Sporadic phaeochromocytomas are rarely associated with germline mutations in the von Hippel-Lindau and RET genes.

OBJECTIVE: von Hippel-Lindau (VHL) disease and multiple endocrine neoplasia type 2 (MEN2) are autosomal dominant cancer syndromes. In both conditions, phaeochromocytoma is a prominent feature. It has recently been suggested that phaeochromocytoma can be the presenting and sole clinical manifestation of these multi-organ syndromes. The aim of this study was to ascertain the incidence of VHL and MEN2 among patients with sporadic phaeochromocytoma by mutational analysis. PATIENTS: Twenty-seven unrelated patients with biochemically and/or anatomically proven sporadic phaeochromocytoma were evaluated. DESIGN AND MEASUREMENTS: Constitutional DNA obtained from the patients was analysed by single stranded conformational analysis (SSCP) for mutations within the VHL gene coding sequence and by denaturing gradient gel electrophoresis (DGGE) for predominant mutations in exons 10, 11 and 16 of the RET proto-oncogene. The incidence of patients positive for either VHL or RET germline mutations was assessed. RESULTS: Twenty-six of 27 patients had normal SSCP patterns in all three VHL gene exon segments and only one patient, with an atypical clinical presentation, had an aberrant pattern in exon 3 which upon DNA sequencing was shown to harbor a G to A transversion mutation at nucleotide 695. All patients had normal RET exon 10, 11 and 16 DGGE migration patterns. CONCLUSION: Most, if not all, patients with typical unilateral sporadic phaeochromocytoma do not have von Hippel-Lindau disease or MEN2. Thus, clinical and/or molecular investigation for von Hippel-Lindau disease and MEN2 in this patient population does not appear to be indicated.

Four peroxidase Loci in red-fruited tomato species: genetics and geographic distribution.

The banding patterns of certain anodal peroxidase variants of red-fruited tomato species are governed by alleles at four loci-two alleles per locus. Alleles at three loci code for modified enzyme migration patterns and are codominant in heterozygotes; those at the fourth locus code for presence or absence of a band. No evidence of linkage was detected in preliminary tests between four of the six possible combinations of loci. All variant alleles-i.e., those not represented in the standard genotype of Lycopersicon esculentum-exist in the wild L. pimpinellifolium from coastal Peru; all but Prx-3(n) are also known in L. esculentum from the sympatric region but are rare or absent elsewhere. Between the distributions of alleles of Prx-1 and those of Ge, the gamete-eliminator locus, a significant association exists, which probably does not owe to genetic linkage. The tendency of alleles of Prx loci, as well as those of cm, Ge, h, and Od, to be shared between wild and cultivated taxa in the sympatric region but seldom elsewhere, in addition to published correlated evidence, suggests that the wild alleles tend to substitute in cultivated forms as a result of introgression. In respect to the number of common alleles, cultivated tomatoes more closely resemble the wild L. esculentum var. cerasiforme than L. pimpinellifolium.

Histone H1 proteins act as receptors for the 987P fimbriae of enterotoxigenic Escherichia coli.

The tip adhesin FasG of the 987P fimbriae of enterotoxigenic Escherichia coli mediates two distinct adhesive interactions with brush border molecules of the intestinal epithelial cells of neonatal piglets. First, FasG attaches strongly to sulfatide with hydroxylated fatty acyl chains. This interaction involves lysine 117 and other lysine residues of FasG. Second, FasG recognizes specific intestinal brush border proteins that migrate on a sodium-dodecyl sulfate-polyacrylamide gel like a distinct set of 32-35-kDa proteins, as shown by ligand blotting assays. The protein sequence of high performance liquid chromatography-purified tryptic fragments of the major protein band matched sequences of human and murine histone H1 proteins. Porcine histone H1 proteins isolated from piglet intestinal epithelial cells demonstrated the same SDS-PAGE migration pattern and 987P binding properties as the 987P-specific protein receptors from porcine intestinal brush borders. Binding was dose-dependent and shown to be specific in adhesion inhibition and gel migration shift assays. Moreover, mapping of the histone H1 binding domain suggested that it is located in their lysine-rich C-terminal domains. Histone H1 molecules were visualized on the microvilli of intestinal epithelial cells by immunohistochemistry and electron microscopy. Taken together these results indicated that the intestinal protein receptors for 987P are histone H1 proteins. It is suggested that histones are released into the intestinal lumen by the high turnover of the intestinal epithelium. Their strong cationic properties can explain their association with the negatively charged brush border surfaces. There, the histone H1 molecules stabilize the sulfatide-fimbriae interaction by simultaneously binding to the membrane and to 987P.

