Cardiac glycosides. 1. A systematic study of digitoxigenin D-glycosides.

A series of digitoxigenin glycosides was studied: five with beta-D-sugars varying stepwise in sugar structure from beta-D-digitoxose to beta-D-galactose, including one beta-D/alpha-D pair. I50 values for these glycosides and digitoxigenin were determined with hog kidney Na+, K+-ATPase. These data suggest a major and unexpected role for 4'-OH conformation in the sugar. All the glycosides with an equatorial 4'-OH were more active than the two with the 4'-OH axial [digitoxigenin beta-D-galactoside (6) I50 = 6.45 X 10(-8) M; digitoxigenin 2'-deoxy-alpha-D-ribo-hexopyranoside (alpha-3a) I50 = 9.33 X 10(-8) M; digitoxigenin I50 = 1.17 X 10(-7) M]. Stereochemistry of the 3'-OH had much less of an activity role than that of the 4'-OH, in contrast to existing models of "sugar-site" binding.

A new pregnane glycoside and a furostanol glycoside from Digitalis cariensis.

Chemical investigation of the aerial parts of Digitalis cariensis Boiss. ex Jaub. & Spach resulted in the isolation of a new pregnane glycoside, cariensisoside (1) and a furostanol glycoside, uttroside A (2), along with the two known phenylethanoid glycosides, lugrandoside (3) and maxoside (4). On the basis of spectral (UV, IR, NMR, MS) and chemical methods, compounds 1 and 2 were identified as digifologenin-3-O-beta-glucopyranosyl-(1-->4)-beta-oleandropyranoside and 3-O-(beta-lycotetraosyl)-26-O-(beta-glucopyranosyl)-(25R)-22 alpha-methoxy-5-furostane-3 beta,26-diol, respectively.

The retarded rate of acid-catalyzed solvolysis of glycoside bonds between reducing-end glucose residue and ceramide in glycosphingolipids compared with that of glycoside bonds between hexopyranosides.

Rates of acid-catalyzed solvolysis of glycoside bonds in glycosphingolipids were compared to establish a basis for conducting saccharide analysis. Permethylated globotetraosylceramide and asialogangliotriaosylceramide as model compounds for methylation and sugar composition analysis, respectively, were solvolyzed under acidic conditions and the sugar components thus obtained were determined at specified times by gas liquid chromatography, after they had been derivatized. Reducing-end glucose residues in both compounds were liberated more slowly than other sugar residues. Glycoside bonds between reducing-end glucose and ceramide in glycosphingolipids would thus appear to be more resistant towards acid-catalysed solvolysis than other glycoside bonds between hexopyranosides.

Medicinal flowers. VI. Absolute stereostructures of two new flavanone glycosides and a phenylbutanoid glycoside from the flowers of Chrysanthemum indicum L.: their inhibitory activities for rat lens aldose reductase.

Two new flavanone glycosides, (2S)- and (2R)-eriodictyol 7-O-beta-D-glucopyranosiduronic acids, and a new phenylbutanoid glycoside, (2S, 3S)-1-phenyl-2,3-butanediol 3-O-beta-D-glucopyranoside, were isolated from the flowers of Chrysanthemum indicum L. cultivated in China together with eight flavonoids. The absolute stereostructures of the new compounds were determined on the basis of chemical and physicochemical evidence. Both of the new flavanone glycosides were found to show inhibitory activity for rat lens aldose reductase.

Analogues of disaccharides and glycosides containing a cyclic guanidinium structure show varying inhibitory effects on glycoside hydrolases.

