Transcriptional regulation of expression of the rainbow trout albumin gene by estrogen.

Estrogens modulate the expression of many liver-specific genes in oviparous species. For instance, expression of the estrogen receptor and vitellogenin genes is strongly up-regulated by estradiol in rainbow trout liver. Using hepatocyte primary cultures, we demonstrate that trout albumin (Alb) gene is also regulated by this hormone. Indeed, treatment of hepatocytes with 1 microM estradiol led, after 24 h, to a dramatic decrease in Alb mRNA level. To investigate the mechanism of this down-regulation, run-off experiments were performed and mRNA half-lives were determined in the presence and absence of estradiol. The results show that the down-regulation of Alb mRNA expression by estrogens occurs only at the transcriptional level.

An in vitro investigation of the role of high molecular weight human salivary sulphated glycoprotein in periodontal wound healing.

High molecular weight human salivary sulphated glycoprotein (SGP) inhibits attachment of fibroblasts to cementum in vitro and thus may enhance periodontal wound healing by repair with long junctional epithelium. However, competitive inhibition by serum constituents might prevent adequate binding of SGP for this effect to occur in vivo. The primary aim of this study was to investigate the co-adsorption in vitro, of SGP, fibronectin (FN), and albumin (ALB) to synthetic hydroxyapatite (HA) from solutions of SGP/FN (62.5/2 micrograms/mL respectively), or SGP/FN/ALB (62.5/2/4 micrograms/mL), or from individual solutions of the agents as controls: Desorption of SGP and FN was studied by preabsorbing HA with SGP or FN and subsequently exposing it to the other agent, or to buffer only as control. Adsorbates were assayed after incubation periods of up to 26 hours using specific ELISAs. SGP displaced previously adsorbed FN after 3 hours and significantly inhibited adsorption of FN and ALB compared with controls. Neither FN or ALB had a significant effect on SGP adsorption. These results are consistent with the possibility that salivary adsorption to cementum during surgery might interfere with repopulation of the root by connective tissue cells and thus contribute to wound healing by repair rather than regeneration. Separate studies taken to validate the ELISA used for SGP determination showed that HA-bound SGP contained all constituents of native SGP as revealed by SDS-PAGE and that ion exchange chromatography of SGP gave 6 fractions (I through VI), of which only the most ionic (VI) was able to inhibit cell attachment in vitro.

Transcytosis of albumin in endothelial cells is brefeldin A--independent.

To determine whether in endothelial cells (EC) the pathways of endocytosis and transcytosis of macromolecules interconnect, the effect of Brefeldin A (BFA) on these processes was tested. To this purpose EC were grown to confluence on plastic culture dishes or on cell culture chamber inserts placed into corresponding wells, so as to obtain a dual chamber system. The cells maintained the typical characteristics of EC and had an electrical resistance in the range of 30-60 Ohm.cm2. Transendothelial transport of albumin conjugated to the fluorochrom Texas Red (Alb-TR) and of horseradish peroxidase (HRP) added to the upper compartment, in the absence or presence of BFA (0-25 micrograms/ml), was evaluated in aliquots collected from the lower compartment. At different time intervals, quantitative data were obtained by fluorimetry and spectrophotometry. In other experiments transcytosis of Alb-TR was examined in the presence of 100 microM forskolin (an inhibitor of BFA effect). The endocytosis of Alb-TR and HRP was evaluated by incubating EC with the probes, and the internalized tracers determined in the cell lysate using the methods described above. The results showed that BFA has no significant effect on transcytosis of albumin and HRP. In contradistinction, BFA (5 micrograms/ml) reduced markedly endocytosis of HRP (by 47%). Forskolin has no effect on transcytosis. The data indicate that the BFA-induced perturbance in the endocytic route does not affect the transcytotic pathway of albumin, and suggest that in EC, transcytosis of macromolecules may represent a shortcut for rapid and direct transport of some plasma molecules across the cell.

