Conformational stability of a model macrocycle tetraamide: an ab initio study.

Ab initio calculations are carried out to investigate the conformational stability of a model macrocyle tetraamide. The four amide groups in the selected model are present in the sequence: -(O=CNH)-Ph-(NHC=O)-CH=CH-(O=CNH)-Ph-(NHC=O)-CH=CH-. In this sequence, two phenyl rings and two ethene groups act as bridges between the amide units. Each amide motif bonds to a phenyl ring through its amide nitrogen and to an ethene group through its amide carbon. Four clearly distinct minimum-energy conformations are found upon full geometry optimization using the B3LYP/6-31+G(d) method. Frequency calculations using the same method confirm that the four conformations are indeed minima in the macrocycle potential energy surface. Relative to the most stable conformer, the other conformations are higher in energy by 0.86, 2.09, and 9.17 kcal/mol, respectively, at the MP2/6-31+G(d,p) level. The stability of the macrocycle conformations is correlated primarily to the existence and strength of intramolecular N-H...O=C hydrogen bonds. Additional stability to the conformations is found to come from weak Ph-H...O=C hydrogen bonding between a carbonyl oxygen and a hydrogen atom of a phenyl group. Solvent effects play an important role in the relative energies of the various conformations, as indicated by the simple SCRF = dipole model calculations for the case of aqueous solution.

Theoretical evidence for C-F bond activation by a fluoro-calix[4]pyrrole-tert-amine macrocycle.

Density functional theory as well as highly correlated ab initio molecular orbital theory was used to explore the possibility of activating C-F bonds in fluoroalkanes by organic macrocycles. The results indicate that the reaction between fluoro-calix [4]pyrrole-tert-amine and CH3F via a Menshutkin displacement mechanism is highly favorable and competitive from a thermochemical point of view with the very efficient C-Cl activation by a simple macrocyclic amine recently reported in the literature (Stanger, L. J.; Noll, B. C.; Gonzalez, C.; Marquez, M.; Smith, B. D. J. Am. Chem. Soc. 2005, 127, 4184).

A new macrocyclic trichothecene, 12,13-deoxyroridin E, produced by the marine-derived fungus Myrothecium roridum collected in Palau.

A new macrocyclic trichothecene, 12,13-deoxyroridin E (1), and three known compounds, roridin E (2), verrucarin A (3), and verrucarin J (4), were obtained as cytotoxic components from the marine-derived fungus Myrothecium roridum, isolated in Palau. 12,13-Deoxyroridin E is the second example of a macrocyclic trichothecene possessing a double bond at C-12-C-13 and was about 80-fold less cytotoxic than roridin E, the epoxide variant.

Aureoverticillactam, a novel 22-atom macrocyclic lactam from the marine actinomycete Streptomyces aureoverticillatus.

During the course of our screening program designed to discover novel anticancer and anti-infective agents from marine microorganisms, a strain of Streptomyces aureoverticillatus (NPS001583) isolated from a marine sediment was found to produce a novel macrocyclic lactam with cytotoxicity against various tumor cell lines. Using extensive MS, UV, and NMR spectral analyses, the structure has been established as compound 1, aureoverticillactam, a 22-atom macrocyclic lactam incorporating both triene and tetraene conjugated olefins.

Seven novel macrocyclic polypeptides from Viola arvensis.

Seven novel macrocyclic polypeptides, designated as varv peptides B-H, have been isolated from the aerial parts of Viola arvensis. Their primary structures have been elucidated by automated Edman degradation and mass spectrometry. They all consist of 29 or 30 amino acid residues, covalently cyclized via the amide backbone and by three internal disulfide bridges. Their amino acid sequences are as follows: varv peptide B, cyclo-(TCFGGTCNTPGCSCDPWPMCSRNGLPVCGE); varv peptide C, cyclo-(TCVGGTCNTPGCSCSWPVCTRNGVPICGE); varv peptide D, cyclo-(TCVGGSCNTPGCSCSWPVCTRNGLPICGE); varv peptide E, cyclo-(TCVGGTCNTPGCSCSWPVCTRNGLPICGE); varv peptide F, cyclo-(TCTLGTCYTAGCSCSWPVCTRNGVPICGE); varv peptide G, cyclo-(TCFGGTCNTPGCSCDPWPVCSRNGVPVCGE); and varv peptide H, cyclo-(TCFGGTCNTPGCSCETWPVCSRNGLPVCGE). The varv peptides B-H exhibited high degrees of homology with the hitherto known macrocyclic peptides varv peptide A, kalata B1, violapeptide I, circulins A and B, and cyclopsychotride A.

Synthesis of the macrocyclic core of laulimalide.

[figure: see text] A stereoselective synthesis of 3, corresponding to the fully functionalized macrocyclic core of the novel microtubule-stabilizing agent, laulimalide, has been completed. Efficient macrolactonization was achieved by a Mitsunobu reaction, installing the sensitive (Z)-enoate, and macrocyclic stereocontrol was then exploited to introduce the methyl group and trans-epoxide.

A rapid access to biaryl ether containing macrocycles by pairwise use of Ugi 4CR and intramolecular S(N)Ar-based cycloetherification.

