Efficient route to 1,3-Di-N-imidazolylbenzene. A comparison of monodentate vs bidentate carbenes in Pd-catalyzed cross coupling.

A new bis(carbene) ligand architecture has been developed and was evaluated in the Suzuki-Miyaura cross-coupling reaction of various aryl halides with phenylboronic acid. Several new bis(carbene) ligands were tested in different carbene:Pd ratios. Pd(OAc)(2) and Pd(2)(dba)(3) were compared for efficiency as a Pd source. It was found that the Pd(OAc)(2)/bis(carbene) system formed a catalyst for the activation of chlorobenzene. [reaction: see text]

Tensile properties and transformation temperatures of Pd added Ti-Ni alloy dental castings.

The effect of palladium (Pd) addition to Ti-Ni alloy as the third element was investigated to improve the super-elasticity of the alloy castings at body temperature for dental application. Ti-50.8Ni (at %) alloy, which exhibited super-elasticity at 310 K in castings, was used for comparison. 5.0, 7.5, 10.0 and 15.0 at % Pd was added to Ti-50.0Ni alloy by the substitution for Ni. The change in the proportion of Ti and Ni was also examined at the fixed Pd addition of 7.5 at %. The properties of the alloys were investigated in tensile test and differential scanning calorimetry (DSC). Ti-42.5Ni-7.5Pd alloy castings showed good super-elasticity among the examined alloys from the viewpoint of residual strain and elongation. Moreover, apparent proof stress could be changeable by the proportion of Ti and Ni with residual strain being kept low. Ti-42.5Ni-7.5Pd alloy castings exhibited better super-elastic flexibility than Ti-50.8Ni alloy, which is proven by lower apparent proof stress and larger elongation. This flexibility appears to be caused by its relatively high martensitic transformation starting temperature point. It is suggested that this flexibility with super-elasticity could widen the clinical application of the alloy casting in dentistry.

Evidence for overlapping substrate specificity between large neutral amino acid (LNAA) and dipeptide (hPEPT1) transporters for PD 158473, an NMDA antagonist.

PURPOSE: The objective of this research was to investigate the substrate specificity of large neutral amino acid carrier (LNAA) and di/tripeptide (hPEPT1) transporters with respect to PD 158473, an NMDA antagonist. METHODS: Cellular uptake studies were carried out using two types of Chinese Hamster Ovary (CHO). CHO-K1 cells represent the wild type with inherent large neutral amino acid (LNAA) activity. CHO-PEPT1 cells were generated by stable transfection of hPEPT1 gene into CHO cells. Therefore, these cells possess both LNAA activity and di/tripeptide transporter activities as a result of the transfection. Cellular uptake of PD 158473 was quantified using a HPLC method previously developed in our laboratory. RESULTS: The utility of the CHO-PEPT1 cell model was demonstrated by determining the uptake kinetics of Gly-Sar, a prototypical dipeptide transporter substrate. Uptake kinetics of PD 158473 displayed two carrier-mediated transport components in CHO-PEPT1 cells, while in CHO-K1 cells the relationship was consistent with classic one component Michaelis-Menten kinetics. These results confirmed the affinity of PD 158473 for both LNAA and di/tripeptide transporters. Further, results from inhibition experiments using these two cell types indicate that the high affinity-low capacity system was the LNAA carrier and the low affinity-high capacity carrier was the di/tripeptide transporter. CONCLUSIONS: This study demonstrates overlapping substrate specificity between LNAA carrier and di/tripeptide transporter (hPEPT1) for PD 158473, an amino acid analog. Establishing Structure Transport Relationship (STR) for this overlap will aid in a design strategy for increasing oral absorption or targeting specific drugs to selected tissues.

Resting dendritic cells induce peripheral CD8+ T cell tolerance through PD-1 and CTLA-4.

