Biological activity of particle exhaust emissions from light-duty diesel engines.

Whole diesel exhaust has been classified recently as a probable carcinogen, and several genotoxicity studies have found particulate exhaust to be clearly mutagenic. Moreover, genotoxicity of diesel particulate is greatly influenced by fuel nature and type of combustion. In order to obtain an effective environmental pollution control, combustion processes using alternative fuels are being analyzed presently. The goal of this study is to determine whether the installation of exhaust after treatment-devices on two light-duty, exhaust gas recirculation (EGR) valve-equipped diesel engines (1930 cc and 2500 cc) can reduce the mutagenicity associated with particles collected during U.S.A. and European driving cycles. Another interesting object was to compare the ability of alternative biodiesel and conventional diesel fuels to reduce the mutagenic activity associated with collected particles from two light duty diesel engines (both 1930 cc) during the European driving cycle. SOF mutagenicity was assayed using the Salmonella/microsome test (TA 98 and TA 100 strains, +/- S9 fraction). In the first part of our study, the highest mutagenicity was revealed by TA98 strain without enzymatic activation, suggesting a direct-acting mutagenicity prevalence in diesel particulate. The 2500 cc engine revealed twofold mutagenic activity compared with the 1930 cc engine (both EGR valve equipped), whereas an opposite result was found in particulate matter amount. The use of a noncatalytic ceramic trap produced a decrease of particle mutagenic activity in the 2500 cc car, whereas an enhancement in the 1930 cc engine was found. The catalytic converter and the electrostatic filter installed on the 2500 cc engine yielded a light particle amount and an SOF mutagenicity decrease. A greater engine stress was obtained using European driving cycles, which caused the strongest mutagenicity/km compared with the U.S.A. cycles. In the second part of the investigation, even though a small number of assays were available, exhaust emission generation by biodiesel fuel seemed to yield a smaller environmental impact than that of the referenced diesel fuel. The results point out the usefulness of mutagenicity testing in the research of both newer, more efficient automotive aftertreatment devices and less polluting fuels.

Female lung cancer and petrochemical air pollution in Taiwan.

To investigate the relationship between petrochemical air pollution and female lung cancer, we conducted a matched case-control study among women who had died in Taiwan from 1990 through 1994. Data about all eligible female lung cancer deaths were obtained from the Bureau of Vital Statistics of the Taiwan Provincial Department of Health. The control group included women who died from nonneoplasms and diseases that were not associated with respiratory problems. We pair-matched the controls to the cases by sex, year of birth, and year of death. Each matched control was selected randomly from the set of possible controls for each case. We used the proportion of a municipality's total population employed in the petrochemical manufacturing industry as an indicator of a resident's exposure to air emissions from the petrochemical manufacturing industry. The subjects were divided into tertiles according to the above indicator. Women who lived in the 2 groups of municipalities characterized by higher levels of petrochemical pollution had a statistically significant higher risk of developing lung cancer than the group that lived in municipalities with the lowest petrochemical air pollution levels (after controlling for possible confounders). The linear trend was also statistically significant (p < .05). The results of this study shed important light on the relationship between the Taiwan petrochemical industry and the resulting risk to human health.

Economic and environmental analysis of retrofitting a large office building with energy-efficient lighting systems.

An evaluation of the economic and environmental costs and benefits that would result if the Zorinsky Federal Building, located in Omaha, Nebraska, USA, converted its current lighting system to a more energy-efficient system (i.e., joined the EPA's Green Lights Program) was conducted. Lighting accounts for 20-25 percent of all electricity sold in the United States. Costs considered in the study included the cost of retrofitting the building's existing lighting system and the cost of disposal of the current lamps and ballast fixtures. Benefits included a reduction of electric utility costs and a reduction of emissions of SO2, NOx, CO2, and CO from electric utility power plants. Environmental and health issues for air pollutant emissions were also addressed. The results showed that significant reductions in utility bills as well as reductions in air emissions would result from a major building converting to a more energy efficient lighting system. The results showed that conversion of this large building would reduce SO2 emissions by 14.6 tons/yr and NOx emissions by 6.3 tons/yr. In addition, the conversion would reduce annual energy costs by approximately $114,000.

Catalyzed UV oxidation of organic pollutants in biologically treated wastewater effluents.