By condensation of 1,3-diamino-2,4-(R)-O-benzylidene-1,3-dideoxy-D-erythritol (3) and 1,3-diamino-2,4-di-O-benzyl-1,3-dideoxy-D-threitol (4) with methyl 2,3,6-tri-O-benzyl-4-deoxy-4-iso-thiocyanato-beta-D-glucopyranosid e (9) the (1-->4)-linked disaccharide analogues 4-deoxy-4-[(4R,5S)-5-hydroxy-4-(hydroxymethyl)-1,4,5,6-tetrahydropyri midin-2- yl[amino-alpha,beta-D-glucopyranose hydrochloride (15) and 4-deoxy-4-[(4R,5R)-5-hydroxy-4-(hydroxymethyl)-1,4,5,6-tetrahydropyri midin- 2-yl]amino-alpha,beta-D-glucopyranose hydrochloride (18) were synthesized. By the same reaction sequence, using 3 and methyl isothiocyanate, the glycoside analogue (4R,5S)-5-hydroxy-4-(hydroxymethyl)-2-methylamino-1,4,5,6- tetrahydropyrimidine hydrochloride (20) was obtained. All compounds possess in their 'glyconic' moiety the flat guanidinium group, mimicking a glucopyranosyl cation. Together with the previously synthesized (1-->6)-linked disaccharide analogues 6-deoxy-6-[(4R,5S)-5-hydroxy-4-(hydroxymethyl)-1,4,5,6- tetrahydropyrimidin-2-yl]amino-alpha,beta-D-glucopyranose hydrochloride (1) and 6-deoxy-6-[(4R,5R)-5-hydroxy-4-(hydroxy-methyl)-1,4,5,6- tetrahydropyrimidin-2-yl]amino-alpha,beta-D-glucopyranose hydrochloride (2), a possible inhibitory effect on the action of alpha-D-glucosidase, beta-D-glucosidase, alpha-D-galactosidase, and beta-D-galactosidase was investigated. All compounds, except 20 with alpha-D-glucosidase where no inhibition could be detected, showed either competitive or mixed competitive inhibition with all enzymes. The effects of the disaccharide analogues were generally weaker as compared to the effect of the previously synthesized configurationally related nitrophenyl glycoside analogues (4R,5S)-5-hydroxy-4-(hydroxymethyl)-2-(p-nitrophenyl)amino-1,4,5,6- tetrahydropyrimidine hydrochloride (21) and (4R,5R)-5-hydroxy-4-(hydroxymethyl)-2-(p-nitrophenyl)amino-1,4,5,6- tetrahydropyrimidine hydrochloride (22). On the basis of experimental results, different binding hydrochloride (22). On the basis of experimental results, different binding modes of competitive inhibitors to the active site of corresponding enzymes are discussed.

Stereoselective synthesis of 2-S-phenyl-2-deoxy-beta-glycosides using phenyl 2,3-O-thionocarbonyl-1-thioglycoside donors via 1,2-Migration and concurrent glycosidation.

[figure: see text] 1,2-Migration and concurrent glycosidation of phenyl 2,3-O-thionocarbonyl-1-thlo-alpha-L-rhamnopyranosides under the action of methyl trifluoromethanesulfonate (MeOTf) afforded in high yields the 3-O-(methylthio)carbonyl-2-S-phenyl-2,6-dideoxy-beta-L-glucopyranosides, ready precursors to the corresponding 2-deoxy-beta-glycosides.

Cytotoxic activity of kaempferol glycosides against human leukaemic cell lines in vitro.

Two kaempferol coumaroyl glycosides (i.e. platanoside and tiliroside) isolated from the methanolic extract of Platanus orientalis L. buds, were examined for their in vitro cytotoxic activity against a panel of human leukaemic cell lines. Platanoside (1) exhibited cytotoxic activity against most of the cell lines tested, while tiliroside (2) was active against two of the nine tested cell lines. Compound 1, was examined for its effect on the uptake of [(3)H]thymidine as a marker of DNA synthesis. Kaempferol was used as a control.

Alkane-1,2-diol-based glycosides and fatty glycosides and wax esters in Roseiflexus castenholzii and hot spring microbial mats.

