Perturbation of conjugation in internally solvated allylic lithium compounds: variation of ligand structure. NMR and X-ray crystallography.

Several allyic lithium compounds were prepared with different potential ligands tethered at C2. These are with CH3OCH2CH2NCH3CH2-, 5 and 1-TMS 6, with (CH3)2NCH2CH2NCH3CH2-, 1-TMS 7, and with ((CH3)2NCH2CH2)2NCH2-, 8 and 1-TMS 9. In all these compounds Li is fully coordinated to the pendant ligand and is sited off the axis perpendicular to the allyl plane at one of the allyl termini as indicated by a combination of X-ray crystallography and NMR spectra. Compounds 5 and 8 are Li-bridged dimers as shown by X-ray crystallography and also dimeric in benzene solution as determined from freezing point determinations. Compounds 6, 7, and 9 are monomeric in THF-d8 or diethyl ether-d10 solution and exhibit one bond 13C1, 6Li scalar coupling at low temperature. Taken together the crystallographic and NMR data indicate that all of these compounds incorporate partially delocalized allylic moieties. Compounds 5 and 8 undergo fast 1,3-Li-sigmatropic shifts that are proposed to take place within low concentrations of monomers in fast equilibrium with prevalent dimers. Averaging with increasing temperature of the one-bond 13C, 6Li coupling constant in 6, 7, and 13 provided the dynamics of bimolecular C-Li exchange with Delta H++ values of 6.7, 12, and 13 kcal x mol(-1), respectively. Averaging of the diastereotopic N(CH3)2 13C resonances of 7 is indicative of fast transfer of coordinated ligand between faces of the allyl plane Delta H++ = 5.3 kcal x mol(-1) combined with slower inversion at nitrogen. Compound 8 exhibits similar effects. It is concluded that variation of the ligand structure changes dynamic behavior of the compounds but has little influence of their degrees of delocalization.

One-Pot synthesis of gamma,delta-unsaturated carbonyl compounds from allyl alcohols and vinyl or isopropenyl acetates catalyzed by [IrCl(cod)]2.

One-pot synthesis of gamma,delta-unsaturated carbonyl compounds from allyl alcohols and vinyl or isopropenyl acetates was achieved through in situ generation of allyl vinyl ethers by the action of the [IrCl(cod)]2 complex followed by Claisen rearrangement of the resulting ethers. For instance, the reaction of trans-2-methyl-3-phenyl-2-propen-1-ol with isopropenyl acetate in the presence of [IrCl(cod)]2 (1 mol %) and Cs2CO3 (5 mol %) at 100 degrees C for 3 h followed by 140 degrees C for 15 h afforded 5-methyl-4-phenyl-5-hexen-2-one in 72% yield. When vinyl acetate was employed in place of isopropenyl acetate, 4-methyl-3-phenyl-4-pentenal was obtained in 83% yield.

Are the 11-oxo-steroids really so hindered towards organometallic compounds?

Allylation of an 11-oxo-steroid, the protected adrenosterone, was studied to examine more closely the steric hindrance of such a ketosteroid. While the beta face exhibits, as well known, a strong steric hindrance, the alpha side appears to be only relatively hindered. A high diastereoselectivity was observed in the addition of crotyl magnesium bromide.

Enhancement of the fungicidal activity of amphotericin B by allicin, an allyl-sulfur compound from garlic, against the yeast Saccharomyces cerevisiae as a model system.

Amphotericin B (AmB) is a representative antibiotic for the control of serious fungal infections, and its fungicidal activity was greatly enhanced by allicin, an allyl-sulfur compound from garlic. In addition to the plasma membrane permeability change, AmB induced vacuole membrane damage so that the organelles were visible as small discrete particles. Although allicin was ineffective in promoting AmB-induced plasma membrane disability, this compound enhanced AmB-induced structural damage to the vacuolar membrane even at a non-lethal dose of the antibiotic. Allicin could also enhance the antifungal activity of AmB against the pathogenic fungus Candida albicans and against Aspergillus fumigatus. In contrast, allicin did not enhance the cytotoxic activity of AmB against cells of human promyelocytic leukemia (HL-60), a vacuole-less organism.