A batch reactor was used to evaluate the efficiency of advanced oxidation process of the organic pollutants in biologically treated wastewater effluents with UV/H2O2. A 450-W high-pressure mercury vapor lamp was used as the light source. During the degradation process, the concentration of the dissolved organic compounds could be increased by more than twofold due to the decomposition of microorganisms. This increase of the dissolved organic compounds was eliminated if the water was filtered before the photodegradation experiments. It is observed that the UV alone could play a role for the oxidation of the organic pollutants; however, the addition of a small amount of hydrogen peroxide promotes the degradation efficiency of organic compounds in wastewater. The best oxidation efficiency was obtained when the water samples were under acidic conditions (pH 5), and the rate of degradation was not enhanced with the increasing H2O2 dosages. The optimum H2O2 dose was between 0.01% and 0.1% for the oxidation processes in this study. The presence of the carbonate/bicarbonate ions in water inhibits the degradation of the organic compounds.

Definition and GIS-based characterization of an integral risk index applied to a chemical/petrochemical area.

A risk map of the chemical/petrochemical industrial area of Tarragona (Catalonia, Spain) was designed following a two-stage procedure. The first step was the creation of a ranking system (Hazard Index) for a number of different inorganic and organic pollutants: heavy metals, polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), polychlorinated biphenyls (PCBs) and polychlorinated aromatic hydrocarbons (PAHs) by applying self-organizing maps (SOM) to persistence, bioaccumulation and toxicity properties of the chemicals. PCBs seemed to be the most hazardous compounds, while the light PAHs showed the minimum values. Subsequently, an Integral Risk Index was developed taking into account the Hazard Index and the concentrations of all pollutants in soil samples collected in the assessed area of Tarragona. Finally, a risk map was elaborated by representing the spatial distribution of the Integral Risk Index with a geographic information system (GIS). The results of the present study seem to indicate that the development of an integral risk map might be useful to help in making-decision processes concerning environmental pollutants.

Toluene removal from air by Dieffenbachia in a closed environment.

Higher plants are likely to play a major role in bioregeneration systems for food, air and water supplies. Plants may also contribute by the removal of toxic organic substances from the air of a closed environment. Dieffenbachia amoena plants were exposed to 0 to 1.2 x 10(6) micrograms toluene m-3 at light intensities of 35 and 90 micromoles m-2 s-1 in sealed chambers. Toluene removal, photosynthesis and respiration were measured. An increased light intensity increased the rate of toluene removal five-fold over the rate at the lower intensity; the kinetics suggest active regulation by the plant. The removal rate saturated at 2700 micrograms toluene h-1 at the lower intensity and failed to saturate at the higher intensity. Toluene exposure inhibited photosynthesis and respiration only transiently and without correlation to toluene concentration. These plants can act as efficient scavengers of toluene in a contaminated environment.

A rapid method for detection of nongenotoxic carcinogens of environmental pollutants using synchronized V79 cells and flow cytometry.

Synchronized V79 cells were treated before entering mitosis with known and suspicious mitotic arrestants and analyzed by flow cytometry and by light microscopy. Colcemid, nocodazole, vinblastine, diethylstilbestrol, triethyl lead and cadmium sulfate caused a dose dependent mitotic arrest of up to 80%, in comparison with 6% for the controls. Mixtures of polycyclic aromatic hydrocarbons and heterocyclic compounds induced a mitotic arrest of 50%-60%. Extracts of airborne particulates revealed a mitotic arrest of 10%-40%. In contrast, benzoquinone and hydroquinone led to a G2-block rather than to a mitotic arrest. Results of flow cytometry measurements correlated well with those obtained by light microscopy. Cell synchronization in combination with flow cytometry seems to be of considerable value as a rapid method for testing nongenotoxic agents with mitotic arresting activity.

Morphofunctional evidence of changes in principal and mitochondria-rich cells in the epidermis of the frog Rana kl. esculenta living in a polluted habitat.

The epidermis of vertebrates is the body's principal barrier against environment and its possible contaminants. The presence of keratins, as well as specific detoxifying molecules or enzyme activities, in the various epidermis layers is believed to be involved in providing protection from harmful environmental influences. Anuran integument is poorly hornified and thus permeable to some endogenous and exogenous compounds and thus serves as a good bioindicator of overall environmental conditions. In the present investigation, we studied the epidermis of Rana kl. esculenta adult specimens collected at two different rice fields, relatively unpolluted and heavily polluted, respectively. Environmental pollution was assayed by chemical analysis performed on both sediments and animals. We evaluated the structural aspects of the epidermis at both light and electron microscopy levels and the pattern of keratinization by immunohistochemistry. Furthermore, we studied the activities of some enzymes (acid and alkaline phosphatase, nitric oxide synthase-related nicotinamide adenine dinucleotide phosphate dehydrogenase, glucose-6-phosphate dehydrogenase, catalase, nonspecific esterases, and succinic dehydrogenase) involved mainly in membrane transport, xenobiotics, and oxidative metabolism. Compared with controls, in polluted animals we found the following results: (1) an increase in pollutant levels (i.e., cadmium, mercury, and lead); (2) less keratinized superficial cells in the epidermis; and (3) changes in most enzyme activities in keratinocytes and mitochondria-rich cells (particularly glucose-6-phosphate dehydrogenase and esterases, both important to counteract oxidative and toxic stress). Taken as a whole, the present data indicate the morphofunctional plasticity of the frog epidermis in response to environmental contamination.

