The quality of meat of lambs fed with feeds containing lasalocid.

The aim of the research was to determine the quality of meat of lambs fed with feeds containing lasalocid. It was ascertained that lasalocid added in the quantity of 50 mg per animal and day to the feed during the whole period of fattening considerably increased the protein content in the longest back muscle in lambs. The addition of 50 mg lasalocid to the feed considerably lowered the fat content in the muscle. The muscle of lambs receiving 50 mg of lasalocid had the best water binding capacity and thus was most suitable for the production purposes. Lasalocid influenced significantly and advantageously the intensity of colour of the muscle.

Bone formation in cranial, mandibular, tibial and iliac bone grafts in rats.

Several studies have suggested that grafts from membranous derived bone (e.g., calvarial grafts) retain their volume better than those from endochondral derived bone (e.g., iliac bone grafts). Increased osteogenesis in grafts of the former type has been offered as the explanation. However, simple volume measurements of the recovered grafts do not differentiate between viable and dead bone. We studied fresh syngeneic full-thickness bone grafts from calvaria, mandibula, tibia diaphysis, and iliac bone implanted in the back muscles of young Lewis rats. Bone formation in grafts recovered 3 weeks postoperatively was quantitatively evaluated by strontium 85 uptake analyses. We found that the strontium 85 uptake was greater in calvarial and mandibular grafts than in tibial grafts. No difference was found among calvarial, mandibular, and iliac grafts or between tibial and iliac grafts. We conclude that the anatomical area of harvest is important regarding new bone formation in syngeneic bone grafts. However, the results do not support the contention that better maintenance of volume of calvarial grafts compared with iliac bone grafts is due to enhanced osteogenesis in the former.

Long-term strength assessment of postoperative diskectomy patients.

A test battery was used to measure abdominal and back muscle strength and endurance in 20 patients at least 1 year after surgical diskectomy. There were ten men and ten women. Data on isometric and isokinetic performance were compared with previously reported normal values. The data showed that every strength parameter tested except male isokinetic flexion strength showed at least a 30% decrease when compared with normal values. Compared with strength data on these same patients immediately after surgery, a 2-tailed Student t test demonstrated a significant difference only in male isokinetic strength (P less than 0.10) and in female isokinetic strength (P less than 0.05). No other difference was found in isometric strength or endurance compared with data obtained from these patients 4 to 6 weeks postoperatively. From these data, it is obvious that more intensive physical therapy is necessary to improve postoperative strength in patients undergoing surgical diskectomy. These data allow for the design of specific isometric, isokinetic, and endurance rehabilitation programs for the postoperative diskectomy patient.

Acute pancreatitis induced by traditional acupuncture therapy.

Although rare, it is possible for acute pancreatitis to develop after blunt or penetrating abdominal trauma. We report here a very rare case of acute pancreatitis induced by traditional acupuncture therapy. A 42-year-old woman with a low body mass index had suffered from functional dyspepsia for one year. She visited an acupuncture clinic and underwent long needle (13 cm) and gold thread needle (3 mm) acupuncture therapy. Five hours later, she presented at our emergency room complaining of severe periumbilical pain. Levels of serum amylase (1162 U/l, normal <220 U/l), and lipase (5195 IU/l, normal <60 IU/l) were high, and an abdominal computed tomography scan showed a diffusely swollen pancreas and ill-defined infiltration of the peripancreatic fat, indications of possible acute pancreatitis. Multiple small acupunctured gold thread needles were also found on the anterior abdominal wall and back muscles. After excluding other possible causes of pancreatitis, traumatic pancreatitis was diagnosed as an adverse effect of the long acupuncture needle therapy.

Detection of fetal lactate with two-dimensional-localized proton magnetic resonance spectroscopy.

BACKGROUND: Antenatal surveillance is inefficient for accurately detecting fetal compromise. A noninvasive technique for assessing fetal metabolic status would be useful for clinical management. CASE: Fetal magnetic resonance spectroscopy was performed at 20 weeks in a pregnancy complicated by severe intrauterine growth restriction to determine if lactate, a metabolite associated with fetal hypoxia, was present. Two-dimensional, single-slice proton magnetic resonance spectroscopy was carried out at 1.5 T using a volume-selective, double-spin echo technique. Lactate was detected in fetal back muscle. Fetal death occurred the next day. CONCLUSIONS: Although this initial report is purely experimental, further development of this technique may prove to be a valuable noninvasive tool in the management of suspected fetal hypoxia.

Stereoscopic structure of sensory nerve fibers in the lumbar spine and related tissues.

