Immunocytochemical localization of 5'-nucleotidase in myelinated peripheral nerves from the rat.

In sciatic and trigeminal nerves from the rat, 5'-nucleotidase immunostaining was observed on the surfaces of the myelinated fibers and in the membranes encircling the outermost loops of the myelin sheaths, the paranodal loops, and perhaps the inner loops, but neither in the compact myelin nor in the axoplasm. These results, which were consistent with previous biochemical data regarding sciatic nerve, suggest that the function of 5'-nucleotidase in myelinated fibers in the peripheral nervous system may be to promote diffusion of adenosine between the glial and neuronal compartments.

Evidence for a metalloprotein structure of plasma membrane 5'-nucleotidase.

To point out the metalloprotein structure of bovine liver plasma membrane 5'-nucleotidase, we studied the inhibition mechanism of the purified enzyme by EDTA: this apparently non-competitive inhibition seems to be dependent on EDTA concentration, pH, temperature and incubation time. When the restoration of activity was assayed by addition of divalent cations or by gel filtration, the inhibition became progressively irreversible with time. Incubation of the enzyme with [14C]EDTA allowed us to observe, after gel filtration as well as after sucrose gradient ultracentrifugation, that the chelating agent is bound to 5'-nucleotidase.

5'-Nucleotidase↗

Activity of cytosolic 5'-nucleotidase in regenerating rat liver after partial hepatectomy.

The specific activity of the cytosolic 5'-nucleotidase in regenerating liver increased to 175% of the control level of sham-operated animals during the 2nd and 3rd day and remained elevated most of the experimental period. The total cytosolic 5'-nucleotidase activity of the regenerating liver reached the level of control rats between 2 and 3 days after the operation. The variation pattern of the enzyme, which was distinctly different from variations of other known phosphohydrolases, was strikingly similar to that of the salvage enzyme hypoxanthine/guanine phosphoribosyltransferase.

5'-Nucleotidase↗

Comparison of 5'-nucleotidase activities of isolated plasma membranes of two ascites cell variants.

The glycogen-containing ascites cell line was found to have a 3-5 times higher 5'-nucleotidase specific activity than the glycogen-free variant, resulting in different substrate affinity constants of Km = 0.14 mM and Km = 0.69 mM respectively. These activity differences were due to true 5'-nucleotidase as shown by its inactivation through specific inhibitors such as concanavalin A and alpha, beta-methylene adenosine diphosphate. Substrate specificity of the enzyme was similar in both cell lines, but differences were observed with respect to the pH optimum and stability.

Topography, purification and characterization of thyroidal 5'-nucleotidase.

1. Subcellular studies of bovine thyroid indicate that 5'-nucleotidase is predominantly associated with plasma membranes, although a considerable part of this ectoenzyme is also found internalized. 2. The enzyme displaying the features of a glycoprotein has been purified 1400 times by detergent solubilization and two subsequent affinity chromatographic steps. 3. Thyroidal 5'-nucleotidase can be classified as an unspecific metallo-dependent 5'-ribonucleotide phosphohydrolase. The native enzyme exists as a dimer (MW 150 kDalton), composed of two similar or identical subunits.

Levamisole inhibition of alkaline phosphatase and 5'-nucleotidase of bovine milk fat globule membranes.

1. The effect of levamisole (LMS) on alkaline phosphatase (EC 3.1.3.1) and 5'-nucleotidase (EC 3.1.3.5) activities of bovine milk fat globule membranes (MFGM) was examined. 2. LMS inhibited MFGM alkaline phosphatase activity in a concentration-dependent manner with 50% inhibition produced by 49 +/- 23 microM LMS. 3. 5'-Nucleotidase was resistant to LMS inhibition with 30.9% inhibition produced by 10 mM LMS, the highest concentration tested. 4. LMS was an uncompetitive inhibitor of MFGM alkaline phosphatase with a Ki of 45 +/- 6 microM. 5. The extent of LMS inhibition of alkaline phosphatase was dependent on the substrate utilized in the assay. 6. The effect of LMS on bovine MFGM alkaline phosphatase was similar to LMS effects on other mammalian alkaline phosphatases of liver/kidney/bone/placental isoenzyme origin.

A colorimetric assay for the determination of 5'-nucleotidase activity.

A rapid, simple, quantitative and sensitive assay for the determination of 5'-nucleotidase has been developed. The method can be applied to both soluble and membrane bound forms of the leukocyte enzyme. Enzyme activity is determined by colorimetric estimation of NH3 released from adenosine, the product of 5'-nucleotidase activity in the presence of adenosine deaminase. The assay may be performed in microtitre plates and read with an automatic multiscan spectrophotometer. Thus it can be applied to a large number of samples for routine medical and research purposes.

