[Radiation safety during work on the ANET-V therapeutic apparatus with 252Cf sources].

The Soviet ANET-W intracavitary neutron therapy apparatus is charged with 252Cf sources, the total mass of the radionuclide being 2100 micrograms. The radiation exposure of the staff has been studied and the results presented. Possible emergency situations have been analysed, methods of their prevention and liquidation worked out. Induced radioactivity in the patient's body has been estimated. Radiation dose levels for the medical staff are 10-15 times lower for the ANET-W apparatus than those for 252Cf sources administered by the manual series system but 10 times higher than those for the AGAT-W gamma-apparatus. The design, blocking system and emergency reset of the sources provide for safe work on the ANET-W apparatus. During liquidation of an emergency situation irradiation of the staff does not exceed 0.3 of the annual maximum tolerance dose. The level of induced activity in the patient's body is insignificant as in 10.5 min it does not exceed background values.

[Apparatus for heart rate biofeedback training].

Present study was undertaken to develop an apparatus for heart rate biofeedback training in clinical basis. The plethysmographic transducer was used for detection of pulses. The main part of this apparatus is composed of three systems in function, that is, an audiovisual feedback system of heart rate in analogue and digital manners, a reinforcement system of exteroceptive stimuli of light and/or sound and operant biofeedback system. Experimental inspection of the apparatus showed no problem except a case of angiospasm of fingers. Clinical trials revealed that this apparatus was available to the biofeedback control of heart beats in both normal volunteers and some cardiac patients.

[Changes in the subvalvular apparatus and left ventricular function in pure mitral stenosis].

Disordered left ventricular function and changes in the sub-valvular apparatus appear to be associated in patients with isolated mitral stenosis (MS). Left ventricular cine-angiography has demonstrated two groups of patients, according to the presence (group II) or absence (group I) of changes in the sub-valvular apparatus. There was no significant difference between the two groups in terms of age, heart rate, left ventricular end diastolic pressure, surface area of the mitral valve, or ventricular ejection time. However, group II patients had a lowered ejection fraction (EF), systolic ejection volume (SEB), speed of shortening of circular fibres (VCF), and systolic work index (SWI) compared with group I cases (respectively 0.49 +/- 0.06 and 0.58 +/- 0.04 for the EF, 36 +/- 7 ml.m-2 and 41 +/- 7 ml.m-2 for the SEV, 0.85 +/- 0.12 circ.s-1 and 1.09 +/- 0.16 circ.s-1 for the VCF, 45 +/- 11 gm.m-2 and 57 +/- 11 gm.m-2 for the SWI. Nevertheless, it appears that changes in the sub-valvular apparatus indicate rather than cause dysfunction of the left ventricle, which occurs in a certain number of patients who have a normal sub-valvular apparatus.

[Highly-efficient apparatus for two-dimensional gel electrophoresis].

An apparatus for two-dimensional gel electrophoresis described enabled to analyze 18 samples simultaneously. The apparatus included a device for electrophoresis in small columns of a gel (A), a device (B) for formation of 18 gel plates and a device (C) for staining and washing of the plates. In order to choose the most optimal conditions for fractionation all the parts of the apparatus are separated by reciprocally perpendicular partitions into sections, which allow to use various buffer systems simultaneously. Results of heart muscle proteins fractionation, carried out in the apparatus on the basis of O'Farrell and Laemmly procedures, are presented.

The functional analysis of the flagellar apparatus in green algae.

The flagellar apparatus of green algae is a complex organelle of great structural diversity. Diversity exists with respect to numbers of flagella, flagellar appendages (e.g., scales), origin of flagella in relation to cell symmetry, basal body and transitional region ultrastructure, fibrous structures interconnecting basal bodies and flagellar root systems. The flagellar apparatus performs several different functions in a single cell and also probably functions differently in different green algal genera. In addition to being an organelle which moves the cell through an aqueous environment the flagellar apparatus reacts to the mechanical, chemical, gravitational and light stimulation of the cell. It also plays an important role during sexual fusion of isogamous, anisogamous and oogamous species of green algae. Furthermore it contains several microtubule-organising centres (MTOCs) and in most flagellated green algae these MTOCs organise the whole microtubular system of the cell. In this review the different strategies which are currently employed to evaluate the functional significance of various structures of the flagellar apparatus in green algae are discussed. Emphasis is given to those structures which are not directly engaged in generating the motive force of flagella.

