Lymphocyte subsets, TNF alpha and interleukin-4 levels in treated and untreated subacute sclerosing panencephalitis patients.

Immunologic studies in relation to clinical status might help to understand the pathogenesis of subacute sclerosing panencephalitis (SSPE) and the effect of treatment. We measured lymphocyte subsets and intracellular TNFalpha and interleukin-4 levels in peripheral blood in SSPE patients. Patients had elevated percentages of CD8+ cells compared to age-matched control children. Rapidly progressive course was associated with increased CD4+ cells. Treatment with interferons and inosiplex altered the percentage of CD3+, CD4+ and CD19+ cells. TNFalpha and interleukin-4 levels had no correlation with course or treatment. The proportions of lymphocyte subsets appear to have a role in the evolution or manifestations of SSPE, if not in the pathogenesis.

T lymphocyte subsets of the infiltrating cells in the salivary gland and kidney of a patient with Sjögren's syndrome associated with interstitial nephritis.

Lymphocyte subsets in the salivary gland and kidney were examined in a 38 years-old female patient with Sjögren's syndrome associated with interstitial nephritis by PAP immunoperoxidase method using monoclonal antibodies. Predominant cells of the infiltrating cells in both tissues were T lymphocytes and most of them were Ia+, OKT4+ cells (activated helper/inducer T lymphocytes). A small number of T lymphocytes were OKT8+ (suppressor/cytotoxic T lymphocytes). Moreover, we found the OKT8+ cells invading the salivary duct epithelial cells. There was no difference in the proportion of lymphocyte subsets of the infiltrating cells between the salivary gland and kidney. A similar pathologic mechanism of tissue damage, therefore, was suggested in both tissues.

T lymphocyte subsets and antilymphocyte antibodies in lupus nephritis.

T lymphocyte subsets were determined in 12 patients with untreated systemic lupus erythematosus (SLE) and in 14 healthy controls. Six out of 8 (75%) patients with lupus nephritis had reduction in the percentage of T helper cells and low helper: suppressor cell ratios compared with controls. None of the 4 patients without nephritis had low ratios. Cold-reactive anti-lymphocyte antibodies cytotoxic to both the helper and the suppressor cells were detected in 7 of the 8 patients who had nephritis. Low T helper: suppressor cell ratio in SLE seems to correlate with the presence of active nephritis.

[Effects of cordyceps sinensis on T lymphocyte subsets and hepatofibrosis in patients with chronic hepatitis B].

In order to find an effective drug to cure patients with chronic hepatitis B, cordyceps sinensis had been used to treat 25 patients with chronic hepatitis B. The comprehensive index, including T lymphocyte subsets (CD4, CD8), hyaluronic acid(HC) and precollagen type III(PC III), were observed before and after treatment. After 3 months of treatment, CD4 and CD4/CD8 ratio increased significantly(P < 0.05), while HA and PC III decreased significantly(P < 0.05) compared with the control. The results suggest that the beneficial effects might be obtained by using cordyceps sinensis to adjust the T lymphocyte subsets level and to treat hepatic fibrosis on patients with chronic hepatitis B.

[Peripheral lymphocyte subsets in patients with Vogt-Koyanagi-Harada syndrome (VKH)].

The peripheral lymphocyte subsets were analyzed in 37 patients with VKH and 32 normal controls by indirect fluorescein assay, using monoclonal antibodies. The percentages of OKT3+, OKT4+, and OKT8+ T-cells in active patients were found to differ insignificantly from those in quiescent patients, as were those in patients of the VK type from those in patients of the Harada type (P > 0.05), whereas the percentage of OKT4+ was significantly higher, that of OKT8+ was lower, and the OKT4/OKT8 ratio was higher in the recurrent patients than in the controls (P < 0.01). There was statistical difference in the ratio of OKT4/OKT8 between the recurrent and the non-recurrent patients (P < 0.01), while the ratio differed insignificantly between the two types of recurrent patients. The results suggested that the recurrence of VKH could be related with imbalance between the T-cell subsets.

[Peripheral T-lymphocyte subsets changes in patients with endocrine and metabolic diseases].