Human semen prostaglandins do not affect sperm motility and migration.

Prostaglandin (PG) E2, PGF2 alpha and testosterone (T) concentrations were determined in the seminal plasma of 57 fertility-clinic patients. Results (mean less than SE) were: PGE2, 3.44 +/- 0.38 mu/ml; PGF2 alpha, 138 +/- 0.33 micrograms/ml; T, 429.54 +/- 14.01 pg/ml. The material was divided into 4 groups: normospermic, oligozoospermic, asthenozoospermic and azoospermic. No statistically significant differences were found among the values of the different groups, except for PGE2, which was found to be increased in the asthenozoospermic group. Sperm migration patterns were assessed in each semen sample, using the Kremer capillary tube test and human cervical mucus. Considering each semen sample individually, no correlations whatever were found among sperm count, motility and migration parameters and the respective concentrations of both PGs and T, nor was there a correlation among PGE2, PGF2 alpha, and T values of the same sample.

Differentiation of the sibling species Biomphalaria occidentalis and Biomphalaria tenagophila by the electrophoretic patterns of their hemoglobin.

A simple and rapid method for differentiating the sibling species Biomphalaria tenagophila and Biomphalaria occidentalis by agarose gel electrophoresis (AGE) is described. Snail hemolymph is used as the test sample and the red coloration of the hemoglobin fraction permits visualization of the migration patterns without resorting to specific stains. Moreover, hemolymph samples may be obtained without killing the snail, thus permitting its use for other studies or for breeding.

Integration of newly born dentate granule cells into adult brains: hypotheses based on normal and epileptic rodents.

The granule cells of the dentate gyrus are a population of neurons continuously generated throughout life. In the rat, the morphological development of newly born granule cells generated in the adult share many similarities with granule cells generated during development. These include a specific migration pattern, orientation and progression of neurite outgrowth. It appears as though varied dendritic morphology occurs depending on the position of the granule cells within the granule cell layer. A hypothesis for granule cell migration and differentiation of their dendritic processes is proposed based on normal and epileptic rats. In this hypothesis, the granule cells are generated in the subgranular zone, and then they migrate into the granule cell layer. During this migration, the sequence of neurite outgrowth is described, where the newly born granule cell first sprouts rudimentary processes. One of these processes, the basal dendrite, is transiently present on developing rodent granule cells in rats. However, in seizure-induced rats the basal dendrite often fails to retract, which leads to the formation of hilar basal dendrites, and also perhaps, ectopic granule cells in the hilus. In this review, granule cell development is discussed with relevance to the creation of the recurrent excitatory circuitry in rodent models of temporal lobe epilepsy.

An examination of population changes in Alabama's Black Belt counties: 1960-1970 and 1970-1980.

"Population changes in 11 Alabama Black Belt counties are examined by comparing annual average birth, death, and migration rates during the 1960s with those of the 1970s. The specific focus of the study is migration patterns among the nonwhite segment of the population. Data from the 1980 and earlier censuses are utilized, along with birth and death data from Alabama vital statistics reports." The results show "that net migration losses among nonwhites declined substantially during the 1970s compared with the 1960s and that in two Black Belt counties there was a reversal from heavy net losses to moderate net gains. Whites, on a county-to-county basis, however, were generally characterized by significantly higher rates of loss during the 1970s than in the 1960s."

Failure of the uncoated titanium ProxiLock femoral hip prosthesis.