[Protective effects of Chailing Guiqi Decoction combined with lumbrukinase on renal function in rats with adriamycin nephropathy].

OBJECTIVE: To study the protective efffects of Chailing Guiqi Decoction (CLGQD) combined with lumbrukinase on renal function in rats with adriamycin nephropathy. METHODS: Thirty-six SD rats were randomly divided into four groups: normal control group, untreated group, simvastatin-treated group and CLGQD -treated group. Adriamycin nephropathy was induced by intravenous injection with 5 mg/kg adriamycin. After seven-day treatment, quantitative measurement of 24-h urine protein was determined with trichloroacetic acid, and serum total protein (TP), albumin (Alb), total cholesterol (TC), triglycerides (TG), high-density lipoprotein (HDL), low-density lipoprotein (LDL), creatinine (Cr) and blood urea nitrogen (BUN) were assessed using automatic biochemistry analyzer. The pathomorphological changes of renal tissues were observed with light and electron microscopes. RESULTS: In the untreated group, the 24-h urine protein excretion, serum TC, TG, LDL, Cr and BUN were significantly higher than those in the normal control group (P<0.05 or P<0.01), while the serum TP, Alb, HDL were significantly lower than those in the normal control group (P<0.01). In the CLGQD-treated group, the 24-h urine protein excretion, serum TC, TG, LDL, Cr and BUN were significantly lower as compared with those in the untreated group (P<0.05 or P<0.01), while the serum TP, Alb and HDL were significantly higher as compared with those in the untreated group (P<0.05 or P<0.01). The pathomorphological findings of the renal tissues under the light microscope in the untreated group showed focal segmental glomerulosclerosis in a few of glomerulus, degenerated and swelled proximal tubular epithelial cells, proteins in cast formation in some renal tubules and scattered fibrosis in interstitial tissues of the kidney, while the electron microscope images showed the fusion of foot processes in glomerular epithelial cells. The pathomorphological changes in the CLGQD-treated group were slighter than those in the untreated group. CONCLUSION: CLGQD combined with lumbrukinase can reduce proteinuria, regulate lipid metabolism, protect renal function, and delay progressive renal damage in rats.

[Clinical observation on treatment of IgA nephropathy with Huobahuagen Tablets and irbesartan].

OBJECTIVE: To observe the effects of Huobahuagen Tablets combined with irbesartan on the risk factors of IgA nephropathy. METHODS: Sixty-two patients diagnosed as IgA nephropathy were randomly divided into control group and treatment group. Thirty patients in the control group were treated with Huobahuagen Tablets (5 tablets po t.i.d.), and 32 patients in the treatment group were treated with irbesartan (150 to 300 mg po q.d.), besides the same treatment as the control group. After 3 months of treatment, the levels of blood pressure (BP), 24 h urine protein (Upr), urinary red blood cells (URBC), blood triglycerides (TG), total cholesterol (TC), albumin (Alb), alanine transaminase (ALT), white blood cells (WBC) and serum creatinine (Scr) were observed. RESULTS: After treatment, the levels of Upr, URBC and Scr in both groups were decreased, as compared with those before treatment (P<0.05 or P<0.01), and the levels of BP, Upr, URBC and Scr in the treatment group were lower than those in the control group (P<0.01). The levels of Alb in both groups were increased, as compared with those before treatment (P<0.05 or P<0.01), and the level of Alb in the treatment group was higher than that in the control group (P<0.05). CONCLUSION: Huobahuagen Tablets, when used together with irbesartan, may improve the renal function of the patients with IgA nephropathy and slow the deterioration of the disease by reducing BP, Upr, URBC and Scr.

Retinol-binding protein, acute phase reactants and Helicobacter pylori infection in patients with gastric adenocarcinoma.

