Lipopolysaccharides of flagellated and non-flagellated Proteus vulgaris strains.

Lipopolysaccharides from two strains of Proteus vulgaris were analyzed. One strain (08) was motile, giving swarming growth on solid media and the other (04)--non-flagellated, not able to swarm. Both lipopolysaccharides appeared to be heterogeneous and were separated into two fractions each. Yield, chemical composition and SDS-polyacrylamide gel electrophoresis showed differences between fractions in percentage content of 0-specific and R-core polysaccharides. Relation between ability to swarming growth of Proteus strains and heterogeneity of their lipopolysaccharides is also discussed.

[Antibiotic sensitivity of bacteria in the genus Proteus in relation to the source of isolation and specific classification].

One hundred and ninety-one Proteus strains isolated from patients with various pathological processes were studied with respect to their sensitivity to 14 antibiotics. It was shown that their resistance depended on the isolation source and microbial species. Amikacin proved to be the most active agent. It was efficient against all the Proteus species, irrespective of the isolation source. Among the drugs efficient against P. mirabilis and P. vulgaris there were gentamicin, tobramycin and sisomicin sulphates.

[Resistance of E. coli and bacteria of the genus Proteus to the thermal processing of perishable meat products].

Experiments were carried out in six meat combines to establish the devitalization of Escherichia coli and bacteria of the Proteus genus at the thermal treatment of perishable meat products. Seventy series of experiments were conducted with 16 types of meat products of a different structure (homogenic and heterogenic), varying composition unequal diameter of the casings (3-4 am, 5-6 am, and 7-8 am), and different type of the latter--natural, paper, and cutis-lika. Twenty-four-hour agar cultures at a density of 10(5)-10(7) bacteria per gram of meat mass were used. In a series of experiments the temperature was dermined of the finished product when it reached 68 degrees C; in another group--after smoking and at various intervals during cooking; and in a third group of experiments the temperature was fillowed up during the entire thermal process, using thermo pairs. Studied was the heat resistance of the organisms tested under laboratory conditions, using the thermal curve in the production of meat products. The samples were investigated at the 24th, and 48th hour, and on the 5th day of room temperature storage. Results showed that devitalization of these organisms set in at 63 degrees C (E. coli) and 60 degrees (Proteus). The composition and homogeneity of the meat mass and the diameter of casings were found to have no effect on the heat resistance of these organisms.

Immunochemical studies on free lipid A from Proteus mirabilis 1959.

The chemical composition and serological activity of free lipid A from Proteus were studied. Only two fatty acids: myristic acid and 3-hydroxymyristic acid were detected. When calculated for glucosamine disaccharide unit, 2 moles of ester-linked and 1 mole of amide-linked fatty acid are present. Amino group of glucosaminyl residue is substituted by 3-hydroxymyristic acid. The occurence of an uncommon (4-aminoarabinose) substituent, attached outside the backbone, was noticed. The results of serological investigation indicate the great similarity between antigenic determinants of Proteus lipid A and Salmonella-type of lipid.

Immunochemical studies on the O-specific side chains of the heterogeneous lipopolysaccharide from Proteus mirabilis O27.

Chemically and serologically identical O-specific fractions were isolated from two different lipopolysaccharide (LPS) preparations of Proteus mirabilis O27. The release of a labile constituent in mild hydrolysis deprived those fractions of their serological activity. The compound was identified as N-acetyl-D-glucosamine and its terminal position in the molecule was established by methylation analysis. The non-carbohydrate constituent of the specific fractions: lysine and alanine are linked via their amino group. Terminal residues of N-acetylglucosamine, have to be considered as immunodeterminants of Proteus mirabilis O27. The role of other carbohydrate and non-carbohydrate constituents for the serological specificity of P. mirabilis O27 is discussed.

[Mortality of rare purulent meningitis, (Staphylococcus, Listeria Pseudomonas, Proteus, Klebsiella, E. coli, Salmonella). An epidemiologic study using the literature from the last 30 years].

3056 cases of purulent meningitis caused by seven rare microorganisms (Staphylococci, Listeria, Pseudomonas, Proteus, Klebsiella, E. coli, Salmonella) were collected from the West European and North American literature of the last 30 years. The average lethality has been calculated for the periods ranging from 1948 to 1962 and from 1963 to 1979 in order to compare the results due to the use of penicillin and older antibiotics with those accomplished with ampicillin and the aminoglycosides. After 1963 the death-rate of each type of meningitis decreased considerably (except for Salmonella meningitis): the lethality of the whole group fell from 48% to 33%, in the subgroup of gram-negative meningitis from 55% to 38%, and in the group of neonatal gram-negative meningitis from 67% to 52%. The best results were seen in Proteus meningitis where lethality decreased from 55% to 15%. In E. coli meningitis there was a reduction from 60% to 43% only. Using the chi-square test all these differences were highly significant (p less than 0.001).

