Phagocytic rabbit cell lines expressing class II MHC products: establishment of cell lines by viral transformation.

Continuous rabbit macrophage-like cell lines were established after in vitro infection of spleen cells with either Simian virus 40, lymphotropic papovavirus or herpesvirus sylvilagus. These cell lines are characterized as morphologically similar to mature macrophages, esterase positive, and have unrearranged immunoglobulin genes. They possess macrophage functionality because they are highly phagocytic and are able to mediate an antibody dependent cell mediated cytotoxicity assay on chicken red blood cells. Northern blot analysis of total cellular RNA with a DQ alpha probe indicates that the cell lines constitutively express message for class II gene products. In addition, cell sorter analysis indicates that two of these lines display class II antigen at the cell surface.

Quantitative assessment of the use of continuous wavelet transform in the analysis of the fundamental frequency disturbance of the synthetic voice.

The aim of this work is to investigate quantitatively the capability of the Continuous Wavelet Transform (CWT) as a tool to estimate (calculate) Jitter and Shimmer, assessing the error between these indices calculated in each Wavelet decomposition and the ones for the original signal, for several dilatation levels. Two synthetic vowels /a/ were generated with the fundamental frequencies of 120 Hz for male and 220 Hz for female, by an autoregressive 22 coefficient all-pole model, and Jitter and Shimmer were introduced to the signal using five different percentage variations. The signals were decomposed by CWT in eight levels of dilatation (1, 2, 4, 8, 16, 32, 64 and 128), using the Mexican Hat, Meyer and Morlet real bases. Jitter and Shimmer were calculated for the original signals and all eight levels of decompositions and then the errors between the indices in the decompositions and the original signals were calculated. It can be concluded that CWT can be used as a tool for pre-processing the signal to measure Shimmer preferentially, and Jitter, instead of using the original signal to do that. The Mexican Hat base provided the lowest errors for Shimmer analysis, where the best dilatation level was 8 (error below 0.1%). In addition, the errors associated with Shimmer index, in general, are lower than the ones associated with Jitter index.

BCI Competition 2003--Data sets Ib and IIb: feature extraction from event-related brain potentials with the continuous wavelet transform and the t-value scalogram.

The t-CWT, a novel method for feature extraction from biological signals, is introduced. It is based on the continuous wavelet transform (CWT) and Student's t-statistic. Applied to event-related brain potential (ERP) data in brain-computer interface (BCI) paradigms, the method provides fully automated detection and quantification of the ERP components that best discriminate between two samples of EEG signals and are, therefore, particularly suitable for classification of single-trial ERPs. A simple and fast CWT computation algorithm is proposed for the transformation of large data sets and single trials. The method was validated in the BCI Competition 2003, where it was a winner (provided best classification) on two data sets acquired in two different BCI paradigms, P300 speller and slow cortical potential (SCP) self-regulation. These results are presented here.

Fringe-density estimation by continuous wavelet transform.

For many phase extraction algorithms, a priori knowledge of a fringe-pattern density distribution is beneficial for later processing. A fringe-density estimation method based on a continuous wavelet transform (CWT) is proposed. For a one-dimensional signal the instantaneous frequency detected at the CWT ridge is directly adopted as a measure of the local fringe density. For a two-dimensional signal the instantaneous frequency components in both the x and the y directions are detected. Their reliability is evaluated by the CWT coefficient magnitude, based on which an approximate density value is given. The capability for noise reduction and the accuracy of the method are discussed.

Continuous wavelet transformation applied to the simultaneous quantitative analysis of two-component mixtures.