AIM: To determine the serum levels of c-reactive protein (CRP), transferrin (TRF), a2-macroglobulin (A2M), ceruloplasmin (CER), a1-acid glycoprotein (AAG), pre-albumin (P-ALB) and retinol-binding protein (RBP) in gastric carcinoma patients and to explore their possible correlation with underlying Helicobacter pylori (H pylori) infection. METHODS: We measured the serum levels of CRP, TRF, A2M, CER, AAG, P-ALB, and RBP in 153 preoperative patients (93 males; mean age: 63.1+/-11.3 years) with non-cardia gastric adenocarcinoma and 19 healthy subjects. RESULTS: The levels of CRP, CER, RBP, and AAG in cancer patients were significantly higher than those in healthy controls (P<0.0001), while no difference was found regarding the TRF, P-ALB, and A2M levels. Cancer patients with H pylori infection had significantly lower RBP values compared to non-infected ones (P<0.0001) and also higher values of CRP and AAG (P = 0.09 and P = 0.08, respectively). CONCLUSION: High serum levels of CRP, CER and AAG in cancer patients do not seem to be related to H pylori infection. Retinol-binding protein seems to discriminate between infected and non-infected patients with gastric carcinoma. Further studies are needed to explore if it is directly involved in the pathogenesis of the disease or is merely an epiphenomenon.

Increased prevalence of intestinal inflammation in patients with liver cirrhosis.

AIM:To investigate the pathophysiology of the digestive tract in patients with liver cirrhosis.METHODS:In 42 cirrhotic patients and 20 control subjects, the following fecal proteins were measured by enzyme-linked immunosorbent assay: albumin (Alb), transferrin (Tf), and alpha(1)antitrypsin (alpha(1)-AT) as a marker for intestinal protein loss, hemoglobin (Hb) for bleeding, PMN-elastase for intestinal inflammation, and secretory IgA for intestinal immunity.RESULTS:The fecal concentrations of Hb, Alb, Tf,alpha(1)-AT, and PMN-elastase were increased in 13 (31%), 8(19%), 10(24%), 6(14%), and 11 (26%) cases among 42 patients, respectively. Fecal concentration of secretory IgA was decreased in 7 (17%) of 42 patients. However, these fecal concentrations were not related to the severity or etiology of liver cirrhosis. The serum Alb level was significantly decreased in patients with intestinal protein loss compared to that in patients without intestinal protein loss.CONCLUSION:These findings suggest that: (1)besides the well-known pathological conditions, such as bleeding and protein loss, intestinal inflammation and decreased intestinal immunity are found in cirrhotic patients; (2)intestinal protein loss contributes to hypoalbuminemia in cirrhotic patients, and (3) intestinal inflammation should not be over looked in cirrhotic patients, since it may contribute to or cause intestinal protein loss and other various pathological conditions.

Effects of electroporation on primary rat hepatocytes in vitro.