Surface markers, 5'-nucleotidase activity, and in vitro functions of lymphocytes from patients with primary humoral immunodeficiency.

Immunologic surface markers, 5'-nucleotidase activity, and in vitro cell functions were determined on peripheral blood mononuclear cells from 20 patients with primary humoral immunodeficiencies. Imbalances of T-cell subsets were a frequent finding and often associated with a deficiency of helper T lymphocytes, predominantly in common variable immunodeficiency. The magnitude of the blastogenic response to mitogens was subnormal in the majority of the samples and showed no correlations to the proportions of helper, suppressor T cells or monocytes in the cultures. Moderate to marked suppression of the pokeweed mitogen-driven allogenic B-cell maturation was mostly mediated by T lymphocytes but modified by monocytes in some patients. In identical twins, T lymphocytes selectively suppressed IgA production. Both in sex-linked agammaglobulinemia and common variable immunodeficiency the activity of 5'-nucleotidase was significantly lower than in controls. This deficiency was mediated by T lymphocytes and showed no correlations to imbalances of T-cell subpopulations or to alterations of lymphocyte functions.

Surface membrane localization of 3'- and 5'-nucleotidase activities in Leishmania donovani promastigotes.

Distinct 3'- and 5'-nucleotidase activities were localized to the surface membrane of Leishmania donovani promastigotes using enzymatic reactions with live intact cells and fine structure enzyme-mediated cytochemical reactions with intact cells, cell homogenates, and isolated surface membranes. The results indicated that virtually all activity of both enzymes was restricted to the parasite surface membrane. Cytochemical localization results demonstrated that both activities were externally disposed on the intact organism, and were restricted to the external face of the isolated parasite surface membrane. The two activities were cytochemically differentiated by their substrate specificities and sensitivity to several inhibitors. On the basis of their cytochemical sensitivity to glutaraldehyde treatment, the two nucleotidase enzymes were distinguished from another external surface membrane enzyme, a nonspecific acid phosphatase. Results of the cytochemical assays further demonstrated the biochemical and structural asymmetry of the isolated parasite surface membrane with regard to the localization and distribution of the active sites of these two enzymes.

5'-nucleotidase from bull seminal plasma.

5'-Nucleotidase (5'-ribonucleotide phosphohydrolase, EC 3.1.3.5) occurs in bull seminal plasma in multiple forms. The heterogeneity does not reflect the existence of true isoenzymes, but is due to the association of the enzyme with particulate material and to molecular aggregation phenomena. Addition of detergents to native bull seminal plasma prevents molecular aggregation, solubilizes the particulate form of the enzyme, and results in the appearance of a single molecular form of the enzyme. Enzyme purification can be achieved after three chromatographic steps which involve negative adsorption of 5'-nucleotidase activity on DEAE-Sephadex A-50 followed by two affinity chromatographies on concanavalin A-Sepharose 4B and ADP-agarose. The enzyme appears to be a dimeric glycoprotein. Some properties of the enzyme, including substrate specificity and the effects of hydrogen ion concentration and of various divalent cations, are reported.

Effects of dithiothreitol, dithioerythritol and chelating agents on 5'-nucleotidase from bull seminal plasma.

5'-Nucleotidase from bull seminal plasma is inhibited by dithiothreitol and dithioerythritol. These reactives proved to dissociate the dimeric glycoprotein 5'-nucleotidase of Mr 160 000 into two subunits of apparent Mr 80 000, indicating that the subunits are held together by interchain disulfide bridges. HPLC determinations of cysteic acid and carboxymethylcysteine protein derivatives resulted in 50 +/- 3 half-cystine plus cysteine residues, while 1.9 +/- 0.4 free cysteine residues were estimated by HPLC analysis. The enzyme is inhibited by EDTA and EGTA, and the inhibition appears to be of the non-competitive type for both the chelating agents. Experiments for the enzyme activity recovery by MgCl2 and CaCl2 additions, after the EDTA and EGTA treatments in the presence of 8 M urea, are reported.

5'-Nucleotidase↗

Chemical modification of essential carboxyl group and histidine residue in the plasma-membrane 5'-nucleotidase.

An investigation, using specific chemical reagents, of the amino acids involved in the catalytic activity of the purified 5'-nucleotidase (5'-ribonucleotide phosphohydrolase, EC 3.1.3.5) from bovine liver plasma membranes, was carried out. The enzyme was irreversibly inactivated by N-ethoxycarbonyl-2-ethoxy-1,2-dihydroquinoline (EEDQ). The inhibition kinetics were of the first-order type and decreased partially in the presence of nucleotides and divalent cations. These results indicate for the first time that a carboxyl group is essential for the catalytic process of 5'-nucleotidase. Moreover, chemical modification by diethylpyrocarbonate also produced inactivation of the enzyme and showed a differential spectrum with a peak at 240 nm characteristic of N-carbethoxyhistidine residues. This inactivation was efficiently released upon decarbethoxylation by hydroxylamine only when the extent of inactivation, due to low concentration of diethylpyrocarbonate, was limited. The time-dependent inactivation followed first-order kinetics and nucleotides afforded significant protection against diethylpyrocarbonate modification. The results indicate the involvement of the histidine residue in catalysis.