[Instantaneous measurement of acoustic reflex latency in daily practice. The apparatus. Results].

Increase in acoustic reflex latency (time between stimulus onset and response) provides a highly specific and sensitive sign for early detection of lesions of the first or second auditory neurones. The performance of a new apparatus, including an Amplaid 702 impedance meter with a specially designed numeric oscilloscope, employed systématically for all audiometric examinations (87 subjects) was compared with that of a reference apparatus (Madsen ZO 73 + Elema Schoenander-jet ink recorder). The new apparatus provided a very simple, precise, and rapid method for measuring acoustic reflex latency. The technique has its limits, however, of critical importance being the determination of the acoustic reflex threshold at close to 1 dB (modification of the apparatus), the means for measuring latency, and the use of three criteria for distinguishing retrocochlear lesions from others. The effect of age on latency, variations in the test-retest results according to the type of deafness, and the influence of averaging techniques are discussed. Typical retrocochlear lesions were rare in this series, and the 5 cases observed are described in detail, followed by a discussion of the advantages and inconveniences of systematic as against selective screening.

Importance of the left ventricular subvalvular apparatus for cardiac performance.

The importance of the subvalvular mitral apparatus for left ventricular performance was studied in eight anesthetized dogs. During extracorporeal circulation St. Jude Medical mitral valve prostheses were implanted preserving the chordae tendineae. Flexible wires were slung around the chordae tendineae and brought to the outside through the left ventricular wall to cut the chordae tendineae by electrocautery in the closed beating heart. The left ventricular diameters were measured by sonomicrometry, left ventricular stroke volume and enddiastolic volume by dye dilution, and left ventricular pressure by catheter tip manometer. Data were collected at different preloads achieved by volume loading with blood before and after the chordae tendineae were cut. The results showed that after the chordae tendineae had been cut left ventricular systolic pressure, heart rate, diastolic and systolic diameters of the left ventricle along the minor axis were not different from the pre-cut values at any left ventricular enddiastolic pressure. However, significant differences were observed for maximum dp/dt (-15%), major axis diastolic diameter (+10%) and systolic shortening (-40%), enddiastolic volume (+18%) at any left ventricular enddiastolic pressure, and stroke volume (-24%) at any enddiastolic volume level. The data demonstrate that the subvalvular apparatus not only maintains physiologic valve function, but contributes significantly to left ventricular performance. The impairment of left ventricular function following removal of the subvalvular apparatus might be aggravated in pre-injured hearts in mitral valve disease. Consequently, the subvalvular apparatus should be preserved in mitral valve replacement whenever possible.

Semi-automatic apparatus for determination of the consistency of microbiological culture media and gel-forming substances (author's transl).

The gel strength is an important factor in the production and preparation of microbiological culture media on the basis of agar-agar, silicagel and gelatine. There is an evident interest in a standardization of this parameter which can influence the number and the form of the colonies, the results of antibiograms and other tests based on diffusion. The apparatus described can be used for simultaneous determination of the gel strength and the elasticity of microbiological and industrial gels. The construction consists of an electromotor-driven supporting table for the specimen and a pressure measuring device. In operation, the gel is moved vertically with a constant speed against the probe of the pressure device. The pressure exerted on the surface of the gel can be read from a dial connected with the probe as well as the pressure at which the surface of the gel is being disrupted (breaking point, gel-strength pressure). The apparatus can be purchased under the name Gelomat from Heinrich Bareiss, 7931 Oberdischingen, West Germany (see Fig. 1 and 2). The results of the measurements can be influenced by various factors such as concentration of the gelating compound, composition and pH of the solvent, time and temperature at which the gel is held before, during and after plate pouring, surface (dry or wet) and heigth of the gel sample, method of mesurement, etc. To account for all these factors, a standard method of measurement is described. The apparatus has a sensibility of ca. 2 g, can measure a gel strength between 0 and 200 g with the standard equipment, and has a standard deviation of less than 1.35 for microbiological agar-agar gels (see Tables 2-4). The time needed for one determination is 40-60 seconds. Compared with other instruments for the same purposes, the Gelomat has following advantages: Simple and handy use, good sensitivity and reproducibility, capability for simultaneous measurement of gel strength and gel elasticity. In the last 4 years, this apparatus has been used with good results in more than 10,000 routine determinations

A fully automated apparatus for a light/dark test measuring anxiolytic or anxiogenic effects of drugs in mice.

