[The effect of the size of the irradiation field on the severity and outcome of the radiation injury of swine skin].

Swine skin areas of 12.56 cm2, 50 cm2 and 100 cm2 were exposed to beta-particles from 90Sr + 90Y (100 Gy). The increase in size of the exposed site caused a considerable increase in the degree of the affection and a change in the regeneration rate. Epithelialization of 12.56 cm2 skin field was completed by the 14th-16th week, and it was absent after 4.5 years in the fields of 50 and 100 cm2.

The carriage and delivery of substances to lymphatic tissues by recirculating lymphocytes. II. Long-term selective irradiation of the spleen and lymph nodes by deposition of indium-114m.

Recirculating lymphocytes were used to carry an isotope of indium, In-114m. The isotope has a half-life of 50 days, and emits both gamma and beta particles, properties that may be useful clinically. In-114m-labelled rat thoracic duct lymphocytes (TDL), injected intravenously, migrated to traffic areas in spleen and lymph nodes. The radioactivity was transferred selectively to a resident, radioresistant phagocytic population located within the marginal zone and red pulp of spleen, where it remained for at least 6 weeks. That the isotope was relatively concentrated in traffic areas, but not elsewhere, suggested that In-114m-TDL could be used as a means of producing total lymphoid irradiation without damaging bone marrow. Recipients of In-114m-TDL developed a profound lymphocytopenia within 2-3 weeks of injection but gained weight at a comparable rate to control rats. The effect of In-114m-TDL injection was also examined in rats injected with a rapidly growing, malignant rat leukaemia. In-114m-TDL delayed the onset of the proliferative phase of the leukaemia from 10 days to 20 days in a dose-dependent fashion. Repeated weekly injections of In-114m-TDL prevented the onset of the leukaemia until the injections ceased after 11 weeks. The potential use of In-114m-labelled lymphocytes in diagnosis (by external imaging) or in treatment (by localized lymphoid irradiation) is considered.

Induction of neoplastic transformation by low-dose-rate exposure to tritiated water.

Exponentially growing mouse BALB/3T3 cells seeded at low density were incubated with various concentrations of tritiated water (3HOH). A dose-dependent increase in the yield of transformants occurred in cells incubated for 100 h with 25 to 500 micrograms/ml of 3HOH. The dose-response curve rose linearly at low doses, with no evidence of a threshold or quadratic component. The transformation frequencies were similar when the same total dose of radiation was given over periods of 5 to 168 h, although survival was slightly lower with 5 h exposure compared with longer intervals. Acute X-ray exposure appeared to be more efficient in inducing transformation than protracted exposure to 3HOH at doses of 200-400 rad, but slightly less effective in the 25-100 rad range. When normalized for survival, 3HOH was more effective than X rays even at the higher doses. This result is very similar to previous findings with [3H]thymidine. A comparison of [3H]thymidine and 3HOH results suggest that the transmutational effect of tritium decay for radioactivity incorporated into DNA may be a significant factor in cytotoxicity but not in transformation; the enhanced effectiveness of both types of tritium exposure for transformation and mutagenesis may result from a higher relative biological effectiveness for the tritium beta particle for these end points.

Randomness of base substitution mutations induced in the lacI gene of Escherichia coli by ionizing radiation.

The lacI system of Escherichia coli provides a method for monitoring mutational events at a large number of sites. Using this system, we have previously determined the mutational spectra for gamma-ray and beta-particle emissions resulting from the decay of tritium. Analysis of these mutational spectra reveals that base substitution mutations induced by ionizing radiation are distributed nearly randomly throughout the lacI gene and include all detectable substitution events. The distribution of ionizing radiation-induced mutagenesis is similar to the low frequency of occurrence mutational events induced by other SOS-dependent mutagens. The lack of an apparent nonrandom or high frequency of occurrence component seen with other SOS-dependent mutagens can be best explained as the result of the random interaction of ionizing radiation with the DNA bases leading to production of a variety of base substitutions.

The effect of section thickness and embedding media on the observed S-phase labelling index of artificially selected cell populations from neonatal mouse liver and spleen.