T cells recognizing self proteins exist without causing autoimmunity in healthy individuals. These autoreactive T cells are kept in check by peripheral tolerance. Using a model for peripheral CD8(+) T cell tolerance resulting from antigen presentation by resting dendritic cells in vivo, we show here that CD8(+) T cell tolerance operates through T cell-intrinsic mechanisms such as deletion or functional inactivation. Peripheral CD8(+) T cell tolerance depended on signaling via the costimulatory molecule PD-1, as an absence of PD-1 converted tolerance induction into priming. Blocking of the costimulatory molecule CTLA-4 resulted in impaired tolerance and enhanced the effect of the absence of PD-1, suggesting that PD-1 and CTLA-4 act synergistically. Thus PD-1 and CTLA-4 are crucial molecules for peripheral CD8(+) T cell tolerance induced by resting dendritic cells.

Direct IR observation of vibrational properties of carbonyl species formed on Pd nano-particles supported on amorphous carbon: comparison with Pd/SiO2-Al2O3.

By diluting optically opaque carbon-supported Pd particles in silica Aerosil we succeeded in observing the IR bands of adsorbed carbonyls and extracting information on the particle dispersion. Comparison with literature single crystal data and with silica-alumina supported Pd allowed us to make an assignment in terms of linear and 2-fold bridged carbonyls formed on Pd(111) and Pd(100) faces. Two Pd/C samples have been investigated. The relative intensities of the two carbonyl families observed on the two samples are consistent with the Pd dispersion independently measured with CO chemisorption, TEM and EXAFS analysis.

Insertion reactions of alkynes and organic isocyanides into the palladium-carbon bond of dimetallic Fe-Pd alkoxysilyl complexes.

Insertion of MeO(2)C-C[triple bond]C-CO(2)Me (DMAD) into the Pd-C bond of the heterodimetallic complex [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]d(dmba-C)] (2) (dppm = Ph(2)PCH(2)PPh(2), dmba-C = metallated dimethylbenzylamine) and [(OC)(3){(MeO)(3)Si}F[upper bond 1 start]e(mu-dppm)P[upper bond 1 end]d(8-mq-C,N)] (3) (8-mq-C,N = cyclometallated 8-methylquinoline) yielded the sigma-alkenyl complexes [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]d{C(CO(2)Me)=C(CO(2)Me)(o-C(6)H(4)CH(2)NMe(2))}] (7) and [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]d{C(CO(2)Me)[double bond, length as m-dash]C(CO(2)Me)(CH(2)C(9)H(6)N)}] (8), respectively. The latter afforded the adduct [(OC)(3){(MeO)(3)Si}F[upper bond 1 start]e(mu-dppm)P[upper bond 1 end]d{C(CO(2)Me)=C(CO(2)Me)(CH(2)C(9)H(6)N)}(CNBu(t))] (9) upon reaction with 1 equiv. of Bu(t)NC. The heterodinuclear sigma-butadienyl complexes [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]d{C(Ph=C(Ph)C(CO(2)Me)=(CO(2)Me)(o-C(6)H(4)CH(2)NMe(2))}] (11) and [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]d{C(Ph)=C(CO(2)Et)C(Ph)=C(CO(2)Et)(CH(2)C(9)H(6)N)}] (13) have been obtained by reaction of the metallate K[Fe{Si(OMe)(3)}(CO)(3)(dppm-P)] (dppm = Ph(2)PCH(2)PPh(2)) with [P[upper bond 1 start]dCl{C(Ph)=C(Ph)C(CO(2)Me)=C(CO(2)Me)(o-C(6)H(4)CH(2)N[upper bond 1 end]Me(2))}] or [P[upper bond 1 start]dCl{C(Ph)=C(CO(2)Et)C(Ph)=(CO(2)Et)}(CH(2)C(9)H(6)N[upper bond 1 end])], respectively. Monoinsertion of various organic isocyanides RNC into the Pd-C bond of 2 and 3 afforded the corresponding heterometallic iminoacyl complexes. In the case of complexes [(OC)(3){(MeO)(3)Si}F[upper bond 1 start]e(mu-dppm)P[upper bond 1 end][upper bond 1 start]d{C=(NR)(CH(2)C(9)H(6)N[upper bond 1 end])}] (15a R = Ph, 15b R = xylyl), a static six-membered C,N chelate is formed at the Pd centre, in contrast to the situation in [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]d{C(=NR)(o-C(6)H(4)CH(2)NMe(2))}] (14a R = o-anisyl, 14b R = 2,6-xylyl) where formation of a mu-eta(2)-Si-O bridge is preferred over NMe(2) coordination. The outcome of the reaction of the dimetallic alkyl complex [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]dMe] with RNC depends both on the stoichiometry and the electronic donor properties of the isocyanide employed for the migratory insertion process. In the case of o-anisylisocyanide, the iminoacyl complex [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]d{C(=N-o-anisyl)Me}] (16) results from the reaction in a 1 : 1 ratio. Addition of three equiv. of o-anisylisocyanide affords the tris(insertion) product [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]d{[C(=N-o-anisyl)](3)Me}] (18). After addition of a fourth equivalent of o-anisylNC, exclusive formation of the isocyanide adduct [(OC)(3){(MeO)(3)Si}F[upper bond 1 start]e(mu-dppm)P[upper bond 1 end]d{[C(=N-o-anisyl)](3)Me}(CN-o-anisyl)] (19) was spectroscopically evidenced. In the complex [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]d{[C(=N-o-C(6)H(4)COCH(2))](2)Me}] (20), the sigma-bound diazabutadienyl unit is part of a 12-membered organic macrocyle which results from bis(insertion) of 1,2-bis(2-isocyanophenoxy)ethane into the Pd-Me bond of the precursor complex [(OC)(3)F[upper bond 1 start]e{mu-Si(OMe)(2)([lower bond 1 start]OMe)}(mu-dppm)P[lower bond 1 end][upper bond 1 end]dMe]. In contrast, addition of two equivalents of tert-butylisocyanide to a solution of the latter afforded [(OC)(3){(MeO)(3)Si}F[upper bond 1 start]Fe(mu-dppm)P[upper bond 1 end]d{C(=NBu(t))Me}(CNBu(t))] (21) in which both a terminal and an inserted isocyanide ligand are coordinated to the Pd centre. In all cases, there was no evidence for competing CO substitution at the Fe(CO)(3) fragment by RNC. The molecular structures of the insertion products 8 x CH(2)Cl(2) and 16 x CH(2)Cl(2) have been determined by X-ray diffraction.