We developed a fully automated light/dark apparatus which detects locomotion, rearing and time spent in light and dark zones, and shuttle crossing of mice. This apparatus is controlled by a personal computer and detects these parameters by using infrared beamsensors. We used this apparatus to investigate the effects of the anxiolytics, diazepam (DZP) and pentobarbital (PB); the putative anxiolytic, 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT); and the anxiogenics, picrotoxin, methyl-beta-carboline-3-carboxylate (beta-CCM), and ethyl-beta-carboline-3-carboxylate (beta-CCE), on each parameter measured by a light/dark test in mice. DZP (5 mg/kg) and 8-OH-DPAT (0.1 and 0.2 mg/kg) significantly increased the time spent in the light zone. PB (10 mg/kg) also significantly increased the number of shuttle crossings between the dark and light zones. Conversely, picrotoxin (1 and 2 mg/kg) significantly decreased locomotion, rearing and time spent in the light zone. beta-CCM (10 mg/kg) and beta-CCE (5 mg/kg) also significantly decreased rearing and time spent in the light zone. Although the same dose of beta-CCE significantly decreased shuttle crossing and rearing in the dark zone, beta-CCE did not change locomotion in either zone. Our results indicate that this apparatus is useful for the assessment of the anxiolytic and anxiogenic activities of drugs in mice.

Portable data acquisition and control apparatus for implanted drug infusion pump interrogation.

Now that implantable drug infusion pumps are well established clinically, methods for diagnosing suspected pump failures are needed. The authors previously constructed a benchtop data acquisition and control apparatus to assist our work in developing new pump technology. Although this device is technically capable of in vivo pump monitoring, it is cumbersome. Thus, they recently created a portable interrogation unit with more limited features. This portable pump interrogation apparatus consists of a 32 bit MS-DOS labtop computer, data acquisition software, an analog/ASCII interface, a pressure transducer, and appropriate fluid conduits. Communication between the device and the implanted pump is via a percutaneous needle puncture of the drug reservoir refill septum. This procedure is identical to that employed in a standard pump refill. Pump performance is evaluated by incrementally filling the pump reservoir while simultaneously measuring reservoir pressure. The resulting data are presented on the computer screen as a plot of pressure versus volume that quickly and simply either eliminates or confirms the reservoir pressure source as a failure mode. Diagnostic runs are saved on file for archival purposes. Their benchtop apparatus has been a valuable and reliable tool over many years of use. The authors believe that their portable apparatus will be equally beneficial.

Freud's speech apparatus and spontaneous speech.

Freud's first book published in 1891 was a monograph entitled On Aphasia. In it he challenges the main authorities of the time by asserting that their manner of understanding aphasias is no longer tenable. Freud proves their theories wrong and presents his own conception of a speech apparatus. The apparatus is the foundation of his clinical and theoretical explanations about the speech function and its pathological manifestations. He built the model of an apparatus capable of explaining spontaneous speech, a function that the competing models of his contemporaries could not fully integrate. Freud's speech apparatus is the first of several models he created to facilitate the understanding of psychic functions. This paper is part of a series dedicated to an in-depth study of the earliest model which would provide the foundations for the understanding of its reappearance in Freud's later theories, and in particular in his analytic technique.

[The role of the basic skeletal system in the stomatognathic apparatus. A critical review of the literature].

The authors make a literature review on the different interpretations of the stomatognathic apparatus and try to identify any component of it and to underline the importance of the basal skeletal structures (maxilla and mandible). In this study the authors examined three female patients affected by different pathologies (dysfunctional, malformative, and traumatic) treated at the Maxillo-Facial Surgery Department of "La Sapienza" University of Rome; in all cases the basal skeletal structures were involved with a total functional alteration of the stomatognathic apparatus. The primary aim of any maxillo-facial surgical treatment is the restoring of skeletal structural harmony with a re-equilibrium of all the components of the apparatus. In conclusion the authors confirm the importance of the basal skeletal system which is strictly connected to all other elements, for the physiologic structural development and functional harmony of the stomatognathic apparatus.

