New staining techniques for the Langerhans cell.

Three new techniques are described for staining the Langerhans cell in whole mounts of fresh human and guinea pig epidermis. These employ paraphenylenediamine, gold sodium thiomalate and cobalt chloride, respectively, and require appropriate epidermal separation with EDTA, ammonium thiocyanate or sodium bromide. Used in conjunction with a modified adenosine triphosphatase stain, these techniques provide greater capability for observing the Langerhans cell in disease states than can be achieved by any single stain. A combined stain with adenosine triphosphate and gold is also described.

[Formation of impulses in an excitable medium].

The paper deals with the evolution of initial perturbation in an active distributed system, described by non-linear equations of a diffusion type. Division of all movements into the less than slow greater than and less than fast greater than ones in time and space makes it possible to give a simple analytical description of all the stages. The following cases are possible: a) Initial distribution limited in space falls into two diverging impulses, each of them consists of two sharp fronts connected by slow movements. While propagating each impulse acquires a stationary form; b) Meeting fronts are formed in the wave, the result is that the initial perturbation disappears in a finite time; c) A sharp front with zero rate of propagation is initiated; its slow evolution may lead to an autooscillation process. The solutions obtained are applicable to the description of concentration waves in oscillatory chemical reactions.

[Significance of the chronic abuse of bromoureide containing hypnotics. Simple chemical methods for its detection (author's transl)].

The abuse of bromoureide (= bromocarbamide) containing sedatives gains increasing clinical relevance. Up to 50% of acute intoxications nowadays are caused by suicidal ingestion of bromocarbamides. The symptoms of the chronic intoxications are due to ionised bromide split off from the organic molecule and slowly accumulating in the organism. Due to the extremely long biological half-life of the bromide ion the chronic ingestion of bromocarbamides can be demonstrated easily and for long periods by determining the concentration of free, i.e. ionised bromide in serum or saliva. Simple and rapid methods suitable for screening purposes are discussed and the results of routine determinations of serum bromide in a psychiatric-neurological department are presented.

Effects of simulated microgravity on metabolic activities related to DNA damage and repair in lymphoblastoid cells.

We adopted a simple experimental framework to follow the dependence of structural aberrations and the modifications in selected metabolic processes correlated with the exposure of cells to microgravity. Alterations to the cellular metabolism induced by exposure to microgravity are evidentiated in the modification of PARP activity (strongly dependent to the presence of DNA damages and to the altered gene expression), in the modification of the repair ability and in the cell's energy homeostasis (NAD and ATP). Cells are exposed continuously to microgravity in a Random Positioning Machine (RPM) in complete medium for 48 hours. At the end of this period a part of these cells are immediately analysed for the parameters reported above and the remaining were furtherly incubated in standard laboratory conditions to document eventual defects during the phases of the recovery process. A part of cells, just after exposure to microgravity, were also subjected to treatment with a strong damaging agent, KBrO3, and these cells were subsequently analyzed. This final treatment was meant to amplify the eventual deficiencies experienced by microgravity-exposed cells in the DNA repair process also in dependence with the alterated metabolic conditions resulting after the exposure to microgravity.

Cell proliferation in liver of Mmh/Ogg1-deficient mice enhances mutation frequency because of the presence of 8-hydroxyguanine in DNA.

The Mmh/Ogg1 gene product maintains the integrity of the genome by removing the damaged base 8-hydroxyguanine (8-OH-G), one of the major DNA lesions generated by reactive oxygen species. Using Ogg1-deficient mice, we sought to establish if cells having high amounts of 8-OH-G have the ability to proliferate and whether the mutation frequency increases after proliferation in vivo. When KBrO(3), a known renal carcinogen, at a dose of 2 grams/liter was administered to Ogg1 mutant mice for 12 weeks, the amount of 8-OH-G in liver DNA from treated Ogg1(-/-) mice increased 26.1 times that of treated Ogg1(+/+) mice. The accumulated 8-OH-G did not decrease 4 weeks after cessation of KBrO(3) treatment. Partial hepatectomy was performed on Ogg1(+/-) and Ogg1(-/-) mice after being treated with KBrO(3) for 12 weeks. The remnant liver from Ogg1(-/-) mice treated with KBrO(3) regenerated to the same extent as nontreated Ogg1(+/-) mice. In addition, 8-OH-G was not repaired during cell proliferation by partial hepatectomy, indicating that there is no replication coupled repair of preexisting 8-OH-G. The mutation frequency after the regeneration of liver from treated Ogg1(-/-) mice showed a 3.5-fold increase compared with before regeneration. This represents a mutation frequency 6.2 times that of normal levels. The proliferation of cells having accumulated amounts of 8-OH-G caused mainly GC-->TA transversions. These results showed that inactivation of the Ogg1 gene leads to a higher risk of cancer because cells with accumulated 8-OH-G still retain the ability to proliferate, leading to an increase in the mutation frequency.

[Determination of trace nitrite by KBrO3-NO2(-)-zincon catalytic system].

A new catalytic kinetic spectrophotometric method for the determination of trace nitrite based on the oxidation reaction between KBrO3 and Zincon in H3PO4 medium has been developed. The optimum conditions for the rate of the reaction were studied, and the rate equation of the reaction was found to be -dcZincon/dt = k'cZincon.cBrO3-.CNO2-. The apparent activation energy and the apparent rate constant were 33.13 kJ.mol-1 and 1.32 x 10(-3).s-1, respectively, the detection limit was 6.0 x 10(-10) g.mL-1, and the range of determination was 0.60-30.00 micrograms.L-1. The method has been used to determine trace nitrite in water with satisfactory result.

Susceptibility of Candida albicans to peroxidase-catalyzed oxidation products of thiocyanate, iodide and bromide.

The susceptibility of Candida albicans (ATCC 10231 and wild strains) to hypo(pseudo)halous ions (OSCN-, OBr-, OI-) produced by the lactoperoxidase system was tested. Six strains of Candida albicans were isolated from swabs taken from the mouths of children with orthodontic appliances and selected on Sabouraud-Chloramphenicol-Actidione agar plates. The survival rate of Candida blastospores after a 30 min exposure to lactoperoxidase system ranged from 79 to 105% in the presence of 615 microM thiocyanate, from 56 to 88% in the presence of 345 microM bromide and from 0 to 4% in the presence of 250 microM iodide. Results showed that only OI- could exert a strong inhibiting effect in vitro on Candida albicans at physiological concentrations. Nevertheless, the activity of the hypoiodite generating system in saliva was found to be under the control of thiocyanate concentration.

[Color reaction of chlorhexidine and proguanil with hypobromite].

Colour reaction of chlorhexidine and proguanil with hypobromite The antimalarial agent proguanil reacts with hypobromite to yield the red coloured (E)-3-[(4-chlorophenyl)imino]-N-isopropyl-3H-1,2,4-triazol-5-amine (6B). The structure of 6B was proven by X-ray. The red colour obtained by the test for the disinfectant chlorhexidinedihydrochloride Ph. Eur. with hypobromite is probably attributable to a corresponding chromophore.