AIM: To investigate the effects of electroporation on primary rat hepatocyte and to optimize the electroporation conditions introducing foreign genes into primary hepatocytes. METHODS: A single-pulse procedure was performed at low voltage (220-400 V) but with high capacitance (500-950 microF). Hepatocytes were divided into 4 groups according to the electroporation conditions: group I, 220 V and 500 microF; group II, 220 V and 950 microF; group III, 400 V and 950 microF,and group IV. The control group was freshly isolated hepatocytes and directly cultured under the same conditions as those of electroporation groups. The effects of electroporation on primary rat hepatocytes were detected by trypan blue exclusion (TBE) and MTT analysis. Besides, albumin (Alb), alanine transaminase (ALT) and lactate dehydrogenase (LDH) in the supernatants of cultured hepatocytes were measured by biochemical assay. RESULTS: Between day 1 and day 15 after incubation, primary rat hepatocytes of each electroporation group appeared normal, being the same with those of control group. TBE staining showed that slight hepatocyte damage and high survival rate were found in the electroporation groups and the control group. Cultured for 3, 7, 11 and 15 days, hepatocyte viability was approximately 92.6+/-2.5 %, 89.5+/-3.3 %, 82.0+/-3.5 % and 74.3+/-1.2 %, respectively. MTT analysis indicated that the viabilities of hepatocytes had no significant difference between each electroporation group, and those were similar to that of control group. At the 36th hour after electroporation, Alb, ALT and LDH in the supernatants of control group were 5.3+/-0.1 g x L(-1), 183.7+/-8.4 nkat x L(-1) and 896.8+/-58.5 nkat x L(-1); those of group II were 5.7+/-0.1 g x L(-1), 215.4+/-16.7 nkat x L(-1) and 1063.8+/-51.8 nkat x L(-1); and those of group III were 5.8+/-0.2 g x L(-1), 217.1+/-8.4 nkat x L(-1) and 1063.8+/-10.0 nkat x L(-1). Statistically, the proteins of group II and group III were significantly higher than those of control group (P<0.05), whereas the protein production of group I, Alb, ALT and LDH were 5.3+/-0.2 g x L(-1), 205.4+/-3.3 nkat x L(-1) and 1035.4+/-116.9 nkat x L(-1), were similar to those of control group. At the same time, TBE and MTT analysis indicated that there was no significant cell viability difference between electroporation groups and control group. CONCLUSION: This single-pulse electroporation procedure performed at low voltage (220-400 V) but with high capacitance (950 microF) is one of the optimal choices to introduce foreign genes into primary rat hepatocyte.

Studies on an antibiotic, albocycline VII. Minor components of albocycline.

As an approach to the search for new potentially useful macrolide antibiotics, we explored the minor components of albocycline (ALB) from the culture broth of Streptomyces bruneogriseus. Eight minor components were isolated and their structures were confirmed as 1 approximately 8. Unexpectedly, they were not glycosidic compounds but only oxidation or reduction products of ALB. Three or four of them will serve as a useful intermediate to introduce amino sugar moiety into ALB skeleton.

[Electron microscopic observation on cysticercus cellulosae treated with mienangling in vitro].

OBJECTIVE: To observe the effect of Mienangling (MNL) in treating cysticercus cellulosae in vitro. METHODS: The changes of cysticercus cellulosae treated in vitro with the MNL extracts, praziquantel (PQT) and albendazole (ALB) were studied under the transmission and scanning electron microscopes. RESULTS: Under the scanning electron microscope, the authors can see that all of the cysticercus's scoleces did not grow in the MNL and PQT groups, cyst walls shrivelled, tegument's ectoplasts were damaged, microtriches's ultrastructures were not clear or exfoliated, some corpuscular matter densely was distributed in the surface; 46.7% of cysticercus cellulosaes in the ALB group had been excystated, the rostellar hooks of rostella developed normally, four sucker's surface wrer hollow, the necks were eroded, and grooves of sonite disappeared. Under the transmission electron microscope, we can see that the tegument, basement membrane, muscular layer and parenchyma in the groups of MNL and PQT were all clearly damaged, but the harmful effect of MNL to parenchyma's nerve cords is more serious than the PQT. The damage of all layers of the cysticercus cellulosae in the ALB group is smaller in degree than the MNL and the PQT. CONCLUSION: The MNL has marked damaging effect to cysticercus cellulosae.

Physical and meiotic mapping of the region of human chromosome 4q11-q13 encompassing the vitamin D binding protein DBP/Gc-globulin and albumin multigene cluster.

The vitamin D binding protein/Gc-globulin (DBP) gene is a member of a multigene cluster that includes albumin (ALB), alpha-fetoprotein (AFP), and alpha-albumin/afamin (AFM). All four genes have structural and functional similarities and map to the same chromosomal regions in humans (4q11-q13), mice, and rats. An accurate physical map of the region encompassing these genes is a prerequisite for study of their respective transcriptional regulation and identification of potential shared regulatory elements. By refining the physical and meiotic maps of the 4q11-q13 region and creating a local PAC contig, the order and transcriptional orientations of these four genes were determined to be centromere-3'-DBP-5'-5'-ALB-3'-5'-AFP-3'-5'-AFM3'-telomere. The ancestral DBP gene was separated from the ALB gene by >1.5 Mb. This organization and spacing establishes a foundation for ongoing functional studies in this region.