In this paper we developed a graphical method based on Haar (HA) and Mexican (MEX) one-dimensional continuous wavelet transforms and we applied it to a mixture of hydrochlorothiazide (HCT) and spironolactone (SP) in the presence of strongly overlapping signals. Keeping in mind to obtain an appropriately transformed spectrum, we tested several values of the scaling parameter a and the point number of the analysed spectrum in the concentration range of 2-22 microg/ml for both active compounds. The optimal values of the scale parameters and the corresponding frequencies were found to be a = 32 and 0.031 for HA and a = 30 and 0.008 for MEX corresponding to 400 points. HA and MEX methods based on a zero crossing technique were applied to the analysed signal and their regression lines at the selected points were obtained. The validation of the above methods was carried out by analysing different synthetic mixtures containing HCT and SP. MATLAB 6.5 software was used for one-dimensional wavelet analysis and the basic concepts about wavelet method were briefly explained. The method developed in this paper is rapid, easy to apply, inexpensive and is suitable for analysing the overlapping signals of compounds in their mixtures without any chemical pre-treatment.

Tissue characterization using the continuous wavelet transform. Part I: Decomposition method.

In this paper, a novel decomposition of the RF ultrasound signal into its coherent and diffused components is proposed. This decomposition is based on thresholding the energy of the continuous wavelet transform of the RF signal using appropriate wavelets. The two components are modeled separately, and the model parameters are estimated. Previous work [1] required assumptions about the periodicity of the coherent scatterers in the tissue. These assumptions are not necessary in this work. The decomposition algorithm is tested on simulated RF images. The accuracy of the estimated parameters is presented as well as the performance of the algorithm in low coherent-to-diffuse components' energy ratios (SNR).

Fractional wavelet analysis for the simultaneous quantitative analysis of lacidipine and its photodegradation product by continuous wavelet transform and multilinear regression calibration.

Fractional wavelet transform (FWT) was applied to the original absorption spectra of lacidipine (LAC) and its photodegradation product (LACD), and the resulting FWT spectra were processed by continuous wavelet transform (CWT) and multilinear regression calibration (MLRC) for the simultaneous quantitative analysis of both products in their binary mixtures. These methods do not require any chemical separation step and chemical complex reaction to obtain a detectable signal for the degradation product. By using the Mexican hat function, 2 calibration functions for LAC and LACD were obtained by measuring the CWT transformed signals at 416.1 nm for LAC and 414.6 nm for LACD, after FWT processing of the original absorption spectra. The calibration graphs were linear in the concentration range of 5.08-40.64 microg/mL for LAC and 0.51-8.16 microg/mL for LACD. The limit of detection and the limit of quantitation were found to be 0.289 and 0.956 microg/mL for LAC and 0.036 and 0.118 microg/mL for LACD, respectively. For comparison, the MLRC algorithm was applied to the linear regression functions for the individual drug and its photoproduct. In this approach, a set of linear regression functions was obtained from the relationship between concentrations and FWT signals in the wavelength range 411.0-412.4 nm. Both methods were applied to the quantitative evaluation of LAC and LACD in laboratory and pharmaceutical samples, and produced very satisfactory results.

Liver morphology in guinea pigs fed pyrolysis products of a polychlorinated biphenyl transformer fluid continuously for 90 days.

After a fire involving a transformer, the State Office Building in Binghamton, New York, was contaminated with soot containing polychlorinated biphenyls, biphenylenes, naphthalenes, dioxins, and dibenzofurans. The toxicity of the soot and its effect on liver morphology after prolonged (subchronic) exposure were determined for both sexes of Hartley guinea pigs, which were fed soot continuously for 90 days. By light microscopy the observed alterations of the liver were predominantly centrilobular; they included hepatocyte hypertrophy, steatosis, increased glycogen and iron, focal necrosis, and bile duct proliferation with fibrosis. Cytoplasmic vacuoles and acidophilic hyalin-like bodies were observed. Electron microscopy of hepatocytes showed proliferated smooth endoplasmic reticulum (SER), cytoplasmic vacuoles, concentric membrane arrays (CMAs), glycogen bodies, and microdroplets of fat, often without limiting membranes. The vacuoles frequently contained membrane fragments and had a halo-like periphery composed of proliferated membranes. Cell debris, membrane fragments, and small CMAs were observed in the sinusoids. Membrane fragments were also observed in the bile canaliculi and bile ducts. Intoxicated bile duct cells contained more cytoplasmic myelin whorls and altered mitochondria. In contrast to the previously reported study of a single dose, these liver alterations showed a strong dose dependence, emphasizing the importance of time and method of administration. The cytoplasmic vacuoles, which were not pronounced in the previous study, are here a prominent alteration, probably originating from outpouchings of canaliculi and sinusoidal membranes. A hypothesis for the mechanism of hepatocyte detoxification based on the proliferated SER and ejection of membrane fragments is proposed.