STUDY DESIGN: Neurotracer was applied to various sites in the rat lumbar spine and related tissues. The segmental distribution of labeled neurons in dorsal root ganglia (DRG) was investigated. OBJECTIVES: To clarify the stereoscopic structure of afferent fibers innervating the lumbar spine and related tissues. SUMMARY OF BACKGROUND DATA: Afferent fibers in the rat L5-L6 lumbar intervertebral disc are reported to originate from neurons in L1 and L2 DRG. However, anatomic studies determined that each dorsal ramus of the spinal nerve sends nerves to dorsal elements of the corresponding lumbar vertebra. METHODS: Fluorescent neurotracer DiI (1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate) was applied to various sites of the lumbar spine and related tissues in the L2, L5, and L6 levels in rats. DRG were sectioned 3 weeks after DiI application. Rostrocaudal distribution of DiI-labeled neurons was investigated. RESULTS: At L5, DiI-labeled neurons were prominent in DRG L3 for the lamina, L2 for the spinous process, L2 for the back muscle fascia, and L1 for the skin. Dorsal elements are therefore innervated by neurons in more rostral DRG. In the transverse plane, the more distant from the DRG a site was, the more rostral the DRG innervating the site. This structure suggested a concentric innervation pattern in the transverse plane. CONCLUSION: Stereoscopically, the peripheral innervation territory of a lumbar DRG is conical, with the apex at the ganglion and the base circumference located on the dermatome. The lumbar spine itself is involved in the conical innervation territories of DRG.

Spinal manifestations in a patient with congenital insensitivity to pain.

Spinal manifestations in congenital insensitivity to pain are relatively uncommon and easily misdiagnosed. We report on a patient with absent protective pain sensation, who developed spinal neuropathic arthropathy. At age 11 years, he presented with a destructive lesion at the L1-L2 level, causing him tingling sensation in both lower limbs. He was treated with combined anteroposterior spinal fusion from T12 to L3 and had full recovery. Five years later, he presented with a long history of clicking in his low back, muscle weakness and paresthesia in both lower extremities during walking, and evidence of Charcot arthropathy at the L4-L5 level, resulting in junctional kyphosis and canal narrowing. Posterior spinal arthrodesis from L3 to the sacrum was performed, due to lack of patient and parental consent for combined anterior decompression/posterior fusion. The patient resumed normal muscle function and his previous level of activities. Spinal complications should be anticipated in this condition and create diagnostic and therapeutic dilemmas. However, surgical management can produce favorable clinical results.

Optimization of bone tissue engineering in goats: a peroperative seeding method using cryopreserved cells and localized bone formation in calcium phosphate scaffolds.

BACKGROUND: Bone tissue engineering by combining cultured bone marrow stromal cells with a porous scaffold is a promising alternative for the autologous bone graft. Drawbacks of the technique include the delay necessary for cell culture and the complicated logistics. We investigated methods to bypass these drawbacks. Furthermore, we investigated the localization of bone formation inside the scaffold. METHODS: Bone marrow stromal cells from seven goats were culture expanded and cryopreserved. One week before surgery, some of the cells were thawed, cultured, and seeded on porous calcium phosphate scaffolds. The constructs were cultured for another week until implantation. The remaining cryopreserved cells were thawed just before implantation and peroperatively resuspended in plasma before combining with the scaffold. Scaffolds impregnated with fresh bone marrow, devitalized cultured constructs, and empty scaffolds served as controls. All samples were implanted in the back muscles of the goats for 9 weeks. RESULTS: Histologic examination showed minimal (<1%) bone in the empty and devitalized scaffolds, 4.2 +/- 5.1 bone area percent in the bone marrow samples, and significantly more bone in both the cultured and peroperatively seeded constructs (11.7 +/- 2.5 and 14.0 +/- 2.0%). The peripheral 350 microm of the implants contained significantly less bone. CONCLUSION: Peroperative preparation of osteogenic constructs with cryopreserved cells is feasible. These constructs yield substantially more bone than the scaffolds alone or scaffolds impregnated with fresh bone marrow. Bone deposition is much less on the scaffold periphery.

Bioactive and bioresorbable cellular cubic-composite scaffolds for use in bone reconstruction.