Synergistic fungicidal activity of Cu(2+) and allicin, an allyl sulfur compound from garlic, and its relation to the role of alkyl hydroperoxide reductase 1 as a cell surface defense in Saccharomyces cerevisiae.

Cu(2+) showed a dose-dependent fungicidal activity against Saccharomyces cerevisiae cells, and its lethal effect was extremely enhanced in the presence of allicin, an allyl sulfur compound from garlic. The fungicidal activity of Cu(2+) was unaffected or rather attenuated by other sulfur-containing compounds such as N-acetyl-cysteine, l-cysteine or dithiothreitol. Ca(2+) could absolutely protect against the lethal effect of Cu(2+) itself, but showed no protection against the fungicidal activity of Cu(2+) newly generated in combination with allicin. Cu(2+) accelerated an endogenous generation of reactive oxygen species (ROS) in S. cerevisiae cells at a lethal concentration, but such intracellular oxidative stress induction was not observed during cell death progression upon treatment with Cu(2+) and allicin. A surfactant, sodium N-lauroyl sarcosinate (SLS), enhanced the solubilization of a few proteins including alkyl hydroperoxide reductase 1 (AHP1) in intact cells, accounting for the absence of this protein in the extract from allicin-treated cells. Allicin-treated cells were rendered extremely sensitive to the subsequent Cu(2+) treatment as in the case of SLS-treated cells. Allicin-treated cells and SLS-treated cells similarly showed an increased sensitivity to exogenously added tert-butyl hydroperoxide (t-BOOH), an organic peroxide that is detoxified by the action of AHP1. Our study suggests that allicin influences the mode of cell surface localization or the related function of AHP1 as a defense against phospholipid peroxidation by the external action of Cu(2+).

Mechanisms by which garlic and allyl sulfur compounds suppress carcinogen bioactivation. Garlic and carcinogenesis.

Overall, a host of studies provides compelling evidence that garlic and its organic allyl sulfur components are effective inhibitors of the cancer process. These studies reveal that the benefits of garlic are not limited to a specific species, to a particular tissue, or to a specific carcinogen. Several mechanisms are likely to account for this protection. Notable among these is a depression in nitrosamine formation and a reduction in carcinogen bioactivation. The benefits provided by garlic must be viewed as part of the entire diet, since several dietary constituents can influence the degree of protection. More than one compound is responsible for the anticancer properties associated with garlic. Future research should focus on how genetic variability and daily environmental factors influence the anticancer benefits attributed to garlic and its allyl sulfur components.

Formation of silazirconacyclopentenes from zirconium-silene complex and alkynes and their reactivities.

During the course of our study on the formation of a complex having a zirconium-silicon bond, we found that zirconium-silene complex 7 was formed from Cp2ZrCl2 and Me2PhSiLi. In the presence of alkyne, diarylalkyne reacted with silene coordinated with zirconium to give silazirconacyclopentene 8. On the other hand, dialkylalkyne inserted into a zirconium-silene complex gave silazirconacyclopentene 9. Hydrolysis of 8 or 9 afforded vinylsilane 13 or allylsilane 16. Transmetalation of zirconacycle 8 into copper in the presence of allyl halide gave a bis-allylated compound in high yield, indicating that alkylation occurred on the alkyne carbon and the methyl group of silicon. From bis-allylated compounds, eight-membered ring compounds having silicon were obtained in high yield using olefin metathesis.

Palladium(0)-catalyzed enantioselective O,S-rearrangement of racemic O-allylic thiocarbamates: a new entry to enantioenriched allylic sulfur compounds.