Following an intraperitoneal injection of tritiated thymidine to neonatal mice, livers and spleens were removed and their labelling indices were derived autoradiographically. This was done in a number of ways: (1) from tissue imprints on gelatinised glass slides; (2) from tissue embedded in JB4 plastic sectioned at thicknesses of 2, 5 and 7 micron; and (3) from tissue embedded in paraffin wax and sectioned at 7 micron. The results show that the indices from the JB4 embedded sections increase as the section thickness decreases, and that this relationship persists down to the notional section thickness of zero in the tissue imprints (in which all the cells are in contact with the autoradiographic emulsion). Indices from the 7 micron paraffin wax embedded sections are surprisingly close to the values from the imprints, are higher than indices from the 5 and 7 micron JB4 embedded sections, and are not significantly different (at the 2% level) from those from 2 micron JB4 embedded sections. Possible reasons for these results are discussed in respect of the autoradiographic process and in relationship to various mathematical correction factors which have been proposed to take account of beta-particle self-absorption in thick sections. It is concluded that none of these correction factors is of value and that the embedding medium has an important effect on the observed labelling indices. Comparisons between labelling indices, therefore, should be made only when they are derived from similarly embedded material at the same section thickness.

[The 32P test in ophthalmology].

Rapidly proliferating tissue incorporates more phosphorus than normal. Thus, following administration of radioactive phosphorus (32P), tumors emit increased amounts of radiation which can be detected. With the proper test technique, a correct diagnosis rate of 96-100% is quoted in the literature. By comparison, between 4 and 20% of all eyes enucleated for suspected malignant melanoma without prior 32P testing contained no neoplasm histologically. The radiation exposure is minimal due to the short half-life of 32P and the low penetration of beta-particles. However, the test usually involves a small operation (conjunctiva and Tenon incision). Testing with gamma rays, however, is not yet wholly satisfactory due to the lack of a suitable gamma-ray detector.

Spectra of base substitution mutations induced in Escherichia coli by tritiated water and the decay of incorporated tritiated thymidine.

The biological effects of tritiated water and of [6-3H]thymidine or [methyl-3H]thymidine incorporated into DNA were compared with those induced by 60Co gamma rays. The killing efficiencies of tritiated water and the tritium-labeled bases were very similar, between 1.8 and 2.0 in terms of the RBE of 60Co gamma rays when compared with the absorbed dose to the bacterial nucleus. The frequency of His+ revertants induced by the decay of [6-3H]thymidine was 3.5 times higher than that induced by [methyl-3H]thymidine or tritiated water; these revertants were most often the result of A:T leads to G:C transitions. In comparison, the other treatments efficiently induced both transitions and transversions. The mutational spectrum resulting from the decay of tritiated water was also determined in the lacI forward-mutagenesis system of Escherichia coli. Transitions predominated at the low dose (2.5 krad), while both transitions and transversions were recovered after a high dose (18 krad). These results are very similar to those observed with 60Co gamma rays and are consistent with the hypothesis that mutagenesis resulting from the decay of [6-3H]thymidine is the result of a position effect, while mutagenesis resulting from the decay of [methyl-3H]thymidine and tritiated water is due to beta-particle ionization.

Radioisotopes in the treatment of ovarian cancer.

Ovarian cancer, currently, is the leading cause of death from malignancies arising in the female genital tract. Investigators and collaborative groups are focusing on this problem in an attempt to improve patient survival. The material reviewed has attempted to gather the data that relate to the use of radioisotopes in the treatment of ovarian cancer. The conclusions reached from this review are: Dose. The initial therapeutic dose of 198Au was empirically determined by Muller. This empirically determined dose was then used to estimate a dose of 32P, based on certain assumptions about these radiocolloids that have been shown to be erroneous. It would appear that the administered dose of 100-150 mCi of 198Au yields a significantly greater tissue dose than 10-15 mCi of 32P. Complications. Complications associated with 198Au are a result of a possibly excessive dose, delivered over a shorter period of time, with a significant gamma component. It is clear that in greater than 99 per cent of disintegrations each beta particle is associated with a gamma component. Distribution, dose distribution, and dose rate. These three critical factors have recently been evaluated for intraperitoneal 32P. It would appear that distribution is non-uniform, and thus dose distribution is not homogeneous, and that dose rate is low and may be ineffective in controlling ovarian cancer. Results of treatment. It would appear from a review of the literature that no prospective randomized well-controlled study as indicated the effectiveness of 32P over other treatment modalities. There is only circumstantial evidence for its effectiveness. The clinical applicability of 32P in the treatment of Stage I ovarian cancer, as part of a treatment protocol, awaits the results of well controlled prospective clinical trials.

[Alpha-recoiling behaviour of 222Rn on 226Ra-sources].