The lipid composition of Roseiflexus castenholzii, a thermophilic filamentous phototrophic bacterium related to uncultivated filamentous phototrophic bacteria that predominate in hot spring microbial mats, is reported. R. castenholzii lipid extracts were dominated by components characterized by alkane-1-ol-2-alkanoate moieties glycosidically bonded to a C(6) sugar. Similar fatty glycosides, with an additional fatty acid esterified, were detected by HPLC-MS. R. castenholzii also produces a suite of wax esters ranging from 37 to 40 carbon atoms in length. In lipid extracts from two nonsulfidic hot spring microbial mats, similar alkane-1,2-diol-based lipids were detected in minor amounts. R. castenholzii lipids are compared to lipids of mats and other thermophilic mat isolates.

Anti-solasodine glycoside single-chain Fv antibody stimulates biosynthesis of solasodine glycoside in plants.

We constructed a recombinant antibody fragment--single chain fragment-variable (scFv) antibody--derived from hybridoma cell lines to control the concentration of solasodine glycosides in hairy root cultures of Solanum khasianum transformed by the anti-solamargine (As)-scFv gene. The properties of the As-scFv protein expressed in Escherichia coli were almost identical to those of the parent monoclonal antibody (MAb). Up to 220 ng recombinant As-scFv was expressed per milligram of soluble protein in transgenic hairy root cultures of S. khasianum. The concentration of solasodine glycosides was 2.3-fold higher in the transgenic than in the wild-type hairy root, as reflected by the soluble As-scFv level and antigen binding activities. These results suggested that the scFv antibody expressed in transgenic hairy roots controlled the antigen level, thus representing a novel plant breeding methodology that can produce secondary metabolites.

Astonishing diversity of natural surfactants: 2. Polyether glycosidic ionophores and macrocyclic glycosides.

Polyether glycosidic ionophores and macrocyclic glycosides are of great interest, especially for the medicinal and pharmaceutical industries. These biologically active natural surfactants are good prospects for the future chemical preparation of compounds useful as antibiotics, anticancer agents, or in industry. More than 300 interesting and unusual natural surfactants are described in this review article, including their chemical structures and biological activities.

Synthesis of di- and tri-saccharides with intramolecular NH-glycosidic linkages: molecules with flexible and rigid glycosidic bonds for conformational studies.

Attempted dephthalimidation of the trisaccharide 1-O-acetyl-3,4-di-O-benzyl- 2,6-di-O-(3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-beta-D-glucopyranosyl )-alpha-D-mannopyranose (1) and its derivatives 2 and 3, as well as the disaccharide 1-O-acetyl-3,4,6-tri-O-benzyl-2-O-(3,4,6-tri-O-acetyl- 2-deoxy-2-phthalimido-beta-D-glucopyranosyl)-alpha-D-mannopyranose (13), with hydrazine hydrate in ethanol at 80 degrees C, produced the trisaccharide-6-O-(2-acetamido-3,4,6-tri-O-acetyl-2-deoxy-beta-D- glucopyranosyl)-3,4-di-O-benzyl-beta-D-mannopyranose-3',4',6'-tri-O-a cet yl- beta-D-glucopyranose 1,2'-N:1',2-O-dianhydride (4) and 3,4,6-tri-O-benzyl-beta-D-mannopyranose 3',4',6'-tri-O-acetyl-beta-D-glucopyranose 1,2'-N:1',2-O-dianhydride (14), respectively, containing an intramolecular NH-glycosidic linkage. The conventional deblocking of compounds 4 and 14 gave the completely deblocked trisaccharide 6-O-(2-acetamido-2-deoxy-beta-D-glucopyranosyl)-beta-D-mannopyranose beta-D-glucopyranose 1,2'-N:1',2-O-dianhydride (6) and the disaccharide beta-D-mannopyranose beta-D-glucopyranose 1,2'-N:1',2-O-dianhydride (16), respectively, containing an intact intramolecular NH-glycosidic bond. The unusual intra NH-glycosyl character makes the linkage rigid, and therefore these compounds should not only be useful for NMR studies but also as substrates or inhibitors of GlcNAc-transferases.