The p42/44mitogen-activated protein kinase inhibitor PD 98059, but not U 0126, increases a K+ current in cardiomyocytes.

1. The effects of the mitogen-activated protein kinase (MAPK) inhibitors PD 98059 and U 0126, useful tools to investigate MAPK involvement in intracellular signal transduction pathways, were assessed on cardiomyocytes. 2. In rat freshly isolated ventricular myocytes, under current-clamp conditions, PD 98059 (40 micro mol/L) shortened the action potential. Under whole-cell patch-clamp, this compound slowly induced a fast activating sustained outward K+ current that was sensitive to 1 mmol/L Ba2+, 100 micro mol/L Gd3+, 3 mmol/L 4-aminopyridine and 100 micro mol/L tetracain. The PD 98059-induced current was prevented by 40 micro mol/L AACOCF3, a cytosolic phospholipase A2 inhibitor. 3. U 0126 (1 micro mol/L), a recently developed highly potent p42/44 MAPK inhibitor, did not alter K+ currents. 4. PD 98059, but not U 0126, increased arachidonic acid content, probably as a consequence of its reported cyclo-oxygenase inhibitory effect. 5. These observations indicate that PD 98059 activates a TREK-1 like current. Thus, this MAPK inhibitor has to be used with caution because alterations in cell metabolism can be secondary to changes in electrophysiological behaviour.

The FGFR1 inhibitor PD 173074 selectively and potently antagonizes FGF-2 neurotrophic and neurotropic effects.