By using electroplated 226Ra-sources, the ejection and injection behaviour of alpha-recoil atoms, 222Rn, which generates on the source surface during alpha-decay process of its parent 226Ra, has been basically investigated. The thinly and uniformly distributed 226Ra-sources were prepared with high deposition yield by the molecular plating technique, in which radium was deposited on a stainless-steel cathode from an isopropanol media by adopting a rotating platinum anode, applied potential of 900 V, and deposition time of 40 min. The ejection efficiency of 222Rn was determined by using the relationship between the 226Ra strength on source and the 222Rn-one transferred on a stainless steel collector in vacuum. It was found that the ejection (or injection) efficiency was greatly influenced on the source preparation methods. The highest ejection efficiency (up to 30%) could be obtained from the electroplated source, whereas the same source could be resulted in the significant decrease of this value (till 2%) after simple ignition treatment. The retention efficiency of 222Rn injected on some collector materials was evaluated to be more than 70% over the tested collectors. From these results, it was concluded that the recoil effects should be most important to interpret the emanation phenomena even in nature.

Radiotoxicity of thallium-201 in mouse testes: inadequacy of conventional dosimetry.

When TI-201 is concentrated in mouse testes, the low-energy Auger electrons following its electron-capture decay are found to be much more effective in causing loss of testicular weight and reduction of sperm heads than the energetic beta particles from similarly distributed TI-204. These results are contrary to expectations based on conventional dosimetry of tissue-incorporated radionuclides, and point to possible underestimation of risks by the currently adopted dosimetric procedures, especially in the case of radionuclides decaying by electron capture and internal conversion.

Counting efficiency of some commercially available liquid scintillators compared.

We investigated four commercially available liquid scintillators as to overall counting efficiency and quenching effect of added phosphate-buffered isotonic saline. They differed significantly in these respects. Evidently, characteristics of the scintillator used in radioimmunoassays where beta-particle emitters are counted should be carefully examined if results are to be optimal.

[The cytogenetic effects in natural populations of Crepis tectorum exposed to chronic irradiation in the region of the Chernobyl Atomic Electric Power Station. An analysis of the frequency of chromosome aberrations and karyotype changes in the 3rd and 4th years after the accident].

In three natural Crepis tectorum populations from 30-km zone of the Chernobyl NPP cytogenetic analysis of plant progenies was carried out. Germination capacity was not correlated with the dose rate of chronic irradiation. There was a correlation between frequency of cells with chromosome aberrations and frequency of seedlings with non-identical altered karyotype among which pericentric inversions and reciprocal translocations dominated. 9-chromosome altered karyotypes were found. Homozygous plants with altered karyotypes were not observed. Several heterozygous plants segregated homozygous seedlings with altered karyotype were found. In comparison with the second year after the accident in the third and fourth years there was stronger deviation from linear dependence between the cytogenetic effect and the dose rate of irradiation.

[The value of Sm-153-EDTMP for treatment of metastatic bone pain and improving quality of life].

Sm-153-EDTMP (ethylenediaminetetramethylene phosphonic acid), a beta-particle and gamma-emitting radionuclide, was used for pain control of bone metastasis. Fourty patients with bone metastasis were administrated a single intravenous injection of 29.6 MBq/kg (0.8mCi/kg), and 7 of them received more than one injection. No obvious side effect was observed. The skeletal images obtained with Sm-153-EDTMP were similar to that of Tc-99m-MDP SPECT images. Follow up study (at least 30 days) revealed bone pain relief and improved quality of life in 82.5% of the treated patients.

Samarium-153-EDTMP biodistribution and dosimetry estimation.

Fifty-two patients were treated with single doses of 153Sm-EDTMP in a Phase I escalating dose protocol for palliation of bone pain from metastatic prostate carcinoma. Samarium-153 (T1/2 46.3 hr), maximum beta-particle energies 810 keV (20%), 710 keV (30%), 640 keV (50%), gamma photon 103 keV (28%), was complexed to the tetraphosphonate chelate, EDTMP. Five groups of patients were treated at doses of 1.0, 1.5, 2.0, 2.5, and 3.0 mCi/kg to evaluate toxicity from treatment. Patients were screened prior to treatment and followed after treatment with 99mTc-MDP bone scans. Biodistribution data on this group of patients were acquired and showed rapid uptake of 153Sm-EDTMP into bone with complete clearance of nonskeletal radiotoxicity by 6-8 hr. Also included are complete sets of dosimetry estimations on an additional seven patients who received 0.5 mCi/kg 153Sm-EDTMP Ca++ as part of a multiple dose therapy trial. Estimated radiation absorbed doses to bone surfaces averaged 25,000 mrad/mCi (6686 Gy/MBq), and urinary bladder doses averaged 3600 mrad/mCi (964 Gy/MBq).

Biological effects of 137CsCl injected in beagle dogs.