Nutritional status of Korean peritoneal dialysis patients.

OBJECTIVES: To assess the nutritional status of Korean peritoneal dialysis (PD) patients and to compare with data from Western literature, and to elucidate independent factors determining nutritional status and death. DESIGN: Cross-sectional single-center study. SETTING: Kidney Center, Soon Chun Hyang University Hospital. MATERIALS: Ninety-eight CAPD patients were included. Of these, 54 patients were male, 32 patients were diabetic, mean age was 47.9+/-13.1 years, and mean duration of CAPD was 22.3+/-21.6 months. The patients were followed until death, transfer to hemodialysis (HD) or other units, transplantation, or until 3 years had elapsed after the first evaluation. METHODS: Nutritional status was assessed by subjective global assessment (SGA), biochemical and anthropometric measurements, fat-free edema-free (FFEF) body mass by creatinine (Cr) kinetics, protein equivalent of total nitrogen appearance (PNA), and urea kinetic studies. RESULTS: By SGA score, 53.1% of patients were classified as normal, 44.9% with mild-to-moderate malnutrition, and 2% with severe malnutrition. Patients with malnutrition were significantly older and had higher peritonitis rates, lower serum albumin (Alb), blood urea nitrogen (BUN), serum Cr, FFEF body mass, mid arm muscle circumference, and PNA (p < 0.05). On stepwise multiple regression analysis, the SGA score was negatively correlated with age and peritonitis rate (p < 0.01). At the end of the 3-year follow-up period, 11 patients were still on CAPD, 26 had died, 51 had transferred to HD and 5 to other units, 3 patients had been transplanted, and 2 patients were lost to follow-up. Patients who died during follow-up were older and had higher peritonitis rates and lower total serum protein, Alb, Cr, and FFEF body mass when compared to those who survived (p < 0.05). Independent predictors of death were age, peritonitis rate, and serum Alb (p < 0.01). CONCLUSION: Malnutrition was as common in Korean PD patients as reported in the Western literature. Our data suggests that, to prevent malnutrition and early death, it is important to reduce the peritonitis rate, to improve protein intake, and to prescribe an adequate dose of peritoneal dialysis.

Surfactant protein SP-B counteracts inhibition of pulmonary surfactant by serum proteins.

In addition to the primary surfactant deficiency in newborns with respiratory distress syndrome (RDS), in the later course of RDS substantial protein leakage into the alveolar spaces can occur by damage to the alveolocapillary membrane. Acute lung injury results in surfactant dysfunction due in part to inhibition by serum proteins. The aim of this study was to investigate the influence of SP-B on the inhibitory effects of albumin (alb) and fibrinogen (fib) on the surface activity of pulmonary surfactant, using a) surface tension measurement with the pulsating bubble surfactometer in suspensions and b) in surfactant films applying the hypophase exchanger. After hypophase exchange a preformed film of Survanta is very resistant to the inhibitory activity of alb or fib. The surface tensions of suspensions are significantly higher (p <0.001) than the surface tensions of preformed surfactant films if alb or fib were added, e.g., 42 (41 to 43) mN/m vs. 21 (19 to 22) mN/m for Survanta with 20 mg alb/ml. After additional supplementation of Survanta with SP-B the surface activity of Survanta/1% SP-B films did not show inhibition by fib (2 mg/ml), (surface tension 8 (4 to 13) mN/m). These results indicate that SP-B can play an important role to protect the pulmonary surfactant film from inactivation by serum proteins.