[Compared studies of antimicrobial agents against E. coli, Klebsiella, Citrobacter and Proteus isolated from urinary tract infections].

In vitro activities of antibacterial agents against E. coli, Klebsiella, Citrobacter and Proteus which were isolated from patients urinary tract infections at 8 hospitals in Japan, were investigated by agar dilution method from July to October in 1979. The summarized results are as follows. 1. Among oral antibacterial agents, MPC and PPA have showed potent antibacterial activities against E. coli and Klebsiella. Among parenteral antibiotics, CTM was the most active against E. coli and Klebsiella. However, ABPC-resistant E. coli and Klebsiella have appeared to occupy about 40% and 96% of bacteria isolated from urinary tract infections, respectively. 2. In vitro activities of antibacterial agents against Proteus and Citrobacter showed not so potent. 3. Causative organisms in female patients with simple urinary tract infections were mainly E. coli and Klebsiella. 4. Among oral antibacterial agents, PPA have shown similar antimicrobial activities against E. coli isolated from simple and complicated urinary tract infections. ABPC and MPC have been influenced in some degree by these factors. However, parenteral antibiotics are not influenced by these factors. On the other hand, in vitro activities of antibacterial agents against Klebsiella isolated from simple and complicated urinary tract infections were similar.

Structure and epitope specificity of the O-specific polysaccharide of Proteus penneri strain 12 (ATCC 33519) containing the amide of D-galacturonic acid with L-threonine.

O-specific polysaccharide was isolated from Proteus penneri strain 12 (ATCC 33519) lipopolysaccharide (LPS) and studied using NMR spectroscopy, including selective spin-decoupling, one-dimensional NOE, two-dimensional homonuclear correlation spectroscopy, 13C,1H heteronuclear correlation spectroscopy and chemical methods (O-deacetylation, Smith degradation, partial acid hydrolysis followed by borohydride reduction and methylation). The amide of D-galacturonic acid with L-threonine [D-GalA(L-Thr)] was identified as a constituent of the polysaccharide and the following structure of the tetrasaccharide repeating unit was established: [formula: see text] where the degree of O-acetylation at either position varies over 20-40%. Serological study with LPS, its degradation products and related synthetic glycoconjugates (2-acrylamidoethyl glycosides of amides of alpha-D-GalA with L-amino acids copolymerised with acrylamide) showed that D-GalA(L-Thr) plays an important role in manifesting the serological specificity of the P. penneri 12 O-antigen. Serological cross reactions between LPSs of P. penneri 12 and Proteus mirabilis S1959, R14/S1959 (transient-like form), O23 and O28 are discussed.

[Proteus syndrome. 8 cases].

INTRODUCTION: Proteus syndrome most often includes: corporal hemi-hypertrophia, gigantism of the extremities with hyperplasia of the palms and soles, vascular abnormalities and varied types of other hamartomatous tumours. The dysmorphic growth generates functional orthopaedic and orthognatic disabilities that increase with age. OBSERVATIONS: We report, on eight cases, our experience in management of Proteus syndrome abnormalities and the benefits of new techniques in imaging and interventional vascular radiology. The correction of limb orthopaedic abnormalities is complicated with elephantiasic enlargement of soft tissues. In some patients the absence of a deep venous network contra-indicates transcutaneous occlusion of dysplatic vessels. Facial asymmetry and orthognatic anomalies are better managed after permanent teeth have appeared. COMMENTARIES: The management of these disabilities is carried out by staff of different specialities but should be organized by a practitioner who regularly follows the child and detects early the functional disturbances. Aesthetic corrections are performed later.

[Susceptibility of Proteus, Morganella and Providencia strains isolated from patients with urinary tract infections on bactericidal activity of normal human serum].

Usefulness of the test determining bactericidal activity of normal human serum was investigated with 50 strains of Proteus, Morganella and Providencia isolated from patients with urinary tract infections (UTI) and with 50 strains isolated from feces. It was found that strains from UTI were more frequently resistant to the action of normal human serum (50% resistant) in comparison with strains isolated from feces (30% resistant). Strains of Proteus belonging to four species were differing greatly in their susceptibility to normal human serum. They can be ranked as followings: P. mirabilis (49% of resistant strains), P. vulgaris (62%), P. morganii (72%) and P. rettgeri (100%). In studies on interaction subinhibitory concentrations of cefotaxime and normal human serum in bactericidal reaction, a synergism was found only with some strains.