Comparative study of the continuous wavelet transform, derivative and partial least squares methods applied to the overlapping spectra for the simultaneous quantitative resolution of ascorbic acid and acetylsalicylic acid in effervescent tablets.

The simultaneous spectrophotometric determination of ascorbic acid (AA) and acetylsalicylic acid (ASA) in effervescent tablets in the presence of the overlapping spectra was accomplished by the continuous wavelet transform (CWT), derivative spectrophotometry (DS) and partial least squares (PLS) approaches without using any chemical pre-treatment. CWT and DS calibration equations for AA and ASA were obtained by measuring the CWT and DS amplitudes corresponding to zero-crossing points of spectra obtained by plotting continuous wavelet coefficients and first-derivative absorbance values versus the wavelengths, respectively. The PLS calibration was constructed by using the concentration set and its full absorbance data consisting of 850 points from 220 to 305 nm in the range of 210-310 nm. These three methods were tested by analyzing the synthetic mixtures of the above drugs and they were applied to the real samples containing two commercial pharmaceutical preparations of subjected drugs. A comparative study was carried out by using the experimental results obtained from three analytical methodologies and precise and accurate results were obtained.

Lactate editing and lipid suppression by continuous wavelet transform analysis: application to simulated and (1)H MRS brain tumor time-domain data.

Determination of lactate concentrations in vivo is required in the noninvasive diagnosis, staging, and therapeutic monitoring of diseases such as cancer, heart disease, and stroke. An iterative filtering process based on the continuous wavelet transform (CWT) method in the time domain is proposed to isolate the lactate doublet signal from overlapping lipid resonances and estimate the magnetic resonance spectroscopy (MRS) parameters of the lactate methyl signal (signal amplitude, chemical shift, J-coupling and apparent transverse relaxation time (T*(2))). This method offers a number of advantages over the multiple quantum (MQ) and difference spectroscopy approaches, including: 1) full recovery of the lactate methyl signal, whereas the MQ methods usually detect 50% of the signal intensity; 2) in contrast to MQ methods, the lipid signal is retained together with J-coupling data on the lactate peak; 3) the CWT method is much less sensitive to motion artifacts than difference spectroscopy. Application of the method to simulated and real (1)H MRS data collected from human blood plasma and brain tumors demonstrated that this filter provides accurate estimates of the MRS parameters of the lactate doublet and efficiently removes lipid contributions.

Bileaflet mechanical valve sound analysis using a continuous wavelet transform.

Bileaflet mechanical valve closing sounds have splits, the duration of which is not constant in normally functioning valves. However, no reports have discussed the influences of valve malfunction on the split interval, neither have any studies discussed the fact that mechanical valve closing sound signals must be analyzed using a time-frequency analysis because they are nonstationary signals. The continuous wavelet transform (CWT), a time-frequency analyzing method using mother wavelets modified by scale numbers, was selected in this study for analyzing bileaflet valve closing sounds because it is easy to understand and has no limitations such as the cross-terms in the Wigner-Ville distribution or the tradeoff between time and frequency resolutions of the short-time Fourier transform. This study compares the properties of the mother wavelets of various CWTs and selects one that is suitable for detection of the clear split in bileaflet mechanical valve closing sound signals. This article also establishes a standard frequency analyzing system for bileaflet mechanical valve sounds. A preliminary study with chirp Doppler signals for comparing the frequency properties of the mother wavelets of various CWTs suggested that Ishikawa's modified Morlet CWT has better time and frequency resolution at the highest frequency scale. Morlet/power CWT analysis of normal in vivo bileaflet valve closing sounds of the ST. Jude Medical (SJM), ATS, and Carbomedics (CM) valves demonstrated clear splits of very short interval at the highest level of frequency. Detection of the disappearance of the split by using this analytical method may be the key to identifying bileaflet mechanical valve malfunction in outpatient departments.