In this study the authors utilized the OKT monoclonal anti-human T lymphocyte antibodies to determine the normal level of T lymphocyte subsets in the peripheral blood of 30 healthy volunteers, 36 patients with Graves' disease (GD), 4 with hypothyroidism and 30 with diabetes mellitus (DM). It is shown that OKT+3, OKT+4 OKT+8 and OKT+4/OKT+8 ratio of T-lymphocyte subsets in the peripheral blood of 30 healthy volunteers were 72.4 +/- 5.1%, 38.9 +/- 5.2%, 26.8 +/- 4.3% and 1.5 +/- 0.3. In 36 patients with GD OKT+3 was 67.6 +/- 5.8%, OKT+4 was 33.8 +/- 6.6% and OKT+8 was 17.9 +/- 6.1%. These were all significantly lower than those of normal controls (P less than 0.001). OKT+4/OKT+8 ratio was 2.1 +/- 0.6, which was significantly higher (P less than 0.001). The value of OKT+4 and OKT+8 in 10 patients with GD after treatment were markedly elevated. However, the ratio of OKT+4/OKT+8 was significantly decreased (P less than 0.05). OKT+3 of 4 cases of hypothyroidism was 69.7 +/- 3.35%, being similar to that of normal controls (P greater than 0.2). OKT+4 and OKT+8 were 31.73 +/- 4.99% and 18 +/- 2.94% respectively, both of which being markedly decreased (P less than 0.01). The ratio of OKT+4/OKT+8 was 1.97 +/- 0.22, being not significantly elevated (P greater than 0.05). OKT+3, OKT+4 and OKT+8 of 23 cases with DM were 68.8 +/- 7.2% (P less than 0.05), 33 +/- 8.1% (P less than 0.005), and 21.2 +/- 6.5% (P less than 0.001) respectively. All of these were significantly decreased. The ratio of OKT+4/OKT+8 was 1.6 +/- 0.5, being not significantly changed.

[Efficacy of shenqi fuzheng injection combined with chemotherapy in treatment of acute leukemia and its effect on T-lymphocyte subsets, serum IFN-gamma, IL-10 and IL-2].

OBJECTIVE: To investigate the clinical efficacy of Shenqi Fuzheng Injection (SFI) combined with chemotherapy in treatment of patients with acute leukemia and its effect on the levels of T-lymphocyte subsets (CD4, CD8, CD4/CD8) and serum interferon-gamma(IFN-gamma), interleukin-10 (IL-10) and IL-2. METHODS: Sixty-five patients with initial treating acute leukemia were randomly divided into 2 groups, the SFI group (n = 32) treated with SFI plus chemotherapy (CT), the control group (n = 33) treated with CT only. The remission rate, changes of peripheral mature neutrophilic granulocyte (PMNG) count, T-lymphocyte subsets, serum IL-10 and IL-2 before and after treatment were determined. RESULTS: The remission rate in the two groups showed no obvious difference (P > 0.05). After CT for the 1st, 2nd and 3rd weeks, the PMNG count decreased in both groups, showing significant difference as compared with that before CT (P < 0.01 or P < 0.05). The PMNG count at the end of the 3rd and 4th week of CT remounted to higher than that at 1st and 2nd week, and the increment in the SFI group was significantly higher than that in the control group (P < 0.05). The levels of CD4, CD4 /CD8, IFN-gamma and IL-2 all increased in the two groups after treatment (P < 0.05, P < 0.01), however, that of IL-10 was significantly decreased (P < 0.01). The difference between the two groups in these criteria after treatment was also significant (P < 0.05). CONCLUSION: SFI can improve and regulate the immune function of the patients with acute leukemia undergoing CT, it could promote bone marrow cells proliferation and enhance the efficacy.

Flow cytometric analysis of T-lymphocyte subsets in cats.

We report a rapid, reliable method for the immunophenotype analysis of feline lymphocytes. Fluorescein isothiocyanate (FITC) conjugated to murine monoclonal antibodies f43, Fel 7 and fCD8 was used to identify phenotypes corresponding to feline T-cells, CD4+ T cells and CD8+ T cells. For isolation of white blood cells, whole blood lysis was faster, less variable and required much less sample than density gradient separation. To identify feline CD4+ and CD8+ cells simultaneously, directly conjugated FITC-fCD8 and phycoerythrin (PE) fCD4 (Fel 7) were used in two-color analysis. The two T cell sub-populations were non-overlapping. Dual-label and single-label values were not significantly different. Mean lymphocyte subset percentages in conventional and specific-pathogen-free (SPF) cats did not differ significantly. These values were: pan T lymphocytes (f43), 54.8%, CD4+ cells (Fel 7), 33.9%, and CD8+ cells (fCD8), 19.1%. Mean CD4/CD8 ratio was 1.9 in normal cats; the range was 1.2-2.6.

Flow cytometric analysis of lymphocyte subsets in peripheral blood of chronic headache patients.

Lymphocyte subsets from 5 groups of patients with different types of chronic headaches were analyzed by flow cytometry to determine the number of cells bearing the following markers: T11 (E-rosette receptor), T3 (total T cells), T4 (T-helper/inducer cells), T8 (T-cytotoxic/suppressor cells), B1 (B-cells), NKH-1 (Natural killer cells), I2 (Class II histocompatibility antigens, monomorphic determinants) and Mo2 (monocytes). In 4 of 5 of these groups, the absolute number of T8 lymphocytes was significantly lower than that of control group. In addition, the T4:T8 ratio was higher in these 4 groups. These findings may suggest a relationship between the immune system and the cause of chronic headaches.

