[Changes in the vascular wall induced by surgical glues. Experimental study].

The effects on vascular tissues of two different types of surgical glue, gelatin-resorcinol-formaldehyde (GRF) and fibrin (Tissucol) were tested on the rat abdominal aorta. The GRF glue induced destruction of the vascular wall: multiple inclusions of the glue were noted in the media. Conversely, the fibrin glue preserved the normal architecture of the three arterial layers. The use of GRF glue therefore should be avoided on particularly fragile tissues (e.g. coronary arteries), and it seems preferable in such cases to use the fibrin glue.

[Glycine determination according to the European Pharmacopoeia].

The treatment of an aqueous solution of glycine with hypochlorite yields formaldehyde and glyoxylic acid. After acidification and removing formaldehyde by boiling glyoxylic acid reacts with resorcinol to the lactone of bis-2,4-dihydroxyphenyl acetic acid. Under alkaline conditions the lactone is oxidized to the violet oxonol anion and subsequently, after hydrolysis of the lactone, to the yellow fluorescent carboxylate anion.

Philodendron-induced dermatitis: report of cases and review of the literature.

Plants of the genus Philodendron, and the family Araceae, were reported as causing dermatitis as early as 1923. The allergenic compounds in philodendrons are resorcinols, which share common chemical side chains with the allergenic catechols of Toxicodendron members of the family Anacardiaceae. I report here two cases of allergic contact dermatitis to Philodendron species commonly found in the Hawaiian islands. Philodendron dermatitis, however, is not confined to tropical or subtropical areas. Exposure may occur in nursery workers in temperate climates and in the home environment worldwide.

Xenopus transcription factor IIIA. Evidence for heterogeneity of Zn2+ binding affinities and specific labeling of cysteine 287.

The release of Zn2+ from transcription factor IIIA (TFIIIA) was examined with the metallochromic indicator 4-(2-pyridylazo)resorcinol (PAR) in the absence and presence of p-hydroxymercuriphenylsulfonate (PMPS). With 0.5 mM PAR, approximately 5 eq of Zn2+ were released from TFIIIA, but no Zn2+ release was detected from the 7 S ribonucleoprotein. The PMPS-promoted Zn2+ release from TFIIIA was 8.7 +/- 0.4 eq of Zn2+ of which approximately 4 eq of Zn2+ rebound to TFIIIA upon displacement of the mercurial with excess 2-mercaptoethanol. These results suggest that at least two affinity classes of Zn2+ binding sites exist in TFIIIA, one of which is released to 0.5 mM PAR in the absence of PMPS. Also, 18 of the 23 cysteine residues of TFIIIA reacted with 5,5'-dithiobis-(2-nitrobenzoic acid). The kinetic data of PAR and 5,5'-dithiobis-(2-nitrobenzoic acid) reactions with TFIIIA were similar, and the spectral changes were characterized by at least three exponential terms. Both TFIIIA and the 7 S particle were reacted with the thiol-specific fluorescent probe N-iodoacetyl-N'-(5-sulfo-1-naphthyl)ethylenediamine (1,5-I-AEDANS). Complete trypsin hydrolysis followed by reverse-phase high pressure liquid chromatography analysis of peptide mixtures showed only one fluorescent peak from the AEDANS-labeled 7 S particle whereas numerous fluorescent peaks were observed with AEDANS-labeled TFIIIA. This further indicates exposure of cysteine residues from Zn2+ binding domains in TFIIIA. Cys287 was identified as the site of modification by amino acid sequencing of the isolated fluorescent peptide from the derivatized 7 S particle. Limited papain cleavage of the AEDANS-labeled 7 S particle indicated that the modified cysteine is located within a 34-kDa TFIIIA fragment. Gel retardation and transcription assays showed that TFIIIA, which had been purified from the AEDANS-labeled 7 S particle, was capable of binding to the internal control region of 5 S RNA gene and retained transcription activity. Thus, Zn2+ binding domains and all but 1 cysteine residue are buried in the 7 S particle, thereby facilitating site-specific labeling of TFIIIA.

