N-acetylmuramoyl-L-alanine amidase assay based on specific radioactive labeling of muropeptide L-alanine: quantitation of the enzyme activity in the autolysin deficient Bacillus subtilis 168, flaD strain.

A sensitive and highly reproducible assay for N-acetylmuramoyl-L-alanine amidase (EC 3.5.1.28) was devised, based on specific and homogeneous L-[14C]alanine labeling of the substrate, the peptidoglycan. The method involves partial purification of both the enzyme and the substrate and monitoring the muropeptide cleavage by coupling fluorodinitrobenzene to freed L-alanine NH2 groups. After acid hydrolysis of the substrate, the resulting DNP-L-alanine and L-alanine are separated by TLC, and radioactive counts in relevant spots are determined. Application of the method to the autolysin-endowed strain and an autolysis-deficient flaD-bearing mutant has revealed (i) that the N-acetylmuramoyl-L-alanine amidase behaves like an endoenzyme with an apparent Kcat(s-1) of 40, and (ii) that the residual enzyme activity in the flaD bearing strain amounts to 2.5 (+/- 0.1)% of that of the parent strain.

A newly designed whole microplate automatic harvester for lymphocyte stimulation assays.

A unique automated sampling manifold designed to recover cells grown in standard 96 well microplates from their culture medium is described. Cells are recovered and washed on fiber glass filter discs. Incorporation of radioisotopes into cells, can then be measured by appropriate counting of the filter discs. Typical applications include termination of mixed lymphocyte cultures, assays of mitogen stimulation of lymphocytes and antigen-specific lymphocyte transformation and assays of interferon activity. The harvester can also be used in other biological systems where collection and washing of precipitates is desired.

[Haemagglutination inhibition test: improvements, limits and advantages of the method. Comparison with neutralization test].

In this study, we report some technical improvements which enable us to envisage wider use of the haemagglutination test and haemagglutination inhibition of the rabies virus. Moderated trypsination of sensitive red blood cells and their counting are important parameters. An interesting application of the test was the haemagglutination competition enabling the amount of soluble glycoprotein (obtained during the fractionating of the virus components) to be evaluated. From antibody kinetics of vaccinated dogs, a study of the correlation which exists between the IHA titres, SN mice titres and SN-RFFIT titres was carried out. Satisfactory correlation was obtained between these three techniques.

Efficacy of moxidectin pour-on against gastrointestinal nematodes and Dictyocaulus viviparus in cattle.

Twenty crossbred beef heifer calves were used for an efficacy evaluation of 0.5% moxidectin pour-on against gastrointestinal nematodes and Dictyocaulus viviparus. The latter parasite and Bunostomum phlebotomum were the target species. The calves were exposed to natural infection on pasture from late September to December 22. Additionally, all calves were experimentally infected with B. phlebotomum at 71 and 29 days before treatment and with D. viviparus at 29 days before treatment. The 20 calves were randomly allocated into two groups of ten, based on presence of patent lungworm and hookworm infections on days -6 and -1. Treatments were as follows: Group A, moxidectin 0.5% pour-on (PO) at 500 micrograms kg-1 BW; Group B, moxidectin vehicle (controls) by PO application. General strongyle and hookworm egg counts and lungworm larval counts were reduced to zero at 13 days after moxidectin treatment; treatment effect was significant (P < 0.05) only for the strongyle and hookworm counts. Percentage reduction for adult hookworms and mature-immature adult lungworms was 100.0 and also for adults and L4 Haemonchus placei and Ostertagia ostertagi and Trichostrongylus axei adults, adult males of Cooperia pectinata and C. spatulata, and Oesophagostomum radiatum adults. Efficacy values for C. punctata adult males, Cooperia spp. adult females, and Cooperia spp. L4 were > 99.9%, > 99.9%, and 92.4%, respectively. All efficacy values were significant (alpha = 0.05, 1-sided) except for Cooperia spp. L4.

Evidence of H-Y antigen on human B lymphocytes.

H-Y antigen was studied in human peripheral blood lymphocytes from male and female subjects by indirect immunofluorescence. In males, 20% of lymphocytes were found to be H-Y+, compared with 9% in females. B- and T-cell-enrichment analysis showed a close correlation between the male B-lymphocyte count and the H-Y+-cell count. The method described is particularly applicable to the measurement of H-Y antigen in cases of ambiguous sex differentiation since it offers direct demonstration of H-Y antigen in the cells of the individual under examination.

Near-field reflection backscattering apertureless optical microscopy: application to spectroscopy experiments on opaque samples, comparison between lock-in and digital photon counting detection techniques.