Corticosteroid receptors and lymphocyte subsets in mononuclear leukocytes in aging.

Plasma cortisol and aldosterone levels and number of related receptors in mononuclear leukocytes were measured in 49 healthy aged subjects (62-97 yr) and in 21 adult controls (21-50 yr). In all subjects, in addition, lymphocyte subsets were determined as an index of corticosteroid action. The mean number of type I and type II receptors was significantly lower in aged subjects than in controls (respectively, 198 +/- 96 and 272 +/- 97 receptors/cell for type I, and 1,794 +/- 803 and 3,339 +/- 918 for type II receptors). Plasma aldosterone and cortisol and lymphocyte subsets were not different in the two groups. All of the parameters were also tested for correlation, and a significant inverse correlation was found between age and type I and type II receptors when all subjects were plotted and between aged and CD4 and age and CD4/CD8 in the aged group. These data show that aged subjects have reductions of corticosteroid receptors that are not associated with increase of related steroids and that this situation probably represents a concomitant of the normal aging process.

B lymphocyte subset patterns and their significance in idiopathic glomerulonephritis.

The proportion of B lymphocyte subsets with surface immunoglobulin G (sIgG) was significantly increased in minimal change nephrotic syndrome (MCNS), membranous nephropathy, IgA nephropathy and mesangiocapillary glomerulonephritis (MCGN) and with sIgA in IgA nephropathy and MCGN, and with sIgE in MCNS. Increased subsets in membranous nephropathy, IgA nephropathy and MCGN corresponded to the immunoglobulins deposited in the glomeruli, and the increased subset of sIgE in MCNS was correlated with the elevation of serum IgE. These results suggest that each disease studied has a characteristic subset pattern of B lymphocyte response. This may have an important role in determining the histological type of idiopathic glomerulonephritis.

The effects of epidural block on the distribution of lymphocyte subsets and natural-killer cell activity in patients with and without pain.

Although epidural anesthesia prevents immune suppression during surgery, no reports have elucidated how epidural block affects immune response in nonsurgical patients. We examined changes in proportion of lymphocyte subsets and in natural-killer (NK) cell activity in patients with and without pain. Fifteen patients with pain (Pain group) and 15 preoperative patients without pain (Preoperative group) received three different treatments in random order: epidural block with 7 mL 1% lidocaine, epidural injection of an identical volume of normal saline, and IV injection of 1 mg/kg lidocaine. Blood samples were drawn before and after 30, 60, and 120 min of treatment. During epidural block at 30 and 60 min, both groups showed significantly decreased epinephrine, norepinephrine, and cortisol levels, and the proportion of NK cells decreased, whereas the CD4+/CD8+ ratio increased significantly. NK cell activity in both groups decreased significantly at 30 and 60 min. At 120 min, the variables had all returned to preblock values. During treatments with saline and IV lidocaine, neither group showed significant changes in any of the above variables. We conclude that epidural block causes a transient and significant alteration of lymphocyte subsets and NK cell activity regardless of pain status.

CD4+ and CD8+ T-lymphocyte subsets in Zambian children.

Age-related changes in lymphocyte subsets in HIV-uninfected Zambian children are described. The total lymphocyte count and numbers of CD4+ and CD8+ T-lymphocytes declined with increasing age, while the percentage of CD4+ and CD8+ T-lymphocytes changed little during childhood. Girls between the ages of 12 and 71 months had a higher percentage of CD4+ T-lymphocytes and a higher CD4:CD8 ratio than did boys of a similar age.

Immunohistological localization of human lymphocyte subsets.

The technical aspects of immunohistological detection of lymphocyte surface antigens were studied in frozen and fixed, embedded tissues employing several immunoperoxidase and immunofluorescent methods. It was found that routine fixation and embedding destroyed immunostaining of surface immunoglobulin and T-cell surface antigen. Cytoplasmic immunoglobulin was preserved in fixed, embedded tissues. A comparative study of nonspecific staining in various tissues using different techniques revealed that immunoperoxidase methods generally gave lower nonspecific staining than immunofluorescent methods. The labeled avidin biotin method achieved the lowest nonspecific staining and appears to be superior to the other methods studied. A preliminary study of T lymphocyte subsets in human B-cell lymphomas and reactive lymph nodes was also performed. It was found that T-cell immunostaining patterns were altered in B-cell lymphomas in comparison to reactive lymph nodes, suggesting a more generalized disturbance of immunoregulation in these tumors.

Peripheral T lymphocyte subsets in rapidly progressive periodontitis.