[Electrocardiographic signal feature extraction and its instrument development based on continuous wavelet transform].

This paper introduces a virtual recording and analysis instrumentation system for recording and identifying electrocardiographic (ECG) signals. The system developed is aimed at constructing a PC-based virtual instrumentation which enables to record. investigate and measure the ECG signal of 12 leads simultaneously and perfectly in order to improve the measuring precision of ECG. Based on the proper feature in time domain of Mexican hat wavelet expressed by positioning and analysis precision for QRS complex, the instrumentation system uses continuous wavelet transform(CWT) and uses the Mexican hat as the wavelet base to measure precisely the characteristic information and generate the precise characteristic parameters of ECG. The analysis of measured ECG signals in hospital demonstrated that even in the condition with serious noise interference, the method presented is still easily to describe the characteristics of ECG on line precisely which makes the instrumentation system valuable in practical application.

Surface contouring by optical edge projection based on a continuous wavelet transform.

A novel optical edge projection method for surface contouring of an object with low reflectivity is presented. A structured light edge is projected onto a dark surface, and the image is captured by a CCD camera. The surface profile of the object is then evaluated by an active triangular projection technique, and a whole-field three-dimensional contour of the object is obtained by scanning the optical edge over the entire object surface. An edge detection method based on a continuous wavelet transform (CWT) is employed to determine the location of the optical edge. The method of optical edge detection is described, and characteristic details of gray-level distribution along the edge are analyzed. It is shown that the proposed wavelet edge detection method is not dependent on any threshold values; hence the true edge position can be determined without subjective selection. A black low-reflectivity object surface made from woven carbon fiber is measured, and the experimental results show that the profile of a woven carbon fiber can be obtained by the proposed method.

Induction of transformation and continuous cell lines from normal human mammary epithelial cells after exposure to benzo[a]pyrene.

Rapidly growing primary cultures of normal human mammary epithelial cells (HMEC) were exposed to 1 microgram of benzo[a]pyrene (B[a]P) per ml for two or three 24-hr periods. The B[a]P-treated populations consistently contained cells displaying a longer period of active growth in culture compared to the untreated control cells. Widespread heterogeneity in morphology and growth patterns was evidenced in these "extended life" (EL) cultures, with multiple sequential changes in these parameters occurring during the course of their life in culture. Two apparently immortal continuous cell lines have thus far emerged from these EL cultures. These lines have been characterized to be of human mammary epithelial origin and derived from the originally treated HMEC specimen. The continuous lines do not appear to be malignantly transformed as they do not cause tumor formation in nude mice and show little or no anchorage-independent growth. Nonetheless, they have acquired several properties characteristic of tumor-derived HMEC, which distinguish them from their normal progenitors. These cell lines, as well as the EL strains, may provide useful substrates for studies to determine what agents can induce further transforming events. Additionally, analysis of the multiple steps occurring in the El cultures, as well as in the emergence of the continuous cell lines, could potentially elucidate the processes occurring during human epithelial cell carcinogenesis and escape from senescence.

The t-CWT: a new ERP detection and quantification method based on the continuous wavelet transform and Student's t-statistics.