Simulating respiratory loads and tuning of closed-circuit underwater breathing apparatus.

An experimental apparatus for simulating respiratory loads in closed-circuit underwater breathing systems comprises a partly submerged vertical tube having a dual path exit on the submerged end of the tube. One path is controlled by a valve that alters the inertance of the system, changes the natural frequency, and provides the tuning. The vertical tube provides elastic loading to simulate a counterlung or breathing bag in an underwater breathing apparatus (UBA). The experimental apparatus was connected to a breathing machine controlled by a signal generator to simulate a diver's breathing. The benefits of reactance tuning of this apparatus are demonstrated. A mathematical model describes both the simulator and UBA. The simulator can be used with human subjects to study respiratory response to changes in impedance.

An ordered inheritance strategy for the Golgi apparatus: visualization of mitotic disassembly reveals a role for the mitotic spindle.

During mitosis, the ribbon of the Golgi apparatus is transformed into dispersed tubulo-vesicular membranes, proposed to facilitate stochastic inheritance of this low copy number organelle at cytokinesis. Here, we have analyzed the mitotic disassembly of the Golgi apparatus in living cells and provide evidence that inheritance is accomplished through an ordered partitioning mechanism. Using a Sar1p dominant inhibitor of cargo exit from the endoplasmic reticulum (ER), we found that the disassembly of the Golgi observed during mitosis or microtubule disruption did not appear to involve retrograde transport of Golgi residents to the ER and subsequent reorganization of Golgi membrane fragments at ER exit sites, as has been suggested. Instead, direct visualization of a green fluorescent protein (GFP)-tagged Golgi resident through mitosis showed that the Golgi ribbon slowly reorganized into 1-3-micron fragments during G2/early prophase. A second stage of fragmentation occurred coincident with nuclear envelope breakdown and was accompanied by the bulk of mitotic Golgi redistribution. By metaphase, mitotic Golgi dynamics appeared to cease. Surprisingly, the disassembly of mitotic Golgi fragments was not a random event, but involved the reorganization of mitotic Golgi by microtubules, suggesting that analogous to chromosomes, the Golgi apparatus uses the mitotic spindle to ensure more accurate partitioning during cytokinesis.

Modifications of the Golgi apparatus in Saccharomyces cerevisiae lacking microtubules.

BACKGROUND: Disassembly of cytoplasmic microtubules by nocodazole in cultured mammalian cells leads to the disruption of the continuous ribbonlike Golgi apparatus and dispersal of the Golgi elements from their normal juxtanuclear location, close to the microtubule-organizing center (MTOC), toward the cell periphery. Clearing of the drug induces reassembly of the microtubules from the MTOC and reorganization of the Golgi elements into a continuous ribbonlike juxtanuclear structure. In the yeast Saccharomyces cerevisiae, the Golgi apparatus does not form a continuous structure as in mammalian cells but instead constitutes independent units dispersed throughout the cytoplasm. It is the purpose of this article to investigate the role of microtubules in the structure and distribution of the Golgi elements in S. cerevisiae by studying the ultrastructure of cell organelles either in mutant cells deficient in beta-tubulin or in wild-type cells treated with the microtubule-depolymerizing drug nocodazole. METHODS: Two S. cerevisiae yeast strains were used in this study: a control wild-type strain, CUY226 (ade2-101, his3-delta 200, leu2-delta 1, lys2-801, ura3-52 Mat alpha), and a mutant strain, CUY66 (tub2-401, ade2-101, ura3-52, Mat alpha). Nocodazole was added to the wild-type cells cultivated at 30 degrees C, and cells were fixed 5 min, 20 min, and 60 min, respectively, after adding the drug to the culture. Both strains were fixed and examined 5 min, 20 min, and 60 min after shifting the cultures from the permissive temperature of 30 degrees C to the restrictive temperature of 14 degrees C. Cells were fixed in 2% glutaraldehyde, treated for 15 min in 1% sodium metaperiodate, postfixed in reduced osmium, and embedded in Epon. To visualize the three-dimensional configuration of cell organelles, stereopairs were prepared from sections stained with lead citrate and tilted at +/- 15 degrees from the 0 degree position of the goniometric stage of the electron microscope. RESULTS: In mutant cells shifted to restrictive temperature and wild-type cells treated with nocodazole, the main ultrastructural modification was a fragmentation of networks of membranous tubules, which probably correspond to the yeast Golgi apparatus. Secretion granules were still present in growing buds, and they were dispersed in the cytoplasm, which contained in addition numerous small vesicles in the 30-60-nm diameter range. CONCLUSIONS: In normal cells, small vesicles may originate from the endoplasmic reticulum and fuse together to give rise to Golgi networks (Rambourg et al. 1994. Anat. Rec., 240:32-41). If this hypothesis is correct, the observations reported might indicate that intact microtubules orient the flow of small vesicles and favour their fusion into Golgi networks.