Basic fibroblast growth factor (FGF-2) promotes survival and/or neurite outgrowth from a variety of neurons in cell culture and regenerative processes in vivo. FGFs exert their effects by activating cell surface receptor tyrosine kinases. FGF receptor (FGFR) inhibitors have not been characterized on neuronal cell behaviors to date. In the present study, we show that the FGFR1 inhibitor PD 173074 potently and selectively antagonized the neurotrophic and neurotropic actions of FGF-2. Nanomolar concentrations of PD 173074 prevented FGF-2, but not insulin-like growth factor-1, support of cerebellar granule neuron survival under conditions of serum/K(+) deprivation; another FGF-2 inhibitor, SU 5402, was effective only at a 1,000-fold greater concentration. Neither PD 173074 nor SU 5402, at 100 times their IC(50) values, interfered with the survival of dorsal root ganglion neurons promoted by nerve growth factor, ciliary neurotrophic factor, or glial cell line-derived neurotrophic factor. PD 173074 and SU 5402 displayed 1,000-fold differential IC(50) values for inhibition of FGF-2-stimulated neurite outgrowth in PC12 cells and in granule neurons, and FGF-2-induced mitogen-activated protein kinase (p44/42) phosphorylation. The two inhibitors failed to disturb downstream signalling stimuli of FGF-2. PD 173074 represents a valuable tool for dissecting the role of FGF-2 in normal and pathological nervous system function without compromising the actions of other neurotrophic factors.

Acid-base profile in patients on PD.

Acid-base profile in patients on PD. Secular trends in dialysis dose in peritoneal dialysis (PD) and modes of dialysis delivery [automated PD (APD) versus continuous ambulatory PD (CAPD)] require a reexamination of acid-base status in patients treated with these renal replacement modalities. We explored steady-state acid-base profile and its determinants in 175 patients on CAPD and 77 patients on APD. The majority (62% to 70%) of patients had serum bicarbonate levels in the normal range, and a minority (17% to 27%) had values just above 28 mEq/L. Only a small percentage (10% to 12%) of patients in either the CAPD or the APD groups had a serum HCO3 less than 22 mEq/L, an indication of the successful correction of acidosis in most patients. The anion gap was elevated (> 16 mEq/L) in the majority of patients on CAPD and APD and bore an inverse relationship to serum HCO3 and a direct relationship to serum albumin and serum phosphate. In CAPD patients, but not APD patients, a significant inverse relationship was observed between the anion gap and peritoneal permeability as assessed by four-hour D/P(creatinine). The correction of acidosis in PD appears to be predominantly achieved by the continuous supplementation of alkali via dialysis, with residual renal function not differentiating the degree of correction. Steady-state serum bicarbonate in patients on CAPD appeared to be responsive to the underlying peritoneal membrane permeability characteristics of the patient that govern alkali loss and gain, but the higher dialysate volumes in APD appear to override this effect. Higher albumin, blood urea nitrogen (BUN), and phosphate in patients with lower HCO3 suggest a discrepancy between daily acid load and dialysis dose.

State-to-state scattering of D2 from Cu(100) and Pd(111).

Results from state resolved experiments are presented for the interaction of D2(v=1,J=2) with Cu(100) and Pd(111). The reflected molecules were probed using quantum state specific spectroscopy. For D2 scattered from Cu(100) the vibrational survival probability and some transition inelastic probabilities were measured for incident energies from 70-200 meV. The survival probability was found to be larger then that found previously for H2(v=1) scattered from the same surface; these differences are discussed in terms of the lower zero point energy and smaller vibrational energy spacings of D2. D2 translational energy exchange was studied for several different scattering channels and interpreted using simple classical calculations. The survival probability was also measured for D2(v=1) scattered from Pd(111) at one incident energy. Pd is reactive for D2 dissociation and this survival probability was measured to be small and also to be much smaller than that for H2(v=1) under similar conditions. Vibrational relaxation channels were studied for D2 scattering from both Cu(100) and Pd(111). The vibrational relaxation probability on both surfaces was also found to be smaller than that measured for comparable channels for H2. The smaller survival probability and vibrational relaxation probability for D2 on Pd(111) cannot be easily accounted for by the difference in zero point energy and vibrational energy spacings.

