Study of peptide dendrimers having a ferrocene core supported on mercaptoundecanoic acid.

Hydrogen-bonding interactions between the carboxylic acid groups of mercaptoundecanoic acid (MUA) coated gold substrates and the ester surface of peptide dendrimers allows the formation of glutamic acid dendrimers films. Dendrimer films were prepared for generations 1-6 (G1-G6) and analyzed by spectroscopic and electrochemical techniques. Electrochemical studies using cyclic voltammetry and differential pulse voltammetry show that all films except those of G6 were electrochemically active. Lack of activity of G6 films is rationalized by the total encapsulation of the ferrocene redox probe by the dendritic sheath and lack of ion pairing, which prevents its oxidation.

Insights into the mechanism of action of ferroquine. Relationship between physicochemical properties and antiplasmodial activity.

Ferroquine (FQ) is a 4-aminoquinoline antimalarial which contains a quinoline nucleus similar to chloroquine, but a novel ferrocenic group in its side chain. Previous work has demonstrated that this compound has excellent activity against malaria parasites, both in vitro and in vivo, with especially good activity against chloroquine-resistant parasites, but details of its mechanism of action have not previously been reported. In this study, we have investigated the physicochemical properties of FQ for comparison with chloroquine (CQ). Like CQ, FQ forms complexes with hematin in solution (log K = 4.95 +/- 0.05). FQ is an even stronger inhibitor of beta-hematin formation than CQ (IC(50) = 0.78 equiv relative to hematin for FQ vs 1.9 for CQ). These data suggest that the mechanism of action of FQ is likely to be similar to that of CQ and probably involves hematin as the drug target and inhibition of hemozoin formation. However, both the basicity and lipophilicity of FQ are significantly different from those of CQ. The lipophilicity of FQ and CQ are similar when protonated at the putative food vacuole pH of 5.2 (log D = -0.77 and -1.2 respectively), but differ markedly at pH 7.4 (log D = 2.95 and 0.85 respectively). In addition, the pK(a) values of FQ are lower (pK(a1) = 8.19 and pK(a2) = 6.99) than those of CQ (10.03 and 7.94, respectively). This suggests that there will be somewhat less vacuolar accumulation of FQ compared with CQ. Single crystal structure determination of FQ shows the presence of a strong internal hydrogen bond between the 4-amino group and the terminal N atom. This, together with the electron donating properties of the ferrocene moiety, probably explains the decreased pK(a). Interestingly, the decreased accumulation arising from the less basic behavior of this compound is partly compensated for by its stronger beta-hematin inhibition. Increased lipophilicity, differences in geometric and electronic structure, and changes in the N-N distances in FQ compared to CQ probably explain its activity against CQ-resistant parasites.

Ferrocene-based electroactive derivatizing reagents for the rapid selective screening of alcohols and phenols in natural product mixtures using electrospray-tandem mass spectrometry.

The alcohols and phenols of oil of cloves, lemon oil, rose absolut, and oil of peppermint were derivatized with ferroceneoyl azide to generate their ferroceneoyl carbamates. These derivatives are selectively detected at the attomole level, in nanomolar concentrations by electrospray-tandem mass spectrometry (ES-MS/MS) without the need for sample cleanup. The ES-MS/MS analyses of the four essential oils revealed all the expected alcohols, and, in the case of lemon oil, it detected alpha-terpineol as a trace component that was not readily observed by GC-MS. The ES-MS/MS analyses complements the more conventional GC-MS analysis. The ES-MS method has the advantage of speed, selectivity, and sensitivity over GC-MS for detection of a targeted alcohol of a specific mass or structural type. The ES-MS method does not require a chromatographic separation of the components to accomplish its task. In contrast, GC-MS remains the preferred method for the determination of the total constituents of an oil. The ES-MS method may produce artifact ions, especially if the sample is wet and an excess of the ferroceneoyl azide is used; however, the artifacts did not interfere with the analyses.

Chirality-organized ferrocene receptor bearing podand dipeptide chains (-L-Ala-L-Pro-NHPyMe) for the selective recognition of dicarboxylic acids.

[structure: see text] The ferrocene receptor bearing the podand dipeptide chains (-l-Ala-l-Pro-NHPyMe) was found to provide a chirality-organized binding site through two intramolecular interchain hydrogen bonds between CO (Ala) and NH (another Ala) of each podand dipeptide chain. The size-selective and chiral recognition of dicarboxylic acids was achieved by multipoint hydrogen bonds of the binding site.

Calix[4]arenes containing ferrocene amide as carboxylate anion receptors and sensors.

Calix[4]arene derivatives containing amide ferrocene units at the wide rim and ethyl ester groups at the narrow rim, L1-L3, were synthesized and their anion binding and sensing properties were investigated. It was found from (1)H NMR titrations that L1-L3 were able to bind selectively with carboxylate anions. Moreover, cyclic voltammetry and square wave voltammetry showed that L1-L3 were able to act as electrochemical sensors for carboxylate anions. [structure: see text]

Highly regio- and enantioselective Heck reaction of N-methoxycarbonyl-2-pyrroline with planar chiral diphosphine-oxazoline ferrocenyl ligands.