UNLABELLED: New prostheses should be evaluated for stability and clinical performance. In a prospective randomized clinical trial, we implanted 22 titanium (Ti) and 20 hydroxyapatite-coated (HA) ProxiLock femoral hip prostheses during total hip arthroplasty in 42 patients. The patients were followed for 24 months with clinical, radiographic and radiostereometric analysis. Full weightbearing was allowed immediately postoperatively. One patient with a titanium stem was lost to followup. During the first two months, 34 of the 41 stems subsided and/or rotated towards retroversion, regardless of stem type. At the 24-month followup 35 of the 41 prostheses were either fully stabilized (16 HA and 11 Ti stems) or had clinical irrelevant migration (four HA and four Ti stems). Six Ti prostheses showed continuous migrations with maximums of 4.7 mm translation and 12.2 degrees retroversion; four of these were revised, the other two had no clinical complaints. CLINICAL RELEVANCE: The migration pattern we found indicates insufficient primary fixation of the ProxiLock stem in an immediate full weightbearing protocol. The HA coating improves the secondary stability of the prosthesis compared to the uncoated stem. Early migration is associated with an increased risk of possible future loosening and revision, and therefore we discontinued the use of this prosthesis. LEVEL OF EVIDENCE: Therapeutic Level I. Effects of migration on cancer incidence and resources for prevention and treatment in Florida.

Migration adds a complex dimension to the task of those who plan and allocate resources for health care. The authors offer a methodology for estimating the contribution of migration to the incidence of cancer, allow for age- and sex-specific cancer risks, and estimate, by country, the impact of recent migration on the annual incidence of cancer in Florida. Cancer and migration data were used to develop estimates of the number of cancer cases for Florida counties that were attributable to recent migrants. A net gain and loss ratio was calculated for new cancer cases in 1980 resulting from the 1975-80 migration pattern. Florida data was used because that State has one of the highest crude cancer incidence rates in the nation, is one of the most populous States, and has a population growth from migration rather than from natural increase. Preliminary findings on the relationship between cancer health services resources and net cancer rates from migration are discussed. County cancer health services resources had a strong positive relationship to population size, but the impact of migration on cancer incidence was in a curvilinear relationship to population size.

Isoforms of heavy and light chains of cardiac myosins from rat and rabbit.

The light chains of myosin from atrial and ventricular tissues from rat and rabbit were examined by one- and two-dimensional polyacrylamide gel electrophoresis. The myosin heavy chains were electrophoretically isolated, digested after denaturation in sodium dodecyl sulfate with papain and proteinase from S. aureus V8, and the resulting peptides resolved in one-dimensional gel electrophoresis. The peptide patterns of myosin heavy chains from atrial and ventricular tissues of adult rabbits were different, indicating differences in their primary structures. No such differences could be detected in a total of around 180 peptides produced by the two proteinases from the myosin heavy chains of adult rat atrial and ventricular tissues. With regard to light chains, the same migration pattern was observed for atrial and ventricular tissues from both rat and rabbit. The atrial light chains ALC1 and ALC2 migrated with molecular weights lying between those of the ventricular light chains VLC1 and VLC2. In two-dimensional electrophoresis, the corresponding light chains from rat and rabbit co-migrated. An additional light chain was observed in foetal ventricles, which exhibited identical electrophoretic properties to ALC1 from adult atrial tissues. In rat myofibrillar preparations from atrium and ventricle, an unidentified protein (x) occurred in the region of light chain-1 but with a more acidic isoelectric point, which seems to be related to the developmental stage of these tissues and which could not be detected in rabbit heart tissues or in any skeletal muscles.

Phosphorylation by cAMP-dependent protein kinase inhibits the degradation of tau by calpain.

The effects of cAMP-dependent protein kinase (cAMP-PK) phosphorylation on the degradation of the microtubule-associated protein tau by calpain were studied. Purified bovine brain tau that had been phosphorylated by cAMP-PK had a slower migration pattern on sodium dodecyl sulfate-polyacrylamide gels and a more acidic, less heterogeneous pattern on two-dimensional, nonequilibrium pH gradient electrophoresis (NEPHGE) gels compared with untreated tau. Phosphorylation of tau by cAMP-PK significantly inhibited its proteolysis by calpain compared with untreated tau. To our knowledge this is the first demonstration that phosphorylation of tau by a specific kinase results in increased resistance to hydrolysis by calpain. Tau dephosphorylated by alkaline phosphatase migrated more rapidly on sodium dodecyl sulfate-polyacrylamide gels and also showed an altered two-dimensional NEPHGE pattern. Dephosphorylation of tau had no effect on its susceptibility to calpain proteolysis, indicating that regulation of the susceptibility to calpain hydrolysis is due to the phosphorylation of a specific site(s). These results suggest a role for phosphorylation in regulating the degradation of tau. Abnormal phosphorylation could result in a protease-resistant tau population which may contribute to the formation of paired helical filaments in Alzheimer's disease.