Three-dimensional structure of the Golgi apparatus in mouse spermatids: a scanning electron microscopic study.

In this study, the three-dimensional organization of the Golgi apparatus in mouse spermatids was elucidated by preparing testicular tissue with the osmium-DMSO-osmium method and examining it by stereo-scanning electron microscopy. The cis-most saccule was found to be a regular network of anastomotic membranous tubules covered by a single cisterna of ER. The trans-Golgi network was seen to be composed of irregular saccules perforated by pores at the edge. It appears that the anastomosing trans-Golgi network breaks down into strings of connected vesicles which arise from the edge of the saccules during the cap phase of spermiogenesis. Many apparently individual vesicles seen in thin sections through the trans-Golgi network are actually joined in continuous strings. This was the first time that these structures could be visualized directly without three-dimensional image reconstruction. By correlating the morphology of the Golgi apparatus with the stage of acrosome formation, the Golgi cisternae were found to change dynamically in a cis-trans direction from fenestrated saccules to continuous strings of vesicles, which finally dissipated as transport vesicles at the trans aspect. This suggests that the hypothetical model of cisternal maturation, which dictates that cargo moves through the Golgi apparatus without leaving the cisternal lumen and the secretion occurs by progressive maturation of the Golgi cisternae as they move in the cis-trans direction, may be applicable to acrosome formation.

Tridimensional structure of the Golgi apparatus in type A ganglion cells of the rat.

The three-dimensional structure of the whole Golgi apparatus and of its components in type A ganglion cells was examined in thin and thick sections by low- and high-voltage electron microscopy. At low magnification, in 10-micron-thick sections of osmicated cells, the Golgi apparatus formed a broad, continuous perinuclear network. At higher magnification and in thinner sections of cells impregnated with uranyl acetate-lead-copper citrate or postfixed in K-ferrocyanide-reduced osmium, the Golgi apparatus appeared as a heterogeneous structure in which saccular regions characterized by stacks of saccules alternated with intersaccular regions made up of branching membranous tubules which bridged the saccules of adjacent stacks. The saccular regions consisted of the following superimposed elements: a cis-osmiophilic element made up of anastomosing tubules; two or three saccules negative for the phosphatases tested (i.e., nicotinamide adenine dinucleotide phosphatase = NADPase, thiamine pyrophosphatase = TPPase, and cytidine monophosphatase = CMPase); two saccules showing TPPase activity; and one to three trans-sacculotubular elements showing a "peeling-off" configuration, one of which showed CMPase activity. The saccules (phosphatase-negative) on the cis-side of the Golgi stacks showed, in addition to small circular pores, larger perforations in register. The cavities thus formed in the stacks of saccules, called "wells," always associated with small 80-nm vesicles, had a pan shape with the mouth directed toward the cis-face and the bottom closed by a TPPase-positive saccule. In face views of the saccules, the smallest of these perforations showed either a crescent shape, due to the presence of a bud on one side of the perforation, or a circular shape with a single small 80-nm vesicle in the center which was occasionally attached to the saccule by a filiform stalk. Such smaller cavities were considered as the precursors of the larger perforations and eventually of the wells. The small 80-nm vesicles seen in the small cavities or in the wells appeared to form in situ and possibly migrate toward the cisternae of endoplasmic reticulum seen proximal to the cis-face of the stack of saccules. Small 80-nm vesicles were also numerous in the intersaccular regions, along the lateral- and trans-aspects of the Golgi stacks, while larger, 150-to 300-nm vesicles, coated and uncoated, were seen only on the trans-face of the Golgi stacks in proximity to the trans-sacculotubular elements which appear to "peel off" from the Golgi stacks.
