Metabolic profile of FYX-051 (4-(5-pyridin-4-yl-1h-[1,2,4]triazol-3-yl)pyridine-2-carbonitrile) in the rat, dog, monkey, and human: identification of N-glucuronides and N-glucosides.

FYX-051, 4-(5-pyridin-4-yl-1H-[1,2,4]triazol-3-yl)pyridine-2-carbonitrile, is a novel xanthine oxidoreductase inhibitor that can be used for the treatment of gout and hyperuricemia. We examined the metabolism of FYX-051 in rats, dogs, monkeys, and human volunteers after the p.o. administration of this inhibitor. The main metabolites in urine were pyridine N-oxide in rats, triazole N-glucoside in dogs, and triazole N-glucuronide in monkeys and humans, respectively. Furthermore, N-glucuronidation and N-glucosidation were characterized by two types of conjugation: triazole N(1)- and N(2)-glucuronidation and N(1)- and N(2)-glucosidation, respectively. N(1)- and N(2)-glucuronidation was observed in each species, whereas N(1)- and N(2)-glucosidation was mainly observed in dogs. With regard to the position of conjugation, N(1)-conjugation was predominant; this resulted in a considerably higher amount of N(1)-conjugate in each species than N(2)-conjugate. The present results indicate that the conjugation reaction observed in FYX-051 metabolism is unique, i.e., N-glucuronidation and N-glucosidation occur at the same position of the triazole ring, resulting in the generation of four different conjugates in mammals. In addition, a urinary profile of FYX-051 metabolites in monkeys and humans was relatively similar; triazole N-glucuronides were mainly excreted in urine.

Imidazo[1,2-alpha]pyridines. III. Synthesis and bradycardic activity of new 5-imidazo[1,2-alpha]pyridin-6-ylpyridine derivatives.

Structural modification of the cardiotonic agent, loprinone (E-1020, 1), suggested by data that it has a less positive chronotropic effect than milrinone (15), led us to find novel bradycardic agents that were structurally different from homoveratryl amine derivatives. Alkyl-oxy, -thio, and -amino derivatives at the 2-position of the pyridine ring of 1 produced bradycardic activity without a significant effect on blood pressure and myocardial contractility. Aryloxy analogues also decreased heart rate, and members with an electron-withdrawing group at the ortho position of the phenyl ring showed higher activity. Replacement of the imidazo[1,2-alpha]pyridine with pyridine resulted in diminished activity. The mechanism of bradycardic activity of these compounds seems to be direct action on the sinus node.

A comparison between pyridinic and non-pyridinic extraction of allergenic materials.

The effect of addition of pyridine in the extracting solvent of allergenic materials was studied in the following substances. Dermatophagoides farinae, Chenopodium album, Parietaria judaica, Phleum pratense, Poa pratensis, and Secale cereale. Pyridinated extracts differed significantly in protein content, isoelectric focusing profile, and allergenic activity from the non-pyridinated extracts. On the other hand, no difference was detected between both extraction systems when their adsorption capacity to aluminium gels was compared.

Unimpaired metabolism of pyridine dinucleotides and adenylates in Chinese hamster ovary cells during oxidative stress elicited by cytotoxic doses of copper-putrescine-pyridine.

Copper-putrescine-pyridine (Cu-PuPy) effectively dismutates superoxide but is also known to produce H2O2 in a redox cycle with glutathione. The treatment of Chinese hamster ovary (CHO) cells with 0.2 mM Cu-PuPy reduced clonogenic survival to 10(-3) in 50 min and caused significant oxidation and depletion of glutathione and continuous accumulation of protein-glutathione mixed disulfides. Remarkably, other important functional parameters of cell metabolism were not impaired: adenylate pool size, adenylate energy charge and the redox ratios of NADP(H) and NAD(H) remained constant. Moreover, within 200 min the pool size of NADP(H) increased linearly by a factor of four at the expense of the NAD(H) pool, resulting in an 8-fold increase in the ratio of NADPH to glutathione disulfide. Also, Cu-PuPy led to a dose-dependent, persistent inactivation of glutathione reductase, which could be reversed by copper chelators. In contrast to Cu-PuPy, glucose oxidase-generated H2O2 induced oxidation and loss of pyridine dinucleotides, depletion of the adenylate pool and deterioration of the energy charge. Oxidation and depletion of bulk glutathione were comparable to a Cu-PuPy treatment, but formation of protein-glutathione mixed disulfides was significantly less pronounced and reversible. The data indicate that the critical factor in Cu-PuPy cytotoxicity is not its function as catalyst of glutathione oxidation and H2O2 generation, but essentially its disruption of antioxidative cellular defence by inactivation of glutathione reductase. The data further suggest that Cu-PuPy inhibits ADP-ribosylation. This would explain why pyridine dinucleotide and adenylate pools are unaffected, and may be an essential prerequisite for the observation that cells, albeit sublethally damaged and denuded of their antioxidative defence, may be rescued by extending Cu-PuPy treatment.