Migration of dendritic cells within 3-D collagen lattices is dependent on tissue origin, state of maturation, and matrix structure and is maintained by proinflammatory cytokines.

The function of dendritic cells (DC) depends on active migration through three-dimensional (3-D) extracellular matrices. We have analyzed the migration of murine DC from different tissue origins within 3-D collagen lattices through the use of time-lapse videomicroscopy and single-cell tracking. Directly after incorporation, 50-90% of DC from the spleen (spDC) and Langerhans cells freshly isolated from the epidermis (fLC) displayed active motility in these matrices. Whereas mature spDC showed multilateral pseudopod dynamics as well as fast and heterogeneous migration, immature fLC displayed a spherical shape with faint membrane processes and very homogenous, slow migration characteristics. In the absence of external stimuli, migration of both, spDC and fLC, vanished after >36 h due to cell death. Maintaining fLC viability by external granulocyte-macrophage colony-stimulating factor or tumor necrosis factor alpha prolonged migration up to 5 days. During this period fLC transformed into mature cells with large dendrites, thereby developing a heterogeneous migration pattern more similar to spDC. In randomly polymerized collagen matrices cell paths were without preferential orientation. In contrast, in artificially aligned lattices directional paths in accordance with the forced fiber orientation were observed. Thus, migration is an inherent property of DC, largely influenced by tissue origin, degree of maturity, and the 3-D structure of the environment.

Lineage, migration, and fate determination of postnatal subventricular zone cells in the mammalian CNS.

We have been examining the developmental fates and migrational patterns of the immature cells in the subventricular zone (SVZ) of the mammalian forebrain by labeling postnatal rat SVZ cells by stereotactic injection of replication-deficient murine retroviruses bearing reporter genes. SVZ cells migrate into adjacent white matter, cortex, and striatum, and differentiate into astrocytes and oligodendrocytes. In white matter, they largely differentiate into oligodendrocytes, whereas in gray matter, they differentiate into both oligodendrocytes and astrocytes. In vitro, SVZ cells are multipotential, able to generate both types of glia, as well as neurons. We infer that developmental fates are in part controlled by important environmental cues that the cells encounter during their migration.

Effect of geographic migration on SMI prevalence estimates.

The prevalence of serious mental illness (SMI) varies by the socioeconomic characteristics of communities. This variation is presumed to be due to the differential incidence of disorders caused by adverse social factors (social causation) and differential geographic migration (social selection and drift). The objective of this study was to measure the geographic migration patterns of adults treated for SMI. A sample of 11,725 adults with three or more psychiatric hospital admissions between July 1978 and November 1992 was drawn from inpatient records. At least one third migrated to different counties between first and last admission. Migration rates were higher for Whites than for African Americans and higher for unmarried than for married patients. There were no significant differences in migration rates by gender or mental disorder. Patient migration did not parallel shifts in the general population. Patient migration was generally toward medium-size, low-income urban counties with relatively declining general populations, and movement was away from both the most rural and the largest urban counties. Public needs assessments and resource allocation policies may understate the need in communities with net SMI out-migration and overstate endemic need where there is net in-migration. In the long term, these same policies may induce migration through resource allocation decisions.

Rural migration and agrarian reform in Russia: a research note.

This study focuses primarily on trends in rural-urban migration in Russia and the former Soviet Union. "New data suggest that a historic shift in migration patterns is underway in Russia, a change that may have profound long-term effects on agrarian reform and the nature of the Russian countryside. We begin with a short review of past rural migration trends and the rural demographic situation, in part using archival data for an oblast in central Russia. We will then present new data on rural migration. Finally, we assess the implications of rural migratory trends for agrarian reform in Russia."