Studies on the anti-tumor-promoting activity of naturally occurring substances. IV. Pd-II [(+)anomalin, (+)praeruptorin B], a seselin-type coumarin, inhibits the promotion of skin tumor formation by 12-O-tetradecanoylphorbol-13-acetate in 7,12-dimethylbenz[a]anthracene-initiated mice.

Since Pd-II [(+)anomalin, (+)praeruptorin B], a seselin-type coumarin, was found to inhibit tumor promoter induced phenomenon in vitro, the effect of Pd-II on the in vivo tumor-promoting action of 12-O-tetradecanoylphorbol-13-acetate (TPA) in 7,12-dimethylbenz[a]anthracene-initiated mouse skin was investigated. Pd-II, applied 40 min before the TPA treatment, at a dose of 10 mumol/painting, completely suppressed tumor formation up to 20 weeks of tumor promotion, without any toxicity. Besides Pd-II, various anti-tumor-promoter coumarins were found in the traditional Chinese medicine Qian-Hu, from which Pd-II was obtained. These coumarins may be useful for the development of an effective method to prevent cancer.

Suppression of expression and function of negative immune regulator PD-1 by certain pattern recognition and cytokine receptor signals associated with immune system danger.

Stimulation of certain cytokine and pattern recognition receptors enhances adaptive immune responses, and in chronic situations, may play a role in the loss of self-tolerance. We hypothesized that in addition to upregulating positive immune receptors (i.e. co-stimulatory molecules), certain cytokine and pattern recognition signals might downregulate negative immune receptors, removing a potential barrier to lymphocyte responsiveness. The newly identified CD28 family member Programmed Death-1 (PD-1) is an inhibitory receptor involved in peripheral tolerance, as evidenced by the frank autoimmunity and autoantibody formation found in PD-1-deficient mice. Here we report that antigen-receptor induced PD-1 expression on murine B cells is markedly reduced by certain signals associated with immune system danger, including LPS, CpG oligodeoxynucleotides and several pro-inflammatory cytokines, through distinct signaling pathways. We further report for the first time that engagement of PD-1 inhibits cell cycle progression in primary B cells and that modulation of PD-1 expression by CpG or IL-4 significantly reverses such inhibition. Our data suggest a novel mechanism for enhancement of normal immune responses and disruption of normal tolerance mechanisms.

Self-assembly of one-dimensional surface structures: long-range interactions in the growth of Ir and Pd on W(110).

Self-assembly of one-dimensional surface structures is examined by tracking single Ir and Pd atoms on W(110) as they incorporate into chains of Ir and Pd, respectively. Ir adatoms move parallel to the chains, but do not come close to the chain sides; incorporation occurs only at the ends. Pd adatoms also migrate parallel to Pd chains, and incorporate at the ends. Occasionally, in the end region, they do jump to the side of a chain, and migrate there until they find an end site. Incorporation behavior for both Ir and Pd is driven by highly anisotropic, long-ranged interactions between atoms on the surface.

[Pd(CH3COO)2]n from X-ray powder diffraction data.

The water-insoluble title compound, catena-poly[palladium(II)-di-mu-acetato-kappa4O:O'], [Pd(C2H3O2)2]n, was obtained from a nitratopalladium solution and acetic acid as a pale-pink powder. Ab initio crystal structure determination was carried out using X-ray powder diffraction techniques. Patterson and Fourier syntheses were used for atom location and the Rietveld technique was applied for the final structure refinement. The structure consists of palladium acetate complexes connected into polymeric chains running along b, in which two Pd atoms are bridged by two acetate groups that are in a cis configuration with respect to one another. The unique Pd atom lies on a site with 2/m symmetry and the acetate moieties have imposed m symmetry; these are joined into infinite chains running along the b direction. The shortest Pd...Pd distance in the row is 2.9192 (1) A. The planes of adjacent palladium complexes are inclined towards each other, the angle between the planes being approximately 30 degrees.

PIP3EA and PD-168077, two selective dopamine D4 receptor agonists, induce penile erection in male rats: site and mechanism of action in the brain.