Reaction of (+/-)-(E)-pent-3-en-2-ol, (+/-)-(E)-hept-4-en-3-ol, (+/-)-(E)-2,6-dimethylhept-4-en-3-ol, (+/-)-cyclohex-2-en-1-ol, and (+/-)-cyclohept-2-en-1-ol with methyl, ethyl, n-propyl, isopropyl, n-butyl, tert-butyl, and benzyl isothiocyanate gave the corresponding racemic O-allylic thiocarbamates of medium to good thermal stability in good yields. The palladium(0)-catalyzed rearrangement of the (+/-)-(E)-pent-3-en-2-ol-, (+/-)-(E)-hept-4-en-3-ol-, (+/-)-cyclohex-2-en-1-ol-, and (+/-)-cyclohept-2-en-1-ol-derived O-allylic thiocarbamates at room temperature in methylene chloride by using Pd2(dba)3*CHCl3 (dba = dibenzylideneacetone) as precatalyst and (+)-(1R,2R)-1,2-bis-N-((2-(diphenylphosphino)benzoyl)-1,2-diaminocyclohexane as ligand for the palladium atom proceeded quantitatively and gave the corresponding acyclic (R)-configured S-allylic thiocarbamates and the cyclic (S)-configured S-allylic thiocarbamates with ee values ranging from 85% to > or = 99% in yields of 76-94%. Rearrangement of the O-allylic thiocarbamates carrying a methyl group at the N atom not only was the fastest but also proceeded with the highest enantioselectivity. No rearrangement was observed under these conditions in the case of the racemic N-methyl O-allylic thiocarbamate derived from (+/-)-2,6-dimethylhept-4-en-3-ol, which has a branched carbon skeleton. (S)-cyclohex-2-enethiol of 97% ee was obtained through hydrolysis of the corresponding N-methyl S-allylic thiocarbamate. 2-((R)-(E)-1-methylbut-2-enylsulfanyl)pyrimidine of 91% ee and 2-((S)-cyclohex-2-enylsulfanyl)pyrimidine of 97% ee were synthesized in one synthetic operation from the corresponding N-methyl S-allylic thiocarbamates and 2-chloropyrimidine. Similarly, (S)-cyclohex-2-enylsulfanyl)benzene of 97% ee was obtained in one synthetic operation from the corresponding N-methyl S-allylic thiocarbamate through a palladium(0)-catalyzed substitution of iodobenzene in the presence of a base. The palladium(0)-catalyzed enantioselective rearrangement of O-allylic carbamates to S-allylic carbamates has been extended from the solution phase to the solid phase by using a methyl thioisocyanate polystyrene resin. In the case investigated the enantioselectivity of the rearrangement on the solid phase was considerably lower than that in solution.

NMR studies of structure and reorganization dynamics of an [6Li]-allylic lithium compound complexed to [14N,15N]-N,N,N',N'-tetramethylethylenediamine: inversion and ligand lithium exchange.

Proton, 13C, 6Li, and 15N NMR line-shape studies of exo,exo-1-trimethylsilyl-3-(dimethylethylsilyl)allyllithium-6Li complexed to [14N,15N]-N,N,N',N'-tetramethylethylenediamine (TMEDA) 2 as a function of temperature and of added diamine reveal the dynamics of three fast equilibrium reorganization processes. These are (with DeltaH values in kilocalories per mole and DeltaS values in entropic units): mutual exchange of lithium between two 2 molecules (6.3, -21), exchange of TMEDA between its free and complexed states (5.0 and -22), and first-order transfer of complexed ligand between the allyl faces (7.0 and -20). Intermediates that are dimeric in TMEDA are proposed for the first two of these reorganization processes.

Murine strain variation in the natural killer cell and proliferative responses to the immunostimulatory compound 7-allyl-8-oxoguanosine: role of cytokines.