The value of the Duquénois test for cannabis--a survey.

The Duquénois test is employed throughout the world as part of the identification procedure for cannabis. A survey of results published for more than 400 herbally derived and botanical materials originating from some 270 different plant species is presented. A survey of results from some 200 organic compounds is also presented. The data show that the D-L modification is the most specific. There is no published report of an obviously botanical material, apart from cannabis, that gives a positive D-L test. Apart from cannabinoids, some resorcinol derivatives give a positive test. All of the materials are easily distinguished from cannabis by TLC.

[Usage of the gel filtration method for the determination of urinary proteins (author's transl)].

The urinary proteins determination was carried out after purification by gel filtration over Sephadex rosin. The proteins eluaded react with Biuret-reagent to establish the coloured copper-proteins complex. If proteins concentrations is inferior, the proteins can be determinated by the copper-proteins complex isolation with second gel filtration and titrating Cu++ with reactives: dietildithiocarbamate or the PAR (4-(2pyridyl-azo)resorcinol monosodium salt).

[Comparative study of the antimutagenic activity of diphenols].

Comparative study of the ability of three diphenols (pyrocathechol, resorcinol and hydroquinone) to inhibit the mutagenic activity of benzo(a)pyrene was carried out using the test for micronuclei account in mice polychromatic erythrocytes. It was suggested that the antimutagenic activity of diphenols used is mostly due to isomeric effect, i.e. the position of hydroxy-groups in molecules of diphenols.

Nuclear magnetic resonance studies on the spatial relationship of aromatic donor molecules to the heme iron of horseradish peroxidase.

The geometry of hydrogen donor molecules bound to horseradish peroxidase was investigated using nuclear magnetic resonance techniques. Between resorcinol and 2-methoxy-4-methylphenol which showed different optical difference spectra, little difference was observed in the orientation of the molecules bound to horseradish peroxidase: the minimal distances between the enzyme iron and the protons of the phenol rings are in the range of 8.4-11.0 A. This situation was not greatly different for the third compound studied in this paper, benzhydroxamic acid, providing evidence against the view that its side chain coordinates to the heme iron. Furthermore, it was found that transferred nuclear Overhauser effect for the signals of these compounds was observable only when the heme peripheral 8-methyl proton signal was irradiated. These results, together with a hypothetical model of the enzyme structure obtained by computer-aided simulation procedures, suggest that the binding of these donor molecules and competitive inhibitors occur in the vicinity of the heme peripheral 8-methyl group, with hydrophobic interactions probably with Tyr-185 and with hydrogen bond with adjacent amino acid residues such as Arg-183.

Rapid colorimetry of zinc in seminal fluid.

This colorimetric method for determination of zinc in seminal liquid is based on the reaction between zinc and 4-(2-pyridylazo)resorcinol in a buffered solution at pH 9.5. It is very simple, fast (approximately equal to 10 min), and economical. It can be used to determine zinc in "whole" semen, seminal plasma, or isolated spermatozoa. We verified its accuracy for sample concentrations up to 0.4 g/L by comparison with a reference method based on atomic absorption spectroscopy of acidic wet-ashed samples. Calcium and magnesium do not interfere, even in concentrations exceeding the physiological values. Between-run precision at a normal concentration of 0.14 g/L is about +/- 7 mg/L; within-run precision at the same concentration is about +/- 2 mg/L.

[Effect of autoinducers of anabiosis for some microorganisms on rat liver mitochondria respiration].

2-(4-Hydroxyphenyl)ethane-1-ol (tyrosol) and 5-n-alkyl(C19,C21)resorcinols produced by some microorganisms as anabiosis autoregulators (factors d1) inhibit the electron transport and uncouple oxidative phosphorylation in the respiratory chain of rat liver mitochondria: a 50% decrease of the respiratory control is caused by 0.32-0.36 mumol of tyrosol or by 0.21-0.26 mumol of alkylresorcinols per mg of protein. Alkylresorcinols reduce the NADH-dehydrogenase and succinate dehydrogenase activities in mitochondria, whereas tyrosol acts predominantly on the NADH-dehydrogenase activity.