PIP3EA (2-[4-(2-methoxyphenyl)piperazin-1-yl-methyl]imidazo[1,2-a]pyridine) and PD-168077 (N-[4-2-cyanophenylpiperazin-1-ylmethyl]-3-methylbenzamide maleate), two selective dopamine D4 agonists, administered systemically, intracerebroventricularly or into the paraventricular nucleus of the hypothalamus induce penile erection in male Sprague-Dawley rats. A U-inverted dose-response curve was found with either compound when given subcutaneously (1-100 microg/kg) or intracerebroventricularly (0.1-20 microg/rat), but not into the paraventricular nucleus (10-200 ng/rat). The pro-erectile effect of PIP3EA and of PD-168077 occurs concomitantly with an increased nitric oxide (NO) production in the paraventricular nucleus, as measured by the increased concentration of nitrites and nitrates found in the dialysate obtained from the paraventricular nucleus by intracerebral microdialysis. These effects of PIP3EA and PD-168077 were reduced by L-745,870 (3-[4-(4-chlorophenyl)piperazin-1-ylmethyl]-1H-pyrrolo[2,3-b]pyridine trihydrochloride), a selective dopamine D4 receptors antagonist, by omega-conotoxin, a blocker of voltage-dependent Ca2+ channels of the N-type, by S-methyl-thiocitrulline, a neuronal nitric oxide synthase inhibitor, and by d(CH2)5Tyr(Me)2-Orn8-vasotocin, an oxytocin receptor antagonist, given into the lateral ventricles, but not into the paraventricular nucleus. Comparison of the dose-response curves of PIP3EA and PD-168077 revealed that PIP3EA is as potent as PD-168077 when given into the paraventricular nucleus, but more potent when given systemically. However, both compounds are less efficacious (e.g. induce a lower number of penile erection episodes) than apomorphine, a classical mixed dopamine receptor agonist, irrespective of the route of administration. These results confirm previous findings showing that central D4 receptors mediate penile erection and show that dopamine D4 receptor agonists act in the paraventricular nucleus to facilitate penile erection by increasing central oxytocinergic neurotransmission.

A new glucose 6-phosphate dehydrogenase variant (G-6-PD Verona) in a patient with myelodysplastic syndrome.

A 67-year-old woman investigated because of 'myelodysplastic syndrome' was found to have a 4-fold increase in G-6-PD activity in her erythrocytes. The enzyme was partially purified and characterized. On grounds of: (a) reduced electrophoretic mobility, (b) abnormal cathodic band(s) in isoelectrofocusing, (c) increased Michaelis constant for glucose 6-phosphate, (d) abnormal thermostability, and (e) abnormal interaction with the ligand NADPH, we conclude that this is a new structural variant which we designate G-6-PD Verona. G-6-PD Verona was the sole apparent source of G-6-PD activity in the patient's erythrocytes; by contrast, the patient's fibroblasts had only normal G-6-PH (type B). The patient's haematological course terminated into acute myeloid leukaemia. We believe G-6-PD Verona was the result of a somatic mutation in an X-chromosome which took place in a haemopoietic cell clone which subsequently underwent neoplastic transformation.

Protection against cocaine toxicity in mice by the dopamine D3/D2 agonist R-(+)-trans-3,4a,10b-tetrahydro-4-propyl-2H,5H-[1]benzopyrano[4,3-b]-1,4-oxazin-9-ol [(+)-PD 128,907].