F/Cl-exchange on AlCl(3)-pyridine adducts: synthesis and characterization of trans-difluoro-tetrakis-pyridine-aluminum-chloride, [AlF2(Py)4]+Cl-.

Whereas liquid CCl3F reacts with solid AlCl3 exothermically under chlorine-fluorine-exchange already above -20 degrees C, no reaction takes place between CCl3F and the pyridine complexes of AlCl3 (AlCl3.Py, AlCl3.2Py, or AlCl3.3Py) up to 100 degrees C. The desired chlorine by fluorine substitution on the monomer AlCl3-pyridine adducts occurs, however, easily using Me3SiF as fluorinating agent. By reacting AlCl3.3Py with Me3SiF (even up to 10-fold stoichiometric excess) in pyridine as a solvent, only two of the three Cl atoms can be substituted by fluorine, leading in good yield to the new "mixed aluminum halide", AlF2Cl.4Py. Actually, it represents the first example of a stable solid donor-acceptor adduct of an aluminum-III halide with two different halogens of defined stoichiometry. It was characterized by multinuclear solid-state NMR (27Al and 19F), IR spectroscopy, as well as single-crystal structure analysis. The new compound has an ionic solid-state structure with helical trans-octahedral [(Py)4AlF2]+ cations and isolated Cl- anions. The comparison of its 27Al MAS solid-state NMR spectra with those of a compound bearing the analogous [(Py)4AlCl2]+ cation reveals an extreme increase in the quadrupolar coupling constants, from 0.24 MHz in case of the chlorine cation to about 16 MHz in case of the new [(Py)4AlF2]+ cation.

Mechanistic evaluation of the halocyclization of 4-penten-1-ol by some Bis(2-substituted pyridine) and Bis(2,6-disubstituted pyridine)bromonium triflates

The halocyclization reaction of 4-penten-1-ol mediated by various bis(2-substituted pyridine) and (2,6- disubstituted pyridine)bromonium triflates (P(2)Br(+)OTf(-)) was investigated to determine the influence of the substituents on the mechanism of reaction. In all cases, the reaction proceeds via a two-step process where the starting P(2)Br(+) reversibly dissociates to a reactive monosubstituted PBr(+), which then is captured by 4-penten-1-ol to form halocyclized product (2-bromomethyltetrahydrofuran). The dissociation rate constant of P(2)Br(+) (k(d)) is sensitive to the steric bulk at the 2- and 6-positions, and in the case of the 2, 6-dicyclohexylpyridine or 2,6-dicyclopentylpyridine, the P(2)Br(+) species are too unstable to isolate. The partitioning ratio of the reactive intermediate (PBr(+)) between reversal and product formation (k(-)(d)/k(2)) is not particularly sensitive to the nature of the pyridine, the limiting values being 3-7 except in the case of bis(2(-)-menthylpyridine)bromonium triflate where the k(-)(d)/k(2) ratio is approximately 80. The reaction of 4-penten-1-ol and its OD isotopomer with bis(lutidine)bromonium triflate was investigated to determine the deuterium kinetic isotope effect (dkie) on the bromocyclization reaction. The (k(-)(d)/k(2))(H/D) ratio is 1.0, indicating that the rate-limiting step for the bromocyclization is probably formation of a PBr(+)-4-penten-1-ol complex which does not involve substantial changes in the bonding of the OH. The cyclization of 4-penten-1-ol and 4-pentenoic acid mediated by bis(2(-)-menthylpyridine)bromonium triflate produces an enantiomeric excess in the cyclized products of only 2.4% and 4.8% respectively.