[reaction: see text] A series of planar chiral diphosphine-oxazoline ferrocene ligands were effectively applied in the asymmetric arylation of N-methoxycarbonyl-2-pyrroline, and up to 99% ee was observed for the product 3. The regioselectivity of this reaction was strongly affected by the different precursors of the palladium species and the polarity of the solvent.

Binding of amino acids into a novel multiresponsive ferrocene receptor having an ene backbone.

[reaction: see text] Receptor 1 featuring two open arms, multipoint binding sites, and unsaturated linkers on a ferrocene platform shows strong 1:1 binding to unprotected alpha-amino acids (UV-vis, fluorescence, CV, ITC, NMR, and ESI-MS). NMR and ESI-MS studies suggest an encapsulative binding mode involving the alpha,beta-unsaturated carbonyl residue (site for -NH3+, interaction A) and the terminal -OH groups (site for -COO-, interaction B).

Chirality organization of ferrocenes bearing dipeptide chains of heterochiral sequence.

[structures: see text] The symmetrical introduction of two dipeptide chains of heterochiral sequence (-L-Ala-D-Pro-NHPy) into the ferrocene scaffold as a central reverse-turn unit was demonstrated to induce both antiparallel beta-sheet-like and type II beta-turn-like structures simultaneously, affording the chirality-organized structure. The ferrocene bearing only one dipeptide chain (-L-Ala-D-Pro-NHPy) exhibited a left-handed helically ordered molecular assembly through a network of intermolecular hydrogen bonds instead of intramolecular hydrogen bonds.

Induction of gamma-turn-like structure in ferrocene bearing dipeptide chains via conformational control.

[structure: see text] A combination of the ferrocene scaffold as a central reverse-turn unit with the dipeptide chains (-L-Pro-L-Ala-NHPy) was demonstrated to induce both inverse gamma-turn-like and antiparallel beta-sheet-like structures. Only the antiparallel beta-sheet-like structure was formed in the ferrocene bearing the heterochiral dipeptide chains (-L-Pro-D-Ala-NHPy), in which highly organized self-assembly was achieved through a network of intermolecular hydrogen bonds.

A bis-guanidine-based multisignaling sensor molecule that displays redox-ratiometric behavior or fluorescence enhancement in the presence of anions and cations.

[structure: see text]The guanidine bridge in the new structural motif [3,3]ferrocenophane acts as a dual binding site for anions and metal ions. Sensing of anions (Cl(-), F(-), NO(3)(-), HSO(4)(-), and H(2)PO(4)(-)) takes place by redox-ratiometric measurements, whereas metal ion (Zn(2+), Ni(2+), and Cd(2+)) recognition is achieved by fluorescence measurements.

Evaluation of genotoxic and immunotoxic activities of potential glucose biosensor components: ferrocenes.

Three ferrocenes used in glucose biosensor construction were tested in the aspect of genotoxic and immunotoxic activities. All three ferrocenes were not mutagenic in the standard bacterial Ames test. Equally in the Sister Chromatid Exchanges test in human lymphocyte cultures, the genotoxic action of tested ferrocenes could be excluded. However, all three significantly decreased the rate of lymphocyte proliferation and especially diminished the numbers of B-lymphocytes and NK-cells after 72 hours of in vitro culture. Marked differences between the ferrocenes in their immunotoxic activities were noticed, and we were able to select those which would be relatively safe and those which should be avoided in further investigation of the glucose biosensor construction. Our results indicate the necessity to estimate immunotoxic effects as well as genotoxic effects, especially in biosensor components potentially used in vivo.

Entropically driven binding in a self-assembling molecular capsule.

The encapsulation of molecules within larger molecular or supramolecular cages (see ref. 1 for examples) brings about many interesting phenomena, among them the stabilization of reactive intermediates, new forms of stereoisomerism, and templating of the cage itself. Here we describe a self-assembling hydrogen-bonded molecular capsule that encapsulates guest molecules in a reversible, apparently entropy-driven process, giving rise to unusual temperature dependence of the formation process. The positive entropy of formation--at first sight unexpected--seems to be the result of the fact that more than one solvent molecule is included in the 'bare' cage, making their replacement by a single large molecule (here adamantane or ferrocene) entropically favourable.

Electron transfer mediated by glucose oxidase at the liquid/liquid interface.

In order to establish an experimental basis for exploring the reactivity of membrane-bound redox enzymes using electrochemistry at an organic/aqueous interface, the reactivity of glucose oxidase adsorbed at the dichloroethane/water interface has been studied. Turnover of glucose in the aqueous phase mediated by dimethyl ferricenium electrogenerated in the organic phase was measured by measuring the feedback current caused by recycling the mediator as the generator electrode approached close to the interface from the organic side. An unexpected self-exchange reaction of the ferrocene at the interface was suppressed by adsorption of a surfactant. The interfacial enzyme reaction could be distinguished from reaction within the bulk of the aqueous phase. Reaction within a protein-surfactant film formed at the interface is conjectured.

Carbon ceramic electrode modified with redox liquid.

A carbon ceramic electrode modified with a redox liquid, butylferrocene, exhibiting in aqueous salt solution electrochemical behaviour resulting from the redox process of the modifier and ion transfer across the liquid-liquid interface has been prepared.