Cocaine abuse is a public health concern with seizures and death being one consequence of overdose. In the present study, dopamine D(3/)D(2) receptor agonists dose dependently and completely prevented the convulsant and lethal effects of cocaine. The D(3)-preferring agonists R-(+)-trans-3,4a,10b-tetrahydro-4-propyl-2H,5H-[1]benzopyrano[4,3-b]-1,4-oxazin-9-ol) [(+)-PD 128,907], (+)-7-hydroxy-dipropylaminotetralin, and the mixed D(3/)D(2) agonists quinpirole and quinelorane were all effective against cocaine toxicity in mice. The anticonvulsant effects of these compounds occurred at doses below those that produced motor impairment as assessed in the inverted screen test. Protection against the convulsant effects of the selective dopamine uptake inhibitor 1-[2-[bis(4-fluorophenyl)methoxy] ethyl]-4-[3-phenyl-propyl]piperazine (GBR 12909) was also conferred by (+)-PD 128,907. The possible selectivity of the effects of (+)-PD 128,907 (3 mg/kg) for these dopaminergic compounds was demonstrated by its general lack of protective efficacy against a host of convulsants acting through other neural mechanisms [pentylenetetrazol, (+)-bicuculline, and picrotoxin, 4-aminopyridine, and t-butylbiclyclophosphoorothionate, N-methyl-d-aspartate, kainate, pilocarpine, nicotine, strychnine, aminophylline, threshold electric shock, and 6-Hz electrical stimulation]. Direct and correlational evidence suggests that these effects were mediated by D(3) receptors. Protection was stereospecific and reversible by an antagonist of D(3) receptors [3-[4[1-(4-[2[4-(3-diethyamino-propoxy)-phenyl]-benzoimidazol-l-yl]-butyl)-1H-benzoimidazol-2-yl]-phenoxy]-propyl)-diethyl-amine; PD 58491] but not D(2) receptors [3[[4-(4-chlorophenyl)-4 hydroxypipeidin-1-yl]methyl-1H-indole; L-741,626]. Anticonvulsant potencies were positively associated with potencies in a functional assay of D(3) but not D(2) receptor function. Together, these findings suggest that the prevention of cocaine convulsions and lethality by (+)-PD 128,907 may be due to D(3) receptor-mediated events.

In vitro susceptibilities of Mycoplasma pneumoniae, Mycoplasma hominis, and Ureaplasma urealyticum to sparfloxacin and PD 127391.

The in vitro activities of two investigational quinolones, sparfloxacin (previously designated AT 4140) and PD 127391, were determined for 30 strains each of Mycoplasma pneumoniae, Mycoplasma hominis, and Ureaplasma urealyticum and compared with those of ciprofloxacin, tetracycline, clindamycin, and erythromycin. Erythromycin was the most active compound against M. pneumoniae (maximum MIC, less than 0.008 microgram/ml). PD 127391 (MICs, less than 0.008 to 0.031 microgram/ml), sparfloxacin (MICs, less than 0.008 to 0.25 microgram/ml), clindamycin (MICs, less than 0.008 to 0.5 microgram/ml), and tetracycline (MICs, 0.063 to 0.25 microgram/ml) were superior to ciprofloxacin (MICs, 0.5 to 2 microgram/ml). Sparfloxacin and PD 127391 were active against M. hominis (MICs, less than 0.008 to 0.031 microgram/ml for each) at concentrations comparable to those of clindamycin (MICs, less than 0.008 to 0.063 microgram/ml) and at concentrations lower than those of ciprofloxacin (MICs, 0.125 to 0.5 microgram/ml). As expected, M. hominis was resistant to erythromycin (MICs, 32 to greater than or equal to 256 micrograms/ml). For U. urealyticum, PD 127391 (MICs, 0.031 to 0.5 microgram/ml) and sparfloxacin (MICs, 0.063 to 1 microgram/ml) were superior to erythromycin (MICs, 0.25 to 4 micrograms/ml), ciprofloxacin (MICs, 0.5 to 8 micrograms/ml), and clindamycin (MICs, 0.25 to 64 micrograms/ml. Both new quinolones were equally active against tetracycline-susceptible as well as resistant strains of M. hominis and U. urealyticum. The possible influence of medium components and/or pH on MICs was evaluated by testing a Staphylococcus aureus reference strain with each antibiotic in SP-4 broth and 10-B broth and comparing the results with published MICs for this strain. MICs determined in 10-B broth for erythromycin were affected most. This study shows that the activities of sparfloxacin and PD 127391 are similar to one another and comparable or superior to those of other drugs used to treat mycoplasmal infections. The MICs of both new quinolones were consistently 2 to several dilutions lower than those of ciprofloxacin for each species.