[Hepatitis B virus infection and aflatoxin exposure in the development of primary liver cancer].

OBJECTIVE: To investigate the role of hepatitis B Virus (HBV) infection and aflatoxin (AF) exposure in the development of primary liver cancer (PLC). METHODS: A 10-year prospective case-control study was carried out in 737 HBsAg carriers and 699 HBsAg negative cases, and the aflatoxin B1 albumin adducts (AFB1-Alb) were detected in the serum of the cohort including 30 HBsAg postive cases and 150 control individuals according to the case-control study model (ratio 1:5) at random in the high prevalance area of PLC. RESULTS: The average year-incidence rate was significantly higher in the HBsAg postive group (824.13/100,000) than in the control group (70.97/100,000, RR = 11.61). There was no significant difference in the incidence rate of other tumors between the two groups (P > 0.05). The serum positive rate of AFB1-Alb was significantly higher in the PLC group (76.67%) than in the control group (48.67%, OR = 3.47), and the serum concentration of AFB1-Alb was also significantly higher in the PLC group than in the control group (P < 0.01). CONCLUSION: HBV infection and AF exposure are important etiological factors in the development of PLC and both result in carcinogenic synergy.

Endocytic uptake of advanced glycation end products by mouse liver sinusoidal endothelial cells is mediated by a scavenger receptor distinct from the macrophage scavenger receptor class A.

Previous studies with peritoneal macrophages obtained from macrophage scavenger receptor class A (MSR-A) knock-out mice showed that the endocytic uptake of advanced glycation end products (AGE) by macrophages was mediated mainly by MSR-A. However, it is controversial whether the endocytic uptake of intravenously injected AGE proteins by liver sinusoidal endothelial cells (LECs) is similarly explained by receptor-mediated endocytosis via MSR-A. The present study was conducted to compare the capacity to endocytose AGE proteins in LECs and peritoneal macrophages obtained from MSR-A knock-out and littermate wild-type mice. The endocytic degradation capacity of MSR-A knock-out LECs for AGE-BSA was indistinguishable from that of wild-type LECs, whereas that of MSR-A knock-out peritoneal macrophages for AGE-BSA was decreased to 30% of that in wild-type cells. Similarly, the endocytic degradation of MSR-A knock-out LECs for acetylated low-density lipoprotein (acetyl-LDL) did not differ from that of wild-type LECs, whereas the endocytic degradation of acetyl-LDL by MSR-A knock-out peritoneal macrophages was less than 20% of that in wild-type cells. Furthermore, formaldehyde-treated serum albumin (f-Alb), a ligand known to undergo scavenger-receptor-mediated endocytosis by LECs, was effectively taken up by MSR-A knock-out LECs at a capacity that did not differ from that of wild-type LECs. Moreover, the endocytic uptake of AGE-BSA by LECs was effectively competed for by unlabelled f-Alb or acetyl-LDL. These results indicate that the scavenger-receptor ligands AGE proteins, acetyl-LDL and f-Alb are endocytosed by LECs through a non-MSR-A pathway.

Serum ascorbic acid and protein calorie malnutrition in continuous ambulatory peritoneal dialysis patients.

Protein calorie malnutrition (PCM) is a major predictor of morbidity and mortality in continuous ambulatory peritoneal dialysis (CAPD) patients. Oxidative stress has been implicated as a contributory cause. The relative lack of ascorbic acid, a plasma antioxidant, has been reported in CAPD patients. We therefore examined the correlation between serum ascorbic acid (SAA) and parameters of PCM in a cross-sectional study of 50 clinically stable adult CAPD patients. The mean age of the study group was 61 +/- 11 years; 48% of the patients were male; and 64% had diabetes. The parameters assessed were SAA, serum albumin (S Alb), body mass index (BMI), subjective global assessment (SGA), and lean body mass (LBM). The results (mean +/- standard deviation) were: SAA, 9.44 +/- 8.79 mg/L; S Alb, 31.6 +/- 4.9 g/L; BMI, 24.4 +/- 3.4; LBM, 0.66 +/- 0.14 kg/kg body weight; SGA, 9 of 50 were considered mildly-to-moderately malnourished (the rest were well-nourished). A significant correlation (p < 0.001) was found between SAA and S Alb only with SAA < or = 9 mg/L. No correlation was seen between SAA (at any level) and BMI or LBM. The SAA was not significantly different between the groups as graded by SGA. The data suggest that the mechanisms for low serum albumin in these patients may be related to the role of ascorbic acid as a plasma antioxidant.

