Colonization of tissue surfaces in the gastrointestinal tract of gnotobiotic animals by lactobacillus strains.

Eight strains of lactobacilli isolated from conventional mice, rats, fowls, and pigs were tested for their ability to colonize squamous epithelia in the gastrointestinal tracts of gnotobiotic animals. Two of the strains isolated from rodents could colonize the nonsecretory epithelium of the stomach of gnotobiotic rodents, and one strain isolated from a fowl could colonize the crop epithelium of gnotobiotic chickens. The specificity of colonization of squamous epithelia by lactobacilli was demonstrated by these in vivo studies; strains of lactobacilli isolated from rodents could only colonize rodent epithelium; the strain from a fowl could only colonize crop epithelium.

Organogenesis from transformed tomato explants.

Tomato was one of the first crops for which a genetic transformation system was reported involving regeneration by organogenesis from Agrobacterium-transformed explants. Since the initial reports, various factors have been studied that affect the efficiency of tomato transformation and the technique has been useful for the isolation and identification of many genes involved in plant disease resistance, morphology and development. In this method, cotyledon explants from in vitro-grown seedlings are precultured overnight on a tobacco suspension feeder layer. The explants are then inoculated with Agrobacterium and returned to the feeder layer for a 2-d period of cocultivation. After cocultivation, the explants are transferred to an MS-based selective regeneration medium containing zeatin. Regenerated shoots are then rooted on a separate selective medium. This protocol has been used with several tomato cultivars and routinely yields transformation efficiencies of 10-15%.

Studies on the enteric absorption of selenium in the chick using localized coccidial infections.

Studies were conducted to determine the primary loci of gastrointestinal absorption of inorganic selenium in the chick using radiotracer techniques and Eimeria sp. infections of known site specificity for the gastrointestinal tract. Results showed that coccidial infections in the anterior regions of the small intestine reduced selenium absorption and potentiated selenium deficiency among chicks fed critical levels of the mineral. Absorption experiments showed that selenium absorption was greatest in the duodenum and anterior ileum, although selenium was bound at least transiently to tissues in the gizzard and crop. It is concluded from these experiments that dietary inorganic selenium is absorbed primarily in the duodenum and anterior ileum. Factors such as enteric disease which disrupt the integrity of the anterior intestinal mucosa can, therefore, reduce the absorption of selenium and other nutrients absorbed in that region, resulting in a potentiation of nutritional deficiencies.

Calcium metabolism in broiler breeder hens. 1. Calcium status of the digestive tract of broiler breeders throughout a 24-hour period.

Dry matter and calcium content of the digestive tract were determined on two groups of broiler breeders during a 24-hr period. The individually caged hens were fed either ad lib or restricted to 122 g of feed during a 4-hr feeding period each day from 0700 to 1100 hr. The hens received artificial light from 0700 to 2300 hr. On Day 14 of the feeding program a total of 100 hens, 10 from each group, were necropsied at selected intervals (0700, 1100, 1900, 2300, 0300 hr), and the contents of the various segments of the digestive tract and feces were collected and analyzed for dry matter and calcium. The calcium analysis also included any endogenous excretions and, therefore, absorption values are apparent. Hens restricted to 122 g feed per day had only 51.7% of the dry matter and 64.2% of the calcium consumed during the 4-hr feeding period remaining in the digestive tract at 1100 hr. By 2300 hr only 20.6% of the feed and 28.1% of the calcium remained. Hens had only approximately 14% (14.2 g) of their total dry matter intake and 16% (.70 g) of the calcium available for absorption from 0300 hr. The ad lib group, with an average intake of 171 g feed per day, had consumed only 26% of their daily feed intake by 1100 hr. Fifty-five percent of the dry matter consumed and 22% of the calcium remained in the digestive tract at 1100 hr. It was concluded that broiler hens restricted to morning feeding do not maintain a constant and equal metering of dry matter or calcium from the crop into the digestive tract throughout a 24-hr period.

Colonization of Lactobacillus acidophilus in gnotobiotic chicks.