The toxicity of intravenously administered 137CsCl in the beagle dog was investigated as part of a program to evaluate the biological effects of internally deposited fission-product radionuclides. The intravenous route of exposure was chosen for simplicity and accuracy because it was known that after intravenous injection, inhalation or ingestion, internally deposited 137CsCl is rapidly absorbed and distributed throughout the body, exposing the whole body to beta-particle and gamma radiations. Fifty-four dogs were injected intravenously with 137Cs to provide one group of six dogs with mean initial body burdens of 141 MBq 137Cs/kg body mass and four groups of 12 dogs each with mean initial body burdens of 104, 72, 52 and 36 MBq 137Cs/kg. Twelve dogs were injected with isotonic saline as study controls. Because the number of study control dogs was small, data from an additional 49 control dogs from other studies at the Inhalation Toxicology Research Institute that were performed over a similar span of years were also used. There was a significant, dose-dependent decrease in survival of the 137Cs-injected dogs. Eleven 137Cs-injected dogs, including all six in the highest initial body burden group, died within 81 days after injection, primarily due to hematopoietic cell damage resulting in severe pancytopenia. An additional 25 dogs had transient hematological dyscrasia but survived for long times. All 137Cs-injected male dogs had marked damage to the germinal epithelium of the testicular seminiferous tubules with azoospermia in the long-term survivors. Benign and malignant neoplasms occurred in a variety of organs in 137Cs-injected dogs, rather than in a single target organ. When individual organs were considered, the incidence of malignant neoplasms was increased in the liver and in the nasal cavity and paranasal sinuses of the 137Cs-injected dogs. There was a 137Cs treatment effect in the incidence of malignant neoplasms (P < 0.001) in male dogs but no 137Cs-related treatment effect in female dogs. However, when malignant mammary neoplasms were excluded from the analysis, there was no gender difference, and there was a dose-related response (P < 0.001) in both males and females for the incidence of malignant neoplasms.

Current studies on biological effects of low level internal irradiation.

The biological effects of relative low level internal irradiation were introduced. 1. Life span studies on carcinogenesis. Pacific Northwest Laboratory observed 3782 rats given a single inhalation of 239Pu O2 at initial lung burden ranging from 0.25 to 180 nCi. Significant life span shortening was found at lung dose > 8 Gy and it was indicated that the presence of a "possible" threshold of about 1 Gy for lung tumor formation. 2. Health effects of radon and its progeny. Both experimental and epidemiological survey were studied. The nominal probability coefficient (fatality) for the public and workers are 7.90 x 10(5) per mJhm-3 (2.77 x 10(-4) per working level month, WLM). 3. Health effects on 3H on postnatal brain development and neurobehavior, genetic effects, carcinogenic effects and adaptive effects of 3H were investigated. 4. Study on the effects of neuroendocrine system under low level irradiation of 75Se (Auger electron emitter) and 35S (beta-particles emitter). The results showed that the neuroendocrine system is very sensitive to small dose of internal irradiation.

Thyroid cancer after radiotherapy for skin hemangioma in infancy.

A Swedish cohort of 14,351 infants exposed to ionizing radiation for skin hemangioma was analyzed with respect to thyroid cancer risk. The subjects were irradiated during the period 1920-1959 and radiotherapy was given with beta particles, gamma rays and/or X rays. The mean absorbed dose to the thyroid was 0.26 Gy (range < 0.01-28.5 Gy). The cohort was matched with the Swedish Cancer Register for the period 1958-1986. Seventeen thyroid cancers were registered (standardized incidence ratio = 2.28; 95% confidence interval 1.33-3.65), and the mean thyroid absorbed dose in those subjects was 1.07 Gy (range < 0.01-4.34 Gy). The effect of different variables on the incidence of thyroid cancer was evaluated with Poisson regression. The excess in thyroid cancer began 19 years after radiotherapy and persisted at least 40 years after irradiation. A statistically significant dose-response relationship was found. The excess relative risk per gray was 4.92 (95% CI 1.26-10.2) and the excess absolute risk was 0.90 x 10(-4) per person-year gray. No significant difference in excess relative or absolute risk could be seen between the sexes.

[Electron microscopical demonstration of polysaccharides in rotifers (Aschelminthes) with the use of OsO4-ferricyanide (author's transl)].

Three species of rotifers were fixed with glutaraldehyde and postfixed in a OsO4-ferricyanide solution. This procedure results in staining of the sarcoplasmic reticulum (SR) and the surface coat of muscle cells as well as in staining of the surface coat of glands and nerve cells. Section-staining with leadcitrate increased the electron opacity of the precipitates, which are interpreted as polysaccharides. The partial decrease of staining of polysaccharides, which sometimes is observed, may be related to processes of function. The precipitates within the SR are considered as glycogen-beta-particles.