We used a novel composite fibre-precipitation method to create bioactive and bioresorbable cellular cubic composites containing calcium phosphate (CaP) particles (unsintered and uncalcined hydroxyapatite (u-HA), alpha-tricalcium phosphate, beta-tricalcium phosphate, tetracalcium phosphate, dicalcium phosphate dihydrate, dicalcium phosphate anhydrate or octacalcium phosphate) in a poly-D/L-lactide matrix. The CaP particles occupied greater than or equal to 70 wt% (greater than or equal to 50 vol%) fractions within the composites. The porosities of the cellular cubic composites were greater than or equal to 70% and interconnective pores accounted for greater than or equal to 70% of these values. In vitro changes in the cellular geometries and physical properties of the composites were evaluated over time. The Alamar Blue assay was used to measure osteoblast proliferation, while the alkaline phosphatase assay was used to measure osteoblast differentiation. Cellular cubic C-u-HA70, which contained 70 wt% u-HA particles in a 30 wt% poly-D/L-lactide matrix, showed the greatest three-dimensional cell affinity among the materials tested. This composite had similar compressive strength and cellular geometry to cancellous bone, could be modified intraoperatively (by trimming or heating) and was able to form cortico-cancellous bone-like hybrids. The osteoinductivity of C-u-HA70, independent of biological growth factors, was confirmed by implantation into the back muscles of beagles. Our results demonstrated that C-u-HA70 has the potential as a cell scaffold or temporary hard-tissue substitute for clinical use in bone reconstruction.

Monoclonal antibody Tor 23 recognizes a determinant of a presynaptic acetylcholinesterase.

A significant proportion of the acetylcholinesterase that is present in the electric organ of Torpedo californica exists as a presynaptic membrane molecule. The monoclonal antibody Tor 23 binds the Torpedo presynaptic nerve membrane where it recognizes a polypeptide of 68,000 daltons. Our present studies indicate that Tor 23 identifies acetylcholinesterase. From the homogenates of Torpedo nerve terminals, Tor 23 immunoprecipitates measurable esterase activity. Esterase precipitation was not observed with no Tor 23 added; nor was it observed with any other test antibodies, including other Tor antibodies, in particular, Tor 70, which binds, as does Tor 23, to the presynaptic nerve membrane. The esterase activity was specific for acetylcholinesterase. Our studies indicate the molecule defined by Tor 23 has the solubility properties described for that of presynaptic acetylcholinesterase: it is soluble in detergent-treated electroplax homogenates and insoluble in high-salt extractions. In sections of Torpedo back muscle, both nerve and endplate acetylcholinesterase can be detected histochemically. Tor 23 localizes to the nerve and is not clustered at the endplate. The utility of the antibody Tor 23 thus includes biochemical and histological analyses of the multiple forms of acetylcholinesterase.

Characterization of the 5-HT receptor subtypes involved in the motor behaviours produced by intrathecal administration of 5-HT agonists in rats.

1. The motor behavioural effects of intrathecal injections of 5-hydroxytryptamine (5-HT) and a variety of 5-HT receptor agonists were examined in adult Wistar rats to establish; (a) which 5-HT receptor subtype/s elicit each behaviour and (b) whether these receptors are located within the spinal cord. 2. Intrathecal injection of 5-methoxy-N,N'-dimethyltryptamine (5-MeODMT), (+/-)-1-(2,5-dimethoxy-4-iodophenyl)-2-aminopropane hydrochloride (DOI) or 2,5-dimethoxy-alpha,4-dimethylbenzene ethamine hydrochloride (DOM) produced dose-related back muscle contractions (BMC) and wet dog shakes (WDS) which were both markedly attenuated by intraperitoneal pretreatment with either ritanserin (1 mg kg-1), ketanserin (0.16 mg kg-1) or mianserin (0.6 mg kg-1) indicating the involvement of 5-HT2 receptors in both these motor behaviours. Both fluoxetine (1-20 mg kg-1, i.p.) and high doses of 5-HT (50 micrograms) following fluoxetine (5 mg kg-1, i.p.) also elicited BMC, further confirming the involvement of 5-HT in this behaviour. 3. Intrathecal 5-carboxamidotryptamine (5-CT) evoked a marked wet-dog shake response without producing any BMC. Intrathecal pretreatment with 8-hydroxy-2-(di-n-propylamino)tetraline (8-OH-DPAT) enhanced, while in contrast 2-methyl-5-HT pretreatment attenuated, 5-HT agonist-induced BMC without affecting WDS. These data suggest that the spinal 5-HT2 receptors mediating BMC are positively modulated by 5-HT1A but negatively influenced by 5-HT3 receptor activation and may be of a different subtype to the supra-spinal 5-HT2 receptors which elicit WDS. 4. A contrast, reciprocal forepaw treading, lateral head weaving, flat body posture and Straub-tail were evoked by 5-MeODMT, 8-OH-DPAT or 5-CT but not by DOI or DOM indicating that these behaviours were not produced by 5-HT2 receptor activation alone. Ritanserin (1 mg kg- 1, i.p.) or ketanserin (0.16mgkg-1, i.p.) pretreatment reduced the reciprocal forepaw treading induced by high intrathecal doses of either 5-MeODMT (25.pg) or 5-CT (50,ug) suggesting that this behaviour may be facilitated by 5-HT2 receptor activation. 5. Intrathecal injection of 5-HT (0.05-50pg, after systemic fluoxetine, 5mg kg 1, i.p.), or 1-(3-chlorophenyl) piperazine (mCPP) produced dose-related forepaw-licking and grooming, neither of which were attenuated by ketanserin (0.16 mgkg-1, i.p.) pretreatment suggesting these behaviours may be mediated by 5-HT1c receptors. In contrast, 2-methyl-5-HT (50 and 100pg) produced sideward tail-flicks, not evoked by any other 5-HT agonist and could therefore be mediated by spinal 5-HT3 receptor activation. 6. These data provide behavioural evidence for the existence of spinal 5-HT2 receptors which produce a novel motor behaviour, BMC. Ligand binding studies and dose-response studies with a range of selective 5-HT antagonists are required to establish whether BMC and WDS are mediated by different subtypes of 5-HT2 receptors.