7-Allyl-8-oxoguanosine (loxoribine) is a di-substituted guanine ribonucleoside which has been shown previously to enhance murine NK activity, B lymphocyte proliferation, and antibody synthesis. In this study we examined the relationship among enhancement of NK activity, proliferation, and cytokine synthesis in the responses of different strains of mice to loxoribine to provide insight into the role of cytokines in these biological activities. The NK response of mice was enhanced both in vitro and in vivo in all strains tested with the exception of the NK-deficient beige (BgBg) mouse. However, there was a marked difference in the degree of NK enhancement noted in other inbred strains, with C3H and CBA mice producing the highest responses, C57BL/6, BALB/c, and DBA/2 strains giving intermediate responses, and SJL mice manifesting low responses. Striking enhancement of NK cell activity was seen in SCID mice. A somewhat different effect was seen in humans. Loxoribine treatment enhanced both the NK and LAK activity of cells from individuals with low and high spontaneous NK activity. The degree of enhancement was similar for both groups, and thus the general hierarchy of NK activity among different donors was maintained. There was less interstrain variation in the murine proliferative response to loxoribine although nude (NuNu) mice showed the highest activity and SJL mice produced substantially lower responses than other strains. All strains produced IL-6, TNF alpha, IFN-alpha/beta, and IFN-gamma when spleen cells were cultured for 48 hr with loxoribine. Interstrain variability of cytokine synthesis displayed no consistent pattern from one cytokine to another, and all failed to correlate with interstrain variability of NK cell activity or B lymphocyte proliferation. When anti-cytokine antibodies were tested for the ability to block the immunostimulatory effects of loxoribine, only anti-IFN-alpha/beta and, to a lesser degree, anti-IFN-beta, partially inhibited NK activation. Similarly, only anti-IFN alpha/beta antibodies partially blocked the proliferative response to loxoribine. In both cases, reconstitution of the responses was achieved by adding back IFN-alpha/beta to cultures containing antibodies against IFN-alpha/beta. These data suggest that, although several cytokines are produced in response to loxoribine, only IFN-alpha and IFN-beta are directly involved in the NK activation and proliferative responses. The pattern of strain variation appears to be reflective of variation in NK cell responsiveness to IFN-alpha/beta.

The immunostimulatory compound 7-allyl-8-oxoguanosine (loxoribine) induces a distinct subset of murine cytokines.

C8- and N7, C8-substituted guanine ribonucleosides comprise a class of molecules with potent immunostimulatory activity for a variety of humoral and cellular immune responses. Although it has been suggested that the immunostimulatory activity may be partially mediated by cytokine production, to date there has been no systematic evaluation of the spectrum of cytokines elicited by these nucleosides. In this study, we examine the cytokines produced by murine spleen cells in response to the di-substituted guanosine analog loxoribine (7-allyl-8-oxoguanosine). First, the levels of cytokine mRNA in spleens from vehicle- or loxoribine-treated mice were compared using PCR analysis with a panel of cytokine-specific primers. Enhancement of IL-1 alpha, TNF-alpha, TNF-beta, IL-6, IFN-alpha, and IFN-gamma mRNA was seen in the spleens of loxoribine-treated mice. IL-12 mRNA responses were more complex, with an increase in the p40 chain and a decrease in the p35 chain. In contrast, no increase was seen for mRNA levels of IL-2, IL-3, IL-4, IL-5, IL-7, or GM-CSF. ELISA assays on the supernatants of loxoribine-treated spleen cells demonstrated that IL-1 alpha, IL-6, TNF-alpha, and IFN-gamma were all produced in a dose-dependent fashion with TNF-alpha produced first, followed by IL-6 and IFN-gamma, and last by IL-1 alpha. IFN-alpha beta activity rose as quickly as TNF-alpha, leveling off at 8 to 12 hr, and was supplanted by a later-occurring surge of IFN-gamma production. IL-1 alpha, IL-6, TNF-alpha, and IFN-gamma were also detected in the sera of mice injected with loxoribine. When antibodies against the relevant cytokines were tested, only anti-IFN-alpha beta inhibited NK activity or lymphocyte proliferation and, in both cases, activity was partially restored by the addition of exogenous IFN-alpha beta. Taken together, these data indicate that loxoribine induces the production of a selective cohort of cytokines all of which have been shown to have immunostimulatory activity. However, only IFN-alpha beta appears to play a role in the enhancement of NK activity and lymphocyte proliferation.

Dependence of synergistic fungicidal activity of Cu2+ and allicin, an allyl sulfur compound from garlic, on selective accumulation of the ion in the plasma membrane fraction via allicin-mediated phospholipid peroxidation.