An apertureless scanning near-field optical microscope (ASNOM) in reflection backscattering configuration is designed to conduct spectroscopic experiments on opaque samples constituted of latex beads. The ASNOM proposed takes advantage of the depth-discrimination properties of confocal microscopes to efficiently extract the near-field optical signal. Given their importance in a spectroscopic experiment, we systematically compare the lock-in and synchronous photon counting detection methods. Some results of Rayleigh's scattering in the near field of the test samples are used to illustrate the possibilities of this technique for reflection backscattering spectroscopy.

Statistical analysis of the fluctuating counts of fecal bacteria in the water of Lake Kinneret.

Counts of E. coli, Enteroccoci and fecal coliforms in four sites around Lake Kinneret (The Sea of Galilee), collected every 2-4 weeks for about 5 years during 1995-2002 showed irregular fluctuations punctuated by aperiodic outbursts of variable magnitude. Because of the haphazard nature of fecal contamination and large intervals between successive counts, these patterns were described by probabilistic models, based on the truncated Laplace or Extreme Value distribution. Their applicability was tested by comparing the predicted frequencies of counts exceeding different levels calculated from the first half of each record with those actually observed in its second half. Despite the records imperfections and minor violations of the underlying models' assumptions, there was a reasonable agreement between the estimated and actual frequencies. This demonstrated that it is possible to translate the irregular fluctuation pattern into a set of probabilities of future high counts. In principle, such probabilities can be used to quantify the water's fecal contamination pattern and as a tool to assess the efficacy of preventive measures to reduce it.

Morphometry of E-PTA stained synapses at the periphery of pathological lesions.

We carried out a novel application of the disector sampling and counting method, in a biopsy material from the pathologic human brain, to estimate the synaptic structural dynamics, quantitatively. Parietal cortex biopsies of adult (mean age: 40.0 years) and old (mean age: 66.2 years) patients having undergone surgical intervention were investigated. The tissue samples were excised at the periphery of meningioma masses. Synaptic contact zones were stained en bloc by the ethanol phosphotungstic acid (E-PTA) preferential technique which selectively enhances both the pre- and post-synaptic paramembranous material separated by a sharp cleft against a very faint background, thus facilitating and objectifying synaptic morphometry. The disector method, associated with currently used morphometric formulas, enabled us to measure the number of synapses/m3 of tissue (numeric density: Nv); the total area of the synaptic contact zones/m3 of tissue (surface density: Sv) and the average synaptic size (S). In old vs. adult patients, Nv decreased by 7.5% (Mean (SEM): Adult 2.0040(0.0452); Old 1.6780(0.0623)), while S increased by 17.5% (Adult 0.0203(0.0026); Old 0.0246(0.0035)). Sv did not show any age-related difference. The same negative correlation between Nv and S has also been reported in physiological aging, and this suggests the active presence of age-related synaptic restructuring mechanisms in the nervous tissue surrounding a tumoral mass.

Drug evolution concept in drug design: 1. Hybridization method.

A novel concept, "drug evolution", is proposed to develop chemical libraries that have a high probability of finding drugs or drug candidates. It converts biological evolution into chemical evolution. In this paper, we present "hybridization" drug evolution, which is the equivalent of sexual recombination of parental genomes in biological evolution. The hybridization essentially shuffles the building blocks of the parent drugs and ought to drug(s); no drug evolution can otherwise occur. We hybridized two drugs, benzocaine and metoclopramide and generated 16 molecules that include the parent drugs, four known drugs, and two molecules whose therapeutic activities are reported. The unusually high number of drugs and drug candidates in the library encourages high expectations of finding new drug(s) or drug candidate(s) within the remaining eight compounds. Interestingly, the therapeutic applications of the eight drugs or drug candidates in the library are fairly diverse as 38 therapeutic applications and 25 molecular targets are counted. Therefore, the library fits as a general chemical library for unspecified therapeutic activities. The hybridization of other two drugs, aspirin and cresotamide, is also described to demonstrate the generality of the method.

Increased lymphocyte replicative index following 2,4-dichlorophenoxyacetic acid herbicide exposure.

OBJECTIVE: Evaluate peripheral blood lymphocyte proliferation (replicative index:RI) and micronuclei frequency (MF) among 2,4-D herbicide applicators. METHODS: Twelve applicators spraying only 2,4-D provided a blood and urine specimen upon enrollment, several urine samples during the spraying season, and a blood specimen at the study's end. Nine controls provided blood and urine specimens upon enrollment and at the study's end. Gas chromatography/tandem mass spectroscopy determined urinary 2,4-D levels and standard in-vitro assays determined RI and MF scores. Applicator RI and MF were compared before and after spraying and with controls. RESULTS: Applicators contributed 45 urine specimens with concentrations ranging from 1.0 to 1700 (microg 2,4-D/g creatinine/L urine) that logarithmically (In) increased as spraying time increased. Applicator RI increased after spraying (p = 0.016), independent of tobacco and alcohol use, and demonstrated a weak dose-response with increasing urinary 2,4-D levels (p = 0.15). Among 2,4-D applicators, pre-exposure complete blood counts and lymphocyte immunophenotypes were not significantly different from post-exposure measurements. CONCLUSION: Urinary 2,4-D concentration, an exposure biomarker, may be associated with lymphocyte replicative index, a cell proliferation biomarker.

