening by univariate analyses. The incidence risk of VTCM during the complete first lactation (305 d) was 10.8%. Ten percent of the diseased animals had more than 1 case of VTCM and 51% of total cases occurred from -7 to 30 d postcalving. The incidence rate of VTCM during the complete first 305-d lactation was 1.13 cases per 100 cow-mo. In total, 18.1% of the animals had elevated SCC at first test milking (mean 21 d) after calving. Veterinarian-treated clinical mastitis at -7 to 30 d postcalving was associated with higher overall incidence of mastitis in the herd and with reproductive disorders (i.e., retained placenta, endometritis, pyometra, dystocia, or twin birth). The risk of elevated SCC increased with increasing percentage of cows in the herd that, some time during the year, had had an increased udder disease score (chronically increased SCC). Other factors associated with increased risk of elevated SCC were increasing amounts of concentrates fed to 11- to 16-mo-old heifers, moving to confined housing the day of calving instead of earlier, and use of restraint measures at milking. In addition, growth rate from birth to weaning, and several feed-related variables (e.g., amount of concentrates and type of roughage given) were associated with VTCM at -7 to 30 d post-calving or elevated SCC at first test milking in the univariate analyses.

Sole ulcers in dairy cattle: associations with season, cow characteristics, disease, and production.

Epidemiological associations, expressed as odds ratios between variables obtained from dairy cow records and sole ulcer occurrence at claw trimmings were estimated with logistic regression analysis on data from 2204 and 1124 cows in lactation 1 and lactations 2 to 9, respectively. Sole ulcer in one foot or more than one foot occurred in 20.0 and 29.7% of cows in lactation 1 and in 23.5 and 24.7% of cows in lactations 2 to 9. The analysis revealed several complicated interactions. Trimming or calving in summer to fall was strongly associated with sole ulcer. Trimming later than 1 to 2 mo after calving was positively associated with sole ulcer depending on milk yield, body weight, or season of calving. If lactation 1 cows were treated for disease (limb, metabolic, digestive, or severe reproductive disorders), sole ulcer in more than one foot occurred earlier in lactation. Milk yield in early lactation and body weight were positively associated with sole ulcer. Heel erosion was associated with sole ulcer depending on season of trimming and parity. Presence of sole ulcer but not heel erosion in a lactation increased the risk of sole ulcer in the subsequent lactation. The interval between trimmings appeared to be important.

Modulatory effects of dietary beta-carotene on blood and mammary leukocyte function in periparturient dairy cows.

Beginning 4 wk prior to predicted calving, 14 Holstein cows per treatment were fed diets 1) unsupplemented (control) or supplemented daily with 2) 300 mg of beta-carotene, 3) 600 mg of beta-carotene, or 4) 120,000 IU of vitamin A. Blood was collected around calving on wk -4, -2, -1, 0 (within 24 h postcalving), 1, 2, and 4 for isolation of lymphocytes and neutrophils and for the analysis of plasma vitamins. Lacteal secretions were collected on wk 0, 1, 2, and 4 for the isolation of phagocytes. Cows supplemented with 600 mg of beta-carotene had higher concentrations of plasma beta-carotene and retinol than did unsupplemented cows. Supplemental vitamin A increased plasma retinol on wk 4 and decreased plasma beta-carotene on wk -1 and 0. Treatment did not affect concentrations of plasma alpha-tocopherol. Blood lymphocyte proliferation in response to concanavalin A, phytohemagglutinin, and pokeweed mitogen during the peripartum period was higher in cows supplemented with beta-carotene than in unsupplemented controls. Phagocytic activity of blood neutrophils was enhanced on wk 1 in cows fed 300 mg of beta-carotene. Intracellular killing by blood neutrophils was enhanced in cows supplemented with beta-carotene (wk 0) and vitamin A (wk 0 and 1). Iodine uptake and nitroblue tetrazolium reduction by blood neutrophils was stimulated in cows supplemented with beta-carotene. Phagocytic activity, iodine uptake, and nitroblue tetrazolium reduction by mammary phagocytes from all cows generally were lower postpartum than on the day of calving. The incidence of retained placenta and metritis was higher for unsupplemented cows than for cows supplemented with beta-carotene. Therefore, dietary beta-carotene can elevate peripartum concentrations of blood beta-carotene, enhance host defense mechanisms by potentiating lymphocyte and phagocyte function, and decrease the incidence of certain reproductive disorders.

Effects of diseases on test day milk yield and body weight of dairy cows from Danish research herds.

