Determination of honokiol and magnolol by micro HPLC with electrochemical detection and its application to the distribution analysis in branches and leaves of Magnolia obovata.

A simple and sensitive method has been developed for determining honokiol and magnolol in fresh Magnolia obovata (M. obovata) by micro high-performance liquid chromatography with electrochemical detection (microHPLC-ECD). Chromatography was performed using a Capcell Pak C-18 UG 120 microbore octadecylsilica (ODS) column, methanol-water-phosphoric acid (65 : 35 : 0.5, v/v/v), as a mobile phase and applied potential at +0.8 V vs. Ag/AgCl. Peak heights were found linearly related to the amounts of honokiol and magnolol injected from 0.67 pg to 2.0 ng (r>0.999). The detection limits (S/N=3) were 0.13 pg, respectively. Honokiol and magnolol of 0.27 ng were detected with relative standard deviation (RSD) of 0.73 and 1.17% (n=5), respectively. Honokiol and magnolol in Magnolia Bark of the Japanese Pharmacopoeia were extracted with 70% methanol, diluted with a mobile phase, and injected into the microHPLC-ECD for determination. Recoveries of honokiol and magnolol in Magnolia Bark exceeded 98.7% with RSD, less than 0.93% (n=5). Determination of the distributions of honokiol and magnolol in bark, phloem, wood, leaf blades, and petioles of fresh M. obovata were made using weight samples of 40-238 mg. This method is useful to determine honokiol and magnolol in M. obovata, which is a candidate for crude magnolia bark for traditional Japanese herbal medicines.

Stilbene derivatives from Pholidota chinensis and their anti-inflammatory activity.

Ethyl acetate extract of Pholidota chinensis L. showed strong NO production inhibitory activity in murine macrophage-like cell line, RAW 264.7, which was activated by a lipopolysaccharide (LPS) and interferon-gamma (IFN-gamma). Fractionation of the active extract led to the isolation of two new stilbene derivatives, 2,3'-dihydroxy-5-methoxy-3,4-methylenedioxydihydrostilbene (Pholidotol A) and 2-hydroxy-5-methoxy-3,4,3',4'-dimethylenedioxydihydrostilbene (Pholidotol B) together with six known stilbene derivatives. Pholidotols A both B and inhibited Nitric oxide (NO) production with an IC(50) value at 24.3 and 17.1 microM, respectively.

Protective effect of an extract of Phyllanthus amarus against radiation-induced damage in mice.

The radioprotective effect of an extract of the plant Phyllanthus amarus (P. amarus) was investigated in adult BALB/c mice. P. amarus extract (750 mg/kg b.wt and 250 mg/kg b.wt) was administered orally to mice for five days prior to whole body radiation (6 Gy) and for one month after radiation. The animals were sacrificed on days 3, 9, 12, and 30 after radiation. P. amarus significantly increased the total W.B.C count, bone marrow cellularity, and alpha-esterase activity as compared to untreated radiation-exposed animals. P. amarus treatment also increased the activity of various antioxidant enzymes, such as superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase (GST), glutathione peroxidase (GPX), and glutathione reductase (GR), both in blood and tissue, which were reduced by radiation treatment. There was also a significant increase in the glutathione (GSH) levels of blood and tissue. Lipid peroxidation levels, which were increased after radiation, were significantly reduced by P. amarus treatment, both in serum and liver. The results collectively indicate that P. amarus extract could increase the antioxidant defense mechanism in mice and there by protect the animals from radiation-induced cellular damage.

Effect of NaCl on in vitro propagation of sweet potato (Ipomoea batatas L.).

In vitro propagation protocols offer a better option for production of quality planting materials in a clonal crop such as sweet potato, which is a food crop of versatile uses. Propagation through axillary shoot proliferation and organogenic and embryogenic regeneration were studied in different genotypes of sweet potato. The addition of NaCl enhanced the rate of multiplication as well as yielded hardy somatic embryos. Optimal doses of NaCl in each mode of propagation were different. The hardy somatic embryos produced in NaCl medium could be stored at 8 degrees C with or without a protective alginate covering. High-frequency germination of stored hardy somatic embryos could facilitate the production of artificial seeds. Plantlets produced with the addition of NaCl in regeneration medium were established in vivo at a high frequency (95-100%). Yield and quality of storage roots developed from artificial seed-propagated plants were comparable with those of source plants.

Detoxification of actual pretreated corn stover hydrolysate using activated carbon powder.

A technique for the removal of acetic acid from an actual pretreated corn stover hydrolysate was investigated. A powdered form of activated carbon previously shown to be effective in the removal of acetic acid from a synthetic hydrolysate was utilized. The method proved to be effective at lowering acetic acid levels while exhibiting minimal adsorption of the desired sugars from the hydrolysate, although at a lower efficiency in the actual hydrolysate than in the synthetic hydrolysate. Results are obtained for temperatures between 25 and 35 degrees C and agitation rates between 150 and 350 rpm in shake flasks. Adsorption isotherm and kinetic rate data are presented. Temperature differences over this range did not have an effect on adsorption characteristics. Five stages of detoxification were necessary to lower acetic acid concentration to the maximum 2 g/L desired for fermentation.

Herbal remedies traditionally used against malaria in Ghana: bioassay-guided fractionation of Microglossa pyrifolia (Asteraceae).

Different extracts from 11 West African plants traditionally used against malaria in Ghana were tested against both the chloroquine-sensitive strain PoW and the chloroquine-resistant clone Dd2 of Plasmodium falciparum. Due to the promising in vitro activity of the lipophilic extract [IC50: 10.5 microg/ml (PoW); 13.1 microg/ml (Dd2)], Microglossa pyrifolia (Lam.) Kuntze (Asteraceae) was chosen for further phytochemical investigation. From active fractions 13 compounds were isolated; their structures were established on the basis of spectroscopic methods. 1-Acetyl-6E-geranylgeraniol-19-oic acid and sinapyl diangelate represent new natural compounds. The two diterpenes E-phytol [IC50: 8.5 microM (PoW); 11.5 microM (Dd2)], and 6E-geranylgeraniol-19-oic acid [IC50: 12.9 microM (PoW); 15.6 microM (Dd2)] proved to be the most active constituents in our test system.

Chemical composition of Tipuana tipu, a source for tropical honey bee products.

Tipuana tipu (Benth.) Kuntze is a tree from the leguminosae family (Papilionoideae) indigenous in Argentina and extensively used in urbanism, mainly in Southern Brazil. The epicuticular waxes of leaves and branch, and flower surface were studied by high temperature high resolution gas chromatography. Several compounds were characterized, among which the aliphatic alcohols were predominant in branch, leaves and receptacle. Alkanes were predominant only in the petals and the aliphatic acids were predominant in stamen. In branches and leaf epicuticular surfaces, six long chain wax esters series were characterized, as well as lupeol and b-amyrin hexadecanoates.