Characterization of eosinophils and detection of eotaxin in skin chamber fluid after challenge with relevant allergen in patients with mild asthma.

BACKGROUND: A selective recruitment of eosinophils to sites of allergic inflammation is suggested to be controlled by regulation of cytokines, chemokines and adhesion molecules. OBJECTIVE: The aim of this study was to examine whether allergen challenge in skin chambers, applied on patients with allergic rhinitis and mild asthma, results in a selective influx of activated eosinophils and detectable levels of cytokines/chemokines related to eosinophil recruitment, such as interleukin (IL)-5 and eotaxin. METHODS: A skin blister was induced on the volar aspect of each forearm; one contained PBS-heparin buffer (control) and the other was challenged with relevant allergen. Peripheral blood was drawn before the allergen was applied to the skin chamber, and the expression of CD9, CD11b and EG2-epitope on intracellular eosinophil cationic protein (ECP) was analysed in eosinophils. Chamber fluid was collected 8 h after allergen application and analysed for differential cell counts, expression of eosinophil activity markers, the presence of ECP, eotaxin, and IL-5. RESULTS: The number of recruited leucocytes was equal in the allergen-challenged chambers and in controls. However, the number of eosinophils was significantly increased in the allergen-challenged chambers, and elevated levels of released ECP were measured. Moreover, the eosinophils recruited were activated, as shown by increased expression of EG2 and CD11b, and decreased expression of CD9, in comparison with blood eosinophils. In the skin chamber fluids, higher levels of eotaxin were detected in the allergen-challenged chambers than in controls, but there were no detectable levels of IL-5. CONCLUSION: We have demonstrated a selective recruitment of eosinophils, and higher levels of released ECP and eotaxin, in skin chambers stimulated with allergen, as compared with control chambers. Allergen challenge in skin chambers is a useful tool for studies of eosinophil recruitment, their state of activation, and their involvement in the allergic inflammatory response.

Time correlated single photon mie scattering from a sonoluminescing bubble

Application of time correlated single photon counting to pulsed Mie scattering enables one to resolve changes in light scattering to better than 50 ps. This technique is applied to the highly nonlinear motion of a sonoluminescing bubble. Physical processes, such as outgoing shock wave emission, that limit the interpretation of the data are measured with a streak camera and microscopy. Shock speeds about 6 km/s have been observed.

In vivo correlation between stratum corneum reservoir function and percutaneous absorption.

A relationship between stratum corneum reservoir function and percutaneous absorption has been established in the hairless rat. Two hundred nanomoles of 10 substances that have a wide range of chemical structures were topically applied for 30 min and the total body distribution was measured after 96 h. The quantity of substance present in the stratum corneum reservoir after 30-min application was measured by liquid scintillation counting after tape-stripping the treated area. A linear relationship exists between the quantity of substance in this reservoir x(nmol X cm-2) and the total amount of radioactivity distributed in the body and excreta y(nmol X cm-2) after 96 h. The relationship is given by: y = 1.644 X x - 0.536 (r = 0.998, p less than 0.001). Apart from the steroids, 80-95% of the compounds were excreted in the urine; and with the exception of thiourea, this elimination was rapid, especially for mannitol and benzoic acid. We confirmed that in terms of penetration there is a factor of 50 between benzoic acid (best) and dexamethasone (worst). Thus the quantity of substance penetrating through intact rat skin can be predicted by measuring the horny layer concentration. The animal data reported here should be verified in humans.

Antiretroviral treatment reverses HIV-induced reduction in the expression of surface antigens on alveolar macrophages in AIDS patients.

MoAbs and immunoperoxidase methods were used to identify antigen-presenting and phagocytic cells and to assess expression of HLA-DR molecules on cells obtained by bronchoalveolar lavage (BAL) from 33 AIDS patients and nine normal volunteers. In 17 patients, not receiving antiretroviral therapy, the expression of HLA-DR molecules (MoAb RFDR1) as well as the percentages of cells expressing RFD1 marker for antigen-presenting cells and RFD7 marker for mature phagocytes were significantly reduced. However, in BAL obtained after commencing treatment with zidovudine (AZT) in 21 patients or with 2',3'-dideoxyinosine (DDI) in five patients, the expression of the markers studied was found to have returned to levels of expression seen in normal lavages. The changes observed were clearly associated with antiretroviral treatment and did not correlate with applications of other drugs, blood CD4 counts or presence of infectious organisms in BAL fluid. As the alterations in the expression of HLA-DR molecules and RFD1 marker on macrophages have been shown to be associated with functional capacities of these cells, the reversal of impaired expression of phenotypic markers on alveolar macrophages in AIDS patients by AZT and DDI signifies an important ability of these drugs to modify immune reactivity and emphasizes the need to monitor such functions in HIV disease.