One-Step conversion of pyridine N-oxides to tetrazolo[1,5-a]pyridines.

Pyridine N-oxides were converted to tetrazolo[1,5-a]pyridines in good to excellent yield by heating in the presence sulfonyl or phosphoryl azides and pyridine in the absence of solvent. Various sulfonyl and phosphoryl azides were screened for reactivity under a standard set of conditions. Diphenyl phosphorazidate was the most convenient reagent and gave high yields. Reaction optimization, scope, and scalability are discussed.

Bis(mu-pyridine-2,6-carboxylato-O,N, O':O)bis[triaquamanganese(II)]-pyridine-2,6-dicarboxylic acid (1/2).

In the structure of the title compound, [Mn(2)(C(7)H(3)NO(4))(2)(H(2)O)(6)].2C(7)H(5)NO(4), a centrosymmetric dinuclear complex, hexaaaquabis(pyridine-2, 6-dicarboxylato)dimanganese(II) and free pyridine-2,6-dicarboxylic acid are present in a 1:2 ratio. In the complex, each Mn(2+) ion is coordinated by three O atoms and one N atom from the pyridine-2, 6-dicarboxylate ligands and by three water O atoms, resulting in a distorted pentagonal bipyramidal coordination. Within the centrosymmetric dinuclear complex, two Mn(2+) ions are bridged by two carboxylate O atoms. The crystal structure is stabilized by hydrogen bonds involving all the H atoms of the water ligands.

Silver(I) complexes of 2-(2-aminoethyl)pyridine: nitrate and perchlorate salts of bis[mu-2-(2-aminoethyl)pyridine-kappa(2)N:N']disilver(I).

2-(2-Aminoethyl)pyridine (2-aep, C7H10N2) acts as a bridging ligand in bis[mu-2-(2-aminoethyl)pyridine-kappa(2)N:N']disilver(I) dinitrate, [Ag2(2-aep)2](NO3)2, and bis[mu-2-(2-aminoethyl)pyridine-kappa(2)N:N']disilver(I) diperchlorate, [Ag2(2-aep)2](ClO4)2. Both salts contain the dinuclear [Ag2(2-aep)2]2+ cation, which possesses a crystallographic inversion center. The Ag...Ag distance is 3.1163 (5) A for the nitrate and 3.0923 (3) A for the perchlorate salt, and may indicate a weak d(10)-d(10) interaction in each case. Essentially linear coordination of the AgI atom is perturbed by weak coordination to the anionic O atoms. These latter interactions organize the dinuclear cations into one-dimensional polymeric chains in the crystals of the two salts.

Synthesis and antiproliferative activity in vitro of new derivatives of 3-aminopyrazolo[3,4-b]pyridine. Part 1. Reaction of 3-aminopyrazolo[3,4-b]pyridine with 1,3-, 1,4-diketones and alpha,beta-unsaturated ketones.

The synthesis of several new pyrazolo[3,4-b]pyridine, pyrido[2',3':3,4]-pyrazolo[1,5-a]pyrimidine and imidazo[1',2':1,5]pyrazolo[3,4-b]pyridine derivatives is described. The obtained compounds were tested for their antiproliferative activity in vitro. One of them, 4-phenyl-2-(3,4,5-trimethoxy-beta-styrylo)pyrido- [2',3':3,4]pyrazolo[1,5-a]pyrimidine (9), revealed cytotoxic properties against the cells of all three human cancer cell lines applied. Another one, 2,4-dimethyl-pyrido[2',3':3,4]pyrazolo[1,5-a]-pyrimidine (2), revealed weak cytotoxic activity only against the cells of human bladder cancer cell line HCV29T. All other compounds tested did not reveal any cytotoxic activity.

Cytogenetic effects of a food mutagen, 2-amino-1-methyl-6-phenylimidazo[4,5-beta]pyridine (PhIP), and its metabolite, 2-hydroxyamino-1-methy-6-phenylimidazo[4,5-beta]pyridine (N-OH-PhIP), on human and Chinese hamster cells in vitro.