Analytical methods for monoterpene glycosides in grape and wine. I. XAD-2 extraction and gas chromatographic-mass spectrometric determination of synthetic glycosides.

Synthetic monoterpene and aromatic beta-D-glucopyranosides, beta-rutinosides and 6-O-(alpha-L-arabinofuranosyl)-beta-D-glucopyranosides and their corresponding alcohols, diluted in a synthetic solution imitating wine, were isolated and separated by selective retention on Amberlite XAD-2. The corresponding recoveries and the conditions for the direct determination of these glycosides by gas chromatography and gas chromatography-mass spectrometry after derivatization were determined. Trifluoroacetylation gave the best results but trimethylsilylation provided complementary results. The separation of some diastereoisomeric monoterpene glycosides was also examined.

Borate complexation of flavonoid-O-glycosides in capillary electrophoresis. II. Separation of flavonoid-3-O-glycosides differing in their sugar moiety.

Capillary electrophoresis was found to give significantly higher efficiency, selectivity and speed than high-performance liquid chromatography for the separation of a mixture of flavonoid-3-O-glycosides differing in their sugar moiety. Boric acid running buffer (0.2 M, pH 10.5) was used for this electrophoretic separation. The migration order in free solution capillary electrophoresis (CE) and the selectivity of these flavonoid-3-O-glycosides can be mainly explained by in situ borate complexation of both the sugar moiety and the cis-1,2-hydroxyl groups on the flavonoid skeleton and, to a lesser extent, by the ionization of hydroxyl groups on the flavonoid skeleton due to alkaline pH conditions. The correlation of the electrophoretic mobilities with the configuration and conformation of the compounds is discussed.

Phenolic glycosides and ionone glycoside from the stem of Sargentodoxa cuneata.

Four phenolic glycosides, cuneatasides A-D (1-4), and one ionone glycoside cuneataside E (5), together with seven known phenolic compounds (6-12) were isolated from the water-soluble constituents of the stem of Sargentodoxa cuneata (Sargentodoxaceae). Their structures were elucidated by spectroscopic analysis. In vitro tests for antimicrobial activity showed compounds 1 and 2 to possess significant activity against two Gram-positive organisms, Staphylococcus aureus and Micrococcus epidermidis.

Studies on antiviral glycosides. Synthesis and biological evaluation of various phenyl glycosides.

A variety of analogues and derivatives of phenyl glycosides were synthesized for examination of their biological activities and of the relationship between structure and antiviral activity. For antiviral activity, a 6-deoxy-6-halogeno-D-glucose residue was most suitable for the carbohydrate moiety and p-alkylphenyl groups for the aglycone moiety. Based on these results, p-(sec-butyl)phenyl 6-chloro-6-deoxy-beta-D-glucopyranoside and p-(sec-butyl)phenyl 6-deoxy-6-iodo-beta-D-glucopyranoside were prepared, and the former compound was found to be the most potent antiviral substance, in this series, against influenza and Herpes simplex virus. The anomeric configuration of phenyl glycosides did not contribute to the antiviral activity.

Phenethyl alcohol glycosides and isopentenol glycoside from fruit of Bupleurum falcatum.

Investigation on the constituents of the fruit of Bupleurum falcatum L. resulted in the isolation of the three new glycosides, phenethyl alcohol 8-O-beta-D-glucopyranosyl-(1-->2)-O-beta-D-apiofuranosyl-(1-->6)-b eta-D- glucopyranoside, phenethyl alcohol 8-O-beta-D-glucopyranosyl-(1-->2)-beta-D-glucopyranoside and isopentenol 1-O-beta-D-apiofuranosyl-(1-->6)-beta-D-glucopyranoside along with five known glycosides, icariside D1, icariside F2, saikosaponin a, saikosaponin c and saikosaponin d. The structures of these compounds were elucidated on the basis of interpretation of chemical and spectral data.