Fibroblastic activities post implantation of cobalt chromium alloy and pure germanium in rabbits.

Different preimplantation surface finishes were applied to surgical vitallium discs and germanium prisms implanted for 20 days within the back muscles of adult rabbits. Histopathologic analysis of the numbers of nuclei of active fibroblasts immediately adjacent to the implants was carried out. The mean apparent volume fractions (MAVF) for the subdermal implant sites were found to depend on the surface cleanliness of the implant, the cleanest or highest-surface-energy surfaces giving the highest MAVF values for active fibroblasts.

Development of muscular endurance and strength from adolescence to adulthood and level of physical capacity in men and women at the age of 34 years.

This study investigated the development of muscular endurance and strength from the age of 16 to the age of 34 and the fitness components at the age of 34 in men and women. A randomly selected group consisting of 220 boys and 205 girls was tested in 1974 and reinvestigated 18 years later. Muscular endurance and strength increased significantly in 4 of the 5 remeasured tests for the men; bench press increased by 58%, two-hand lift by 20%, hand grip by 16% and Sargent jump by 8%. For the women there was a significant increase in bench press and two-hand lift by 25% and 7% respectively, but hand grip and Sargent jump decreased with a few per cent. The hip and trunk flexion test, sit-up, decreased remarkably in both men (58%) and women (78%). At the age of 16 physical capacity was significantly higher in all tests in the men. This was also the situation at the age of 34 except for VO2 max in ml.kg-1.min-1 and muscular endurance in the back muscles. There were no differences in flexibility between men and women except for less flexibility in the hamstring muscles in the men. The result of the balance test was significantly better in the men.

A survey of golf injuries in amateur golfers.

A survey of 461 amateur golfers was undertaken to assess golf injuries. There were 193 respondents of whom 57% reported injuries. Wrist, back, muscle, elbow and knee problems were the most likely ailments to compromise a player's game. Overuse and poor technique were the main aetiological factors. Attention to those factors in addition to maintenance of physique would reduce the incidence of these problems.

Estimation of the brain and spinal cord conduction time in man by means of the somatosensory evoked potentials and F and H responses.

Somatosensory evoked potential (SSEP) recordings from the scalp were performed in 17 healthy subjects. In seven of these SSEP was also recorded at the level of the second lumbar spine. In the other ten F and H responses and the corresponding M responses were studied. By means of the SSEP recordings at the level of the second lumbar spine and the F- and H-responses, the conduction time in the brain and spinal cord, that is central latency, was calculated and the following results were obtained: 16.0 ms with standard deviation (SD) +/- 1.1 ms (by means of SSEPs), 9.5 +/- 2.4 ms (by means of F response) and 13.1 +/- 1.5 ms (by means of H response). Of the three methods used the H response method seems to be the best for clinical purposes: it is easy to perform and statistically it is more stable than the F response recording; moreover the recording can be performed reliably even in persons with thick back muscles and subcutaneous fat, unlike the evoked potential procedure which only with difficulty shows detectable responses at the lumbosacral levels in such persons. Three patients are presented to illustrate the technique; in one of these the recording evoked potentials from the epidural space were recorded.