Age-dependent changes in extracellular proteins, aminopeptidase and proteinase activities in Frankia isolate BR.

To investigate protein secretion by the nitrogen-fixing actinomycete Frankia isolate BR, we designed a rapid DEAE adsorption, salt elution and Biogel P6DG desalination method to concentrate protein from the growth medium. Secreted proteins reached a maximum concentration (5.6 gm l-1) in the medium at growth arrest. Analysis by SDS-PAGE detected up to 63 extracellular polypeptides when Frankia cells were grown under stirred conditions in BAP medium supplemented with phosphatidylcholine and MES buffer and 65 proteins in stirred BAP media alone. The pattern of extracellular polypeptides changed during growth. Several extracellular proteolytic activities were detected and compared with intracellular ones. The substrate specificity of the extracellular and intracellular aminopeptidase activities were the same. Also, the electrophoretic migration patterns of secreted and intracellular aminopeptidases could not be distinguished. Secretion of the proline-specific aminopeptidase FAP proteinase (PF) were secreted: 10 had the same electrophoretic mobility as their intracellular counterparts after SDS-gelatine-PAGE while five (PF - 39.5, PF - 38.5, PF - 36.5, PF - 25.5 and PF - 20.5 kDa) had a different electrophoretic mobility and, therefore, appeared to be exclusively extracellular. At least seven extracellular proteinases appeared to increase coordinately in activity shortly before growth arrest.

Expression and functional properties of four slow skeletal troponin T isoforms in rat muscles.

We investigated the expression and functional properties of slow skeletal troponin T (sTnT) isoforms in rat skeletal muscles. Four sTnT cDNAs were cloned from the slow soleus muscle. Three isoforms were found to be similar to sTnT1, sTnT2, and sTnT3 isoforms described in mouse muscles. A new rat isoform, with a molecular weight slightly higher than that of sTnT3, was discovered. This fourth isoform had never been detected previously in any skeletal muscle and was therefore called sTnTx. From both expression pattern and functional measurements, it appears that sTnT isoforms can be separated into two classes, high-molecular-weight (sTnT1, sTnT2) and low-molecular-weight (sTnTx, sTnT3) isoforms. By comparison to the apparent migration pattern of the four recombinant sTnT isoforms, the newly described low-molecular-weight sTnTx isoform appeared predominantly and typically expressed in fast skeletal muscles, whereas the higher-molecular-weight isoforms were more abundant in slow soleus muscle. The relative proportion of the sTnT isoforms in the soleus was not modified after exposure to hindlimb unloading (HU), known to induce a functional atrophy and a slow-to-fast isoform transition of several myofibrillar proteins. Functional data gathered from replacement of endogenous troponin complexes in skinned muscle fibers showed that the sTnT isoforms modified the Ca(2+) activation characteristics of single skeletal muscle fibers, with sTnT2 and sTnT1 conferring a similar increase in Ca(2+) affinity higher than that caused by low-molecular-weight isoforms sTnTx and sTnT3. Thus we show for the first time the presence of sTnT in fast muscle fibers, and our data show that the changes in neuromuscular activity on HU are insufficient to alter the sTnT expression pattern.

Application of immunohistochemistry to the isolated mucosa of the mouse gastrointestinal tract, with special reference to somatostatin cells.

In the present study, in order to easily grasp whole images of somatostatin (D) cells, the isolated mucosa of the mouse gastrointestinal tract was immunohistochemically treated. The present study revealed that: (1) in the stomach, small-intestinal villi and colon, about 20% of the D cells extrude basal cytoplasmic processes, showing terminal expansions in many cases; on the other hand, in the crypts of the small intestine, few D cells possess basal processes, and (2) in the stomach, there is no determined tendency in the direction of the basal processes of the D cells; on the other hand, in the small-intestinal villi and colon, most D cell basal processes run toward the villus base and colon crypt bottoms. The direction of the basal processes of the D cells in the gastrointestinal tract seems to be mostly in favor of the migration pattern of epithelial cells described previously. It is likely that, if the targets of the D cells are near the D cells, the basal process is not necessary for local secretion of somatostatin. During migration, however, D cells might extrude basal processes to keep relationships with their targets.