The pre- and postdisease interrelationships of energy corrected test day milk yield and body weight of dairy cows caused by mastitis, three reproductive disorders (retained placenta, metritis, cystic ovaries), and seven metabolic disorders (milk fever, ketosis, decreased rumen motility, enteritis, left displaced abomasum, right displaced abomasum, and off feed) were quantified by using mixed models analysis with repeated measures of continuous data. The data were weekly recordings from 4414 lactations collected in three Danish research herds. High milk yield was a risk factor for ketosis and enteritis. Heavier primiparous cows were more likely to contract mastitis. Milk yield was decreased for a disease-specific period for all study diseases except cystic ovaries and right displaced abomasum. Metabolic disorders had a detrimental effect on body weight. The highest weight loss (69 kg) was associated with left displaced abomasum. The persistence of the weight loss differed considerably among study diseases. Almost all weight loss occurred up to and including the initial week after diagnosis, which emphasized the detrimental effect of the subclinical stage. However, weekly measured body weight seemed superior to weekly energy corrected test day milk yield for disease detection only for decreased rumen motility and left displaced abomasum. This study demonstrates the importance of the predisease level for accurate estimation of the loss of milk yield and body weight from disease.

Causes of mortality in chickens that regressed a Rous sarcoma virus-induced tumor.

Rous sarcomas were induced in 6-week-old chickens of several genetically different stocks: inbred lines C, 6(1), 6(3), and 7(2); crosses of inbred lines (6(3) X 7(2)) F4 and (6(1) X 15(1)) F2 X 6(1); and reciprocal crosses (15(1) X 100) F1 X 15(1) and (15(1) X 100) F1 X 100. The resulting tumors were scored for size six times during a 10-week period. Females that had completely regressed their sarcomas were placed in individual laying cages and examined weekly for reappearance of a tumor. After death, the probable cause was determined by necropsy. The major causes of death in the pooled sample of 49 females were fatty liver hemorrhagic syndrome (24.6%), reproductive disorder (14.2%), Marek's disease (12.2%), and lymphoid leukosis (6.1%). Elapsed time between tumor regression and death from any cause ranged from 21 days to 1930 days (5.3 years). One tumor recurred, this in a bird which eventually died with a massive sarcoma in the left wingweb and Rous metastasis in liver tissue. These data provide evidence of specific resistance to neoplastic disease.

Development and epidemiological investigation of a TaqMan-based multiplex real-time quantitative PCR assay for simultaneous detection of five bovine viruses (BVDV, AKAV, BNoV, BEV, and BCoV).

INRODUCTION: Infectious diseases caused by bovine viral diarrhea virus (BVDV), Akabane virus (AKAV), bovine norovirus (BNoV), bovine enterovirus (BEV), and bovine coronavirus (BCoV) significantly threaten the cattle industry, resulting in substantial economic losses. These pathogens often present similar clinical signs, such as diarrhea, vomiting, and reproductive disorders in pregnant cattle, and frequent covert or mixed infections further complicate accurate diagnosis. Therefore, rapid, sensitive, and field‑deployable diagnostic methods are essential for effective disease surveillance and control in the cattle industry. METHODS: In this study, we report for the first time the establishment of a TaqMan‑based real‑time quantitative PCR (qPCR) assay that enables simultaneous detection of these five bovine viruses. Multiple sequence alignment of conserved genomic regions was performed, and virus‑specific primers and probes were designed and optimized using Beacon Designer 7 software. Subsequently, a TaqMan‑based multiplex real‑time qPCR assay was established for simultaneous detection of BVDV, AKAV, BNoV, BEV, and BCoV. The established detection method was applied to 200 clinical samples collected from 10 farms in multiple regions of Jilin Province. RESULTS: The results showed that the detection rates for BVDV, AKAV, BNoV, BEV, and BCoV were 33.50%, 0.50%, 4.50%, 7.50%, and 12.00%, respectively. Mixed infections were detected in 9 samples co‑infected with two of the five pathogens, with an overall mixed infection rate of 4.50%. Compared with conventional PCR, coincidence rates were 100% for BVDV, AKAV, BNoV, BEV, and BCoV. DISCUSSION: These findings indicate that the TaqMan multiplex real‑time qPCR assay developed here demonstrates favorable specificity, sensitivity, and reproducibility. This assay enables efficient detection and surveillance of bovine viruses, offering a reliable technical tool for the diagnosis and control of corresponding viral diseases in cattle.

Evaluation of electroacupuncture on ovariectomized rats: implications of modern scientific mechanisms on acupuncture curing woman perimenopausal syndrome.

Acupuncture has been used for treatment of female reproductive disorders dated back at least 2000 years. It has been known to promote homeostasis, modulate hormonal disturbance and induce ovulation. While the increasing popularity of acupuncture therapy demands a serious debate about its scientific documentation. This article is intended to present the experimental data about the effects of electroacupuncture at Guanyuan (RN 4), bilateral Zhongji (RN 3), Sanyinjiao (SP 6) and bilateral Zigong (EXCA 1) in the ovariectomized rats. The EA stimulation produced from a Model G6805-H EA apparatus, and the stimulation parameters were frequency of 3Hz and intensity about 1-2mA, which was progressed in recent years by our group, so as to provide preliminary scientific background of acupuncture therapy of female perimenopausal syndrome.

Brucella sp. antibodies in polar bears from Svalbard and the Barents Sea.