Assessing cytotoxicity of photosensitized transformation products of 2,4,6-trinitrotoluene (TNT) and atrazine with freshwater microbial assemblages.

In this study of riboflavin-sensitized photolysis of 2,4,6-trinitrotoluene (TNT), the final photoproducts were found to include 2-amino-4,6-dinitrotoluene, 4-amino-2,6-dinitrotoluene, and 3,5-dinitroaniline. After exposure to TNT (10 mg/L) for 90 min, in a river water sample there was inhibition of the viability count of heterotrophic bacterial assemblages by 28.3% and 24.3% and of bacterial heterotrophic mineralization of glucose by 99.5% and 93.6% relative to the light control and dark control groups, respectively. After exposure separately to 10 mg/L 2-amino-4,6-dinitrotoluene, 4-amino-2,6-dinitrotoluene, or 3,5-dinitroaniline, the viability count of heterotrophic bacterial assemblages was enhanced by 30.0%, 98.2%, and 148.7%, respectively, relative to the TNT group in light and by 12.2%, 40.2%, and 36.5%, respectively, relative to TNT group in dark. After exposure to the same aforementioned test chemicals, heterotrophic activity of bacterial assemblages was inhibited by 75.7%, 72.4%, and 56.0%, respectively, relative to light control group, and there were no significant changes in the dark. A similar study was conducted with atrazine (10 mg/L). The main products of atrazine riboflavin-sensitized phototransformation include desethylatrazine (DEA) and desisopropylatrazine (DIA). It was found that DEA and DIA did not inhibit microbial heterotrophic activity. However, after 72 h the viable count was enhanced by 52.8% and 63.3% in the DEA and DIA exposure groups, respectively, in comparison to the control. These results suggested that photosensitized transformation decreased TNT and atrazine cytotoxicity to microbial assemblages in the natural water. Photoproducts of atrazine--DEA and -DIA, and of TNT could become growth substrates for bacterial assemblages in natural water.

Phototoxicology. 2. Near-ultraviolet light enhancement of Microtox assays of trinitrotoluene and aminodinitrotoluenes.

Coexposure of 2,4,6-trinitrotoluene (TNT), 2-amino-4,6-dinitrotoluene (2A), or 4-amino-2,6-dinitrotoluene (4A) to near-ultraviolet (nuv) light (lambda max-354 nm) significantly enhanced their toxicity toward Photobacterium phosphoreum (Microtox bioassay) during 30 min but not 15 min. Based on the slopes of the dose-response lines, the nuv coexposure and dark toxic mechanisms of action for TNT, 2A, and 4A appeared to be similar. nuv coexposure of binary mixtures significantly enhanced (supraadditivity) the toxicity of these compounds to P. phosphoreum. Under normal laboratory lighting, the toxicity of TNT + 2A and 2A + 4A mixtures were supraadditive but the toxicity of TNT + 4A mixtures could be explained by simple addition. Supporting these conclusions, the response curves of alpha-terthienyl, a compound known not to require nuv for toxicity, were similar in the dark and with nuv coexposure. In contrast, angelicin and psoralen, compounds known to require nuv coexposure to damage DNA, gave response curves having different slopes in the dark and with nuv coexposure. The nuv coexposure Microtox assay was able to detect and quantify phototoxicity in psoralen, angelicin, alpha-terthienyl, anthracene, TNT, and aminodinitrotoluenes.

The alarm reaction of coho salmon parr is impaired by the carbamate fungicide IPBC.