Allicin was effective in decreasing the lethal concentration of Cu (2+) against various fungal strains including a plant pathogen, Fusarium oxysporum, so that the minimum fungicidal concentration (MFC) of the ion for the fungus could be reduced to 2 % of that detected without allicin. In Saccharomyces cerevisiae, Cu (2+) was not apparently taken up by cells when added alone at a non-lethal concentration, whereas the ion was efficiently incorporated into cells in the presence of allicin, as in the case of cells treated with the ion at a lethal concentration. Although allicin likely increased cellular permeability to Cu (2+) due to its promotive effect on plasma membrane phospholipid peroxidation, these cell-surface events did not result in endogenous reactive oxygen species (ROS) production, a typical toxic effect of the ion. Cu (2+) was detected in the cytoplasmic fraction of cells that had been treated with the ion at a lethal concentration, whereas the ion was entrapped in the plasma membrane fraction upon their treatment with the ion at a low concentration in combination with allicin. Cu (2+) could be solubilized from the plasma membrane fraction by a procedure for the extraction of hydrophobic proteins rather than the extraction of phospholipids, suggesting its complexation with a plasma membrane protein as a result of allicin treatment. Such a subcellular localization of Cu (2+) resulted in the selective leakage of intracellular K (+), but not in the disruptive damage on the plasma membrane, and was considered to underlie the synergistic fungicidal activity of Cu (2+) and allicin.

Induction of histone acetylation and inhibition of growth of mouse erythroleukemia cells by S-allylmercaptocysteine.

Growth-inhibitory effects on DS19 mouse erythroleukemia cells were seen in the micromolar concentration range with allicin and S-allylmercaptocysteine and in the millimolar range with allyl butyrate, allyl phenyl sulfone, and S-allyl cysteine. Increased acetylation of histones was induced by incubation of cells with the allyl compounds at concentrations similar to those that resulted in the inhibition of cell proliferation. The induction of histone acetylation by S-allylmercaptocysteine was also observed in Caco-2 human colon cancer cells and T47D human breast cancer cells. In contrast to the effect on histone acetylation, there was a decrease in the incorporation of phosphate into histones when DS19 cells were incubated with 25 microM S-allylmercaptocysteine. Histone deacetylase activity was inhibited by allyl butyrate, but there was little or no effect with the allyl sulfur compounds examined in this study. A similar degree of downregulation of histone deacetylase and histone acetyltransferase was observed when DS19 cells were incubated with S-allylmercaptocysteine or allyl isothiocyanate. The induction of histone acetylation by S-allylmercaptocysteine was not blocked by a proteasome inhibitor. The mechanism by which S-allylmercaptocysteine induces histone acetylation remains to be characterized. It may be related in part to metabolism to allyl mercaptan, which is a more effective inhibitor of histone deacetylase.

Mutagenic properties of N-cyclopropyl and N-allyl-N-nitroso compounds. Studies on the nature of alkylating species.

A series of directly acting N-nitroso compounds, N-nitroso-N-allyl urea 6, N-nitroso-N-cyclopropyl urea 7, N-nitroso-acetoxymethyl-allylamine 8, N-nitroso-acetoxymethyl- cyclopropylamine 9, N-nitroso(1- acetoxyethyl )allylamine 10 and N-nitroso(1- acetoxyethyl ) cyclopropylamine 11, which may hydrolize to liberate either cyclopropylating or allylating electrophiles, were synthesized and comparatively investigated for mutagenicity in Salmonella typhimurium TA 1535. Hydrolysis rates in aqueous buffered solution do not differ significantly in the allyl- and cyclopropyl series. Analysis of the hydrolysate of all compounds revealed only the presence of allylalcohol and not cyclopropanol . In contrast to the expected equal potencies, due to chemical rearrangement of the alkylating species from cyclopropylcation to allylcation , the results showed that the cyclopropylating analogs were much more effective mutagens than were the allylating compounds. We conclude that for the cyclopropylating compounds the diazonium ion intermediate - and not the free cation - is the alkylating species, during mutagenesis.

Theoretical structure-activity study of mutagenic allyl chlorides.

Ab initio molecular orbital calculations at the STO-3G and 3-21G levels were used to clarify the mechanism of mutagenic action of various substituted allyl chlorides. Computed molecular properties were compared with experimental mutagenic potentials of these allylic compounds. In agreement with experiment, the computational results suggest that the primary mechanism of action involves the SN1 formation of allylic cations which then react with nucleophilic centers of nucleic acid bases. The usefulness of computed properties in estimating the degree of alkylating activity and mutagenicity was evaluated. In general, stability of the allylic carbocation intermediate and the degree of charge delocalization in the allyl system correlate well with observed mutagenic potentials.