2-Amino-1-methyl-6-phenylimidazo[4,5-beta]pyridine (PhIP) induced structural chromosomal aberrations (CAs) and sister-chromatid exchanges (SCEs) in human lymphocytes and human diploid fibroblasts (TIG-7) at concentrations above 12.5 microgram /ml in the presence of rat S9 mix. PhIP also elevated the frequencies of SCEs in human lymphocytes in the presence of rat S9 at concentrations above 2.0 microgram/ml with dose-dependency. A proximate form of metabolites of PhIP, 2-hydroxy-amino-1-methyl-6-phenylimidazo[4,5-beta]pyridine (N-OH-PhIP), caused CAs in human and Chinese hamster fibroblast cells in the absence of S9 mix at concentrations above 0.75 microgram/ml and 1.25 microgram/ml, respectively, which were 10 times lower than the effective concentration of PhIP. No marked differences were observed in the cytogenetic sensitivity to N-OH-PhIP between human and Chinese hamster cells, except between lymphocytes obtained from different donors.

Inhibitors of VEGF receptors-1 and -2 based on the 2-((pyridin-4-yl)ethyl)pyridine template.

We have developed a series of novel potent ((pyridin-4-yl)ethyl)pyridine derivatives active against kinases VEGFR-1 and -2. Both specific and dual ATP-competitive inhibitors of VEGFR-2 were identified. Kinase selectivity could be controlled by varying the arylamino substituent at the 1,3,4-oxadiazole ring. The most specific molecules displayed >10-fold selectivity for VEGFR-2 over VEGFR-1. Compound activities in vitro and in cell-based assays (IC(50)<100nM) were similar to those of reported clinical and development candidates, including PTK787 (Vatalanibtrade mark). High permeability of active compounds across the Caco-2 cell monolayer (>30x10(-5)cm/min) is indicative of their potential for intestinal absorption upon oral administration.

Zinc(II) complexes derived from pyridine-2-carbaldehyde thiosemicarbazone and (1E)-1-pyridin-2-ylethan-1-one thiosemicarbazone. Synthesis, crystal structures and antiproliferative activity of zinc(II) complexes.

The complexes [ZnCl(2)(HFoTsc)xH(2)O], [Zn(FoTsc)(2)], [ZnCl(2)(HAcTsc)xH(2)O] and [Zn(AcTsc)(2)], where HFoTsc and HAcTsc is pyridine-2-carbaldehyde thiosemicarbazone and (1E)-1-pyridin-2-ylethan-1-one thiosemicarbazone respectively, have been prepared and structurally characterized by means vibrational, and NMR ((1)H and (13)C) spectroscopy. The crystal structures of the complexes [ZnCl(2)(HFoTsc)xH(2)O], [Zn(AcTsc)(2)] and [ZnCl(2)(HAcTsc)xH(2)O] have been determined by X-ray crystallography. The metal co-ordination geometry of [ZnCl(2)(HFoTsc)xH(2)O] and [ZnCl(2)(HAcTsc)xH(2)O] is described as distorted square pyramidal and the two complexes are self-assembled via pi-->pi stacking interactions and intermolecular hydrogen bonds. In these two cases molecular recognition of the hydrogen bonds leads to aggregation and a supramolecular assembly of infinite two-dimensional network. The metal co-ordination geometry of [Zn(AcTsc)(2)] is described as distorted octahedral configuration in a trans-N(2)-cis-N(1)-cis-S configuration. HFoTsc and HAcTsc and the zinc complexes have been evaluated for antiproliferative activity in vitro against the cells of two human cancer cell lines: MCF-7 (human breast cancer cell line), T24 (bladder cancer cell line) and a mouse fibroblast L-929 cell line. The cytotoxic activity shown by these compounds indicates that coupling of HFoTsc and HAcTsc to Zn(II) metal center result in metallic complexes with important biological properties since they display IC(50) values in a microM range similar to or better than that of the antitumor drug cis-platin and are considered as agents with potential antitumor activity candidates for further stages of screening in vitro and/or in vivo.

Anticonvulsant activity of two metabotropic glutamate group I antagonists selective for the mGlu5 receptor: 2-methyl-6-(phenylethynyl)-pyridine (MPEP), and (E)-6-methyl-2-styryl-pyridine (SIB 1893).