[Selected properties of strains of a new species of Proteus penneri from the second American collection].

The second collection of the novel species Proteus penneri consists of 25 strains from which only two have shown rough from properties in the tests differentiating S and R variants of bacteria. The migration pattern of their lipopolysaccharides in gel electrophoresis was leader-like, typical for smooth organisms. 13 out of 25 lipopolysaccharide preparations showed strong-reactivity with anti-0 sera in semi-quantitative precipitation test. Serological similarity between the strains within species Proteus penneri is discussed.

[Immunochemical studies of O-specific polysaccharide of Proteus penneri 42 lipopolysaccharide].

O-specific polysaccharide was obtained an mild acid degradation of Proteus penneri strain 42 lipopolysaccharide and found to contain D-glucose, D-galacturonic acid and 2-acetamido-2-deoxy-D-glucose in molar ratio 2:1:1. Methylation analysis showed that the polysaccharide is linear, one of the glucose residue is substituted at position 2, the second one and the residue of galacturonic acid at position 4, and the 2-acetamido-2-deoxy-D-glucose residue at position 3. On the basis of non-destructive NMR analysis the following structure of repeat unit of the O-specific polysaccharide was established and confirmed independently by methylation analysis: -2)-beta-D-Glc-(1-->4)-beta-D-Glc-(1-->3)-beta-D-GlcNAc-(1-->4)-al fa-D-GalA-(1--> The serological investigation with application of P. penneri strain 42 anti O-serum has shown the activity of homologous preparations of LPS and PS, as well as cross-reactions with heterologous lipopolysaccharides from other Proteus strains.

[Induction of synthesis of extracellular DNAase from Proteus mirabilis under the effect of compounds, blocking DNA replication].

The effects of mitomycin C and nalidixic acid on the biosynthesis of extracellular endodeoxyribonuclease in Proteus mirabilis have been studied. The presence of both antibiotics in the periodic and short-time cultures of washed off cells has increased both the activity of the DNAse and protein yield in cultural liquid and bacterial cells. PAGE-electrophoresis has shown the effect of mitomycin C to increase or induce the synthesis a large number of Proteus mirabilis extracellular proteins.

Spinal canal compromise in Proteus syndrome: case report and review of the literature.

A 24-year-old Vietnamese man with Proteus syndrome presented to the emergency department with progressive paraplegia and underwent urgent operative decompression of an angiolipomatous mass compressing the spinal cord. The associated spinal deformity and difficulty in imaging the mass, together with the paucity of discussion regarding the spinal cord sequelae of this rare syndrome in the literature, made this an extremely challenging case. Spinal cord compromise in patients with Proteus syndrome can result from either infiltration of the spinal canal by an angiolipomatous mass or canal narrowing as a result of vertebral hypertrophy. Regardless of the cause, the prognosis for neural recovery remains poor.

Structural and serological studies of the O-antigen of the bacterium Proteus vulgaris OX2 (serogroup O2) used in the Weil-Felix test.

Based on monosaccharide analysis and 1H- and 13C-NMR spectroscopy, the following structure of the O-specific polysaccharide chain of Proteus vulgaris OX2 lipopolysaccharide (LPS), which defines the O2 specificity of Proteus, was established: [formula: see text] where L-QuiNAc is N-acetyl-L-quinovosamine (2-acetamido-2,6-dideoxy-L-glucose). Various strains of P. vulgaris OX2 used in the Weil-Felix test for serodiagnosis of rickettsiosis (spotted fevers, except for Rocky Mountain spotted fever) were shown to produce LPS with the same O-specific polysaccharide, which differs structurally and serologically from LPS of P. vulgaris OX19 used as antigen for serodiagnosis of typhus and Rocky Mountain spotted fever. O-Acetyl groups present in the polysaccharide are not important for manifesting the immunospecificity. ELISA confirmed that the epitope responsible for the cross-reactivity between sera from patients with Japanese spotted fever and P. vulgaris OX2 cells is located on the P. vulgaris LPS. At the same time, no cross-reaction was observed between rabbit anti-P. vulgaris OX2 antibodies and the spotted fever group (SFG) rickettsial cells. Therefore, human anti-SFG rickettsial antibodies and rabbit anti-P. vulgaris OX2 antibodies may bind to distinct epitopes on P. vulgaris OX2 LPS, and no epitope recognized by rabbit anti-P. vulgaris OX2 antibodies is present on the LPS or any other surface antigen of SFG rickettsiae.
