[Morphological and histochemical reactions of an organism to the administration of Clostridium botulinum toxin. IX. Reactions of the neurons of the abducens nerve nucleus and of the Darkshevich's nucleus to the administration of C. botulinum type B toxin].

Pathomorphological and histochemical study carried out in guinea pigs at the stage of paralysis of the limbs after the oral administration of C. botulinum toxin has revealed the presence of morpho-metabolic changes pertaining to phase 2 of the development of dystrophic processes in the neurons of the nucleus of the abducent nerve and Darkshevich's nucleus. At the myasthenic stage the development of dystrophic processes in the neurons of Darkshevich's nucleus has been found to reach phase 3. The study indicates that C. botulinum toxin possesses different tropism in respect of the neurons of the oculomotor nuclei; this fact correlates with the clinical picture of botulism.

Kidney and liver metallothioneins in rats after administration of an organic compound.

Intubation of rats with alpha-mercapto-beta-(2-furyl)-acrylic acid (MFA) for 5 days at 50 mg/kg caused a 7-fold increase in kidney copper concentration, a 2-fold increase in kidney zinc concentration, and a 20% increase in liver zinc concentration. The proteins which bound the increased metals were purified and identified as metallothioneins by their amino acid compositions. Two isoforms were isolated from each organ. Renal thioneins appeared identical to counterpart hepatic apoproteins, but the former bound Cu and Zn in a 2:1 mole ratio and the latter bound only Zn. Kidney contained over 10 times more metallothionein per g of tissue than did liver. In rats previously administered MFA, injection of cadmium sulfate resulted in rapid displacement of liver metallothionein-bound Zn by Cd under conditions where minimal metallothionein was found in Cd-dosed animals not administered MFA. We conclude that MFA induces metallothionein biosynthesis in kidney and liver of normal rats; this is a novel effect for an organic compound.

[Distribution of functionally differing cells in immunocompetent organs on haloperidol administration].

Haloperidol blockade of the dopaminergic system entails redistribution of functionally different cells. Suppressors of IgM and IgG response migrate, respectively, from the spleen and lymph nodes to the bone marrow whose cell transfer leads to inhibition of immune response. The data obtained indicate common regularities in cell distribution under blockade of the dopaminergic and activation of serotoninergic systems. Therefore, at the basis of inhibition of immunogenesis there lie monotypic processes of suppressor cell migration whatever the system responsible for it.

[Residues of chloramphenicol in the calf organism following parenteral administration of the Czechoslovak preparation Chronicin inj].

Using two biological methods the chloramphenicol content in various tissues of 11 calves aged 3--18 days was determined after a single-dose therapeutical administration of the Czechoslovak preparation of Chronicin inj. in the quantity of 12 mg kg-1 body mass. Of four test strains, solely Micrococcus flavus (ATCC 10240) strain proved suitable. Chloramphenicol was quantitatively determined by reading from transformed standard curves. An evalua tion of the two approximately equally sensitive methods yielded the following results: three days after the injection all calves were positive, at 5 and 10 days, one of three calves in each group was negativ similarly to both calves slaughtered 15 days post injection. The residues appeared most frequently in the liver, at the site of injection, and in the fore and hind musculature. Maximum detectable concentrations were recorded in the liver. Culinary and technological treatment of positive meat and liver samples revealed that 15 minutes' boiling and 30 minutes' roasting destroyed the microbiologically active chloramphenicol residues in the samples. Neither deep freezing nor salting was sufficient for residue inactivation, although in some samples a decrease in biological chloramphenicol activity by circa 20--30% was recorded. a preslaughter withdrawal time of 15 days is recommended.