Germ-free chicks hatched in flexible film gnotobiotic isolators were dosed orally at 2 days of age with a pure culture of Lactobacillus acidophilus. The chicks monoassociated with L. acidophilus were killed at 2 weeks of age to obtain gastrointestinal tract specimens for histological sectioning. Both light microscopy and transmission electron microscopy were performed to determine colonization and adhesion of lactobacilli. This strain of L. acidophilus was found associated with epithelium from three segments of the gastrointestinal tract (crop, proventriculus, and duodenum) of the gnotobiotic chick. The electron micrographs showed not only a close relationship between the Lactobacillus organism and the crop epithelia, but also an attachment through physical contact.

The components of crop productivity: measuring and modeling plant metabolism.

Several investigators in the CELSS program have demonstrated that crop plants can be remarkably productive in optimal environments where plants are limited only by incident radiation. Radiation use efficiencies of 0.4 to 0.7 g biomass per mol of incident photons have been measured for crops in several laboratories. Some early published values for radiation use efficiency (1 g mol-1) were inflated due to the effect of side lighting. Sealed chambers are the basic research module for crop studies for space. Such chambers allow the measurement of radiation and CO2 fluxes, thus providing values for three determinants of plant growth: radiation absorption, photosynthetic efficiency (quantum yield), and respiration efficiency (carbon use efficiency). Continuous measurement of each of these parameters over the plant life cycle has provided a blueprint for daily growth rates, and is the basis for modeling crop productivity based on component metabolic processes. Much of what has been interpreted as low photosynthetic efficiency is really the result of reduced leaf expansion and poor radiation absorption. Measurements and models of short-term (minutes to hours) and long-term (days to weeks) plant metabolic rates have enormously improved our understanding of plant environment interactions in ground-based growth chambers and are critical to understanding plant responses to the space environment.

Biosphere science news roundup. Biosphere 2 completes first two year closure experiment

On September 26, 1993 Biosphere 2 completed it's first two year experiment which included eight "Biospherians." Now that the first experiment is concluded the results can be examined and reported. Future issues of the journal will include more in depth peer reviewed articles on the experiment while this article will give a brief overview. The facility is now between experiments and is undergoing minor upgrades in preparation for the next ten and a half month enclosure scheduled for March 1994. The new crew has been chosen and is "moving in" now. Now that an interval of time has passed since September 26, 1993 and the end of the initial two year closure of Biosphere 2 it is appropriate to take stock of initial accomplishments and problem areas.

Understanding yields in alley cropping maize (Zea mays L.) and Cassia siamea Lam. under semi-arid conditions in Machakos, eastern Kenya.

Six seasons of experiments in Machakos, Kenya, revealed that above about 150 mm of rainfall, maize yields per row in alley cropped "replacement" agroforestry (AF) plots, of Cassia siamea Lam. and maize (Zea mays, cv. Katumani Composite B), may be expected to exceed those in the control (sole maize) plots. Such yields were insufficient to compensate for the area "lost" to the hedgerows. Below about 150 mm the control plots may be expected to perform better. This result was due to competition for water. Greater association of the fine roots of Cassia and maize was observed in the middle of the alleys than near the hedgerows. Photosynthetic consequences of shading were insignificant relative to other factors. In the alleys, reductions of soil temperature due to shade in the western and eastern maize rows were higher than in the middle row. Soil moisture extraction was higher in the AF than in the control plots. In the AF plots, moisture extraction was greater under the central maize rows than under those nearest the Cassia. Yield patterns followed such soil temperature and soil moisture patterns. Maize transpiration and photosynthetic rates were significantly higher in the control than in the AF plots during a below-average rainy season but not during above-average rainy seasons. It is concluded that alley cropping under semi-arid conditions should be approached differently from the system worked on. It must at least provide strong physical protection of crops and/or soils and have a strong economic incentive to be of interest to the farmers.

The situation of liver fluke infestation in cattle in Bali.

