Subtle changes on the surface of polymeric materials on autoclaving have been ignored so far. An increase in cyclic trimer content on the surface of polyethylene terephthalate on autoclaving is reported. This increase may affect the biocompatibility of polyethylene terephthalate. Quantification of the cyclic trimer was done by high performance liquid chromatography.

A standard low temperature steam/formaldehyde autoclave was tested according to the manufacturer's instructions, using a range of test pieces containing Bacillus stearothermophilus spores as the challenge organism. There were failures in killing the challenge organism and the reasons for these are discussed. A description of modifications made to the autoclave is given together with details of an improved operating cycle. The performance of the modified autoclave was greatly improved and conditions were established for reliable and consistent sterilization of all the test pieces. A commercially produced prototype autoclave similarly modified also showed effective sterilization.

BACKGROUND: One form of medical waste known to be capable of transmitting disease is the contaminated sharp. Safe handling and disposal of sharps is an essential element of any infection control program. Many areas allow the on-site treatment of sharps containers. However, little information currently exists as to the most effective sterilization procedures and container designs. METHODS: This study was intended to evaluate the effect treatment with various autoclaves had on bacterial endospores present on strips or needled syringes. Strips contained 1.7 x 10(5) Bacillus stearothermophilus spores; syringes were soiled with equal numbers of spores or with spores plus blood. Syringes were tested capped and uncapped. A gravity-displacement autoclave and a high-vacuum autoclave were used. Strips and syringes were placed within sharps containers three quarters filled with representative materials. Six types of containers were tested. Containers were processed sitting up or on their sides. Processed strips and needles were aerobically cultured at 56 degrees C for 7 days. If sterilization was not accomplished initially, additional exposure time was added. RESULTS: (1) Soiled syringes were more difficult to sterilize than strips. (2) Capping or the presence of blood did not affect sterilization efficiency. (3) Container positioning was important only for the gravity-displacement autoclave. (4) Additional exposure time was required in the gravity displacement autoclave when sterilizing soiled syringes but not strips. (5) High-vacuum autoclaving killed all spore challenges within the normal processing interval. CONCLUSIONS: The data indicate that processing of sharps containers within a gravity-displacement autoclave appears to require extended exposure intervals to achieve sterilization.

A microtiter plate toxicity test based on fluorescence was developed to determine the residual concentration of formaldehyde on medical items after LTSF sterilization. The residual formaldehyde on eight common materials, some of which are used in different clinical instruments and devices were analysed after sterilization with LTSF. Formaldehyde residues were detected on cotton, filter paper, natural rubber, PVC, and silicone-coated latex, but not on polyurethane, silicone or glass. Formaldehyde never exceeded the recommended maximum concentration on clinical devices of about 5 microg/cm2. The results were compared with those obtained by means of a chemical method, the correlation being good (R2=0.9396). The biological method proposed here is fast and can be automated, which means that it could be used as a screening method when there are doubts as to the accumulation of residues on clinical materials or instruments that are going to be sterilized with LTSF.
Oat hulls and corn stover were used to produce chars at approximately 500 degrees C. The carbon concentrations of oat hull char and corn stover chars produced were 72.3 and 68.0 wt.%, respectively. Both activation burn-off and Brunauer-Emmett-Teller (BET) surface area appear to exhibit a linear relationship with respect to activation time of oat hulls. As to corn stover activated carbons, there is no linear relationship between activation time and BET surface area. However, activation burn-off of and activation time appear to relate in a linear manner for the activated carbons produced from corn stover chars. Oat hull is better than corn stover as a raw material for the production of activated carbon.
The two main sugars in the agricultural by-product corn stover are glucose and xylose. Co-fermentation of glucose and xylose at high content of water-insoluble solids (WIS) without detoxification is a prerequisite to obtain high ethanol concentration and to reduce production costs. A recombinant strain of Saccharomyces cerevisiae, TMB3400, was used in simultaneous saccharification and fermentation (SSF) of whole pretreated slurry of corn stover at high WIS. TMB3400 co-fermented glucose and xylose with relatively high ethanol yields giving high final ethanol concentration. The ethanol productivity increased with increasing concentration of pretreatment hydrolysate in the yeast production medium and when SSF was performed in a fed-batch mode.

Simultaneous distillation-extraction was proposed as a preconcentration step for the determination of phenol and its derivatives in aqueous and soil samples. Detection limits of 0.01 mg l(-1) (water) and 0.1 mg kg(-1) (soil) were achieved by gas chromatography-flame ionization detection. The described preconcentration procedure was applied for the primary study of the adsorption equilibrium in a water-soil system serving as a model of phenol behaviors in the environment.