Aromatic substrate molecules bind at the distal heme pocket of myeloperoxidase.

Binding of aromatic substrate molecules to myeloperoxidase has been investigated by EPR spectroscopy and model building. Binding of aromatic substrate molecules, such as phenol, p-cresol, resorcinol, and 4-amino salicylate, replaces the original rhombic high spin EPR spectrum of the ferric enzyme (g = 6.74, 5.18, and 1.97) by another high spin signal (g = 7.04, 4.87, and 1.93) indicating that these substrate molecules bind near the heme center of the enzyme. Salicylhydroxamic acid and benzohydroxamic acid complexes of myeloperoxidase showed EPR spectra composed of high spin (g = 6.99, 4.93, and 1.95) and low spin (2.66, 2.22, and 1.81) signals. The hydroxamic side chains of these two substrates seem to interact with the heme iron. Model building based on the three-dimensional structure of the enzyme (Zeng, J., and Fenna, R. E. (1992) J. Mol. Biol. 226, 185-207) revealed the presence of a hydrophobic pocket at the entrance of the distal heme cavity where the aromatic ring of these substrates can bind. Moreover, the six-membered ring portion of salicylhydroxamic acid and benzohydroxamic acid could bind to this hydrophobic pocket with the hydroxamic side chain placed between the imidazole of the distal His and the heme iron. The EPR results on lactoperoxidase and intestinal peroxidase also suggest the presence of an aromatic substrate binding site similar to that of myeloperoxidase.

[A study on feasibility of the GR phenolaldehyde resinifying agent].

According to the technological requirements of synthesizing resin and the properties of the resinifying agent, glutaraldehyde, resorcinol and sodium hydroxide with an optimal ratio was selected as a new resinifying agent. The GR resin solidified usually within 20 to 30 minutes, which was suitable to clinical use. Histologic study indicated that GR could resinify living tissues and the residual and infected pulpal tissue in root canals. It not only filled the main canals and sealed the apical foramen, but also infiltrated into the accessory canals and fine branches of canals. The permeating distance of GR in dentinal tubules was short. The results showed GR had the basic properties of resinifying agent.

On the reaction mechanism of phenol hydroxylase. New information obtained by correlation of fluorescence and absorbance stopped flow studies.

Steps in the hydroxylation pathway of the flavoprotein phenol hydroxylase with resorcinol as substrate have been studied by a combination of fluorescence and absorbance stopped flow techniques. In the presence of azide, a series of highly fluorescent oxygenated flavin intermediates has been observed, corresponding to those previously detected by absorbance measurements (Detmer, K., and Massey, V. (1985) J. Biol. Chem. 260, 5998-6005). In addition, yet another intermediate has been found as the immediate product of the reaction of the reduced enzyme with O2. This new species is non-fluorescent in the presence of azide, but fluorescent in the absence of monovalent anions and had escaped detection in previous absorbance studies because of the similarity in its rates of formation and conversion to the next intermediate and similarity in their spectra. These two early intermediates are tentatively identified as the anionic and protonated species of the flavin C4a-hydroperoxide or, alternatively, as two conformationally different forms of the enzyme hydroperoxide. The next intermediate, previously referred to as intermediate II, is also highly fluorescent and so is considered unlikely to be due to a complex of a flavin alkoxyl radical and a substituted cyclohexadienyl radical, as proposed by Anderson et al. (Anderson, R. F., Patel, K. B., and Stratford, M. R. L. (1990) J. Biol. Chem. 265, 1952-1957). The conversion of intermediate II to the next intermediate, intermediate III (the C4a-hydroxyflavin), is characterized by a large substrate deuterium isotope effect in the 320-390 nm range, but not by fluorescence or by absorbance at wavelengths > 400 nm. This is ascribed to dissociation from the enzyme of a cyclohexadienone product, leaving the enzyme in its C4a-hydroxyflavin form. The latter eliminates H2O to re-form oxidized flavin, but in a competing reaction, in the presence of excess substrate, forms a very stable complex, which decays orders of magnitude more slowly than the uncomplexed enzyme.