A reappraisal of the subunit molecular mass of chicken liver cytosol 5'-nucleotidase.

The subunit molecular mass of chicken liver cytosol 5'-nucleotidase was earlier reported to be 51 kDa upon sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (Naito, Y. and Tsushima, K. (1976) Biochim. Biophys. Acta 438, 159-168). By immunoblot analyses after SDS-gel electrophoresis, however, a fraction from the liver homogenized in the presence of leupeptin showed multiple bands with molecular masses around 57 kDa, and SDS-extracted proteins directly from the liver exhibited a single 70 kDa component. These results indicate that the 51 kDa peptide observed in the cytosol 5'-nucleotidase preparation might, in fact, be a proteolytic artifact.

ESR study of 5'-nucleotidase from bull seminal plasma.

5'-Nucleotidase of bull seminal plasma has been spin labeled with the sulfhydryl reagent 3-maleimidoproxyl. ESR analysis reveals the presence of two classes of labeled sites. The first is characterized by a long spin label rotational correlation time, from which a protein diameter of about 70 A can be estimated, under the assumption of a spherical shape. The second class is characterized by a shorter correlation time of the covalently bound spin labels and binding of the substrate sodium thymidine 5'-monophosphate to 5'-nucleotidase results in a reduction of their mobility. Low-temperature ESR analysis shows that no paramagnetic ion is bound to the native protein.

Elevated specific activity of 5'-nucleotidase in a spinal cord myelin fraction from shiverer mice. Comparison with other myelin-associated enzymes and myelin proteins.

Myelin partially purified from spinal cords of dysmyelinating mutant (shiverer) mice had almost three-fold the specific activity of 5'-nucleotidase found in the respective myelin fraction from normal mice. The specific activities of two other normally myelin-associated enzymes, 2',3'-cyclic nucleotide-3'-phosphohydrolase and carbonic anhydrase, were only slightly higher in the myelin membranes from shiverers, compared to those from controls. In the mutants, the three enzymes probably occur in oligodendrocyte processes. Hypothetically, the 5'-nucleotidase in the myelin sheath in shiverer and normal mice may be localized in specialized structures.

Isolation and kinetic properties of 5'-nucleotidase from guinea-pig skeletal muscle.

5'-Nucleotidase (EC 3.1.3.5) has been solubilized and purified 1200-fold from guinea-pig skeletal muscle, to a specific activity of 40 U/mg protein. The purified enzyme yields a single protein band on polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. Guinea-pig skeletal muscle 5'-nucleotidase is extremely sensitive to inhibition by nucleoside di- and triphosphates. The inhibition is of the competitive type, and can be reversed only by strong excess of Mg2+. Nucleoside diphosphates are more powerful inhibitors than nucleoside triphosphates. The Ki values for ADP and ATP are 0.036 and 0.28 microM, respectively. The purified enzyme does not require exogenous cations for maximal activity and is inhibited by EDTA. This inhibition is reversed by divalent cations. This indicates that the enzyme contains a tightly bound metal cation.

Activities and some properties of 5'nucleotidase in isolated hamster fat cells.

Kinetic parameters and the possible cellular location of 5'nucleotidase were studied in isolated hamster fat cells. Measurements of the enzyme were performed using 5'IMP instead of 5'AMP as substrate. The apparent Vmax and Km values obtained support the view that two forms of 5'nucleotidase, having different affinities, are present in hamster fat cells. The major form was located in the outer surface of plasma membrane and the other one inside the cell. The effects of ADP, adenosine and inosine on the enzyme were studied. ADP and adenosine were competitive inhibitors. The enzyme was not modified by inosine.

Differences in the release of 5'-nucleotidase and alkaline phosphatase from plasma membrane of several cell types by PI-PLC.

1. We have compared the effect of phosphatidyl inositol specific phospholipase C (PI-PLC) on the attachment of both 5'-nucleotidase and alkaline phosphatase to the liver plasma membrane from different species. 2. Our results demonstrate differences in the susceptibilities of both enzymes to PI-PLC treatment in relation to their origin. 3. These results were confirmed by immunoblotting using polyclonal anti-5'-nucleotidase antibodies. 4. In addition, in a single animal, susceptibility of both enzymes to PI-PLC treatment is different from one tissue to another. 5. The different percentages of released enzymes could be explained either by a polymorphism in the anchoring of these proteins at the cell surface membrane, or by a different steric hindrance or environment at the cleavage site itself.