To determine whether the carbamate fungicide IPBC alters the olfactory-mediated behavioral and physiologic alarm responses of coho salmon parr (Oncorhynchus kisutch), groups of coho were exposed to skin extract (an alarm pheromone source) under a variety of conditions. In the 3min following skin extract exposure, freezing behavior was significantly increased (In the 3 min following skin extract exposure, freezing behavior was significantly increased under darkness (IR lighting) but not ambient lighting (25.3+/-2.6% and 7.5+/-5.7%, respectively; Delta calculated as: [(time (s) after/time (s) before)-1]x100%), and so IR was used for further experiments. Physiologically, following skin extract exposure, plasma cortisol concentration was increased at 0.5h (58.1+/-14.6ng/ml versus 4.32+/-1.31ng/ml, exposed versus control), hematocrit (Hct) was increased at 2h (50.4+/-1.0% versus 41.7+/-1.6%), and leucocrit (Lct) was decreased at 0.5 and 2h (0.534+/-0.114 and 0.13+/-0.01% versus 1.23+/-0.20%). After 0.5h exposures to 0, 1, 10 and 100microg/l IPBC and skin extract, the time spent dashing (>5cm/s) increased significantly (323+/-118%) in the first minute after skin extract exposure, but was absent in IPBC-exposed coho. Freezing behavior increased after skin extract exposure with control and 1microg/l IPBC exposures (11.0+/-3.0% and 17.7+/-11.0%, respectively), but was absent after 10microg/l and decreased after 100microg/l IPBC. Physiologically, Hct and plasma lactate concentration were significantly increased above controls after 1microg/l IPBC exposure (Hct: 45.7+/-1.6% versus 34.0+/-1.6%, lactate: 12.8+/-1.2mM versus 3.30+/-1.2mM). After 10microg/l exposure, IPBC alone elicited a stress response similar to skin extract. However in the 100microg/l treatment group the stress parameters were not different from controls. These findings suggest that the behavioral and physiologic alarm responses of juvenile salmonids may be impaired by acute exposure to > or =1microg/l IPBC.

Comparative eco-toxicity of nanoscale TiO2, SiO2, and ZnO water suspensions.

The potential eco-toxicity of nanosized titanium dioxide (TiO(2)), silicon dioxide (SiO(2)), and zinc oxide (ZnO) water suspensions was investigated using Gram-positive Bacillus subtilis and Gram-negative Escherichia coli as test organisms. These three photosensitive nanomaterials were harmful to varying degrees, with antibacterial activity increasing with particle concentration. Antibacterial activity generally increased from SiO(2) to TiO(2) to ZnO, and B. subtilis was most susceptible to their effects. Advertised nanoparticle size did not correspond to true particle size. Apparently, aggregation produced similarly sized particles that had similar antibacterial activity at a given concentration. The presence of light was a significant factor under most conditions tested, presumably due to its role in promoting generation of reactive oxygen species (ROS). However, bacterial growth inhibition was also observed under dark conditions, indicating that undetermined mechanisms additional to photocatalytic ROS production were responsible for toxicity. These results highlight the need for caution during the use and disposal of such manufactured nanomaterials to prevent unintended environmental impacts, as well as the importance of further research on the mechanisms and factors that increase toxicity to enhance risk management.

Effects of polydiallyldimethyl ammonium chloride coagulant on formation of chlorinated by products in drinking water.

The objectives of this research work was to evaluate the reduction of THM precursors by cationic p-DADMAC and determine the correlations between the chlorine demand and trihalomethane formation in the presence of electrolyte solutions and ambient light. The chlorine demand was found to be significantly reduced provided that the H2SO4 electrolyte was fed to the sample solutions. The amount of CHCl3 formation was also decreased when the Na2SO4 electrolyte was introduced in spite of the levels of light intensity. The p-DADMAC can not only effectively remove the turbidity but also reduce the formation of CHCl3. The optimum dosage of p-DADMAC for reducing the turbidity, TOC and CHCl3 in the humic acid and source water samples was determined and depended upon the nature of organics.

TiO2-induced heterogeneous photodegradation of a fluorotelomer alcohol in air.

Degradation of C4F9C2H4OH in air over TiO2 particles was examined in this first report of gas-solid heterogeneous photochemical degradation of fluorotelomer alcohols (FTOHs), which may be precursors of perfluorocarboxylic acids (PFCAs) in the environment. Photoirradiation (>290 nm) of C4F9C2H4OH in air flowing over TiO2 produced CO2, via C4F9CH2CHO, C4F9CHO, CnF(2n+1)COF (n=2 and/or 3), and COF2, in that order. X-ray photoelectron spectroscopy of the Ti02 surface showed a decrease in the amount of fluorine bonded to carbon and an increase in the amount of F- as the degradation of C4F9C2H4OH in air proceeded. Of the carbon content in the initial C4F9C2H4OH (78.8 ppmv), 90.7% was transformed to CO2, and the predominant fluorine species produced on the TiO2 surface was F-. Fluorotelomer unsaturated acids, which are considered to be toxic and have been observed in the biodegradation of FTOHs, did notform. Increased relative humidity in the air accelerated the decomposition of the reaction intermediates, which led to increased CO2 and F- formation. This result indicates that humidity is a key factor for counteracting FTOHs in indoor air. Although perfluoroalkyl substances such as PFCAs in water reportedly undergo little photodegradation over TiO2, our data show that mineralization of C4F9C2H4OH in air can be achieved with TiO2.