An improved assay of gangliosides separated by thin-layer chromatography.

There were no differences in the recoveries of ganglioside sialic acid from silica gel G thin-layer chromatograms when they were sprayed with resorcinol reagent varying in normality between 3 and 10. However, both the temperature at and the time for which the plates were heated after spraying did affect the recoveries, which can reach 100% when the plates are heated for 6 min at 135degreesC.

Quantitative colorimetric determination of urinary p-aminophenol with an automated analyzer.

We developed an automated colorimetric method for the quantitative determination of p-aminophenol with a Cobas Mira analyzer. The procedure can be used for the biological monitoring of human exposure to aniline. An absorbed aniline dose is extensively oxidized to p-aminophenol, which is excreted in urine mainly as glucurono- and sulfo- conjugates. After enzymatic hydrolysis, we reacted the free compound with resorcinol in the presence of manganese ions to form an indophenol dye, which is measured at 550 nm. Excellent accuracy (102.8%, 103.9%, and 96.8% at 2.5, 50, and 90 mg/L, respectively) and precision (7.7%, 2.1%, and 0.8% CV for within-run and 11.1%, 4.7%, and 4.6% for total reproducibility at 2.5, 50, and 90 mg/L, respectively) were achieved over a linear concentration range of 2.0 to 100 mg/L. The detection limit was 0.9 mg/L and no significant interference (except for o-aminophenol) was found for several investigated drugs and related compounds. The proposed method was used for a stability study and to analyze several samples from an occupational health screen.

Resinifying therapy in endodontics. II--Histological and bacteriological evaluation.

This study was conducted on six rabbits, six rats and six human permanent teeth to evaluate histologically the effect of resin resorcinol in the periapical areas, the bacteriological aspect of the resin and the penetration of resin in the dentinal tubules. After 12 days period of observation plenty of lymphocytes and macrophages were evident along with granulomas which are suggestive of the tendency of the resin to stay longer. It was observed that the resin penetrate approximately three fourth distance inside the dentinal tubules. In the last phase the resin was found to be effective against the eight commonly found micro organisms. Though the histological results indicated that the periapical tissue would take longer time for complete repair, yet clinically the tooth functions well. Resinifying therapy is effective and economical method in root canal treatment.

Gangliosides in human uveal melanoma metastatic process.

The inability of current therapy to prevent metastases arising from uveal melanoma often results in patient mortality. With the goal of developing a treatment for metastasis, gangliosides were studied as potential tumor-associated antigens. Our report describes the production of a metastatic liver variant (MH) from a human uveal melanoma cell line (SP6.5). Cells were injected into nude mouse spleens and liver metastases collected 2 months later. After 21 days of in vitro subculture, the cells were re-injected into normal nude mice spleen; 10 cycles (MH10) were performed. Gangliosides were extracted, purified, chromatographed on HPTLC plates and sprayed with a resorcinol-HCl reagent, the sialic acid spots being quantified by densitometry. Gangliosides were analyzed in each metastatic liver variant and compared with the SP6.5 s.c. tumor. The results showed a significant increase in GM3 and a significant decrease in GD3 and GD2 in the last metastatic variants obtained (MH5, MH8, MH9 and MH1O) compared with the primary s.c. tumor, SP6.5. Such evolution in the ganglioside pattern was maintained throughout the progression of the different liver variants. Our results indicate that precursor ganglioside GM3 and gangliosides GD3 and GD2 could be associated with neoplastic evolution of malignancy of human uveal melanoma in nude mice.