OBJECTIVE: This study was aimed at developing a method for extraction and assessment of event-related brain potentials (ERP) from single-trials. This method should be applicable in the assessment of single persons' ERPs and should be able to handle both single ERP components and whole waveforms. METHODS: We adopted a recently developed ERP feature extraction method, the t-CWT, for the purposes of hypothesis testing in the statistical assessment of ERPs. The t-CWT is based on the continuous wavelet transform (CWT) and Student's t-statistics. The method was tested in two ERP paradigms, oddball and semantic priming, by assessing individual-participant data on a single-trial basis, and testing the significance of selected ERP components, P300 and N400, as well as of whole ERP waveforms. The t-CWT was also compared to other univariate and multivariate ERP assessment methods: peak picking, area computation, discrete wavelet transform (DWT) and principal component analysis (PCA). RESULTS: The t-CWT produced better results than all of the other assessment methods it was compared with. CONCLUSIONS: The t-CWT can be used as a reliable and powerful method for ERP-component detection and testing of statistical hypotheses concerning both single ERP components and whole waveforms extracted from either single persons' or group data. SIGNIFICANCE: The t-CWT is the first such method based explicitly on the criteria of maximal statistical difference between two average ERPs in the time-frequency domain and is particularly suitable for ERP assessment of individual data (e.g. in clinical settings), but also for the investigation of small and/or novel ERP effects from group data.

[RAEB in transformation in continuing complete remission by small doses of cytarabin].

Two patients with refractory anemia with excess of blasts in transformation having entered remission and in continuing complete remission with the small-dose cytarabine therapy (SDCA) are reported. SDCA was the only chemotherapy used in these two patients. Cytarabine was injected subcutaneously every 12 hours at doses 15 mg and 10 mg, respectively, in cases 1 and 2. Case 1, a 60-year-old woman, achieved remission after a 14-day course of SDCA. Thereafter, she received SDCA for seven consecutive days every month or every other months in the first year and every two or three months in subsequent years. Her remission has been maintained for 57 months. Case 2, a 76-year-old man, attained a remission after three courses of SDCA, each consisting of 16, 11 and 14 days, respectively. SDCA was then repeated every month for 6 consecutive days. He is still in remission after 33 months. Whereas SDCA caused severe bone marrow suppression during induction, it was well tolerated when given in remission without necessitating any supportive measure.

A polycatenar mesogen with a perfluorinated moiety showing continuous phase transformation between a smectic A phase and a structured, fluid, optically isotropic phase.

A polycatenar mesogen consisting of a four aromatic ring system with three long alkoxy chains and a bulky perfluorinated substituent has been synthesized and found to show the polymorphism of a cubic phase, a columnar phase and a smectic A phase, including a structured, fluid, optically isotropic phase with the phase transformation between the smectic A phase and the isotropic phase occurring continuously.

Tactile stimulus predictability modulates activity in a tactile-motor cortical network.

Manipulating objects in the hand requires the continuous transformation of sensory input into appropriate motor behaviour. Using a novel vibrotactile device combined with fMRI, the cortical network associated with tactile sensorimotor transformations was investigated. Continuous tactile stimuli were delivered in a random or predictable pattern to the second digit on the right hand of all subjects. To better distinguish sensory and motor processes, subjects were instructed to make proportionate motor gripping responses with their left hand. A consistent cortical network of activation was revealed that included the supplementary motor, dorsal and ventral premotor, posterior parietal, primary and secondary somatosensory and primary motor cortex. Tracking the unpredictable versus predictable tactile stimulus led to greater delays in motor responses and to increased performance errors. Cortical effects due to stimulus predictability were observed in several components of the network, though it was most evident as increased cortical activation in frontal motor regions during tracking of unpredictable tactile stimuli. In contrast to the proposed hypotheses, primary and secondary somatosensory cortices contralateral to tactile input did not reveal enhanced responses during unpredictable tracking. Facilitation during unpredictable tracking was also observed in primary somatosensory cortex contralateral to motor responses, the receptive site for movement-related afference. The present study provides a novel and controlled approach to investigate the loci associated with tactile-motor processing and to measure the task-specific effect of stimulus predictability on network components.