Salivary immunoglobulin-A as a marker of stress during strenuous physical training.

BACKGROUND: Immunoglobulin-A in saliva (SIgA) has been proven to be decreased during periods of stress, a change that also correlates with increased disease risk. HYPOTHESIS: Concentration of SIgA is negatively associated with dietary deprivation, negative moods, and anxiety. METHODS: SIgA was evaluated as a marker of the severity of stress during a 19-day Royal Australian Air Force (RAAF) survival course, during which students experienced hunger, thirst, boredom, loneliness, and extreme heat and cold combined with demanding physical effort. There were 27 men and 2 women who participated. Students kept daily food diaries, from which daily intakes of energy and macronutrients were calculated. Saliva samples were collected on 9 d for the measurement of the ratio of SIgA to albumin (Alb). Students completed a health checklist and the State Anxiety Inventory on the same 9 d and the Profile of Mood States on 3 of the days. RESULTS: Dietary restriction, consumption of alcohol, body mass loss, occurrence of upper respiratory tract infection, and negative emotions were negatively associated with SIgA:Alb. CONCLUSION: SIgA:Alb is a useful marker of the severity of stresses encountered during stressful training.

[Influence of escharectomy and skin grafting during early burn stage on acute-phase response in severely burned rats and its significance].

OBJECTIVE: To investigate the influence of escharectomy and skin-grafting during early burn stage on the production of acute-phase reactants by the liver in severely burned rats. METHODS: Ninety-six Wistar rats were randomly divided into three groups: normal control group (C), escharectomy on postburn day (PBD) 1 group (0) and escharectomy on PBD4 group (F). 30% III degrees (total body surface area full-thickness) burn injury was caused by immersing the back in boiling water for 10 seconds. Escharectomy and skin grafting with cryopreserved allogeneic skin were performed one day or 4 days after the burn injury. ON days 1, 2, 4, 5 and 7 after burn injury blood was withdrawn from the abdominal aorta of 8 rats for each experimental group. The serum contents al-antitrypsin (AT), al-acid glycoprotein (AGP), C-reactive protein (CRP), albumin (Alb), and transferrin (Trf) were measured by turbidimetry. RESULTS: Significant increase in concentrations of positive acute-phase reactant, CRP, AGP, and AT, was observed in group O and group F as compared with normal control, (P < 0.05), while the concentrations of negative acute-phase reactants, Alb and Trf, decreased markedly (P < 0.05). The serum concentrations of AT and AGP in group O began to decrease significantly on PBD 5 and PBD7 respectively in comparison with that of the group F (P < 0.05), however, they still maintained a rather higher levels on PBD7. The concentration of CRP in group O began to decrease on PBD4, and the values on PBD4, 5, and 7 were significantly lower than those of the group F (P < 0.05). The concentration of CRP in PBD1 group reached the normal level on PBD7, however, the concentration of CRP in group F kept increasing till PBD5, and remained significantly higher than that in group C (P < 0.05). The concentration of Alb in group O was significantly higher on PBD7 than that in group F (P < 0.05). The concentration of Trf in group O on PBD5 and PBD7 was significantly higher than those in group F (P < 0.05). CONCLUSION: Escharectomy and skin grafting during early burn stage attenuates acute-phase response after burn injury and are beneficial to the organism.