[reaction: see text] From readily accessible starting materials, macrocycles with an endo aryl-aryl ether bond are synthesized in only two operations by combination of the Ugi four-component reaction and an intramolecular S(N)Ar reaction. The nitro group serves as an activator for the macrocyclization and provides a handle for the introduction of functional group diversity. A Ugi reaction promoted by ammonium chloride in aprotic solvent is documented for the first time.

A chiral 28-membered macrocycle with symmetry and structure similar to that of trans-cyclooctene.

[reaction: see text] A bridged N,N-di(aryl)-1,2,4,5-benzenediimide was synthesized in which restricted rotation led to two diasteriomeric conformations at room temperature. The more stable syn-macrocycle is achiral, whereas the strained anti-macrocycle possesses planar chirality similar to that of trans-cyclooctene. The structure was characterized by X-ray crystallography, and the enantiomers were resolved by chiral chromatography.

Synthesis and structure of a polyphenylene macrocycle related to "cubic graphite".

[structure: see text] Palladium-catalyzed coupling of 1,2-bis(4-bromophenyl)-3,4,5,6-tetraphenylbenzene and the corresponding hexaphenylbenzene bis(boronic acid) gave a mixture of linear and cyclic oligomers of hexaphenylbenzene. An X-ray crystal structure of the tetrameric oligomer showed it to be the polyphenylene macrocycle 4 (C(168)H(112)). The roughly D(2) symmetric macrocycle contains a large central cavity, and it is one of the channel substructures of "phenylogous cubic graphite".

A chiral A2 B2 macrocyclic minireceptor with extreme enantioselectivity.

We describe a novel macrocyclic minireceptor that is assembled from a chiral 1,2-diamine and 5-allyloxyisophthalic acid. After immobilization on HPLC silica, the chiral macrocycle preferentially binds the L-enantiomers of simple amino acid derivatives, with enantioselectivities values up to 3.0 kcal/mol. Regioselective synthesis of antiparallel loops on a macrocyclic scaffold constrained by oxazoles and thiazoles.

The regioselective syntheses and structures are reported for two tris-macrocylic compounds, each possessing two antiparallel loops on a macrocyclic scaffold constrained by two oxazoles and two thiazoles. NMR solution structures show the loops projecting from the same face of the macrocycle. Such molecules are shown to be prototypes for mimicking multiple loops of proteins.[structure: see text]

The first electroresponsive phenylazomethine macrocycles: highly preferential formation and regular molecular packing.

[structure: see text] Highly preferential formation of novel polyphenylazomethine macrocycles was achieved by further addition of TiCl(4) and/or the monomer during the course of the polycondensation. These macrocycles have unique structures based on the (E)/(Z)-conformation of the azomethine bonds, the extremely regular molecular-packing state, and the reversible redox properties by protic acid doping.

Cobalt-mediated macrocyclizations. Facile synthesis of 2-oxo pyridinophanes via [2 + 2 + 2] cycloaddition of alpha, omega-diynes and isocyanates.

[reaction: see text] Cobalt-mediated [2 + 2 + 2] cycloaddition of alpha,omega-diynes and isocyanates provides a direct approach to macrocyclic 2-oxopyridinophanes. This macrocyclization process, which proceeded most efficiently with aliphatic isocyanates, was conveniently performed at a moderate temperature (85 degrees C) without irradiation or syringe-pump addition.

Short and diverse route toward complex natural product-like macrocycles.

[reaction: see text] A general strategy toward macrocyclic compounds using multicomponent reaction (MCR) chemistry, e.g., Passerini and Ugi variants, and ring-closing metathesis (RCM) is introduced. The corresponding bifunctional isocyanides carboxylic acids bearing a terminal olefin are easy to prepare from the corresponding commercially available starting materials. Advantageously, this strategy allows fast access to a diverse conformational space of natural product-like macrocycles and could thus be of interest in the discovery of novel bioactive agents.

Self-organized heteroditopic macrocyclic superstructures.

[structure: see text] The synthesis of heteroditopic macrocyclic ureido receptors and of their NaX complexes is described. NMR studies and determination of the crystal structure show the formation of self-organized dimeric or polymeric superstructures by a cooperative macrocyclic cation complexation, anion-hydrogen bonding, and pi-pi stacking interactions. Membrane transport experiments show a direct relation between the synergetic ion-pair recognition and the transport properties of these molecular information transfer devices.

Proofs of macrocyclization of gable porphyrins as mimics of photosynthetic light-harvesting complexes.

Porphyrin macrocycles composed of five and six units of m-gable imidazolylporphyrinatozinc (1-Zn) were synthesized by self-assembled cyclization followed by ring-closing metathesis linkings. Each porphyrin macrocycle was isolated by GPC chromatography, and their molecular weights were determined by MALDI-TOF mass spectroscopy. These structures represent mimics of light-harvesting complexes in photosynthetic bacteria. [structure: see text]

Novel pi-expanded radialene macrocycles with inner cavity.

[structure: see text] Polyenyne macrocycles with pi-extended [9]- and [12]radialene frameworks have been synthesized. These radialenes exhibit restricted rotation of the aromatic rings, and the D3- and D4-symmetric structures in solutions have been determined by dynamic NMR. The macrocyclic radialenes bear small to medium inner cavities, and the small cavity of the pi-extended [9]radialene can incorporate a silver cation.