The peripheral blood lymphocyte subsets of 10 patients with rapidly progressive periodontitis were investigated with monoclonal antibody. In 4 patients, the T helper/T suppressor ratio was increased. 5 others had a slightly reduced T helper/T suppressor ratio as compared to control group. These findings may indicate a possible cellular immune response in the pathogenesis of rapidly progressive periodontitis.

Influence of selective T-lymphocyte depletion on the lung pathology of gnotobiotic calves and the distribution of different T-lymphocyte subsets following challenge with bovine respiratory syncytial virus.

The depletion of CD8+ T-lymphocytes with a murine monoclonal antibody (mAb) specific for the CD8 molecule delayed the ability of three gnotobiotic calves to clear bovine respiratory syncytial virus (BRSV) from their lungs within 10 days after an experimental challenge with the virus. This protracted infection was associated with an enhanced pneumonic consolidation score (21.6 per cent) compared with seven control calves (7.4 per cent) and a histological lesion of active respiratory epithelial hypertrophy. Three gnotobiotic calves depleted of the CD4+ subpopulation with the appropriate mAb also had enhanced macroscopic lesions (16.6 per cent) but the histological lesion was less active. The depletion of the gamma/delta TCR+ WC1+ subpopulation had no apparent effect on the macroscopic or microscopic pulmonary lesions. Although the depletion of the CD8+ or the CD4+ subpopulations enhanced the pulmonary lesions, no clinical signs of respiratory disease were detected. Immunoperoxidase labelling and image analysis of the lymphocyte subpopulations in lung tissue revealed an increase in the number of CD8+ T cells after the infection of non-depleted, control calves, especially in the lamina propria of the large bronchioles. Calves depleted of individual lymphocyte subsets and infected with BRSV showed no compensatory increase in the remaining subpopulations and no lymphoreticular hyperplasia.

Age-related changes of lymphocyte subsets in normal bone marrow biopsies.

There is a paucity of information in the literature concerning the age-related changes of the lymphocyte subsets in bone marrow (BM), and the available reports disagree about the characteristics of the population studied and the methods for obtaining, handling, and analyzing the samples. The purpose of the present study was to determine the distribution of lymphoid subsets in the BM from infants, children, and adults by analyzing fragments of sternum obtained during cardiovascular surgery. The samples were studied by flow cytometry employing the whole blood lysis method and excluding from the analysis the contamination of the lymphoid window by erythroid precursors. We observed that in the first 4 years of life the B subset represented more than 65% of all cells in the lymphoid window, most of them (80%) exhibiting the immature phenotype CD19+CD100+. Conversely, the T subset was composed of mature CD4+ or CD8+ cells, with the CD4/CD8 ratio being less than 1 in all age groups. With age there was a progressive decrease in the percentage of B cells and an increase of T cells, reaching similar proportions in the BM from adults (33.6% and 34.8%, respectively). Furthermore, the percentage of CD10+ cells in the B subset decreased independently, whereas the CD20 expression increased. The percentage of NK cells did not change with age.

Inflammatory cell profiles and T-lymphocyte subsets in chronic obstructive pulmonary disease and severe persistent asthma.

BACKGROUND: Severe persistent asthma (SPA) and chronic obstructive pulmonary disease (COPD) are both associated with non-reversible airflow limitation and airway neutrophilia. OBJECTIVE: To compare inflammatory cell profiles and T lymphocyte subsets between SPA and COPD patients with similar severity of airflow limitation. METHODS: Sputum induction and lung function tests were performed in 15 COPD patients aged (mean+/-SD) 68+/-8 years, ex-smokers, mean forced expiratory volume in 1 s (FEV1) 45% of predicted (% pred) and 13 SPA aged 55+/-10 years, non-smokers, mean FEV(1) 49% pred. All patients were on inhaled steroid treatment. Eight asthmatics exhibited irreversible airflow limitation. Differential cell count, metachromatic cell count and double immunocytochemistry for the analysis of T lymphocyte subsets were performed on sputum slides. RESULTS: COPD patients had increased sputum neutrophils in comparison with SPA (P<0.03), but similar to SPA with fixed obstruction. In COPD sputum neutrophils negatively correlated with the lung transfer factor for carbon monoxide (KCO) (r=-0.462, P=0.04). SPA showed significantly increased eosinophils and metachromatic cells vs. COPD patients (P<0.04, P<0.007, respectively). Increased CD4/CD8 and decreased CD4-IFN-gamma/CD4-IL4+ cell ratio (P<0.001) were found in SPA vs. COPD. In SPA, CD4/CD8+ cell ratio correlated with sputum eosinophils (r=0.567, P=0.04). CONCLUSION: In spite of treatment with inhaled steroids, SPA and COPD exhibit distinct sputum inflammatory cell patterns, although SPA with fixed airflow limitation and COPD patients have similar numbers of neutrophils.