Improvement in microscope photometry by voltage to frequency conversion: analogue measurement and digital processing.

A system for digital measurement of microscope photometric signals, based on Voltage to Frequency Conversion (VFC) and suitable for automatic data processing, has been devised. Advantages and disadvantages are illustrated with special emphasis on the comparison between photocurrent measuring and photon counting techniques. A statistical analysis of the results allows the calibration of VFC net counts versus detected photons. A biological application is reported; this shows the suitability of the system even to low light levels.

Acoustic microcavitation: enhancement and applications.

Experimental investigation into acoustic microcavitation is extended to include "cavitation activity" in addition to the threshold measurements using acoustic detectors. The primary setup incorporates two detectors: an unfocused, untuned 1-MHz transducer, which serves as a passive detector, and a focused 30-MHz transducer used in pulse-echo mode as the active detector. Cavitation itself is brought about by a focused piezoelectric transducer driven in pulse mode. The active detector is arranged confocally with respect to the cavitation transducer. Both the interrogating pulse and the cavitation pulse arrive simultaneously at the common focus which is the region of cavitation. Cavitation is conducted primarily at 0.75 MHz and 1% duty cycle in clean water using microparticles to seed the events. Cavitation activity appears to be directly proportional to the number density of the particles present in the cavitation medium. The fact that the active detector affects the cavitation process can be further exploited to seek interesting applications leading possibly to submicronic particle counting and testing of surface characteristics of silica particles used in liquid chromatography.

Short term effect of air pollution on mortality in Polish urban populations--what is different?

STUDY OBJECTIVE: To assess, within the multinational European Community funded APHEA project, the relationship between daily ambient air pollution with sulphur dioxide (SO2) and black smoke (BS) and the daily number of deaths from all causes except external, from cardiovascular diseases, and from respiratory diseases taking into account several possible confounding factors. DESIGN: Time series analysis with the application of Poisson regression to the counts of daily number of deaths from selected causes over several years. Data were obtained from the computer files of individual death records in four Polish cities. Criteria developed for all the centres participating in the project were applied to the process of model building. SETTING: Four Polish cities - Cracow (period 1977-89), Lodz (1977-90), Poznan (1983-90), and Wroclaw (1979-89). SUBJECTS: Permanent residents of the cities who died in a city of residence or in a surrounding region during the study period. MAIN RESULTS: There were significant positive associations between mortality from all causes excluding external ones and SO2 and BS in Cracow and in Lodz and between mortality from cardiovascular diseases and SO2 in Cracow alone. In other cities the association was either non-significant or significant but in the opposite direction. No significant positive association was found between respiratory deaths and these pollutants. CONCLUSIONS: Short term effect of air pollution on mortality in Polish urban population cannot be ignored. However, differences observed between the cities suggest that the association may be not so straightforward and therefore there is a need of further studies to identify additional factors that may modify and confound the association.

Harvesting of peripheral blood progenitor cells with different programmes of discontinuous flow systems.

BACKGROUND AND OBJECTIVES: From a technical point of view two problems arise during the collection of peripheral blood progenitor cells (PBPC): first, the volume of the apheresis product is often large, requiring volume reduction prior to cryopreservation. Second, the platelet (PLT) loss due to the harvesting of the buffy coat is an unwanted side effect. With respect to these problems, the present study was designed to compare programs for PBPC collection with discontinous-flow cell separators. MATERIALS AND METHODS: Three different protocols for PBPC harvesting were investigated in 32 patients with malignancies. In the first protocol, the blood cell separator Haemonetics MCS 3p was used. In the second protocol using the same machine, the opening and closure of the stem cell valve was modified in combination with a centri surge to reduce the PLT loss. The MCS+ device with another configuration of the valves was used in the third protocol. PBPC were mobilised by chemotherapy plus cytokine administration or application of growth factor alone. Blood counts and CD34 antigen-expressing cells were determined before apheresis and in the PBPC product. Colony-forming unit granulocyte/macrophage (CFU-GM) were determined in the apheresis product. RESULTS: 55 PBPC collections were carried out with a median end product volume of 105 ml. The median counts for CD34+ antigen-expressing cells and CFU-GM were 1.5x10(6 )and 2.5x10(4)/kg body weight, respectively. PLT loss was significantly lower in protocol III. CONCLUSION: The study reported here revealed that PBPC could be easily collected in a reduced product volume by the intermittent-flow cell separators. No additional centrifugation prior to cryopreservation was necessary to remove the plasma from the apheresis product. Patient's PLT loss was reduced by the centri surge technique but this still has to be improved.
