Development of plant regeneration and transformation protocols for the desiccation-sensitive weeping lovegrass Eragrostis curvula.
A tissue culture protocol, suitable for transformation, was established for the pasture grass Eragrostis curvula. Callus was generated in the dark from leaf and seed tissues on a medium comprising MS salts supplemented with 2 mg/l 2,4-D, 0.01 mg/l BAP and 2% sucrose. Plant regeneration occurred via organogenesis on the same medium with 6% and 3% sucrose for shoot and root formation, respectively. Optimal regeneration (50 plantlets per callus) occurred when light of 45 micromol/m2 per s was used. The yeast Saccharomyces cerevisiae Hsp12 gene was cloned into the Sac1 of the pCAMBIAUbeeQ vector and callus was transformed by biolistic bombardment. Best transformation (30%) occurred when the target tissue was bombarded twice at a distance of 70 mm using a bombardment pressure of 9,100 kPa. Although successful transformation and transcription of the Hsp12 gene occurred, no Hsp12 protein was found present in tissue extracts of transformed grass.
Genome polymorphisms and gene differential expression in a 'back-and-forth' ploidy-altered series of weeping lovegrass (Eragrostis curvula).
Molecular markers were used to analyze the genomic structure of an euploid series of Eragrostis curvula, obtained after a tetraploid dihaploidization procedure followed by chromosome re-doubling with colchicine. Considerable levels of genome polymorphisms were detected between lines. Curiously, a significant number of molecular markers showed a revertant behavior following the successive changes of ploidy, suggesting that genome alterations were specific and conferred genetic structures characteristic of a given ploidy level. Genuine reversion was confirmed by sequencing. Cluster analysis demonstrated grouping of tetraploids while the diploid was more distantly related with respect to the rest of the plants. Polymorphic revertant sequences involved mostly non-coding regions, although some of them displayed sequence homology to known genes. A revertant sequence corresponding to a P-type adenosine triphosphatase was found to be differentially represented in cDNA libraries obtained from the diploid and a colchiploid, but was not found expressed in the original tetraploid. Transcriptome profiling of inflorescence followed by real-time polymerase chain reaction validation showed 0.34% polymorphic bands between apomictic tetraploid and sexual diploid plants. Several of the polymorphic sequences corresponded to known genes. Possible correlation between the results observed here and a recently reported genome-wide non-Mendelian inheritance mechanism in Arabidopsis thaliana are discussed.

Both the eyes and the nose are in constant contact with the external environment and thus are common targets for immune-mediated disease. Progress in understanding the pathophysiology of allergic nasal and ocular disease has greatly enhanced the approach to therapy. Chronic rhinitis may be caused by a variety of immunologic and nonimmunologic mechanisms. Allergic and nonallergic factors often coexist, which complicates diagnosis and therapy. In external ocular disorders, both IgE- and IgG-mediated mechanisms may be involved. Elucidation of these mechanisms will be important in improving diagnosis and in facilitating development of new therapeutic modalities.

Beyond weeping and crying: a gender analysis of expressions of depression.
OBJECTIVE: To explore depression from a gender perspective, by capturing depressed women's and men's formulations of their experiences and understanding of their situation. DESIGN: Qualitative interview study. SETTING: A healthcare centre in northern Sweden. SUBJECTS: Eighteen patients who had been diagnosed with depression and treated for at least 6 months were interviewed in depth, both women and men of different ages and social status. Open questions were posed around the themes of Malterud's key questions, focusing especially on how the informants conveyed their experiences. Interviewing and qualitative data analysis went on simultaneously. RESULTS: The experience of depression held similarities for men and women, but the outward manifestations differed by gender as well as socioeconomic status. Though experiences of high demands underlay the narratives of all informants, home or work had different priority. Men talked more easily about physical distress--often the heart--than about emotions. Women verbalized more readily emotional distress--shame and guilt--while physical symptoms often revolved around the stomach. Men dealt with insecurity by aggrandizing their previous competence, women by self-effacement. CONCLUSION. As clinicians we must listen attentively not only to the manifest but to the avoided or unarticulated. By doing so we might counteract normative gender patterns that highlight the depression of women and conceal that of men.

Crying, weeping or tearfulness in British and Israeli adults.
Participants (aged 20-42 years; N = 448) from Britain and Israel answered questions on their crying behaviour. They were more likely to cry alone than when with others and least likely to cry with people they did not know well. Women cried more often and for longer and with more intensity than men. Britons cried more than Israelis. Explanations in terms of differential inhibition were not supported. The sex difference was especially marked for situations involving criticism from others, anger, or problems with work, where men were particularly unlikely to cry, and least for those situations Darwin described as involving tender feelings. The pattern was related to differences documented elsewhere in which men tend to more active and confrontational styles of behaviour. British-Israeli differences were consistent with the effects of military service. Tearfulness for tender feelings increased with age in all groups. A principal component analysis distinguished crying that is intense, long lasting and difficult to stop from crying that is more diverse in appearance but more controllable. The distinction seems related to whether crying pre-empts or concurs with other ongoing activities.