VX2 carcinoma in rabbits after radiofrequency ablation: comparison of MR contrast agents for help in differentiating benign periablational enhancement from residual tumor.

PURPOSE: To prospectively compare the accuracy of a blood pool agent, SH L 643A, with that of gadopentetate dimeglumine in differentiating benign periablational enhancement from residual tumor in VX2 carcinomas in rabbits after radiofrequency (RF) ablation. MATERIALS AND METHODS: Experiment was approved by the animal care committee. Sequential MR images were obtained before and with SH L 643A (17 000 Da, 0.05 mmol/kg) and, after a 24-hour interval, gadopentetate dimeglumine (546 Da, 0.1 mmol/kg) in 12 rabbits with VX2 carcinoma in the back muscle prior to (n = 12) and early (n = 12), 1 week (n = 8), and 4 weeks (n = 4) after RF ablation. RF ablation was performed with output of 90 W but at less than 300 seconds to ensure incomplete tumor ablation. The pathologic specimens were sectioned in the same plane as MR imaging, and the enhancement ratios (ie, the ratios of postcontrast to precontrast signal intensity) and the microvessel densities of residual tumor and benign periablational enhancement were assessed. RESULTS: With SH L 643A, the peak enhancement ratios of residual tumor (1.64 +/- 0.31 [standard deviation]) were significantly higher than those of benign periablational enhancement (0.97 +/- 0.16) (P < .001). With gadopentetate dimeglumine, the peak enhancement ratios of residual tumor (1.82 +/- 0.33) were not different from those of benign periablational enhancement (1.71 +/- 0.36). In benign periablational enhancement, enhancement ratios with injection of SH L 643A were lower than those with injection of gadopentetate dimeglumine for all time points up to 30 minutes (P < .05). The microvessel density was 25.72 +/- 5.43 vessels per field of view for residual tumor and 10.37 +/- 2.88 vessels per field of view for benign periablational enhancement (P < .001). CONCLUSION: Blood pool contrast agent SH L 643A permits more accurate differentiation of benign periablational enhancement from residual tumor compared with the extracellular agent gadopentetate dimeglumine.

Demonstration of changes in fetal liver erythropoiesis using echo-planar magnetic resonance imaging.

This study investigated the variation in magnetic resonance characteristics of the fetal liver during a time of changing erythropoietic function. Echo-planar imaging was carried out in 25 normal pregnant women at 20 and 26 wk gestation. The signal intensity from regions of the fetal liver, background image, and maternal back muscle and the highest signal intensity from the maternal spinal cord were measured and compared with the signal intensity of amniotic fluid. Data are expressed as ratios, in arbitrary units (median pixel values; interquartile range shown in parentheses), and analyzed with the use of Wilcoxon's signed-rank test. At 20 wk, the signal intensity ratio of liver to amniotic fluid was 0.309 (0.231-0.365). At 26 wk, the ratio was 0.544 (0.429-0.616). The change was highly significant (P < 0.0001). No change in the signal intensity ratios of amniotic fluid compared with other measured parameters was noted. These data are consistent with known changes in fetal liver erythropoiesis occurring between 20 and 26 wk gestation and have potential use in early noninvasive physiological assessment of the fetus.

Postpartum symptoms after antepartum bed rest.

OBJECTIVE: To determine the type and frequency of postpartum symptoms during recovery from antepartum bed rest treatment across 6 weeks postpartum. DESIGN: Longitudinal repeated measures study. SETTING: Three perinatal tertiary care hospitals in two cities in the Midwest. PARTICIPANTS: One hundred six postpartum women who had a singleton high-risk pregnancy and were treated with antepartum bed rest. MAIN OUTCOME MEASURES: A Postpartum Symptom Checklist assessed physiologic and psychological symptoms. RESULTS: Women reported 12.5 symptoms at 2 days postpartum, but symptoms rapidly declined at 1 week. Ninety-three percent reported a mean of 6.6 symptoms at 6 weeks postpartum. Symptoms that continued to be reported at 6 weeks by at least 40% of women were fatigue, mood changes, tenseness, difficulty concentrating, back muscle soreness, dry skin, and headache. Women who had a cesarean delivery reported significantly more symptoms than those who had a vaginal delivery (p = .006). Length of maternal bed rest was significantly correlated with the number of symptoms at postpartum weeks 1, 2, 4, 5, and 6. CONCLUSION: Postpartum symptoms decreased across time but reveal an underlying morbidity that is not resolved by 6 weeks postpartum.
