[Histochemical and electron microscopic study of the formation of trabecular meshwork and changes of glycosaminoglycans].

The development of iridocorneal angle and trabecular tissue was investigated histochemically and electron microscopically. Sixty-three human eyes at from 5 to 22 weeks of gestation were used in this study. For identifying glycosaminoglycans, alcian blue staining and enzyme digestion methods with hyaluronidase and chondroitinase AC and ABC were carried out light microscopically. Electron microscopically, specimens were stained with ruthenium red. In the early stage from 8 to 10 weeks, mesenchymal cells between the primordium of the cornea and the iris became elongated and connected with one another. Desmosome-like junctional complexes were observed in these cells. Schlemm's canal and para-canalicular tissue were observed at 20 weeks and the structure of the meshwork became similar that of adults. From findings of enzyme digestion methods, glycosaminoglycans in trabecular tissue seemed to be mainly hyaluronic acid in the early stage, which was replaced by chondroitin sulfate and dermatan sulfate afterwards. Substances positive for ruthenium red, which seemed to be glycosaminoglycans, was observed in intercellular spaces electron microscopically. They decreased according to development. These findings indicated that the beginning of aqueous outflow might be related to the decrease of glycosaminoglycans in trabecular tissue.

Adrenergic drug effects on cyclic AMP in cultured human trabecular meshwork cells.

Cyclic AMP production in the presence or absence of various adrenergic agonists and antagonists was determined in cultured human trabecular cells using a gammaflow automated radioimmunoassay that allowed multiple simultaneous experiments and reproducible results. Adrenergic agonists and antagonists showed activation and inhibition constants consistent with the presence of beta2-receptors: Ka of isoproterenol < epinephrine < norepinephrine < phenylephrine; Ki of timolol < betaxolol < celiprolol < atenolol. Selective ICI antagonists showed beta2-specificity.

Ultrastructural changes of human trabecular meshwork after photocoagulation with a diode laser.

A diode laser, which emitted infrared radiation at a wavelength of 810 nm, was used to perform trabecular photocoagulation in a human eye due for enucleation for malignant melanoma. For comparison, burns were applied with an argon blue-green laser (488-514.5 nm). With each laser, the treatment spot size was 100 microns and the pulse duration was 0.20 sec. Visible lesions were produced with a power of between 750 mW and 1.2 W with the diode laser, and 500-900 mW with the argon laser. The pattern of damage produced by both modalities was similar and essentially consisted of contraction or expansion of trabecular beams, with trabecular destruction occurring only in relation to high power exposures. These findings confirm that trabecular photocoagulation is not a process that depends upon the wavelength of the incident energy at the two spectral extremes of 488 nm and 810 nm.

[Laser coagulation of the trabecular meshwork in primary open-angle glaucoma].

This paper presents data on 30 phakic eyes with primary open-angle glaucoma treated by argon laser trabecular surgery (Wise's technique). Mean intraocular pressure was reduced from 27.8 mm Hg to 18.3 mm Hg. The mean observation time was 7.2 months. Therapy failed in 3 cases of pigmentary glaucoma. There were no appreciable complications.

[Effects of porcine pancreatic elastase-1 on elastin in human trabecular meshwork--immunohistochemical studies: Report 4. Pseudoexfoliation glaucoma].

The aim of this study was to determine the location of elastin in the pseudoexfoliative material (PSX) in trabecular tissue of pseudoexfoliation (PE) glaucoma and to estimate the influence of porcine pancreatic elastase-1 (PPE) on elastin in PSX. Trabecular tissue obtained from trabeculectomy specimens of PE glaucoma were used. The tissues were embedded in Lowicryl K4M and sectioned for electron microscopy. First, the sections were subjected to protein A-gold immunohistochemical staining to determine the location of elastin in the tissues. Second, the sections were exposed to PPE, before immunostaining, to evaluate the change of immunolocalization of the gold particles in the tissue. The results were as follows. The gold particles indicating the presence of elastin were located in the PSX as well as in elastic fibers. The density of gold particles in PSX was reduced by PPE. These results show that elastin is located in PSX in trabecular tissue of PE glaucoma, and that PPE dissolves the elastin in PSX.