A prevalence of 5.4% of anti-Brucella sp. antibodies was found in plasma samples from 297 polar bears (Ursus maritimus) from Svalbard and the Barents Sea. Plasma was tested by the classical brucellosis tests Slow Agglutination of Wright (SAW), EDTA modified SAW and Rose Bengal test, as well as by an indirect Protein A ELISA. Only samples classified as positive in all tests were regarded as containing anti-Brucella sp. antibodies. A significant west to east increase in the proportion of bears with anti-Brucella sp. antibodies was found, with 3.6% (n = 253) at Svalbard (Spitsbergen, Nordaustlandet, Edgeøya, Barentsøya and Hopen), and 15.9% (n = 44) in the central Barents Sea. Anti-Brucella sp. antibodies were previously found in ringed seals (Phoca hispida) and harp seals (Phoca groenlandica) from the same geographical areas. The ringed seal is an important prey species for the Svalbard polar bear population, and may thus be a source of brucellosis for the bears. There are no indications of reproductive disorders caused by Brucella sp. or other infectious agents in our study polar bear population. Potential impacts of Brucella sp. exposure on individuals or the population are unknown.

[The ten leading work-related diseases and injuries proposed by the National Institute for Occupational Safety and Health (NIOSH)].

Ten leading work-related diseases and injuries were proposed by the NIOSH to be controlled. These diseases and injuries are occupational lung diseases, musculoskeletal injuries, occupational cancers (other than lung), amputations, fractures, eye loss, lacerations, and traumatic deaths, cardiovascular diseases, disorders of reproduction, neurotoxic disorders, noise induced loss of hearing, dermatologic conditions, and psychologic disorders. Current topics regarding these diseases and injuries are discussed.

Pre-operational health status of a rural community living around the Kakrapar Atomic Power Station.

Pre-operational health survey was conducted around Kakrapar Atomic Power Station (KAPS) during 1991-92, just one year before the first Unit of KAPS commenced operation on September 3, 1992. Total seventeen villages were selected around KAPS, which were divided into four clusters. Total population was 14,976, of which 7540 were male and 7436 were female. Door to door 100% survey was conducted with the help of Medical doctor. Health survey was conducted specially to find out the prevalence of radiation related diseases such as congenital deformity, mental retardation, cataract, goiter (grade II & IV), reproductive disorders, and cancer, besides study of morbidity pattern and age and sex specific deaths. Results revealed that prevalent diseases pattern fell well within the range of natural incidences of diseases of rural population.

A novel missense mutation D513G in exon 10 of the cystic fibrosis transmembrane conductance regulator (CFTR) gene identified in a French CBAVD patient. Mutations in brief no. 175. Online.

Congenital bilateal absence of the vas deferens (CBAVD) with obstructive azoospermia is a congenital reproductive disorder that affects one in 10000 male individuals. The observation that many men presenting with CBAVD have mutations in their CFTR genes had led to the proposal that CBAVD may be a primary genital form of cystic fibrosis. We report here one novel mutation located in exon 10 of the CFTR gene. This mutation, named D513G (A-->G at position 1670), has been found in one of 83 patients with CBAVD from France, the analysis of exon 10 using a chemical clamp DGGE assay allowed us to identify three CF mutations AEF508 (37/166; 22%), AE1507 (1/166; 0/6%) and D513G (1/166; 0.6%), and two variants M470V and E528E (1716 G>A). The novel D513G mutation has not been found in more than 200 non-CF chromosomes and in a sample of 300 CF chromosomes from French classical CF patients.

Noninvasive reproductive monitoring in the okapi (Okapia johnstoni).

Fecal progestagen analysis in okapis (Okapia johnstoni) was used for diagnosis of pregnancy and reproductive disorders, including a comparison of urinary and fecal progestagen analysis and endocrine data on the postpartum period. Data were generated on reliability of fecal progestagen analysis in early pregnancy diagnosis, and case reports were compiled involving single animals with missing luteal activity, abortion after twin pregnancy, and abortions due to deficient placental progestagen production. There was approximately 100-200-fold higher progestagen concentration in feces than in urine, thus explaining the high reliability of fecal progestagen evaluations in diagnosing luteal function and pregnancy. The postpartum period was characterized by lactational anestrus of several months duration, and a postpartum estrous cycle about 2-3 wk after parturition was observed in two of eight animals. An animal with five abortions due to deficient placental progestagen production was treated with altrenogest in a subsequent pregnancy and carried the fetus to term.

[Dynamics in the health of women occupied in agricultural sector of the Ukraine].

Work conditions of women engaged into crop-raising and animal husbandry are characterized by variable complex of occupational physical, chemical and biologic hazards caused by the production specificity. The latter results in increased prevalence of chronic somatic diseases and reproductive disorders (menstrual dysfunction, higher incidence of miscarriage, etc.). Further dynamic research based on multifactorial approach are required to study health state of rural population.