From 1975 to 1982, the prevalence of liver fluke infestation in Bali cattle of the island of Bali, Indonesia varied between 29% and 46%, and from 1983 to 1988 prevalence rates varied between 6% and 22%. Changes in prevalence rates were more closely associated with water levels in pastures than with other variables in pastures, such as temperatures and pH. Mass-fasciolicide treatment of traditionally reared Bali cattle was economical. The decrease in the prevalence of liver flukes in Bali cattle since 1983 was associated with (1) the introduction of a rice planting system which alternated rice with other crops, and (2) with mass-fasciolicide treatments conducted by the veterinary services.

Alimentary foreign body in an Amazon parrot.

An Amazon Parrot regurgitated and drooled and had blood in its droppings. Abdominal palpation revealed several small, spherical masses. Positive-contrast radiographs clearly delineated the objects. With the bird under halothane anesthesia and the beak held open, a sponge forceps was used to extract a string of 25 beads from the alimentary tract. Gentamicin and dexamethasone were given IM and recovery was uneventful.

Patch spraying: future role of electronics in limiting pesticide use.

Developments relating to the control of application equipment can deliver improvements in pesticide use by better matching applications to target requirements. This may have components relating to the spatial distribution of a weed, pest or disease or methods by which the target, particularly a crop canopy, can be described with respect to a given application. Changes in application can relate to the dose and/or volume applied, but may also concern the way in which a treatment is delivered in terms of parameters such as spray trajectory angle and droplet size distribution. For many weed species there is evidence of patchy distributions in field situations. Studies have shown that savings of typically up to 40% in herbicide use can be achieved by adopting patch spraying approaches in such situations. Weed patch detection is key to the performance of such patch spraying systems. In widely spaced rowcrops such as vegetables, there is considerable scope for developing fully automated detection systems based on image analysis, and for the development of accurate guidance systems that apply pesticides only to the crop row. In crops with a relatively high plant density, weed detection in the medium/short term is likely to be based on manual discrimination. The costs of labour for manual weed patch mapping have been estimated at less than 1.50 ha(-1) pounds sterling. Potential savings in pesticide use can also be made if applications are matched to crop canopy structure. This is most important in bush and tree crops where savings of up to 75% in pesticide use could be achieved. In crops such as cereals, studies have shown that savings in fungicide use may be possible, particularly at earlier stages of growth by adjusting spray delivery to measured canopy characteristics. Key components of the performance of application systems concern the ability to deliver over a dose rate range of more than 3:1 while maintaining control of variables such as delivery trajectory angle and spray quality. Traceability and the effective monitoring of applications is likely to be a major driver influencing the uptake of more sophisticated control systems. Methods of labelling pesticides with systems that can be read by the application unit will be an important step in the development of recording and data handling systems that will operate safely with the minimum of operator input and enable the environmental advantages of targeted pesticide application to be monitored.

The potential importance of nut removal by rodents from Australian macadamia orchards.

Extensive crop damage due to rodents, predominantly the black rat (Rattus rattus) is a major concern to both the Hawaiian and Australian macadamia industries. Within Australian systems, indicated the importance of adjacent non-crop habitats in the damage process with damage being directly related to the size and temporal stability of these habitats. It was also suggested that the current estimates of crop loss might be underestimates as R. rattus may remove nuts into adjacent non-crop habitats, however, the economic significance of nut removal was not investigated. This study has determined that the crop loss due to nut removal is equivalent to the crop loss due to in-crop feeding within the first row of the orchard, with nuts being removed and consumed under the cover provided by weedy adjacent habitats. Previous studies (White et al.,1997; Horskins et al.,1998; White et al.,1998) investigating the impact of rodents on crop damage have ignored the nut removal component of the damage process and hence rodents have a far greater impact on the Australian macadamia industry than previously suggested. This study also indicates that the cost-benefit of habitat manipulation as a rodent management strategy is much greater than that suggested by . Habitat manipulation is not only a cost-effective means of damage control but is also sustainable, having none of the potentially adverse public health and environmental effects of continual rodenticide use.