The selective mGlu5 antagonists, MPEP, 2-methyl-6-phenylethynyl-pyridine, and SIB1893, (E)-6-methyl-2-styryl-pyridine, have been evaluated as antiepileptic drugs in DBA/2 mice and lethargic mice. Clonic seizures induced by the selective mGlu5 agonist, (R,S)-2-chloro-5-hydroxyphenylglycine (CHPG), 3 micromol intracerebroventricularly (i.c.v.), are potently suppressed by both compounds (MPEP, ED(50)=0.42 [0.28-0.62] mg/kg intraperitoneally (i.p.); SIB 1893 ED(50)=0.19 [0.11-0.33] mg/kg i.p. ). Clonic seizures induced by the mGlu1,5 agonist, 3, 5-dihydroxyphenylglycine (DHPG), 1.5 micromol i.c.v., are less potently suppressed by both compounds (MPEP, ED(50)=22 [13-38] mg/kg i.p., 110 [67-180] nmol i.c.v.; SIB1893, ED(50)=31 [18-54] mg/kg i.p. , 95 [82-110] nmol i.c.v.). Sound-induced seizures in DBA/2 mice are suppressed at 15 min by MPEP and SIB 1893 (MPEP ED(50) clonic seizures=18 [10-32] mg/kg i.p., 93 [69-125] nmol i.c.v.; tonic seizures=6.1 [4.5-8.3] mg/kg i.p., 46 [26-80] nmol i.c.v.; SIB 1893 ED(50) clonic seizures=27 [17-44] mg/kg i.p., 825 [615-1108] nmol i. c.v., tonic seizures=5.4 [3.4-8.6] mg/kg i.p., 194 [113-332] nmol i. c.v.). The ED(50) for MPEP for impaired rotarod performance is 128 [83-193] mg/kg i.p., at 15 min, i.e. a therapeutic index for sound-induced seizures of 5-20. In lethargic mice (lh/lh), a genetic absence model, MPEP, 50 mg/kg i.p., caused a marked reduction in the incidence of spontaneous spike-and-wave discharges. These selective antagonists of mGlu5 block seizures due to activation of mGlu5 at very low systemic doses. At rather higher doses they block convulsive and non-convulsive primary generalised seizures.

Synthesis and biological evaluation of pyridine-modified analogues of 3-(2-aminoethoxy)pyridine as novel nicotinic receptor ligands.

Analogues of the potent nicotinic receptor agonist 3-(2-aminoethoxy)pyridine substituted at the 5' and 6'-positions of the pyridine ring were synthesized and tested in vitro for nicotinic receptor binding activity (displacement of [(3)H](-)cytisine from whole rat brain synaptic membranes). The substituted analogues exhibited K(i) values ranging from 0.076 to 319 nM compared to a K(i) value of 26 nM for compound 1. Among the compounds tested, 5'-vinyl-6'-chloro substituted 1 was the most potent.

Normal coordinate analysis of 4-aminopyridine. Effect of substituent on pyridine ring in metal complexes of 4-substituted pyridines.

The normal coordinate analysis has been performed for 4-aminopyridine (4-apy) assuming C2v molecular symmetry. A Urey-Bradley force field has been used. The force constants are adjusted to fit the observed frequencies for 4-apy and its deuterated species. The vibrational assignment has been made on the basis of the calculated frequencies and potential energy distributions. The calculated frequencies were in good agreement with the observed frequencies. The substituent effect upon the M-nitrogen (ligand) (L = 4-methlypyridine, 4-ethylpyridine, 4-vinylpyridine. 4-aminopyridine and 4-cyanopyridine) and pyridine ring frequencies has also been investigated. The frequency shifts are found to be sensitive to the substituent in the 4-position of the pyridine ring.

Synthesis of potential anticancer agents: imidazo[4,5-c]pyridines and imidazo[4,5-b]pyridines.

The 1,2-dihydropyrido[3,4-b]pyrazines (1) are mitotic inhibitors with significant antitumor activity in mice. Also, the active imidazo[4,5-b]pyridine 3 was shown to cause the accumulation of cells at mitosis. Routes were developed for the synthesis of congeners of 3 by cyclization of 4-(substituted amino)-5,6-diaminopyridines with ethyl orthoformate. Oxidative cyclization of either 4,5- or 5,6-diaminopyridines with aryl aldehydes produced the [4,5-c] and [4,5-b] imidazopyridine ring systems, respectively. The latter reaction with 6-(substituted amino)-4,5-diaminopyridines gave imidazo[4,5-c]pyridine ring analogues of 1. Biological studies indicated that the target compounds were less active than 1 and 3.
